        Anti-oxidative effects of broccoli (Brassica oleracea var. italica ) sprout extract in RAW 264.7 cell and cisplatin-induced testicular damage

        Background: Brassica oleracea var. italica (broccoli), a rich source of antioxidants, can prevent various diseases and improve human health. In this study, we investigated the antioxidative effects of broccoli sprout extract on oxidative stress induced by lipopolysaccharide and cisplatin in cell and organ tissue models. Methods: Antioxidative effect of BSE was evaluated using DPPH and ABTS in RAW 364.7 cells, and effects of BSE on testes were investigated using Cisplatin-induced testicular damage model with an in vitro organ culture system. Results: The DPPH assay showed that the antioxidant activity of the alcoholic broccoli sprout extract was higher than that of the water extract. Additionally, the expression levels of antioxidation-related genes, Nrf2 , Gsr , HO-1, and catalase , were significantly increased in broccoli sprout extract-treated RAW 264.7 cells, and the extract suppressed lipopolysaccharide-induced mitochondrial dysfunction. Based on the results in the RAW 264.7 cell culture, the antioxidative effects of the extracts were investigated in a mouse testis fragment culture. The expression of Nrf2 , HO-1 , and Ddx4 was clearly decreased in cisplatin-treated mouse testis fragments and not in both broccoli sprout extract- and cisplatin-treated mouse testis fragments. In addition, the oxidative marker O-HdG was strongly detected in cisplatin-treated mouse testis fragments, and these signals were reduced by broccoli sprout extract treatment. Conclusions: The results of this study show that broccoli sprout extracts could serve as potential nutraceutical agents as they possess antioxidant effects in the testes.

        Evaluation of Antioxidative Effects of Lactobacillus plantarum with Fuzzy Synthetic Models

        Numerous studies suggest that the effects of lactic acid bacteria (LAB) on oxidative stress in vivo are correlated with their antioxidative activities in vitro; however, the relationship is still unclear and contradictory. The antioxidative activities of 27 Lactobacillus plantarum strains isolated from fermented foods were determined in terms of 2,2-diphenyl-1-picrylhydrazyl, hydroxyl radical, and superoxide radical scavenging abilities, reducing activity, resistance to hydrogen peroxide, and ferrous chelating ability in vitro. Two fuzzy synthetic evaluation models, one with an analytic hierarchy process and one using entropy weight, were then used to evaluate the overall antioxidative abilities of these L. plantarum strains. Although there was some difference between the two models, the highest scoring strain (CCFM10), the middle scoring strain (CCFM242), and the lowest scoring strain (RS15-3) were obtained with both models. Examination of the antioxidative abilities of these three strains in D-galactose-induced oxidative stress mice demonstrated that their overall antioxidative abilities in vitro could reveal the abilities to alleviate oxidative stress in vivo. The current study suggests that assessment of overall antioxidative abilities with fuzzy synthetic models can guide the evaluation of probiotic antioxidants. It might be a more quick and effective method to evaluate the overall antioxidative abilities of LAB.

        추출용매에 따른 브로콜리의 항산화와 항염증에 대한 효과

        Broccoli has a functional substance, sulforaphane that has effects of anticancer, antioxidant, antimicrobial and anti-inflammatory. Sulforaphane, one of the hydrolysis products of glucoraphanin in broccoli, cabbage and kale, was contributed to the role of antioxidant. Broccoli contains a number of bioactive compounds including glucosinolates, S-methyl cysteine sulfoxide and many antioxidants. The ethanol extract (BE), hexane extract (BH),propylene glycol extract (BP) and butylenesglycol extract (BB) of broccoli were used to investigate the antioxidation and anti-inflammatory effects of sulforaphane extracts from broccoli. The high scavenging abilities of DPPH, O2−were observed. Also sulforaphane extracts from broccoli showed the inhibition effect on NO rate. These results demonstrated that sulforaphane extracts from broccoli could be useful as an antioxidation and anti-inflammatory functional ingredient.

        신선육자환 에탄올 추출물의 항산화 활성

        Objective : The Sinseon-yukza-hwan (SSY), a Korean medicinal formula which includes Radix Rehmanniae Preparata and other medicinal herbs, has long been used for treatment of alopecia and gray hair through oral administration. This study is designed to enhance the utilization of natural materials in hair and scalp-related cosmetics. Possibility of SSY as an antioxidant was examined from its 50% ethanol extract. Methods : The antioxidative capacities were evaluated by determining total phenolic and flavonoid contents, 1,1-diphenyl-2-picryl hydrazyl (DPPH), 2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), reducing power and hydroxyl radical scavenging activity. Results : Total polyphenol and flavonoid contents of SSY were 25.53 mg TE, tannic acid equivalent/g and 18.90 mg RE, rutin equivalent/g, respectively, which correlated strongly its antioxidative activity. The DPPH and ABTS free radical scavenging activities of SSY at $0.1mg/m{\ell}{\sim}5mg/ml$ were ranged from 20% to 85% and 10% to 58%, respectively. Also the hydroxyl radical scavenger activity and reducing power increased in SSY-treated group, which were significantly lower in SSY-compared to BHA-treated group. But the highest reducing power was shown as 79% from SSY-treated group, which was higher value than 65% from BHA-treatment. These results showed that SSY extract effectively inhibited the generation of free radicals in the all assay system with dose-dependent manners. Conclusions : Thus, the present study provide preclinical data to support the expanded application of SSY, which could be potential candidates for natural antioxidants.

        Brazilin의 Potato Chip에 대한 항산화 효과

        소목으로부터 분리한 brazilin을 potato chip 제조시 사용하는 튀김유에 첨가하여 항산화 활성 정도를 시험하였다. 이때 현재 사용중인 항산화제도 동일 조건에서 시험하였다. 저장 30일 후 potato chip에서 추출한 유지의 과산화물가는 대조구(control), BHA(50 ppm)-BHT(50 ppm), δ-tocopherol(100 ppm) 및 brazilin(100 ppm)이 각각 134 meg/㎏oil, 84.06 meg/㎏ oil, 117.10 meg/㎏ oil 및 68.56 meg/㎏ oil을 나타내었다. 각 처리구는 대조구에 비하여 BHA(50 ppm)-BHT(50 ppm)가 1.6배, δ-tocopherol(100 ppm)이 1.14배, brazilin(100 ppm)이 1.97배의 항산화 효과를 나타내었다. 위 결과에서 brazilin(100 ppm)의 경우, BHA(50 ppm)-BHT(50 ppm)보다도 과산화 물가가 낮아 과산화물 생성 억제력이 효과적임을 알 수 있다. 또한 유도기간 연장 효과는 Rancimat을 이용한 고온가열시험(180℃, 100 ppm)에서 대조구에 비하여 BHA(50 ppm)-BHT(50 ppm)가 1.4배, brazilin(100 ppm)이 1.53배로 brazilin이 상업용 항산화제보다 보다 항산화 효과가 높게 나타났다. 또한 brazilin(100 ppm)은 palmitic acid(C16:0)/linoleic acid(C18:2) 값의 감소를 억제하는 능력과 conjugated dienoic acid 함량의 증가 억제력이 상업용 항산화제보다 우수하였다. Brazilin was added to frying oil used in the production of potato chips and their antioxidative effects against Caesalpinia sappan L. were evaluated. Additionally, the antioxidative activity was tested under the same conditions that commercial antioxidants are evaluated. The peroxide value of the oil and fat extracted from the potato chips was 134 meg/㎏ oil, 84.06 meg/㎏ oil, 117.10 meg/㎏ oil and 68.56 meg/㎏ oil in the control group, BHA(50 ppm)-BHT(50 ppm) group, δ-tocopherol (100 ppm) group and brazilin(100 ppm) group after storage for 30 days. The antioxidative effect of chips subjected to these treatments were 1.6 times, 1.14 times and 1.97 times greater than that of the control. In addition, the peroxide value was lower in the brazilin(100 ppm) group than in the BHA(50 ppm)-BHT(50 ppm) group and this group also had a superior effect at inhibiting the production of peroxide. Furthermore, an experiment conducted at high temperature using the Rancimat resulted in the antioxidant activity of brazilin(100 ppm) and BHA(50 ppm)-BHT(50 ppm) being 1.53 and 1.4 times greater than that of commonly used synthetic antioxidants. Finally, brazilin(100 ppm) effectively decreased the palmitic acid (C16:0)/linoleic acid(C18:2) value and increased the conjugated dienoic acid content to a greater degree than commercial antioxidants.

        쌀겨발효추출물의 이화학적 특성 및 항산화 작용

        ㈜한국바이오솔루션에서 제공받은 쌀겨발효 추출물(Fermented Rice-Bran, FRB)과 현재 시판 중에 있는 제품의 주원료로 사용되고 있는 미배아대두발효 추출물(Fermented Rice-Soybean, FRS)을 대조군으로 in vitro상에서의 이화학적 특성과 생리활성 및 항산화작용에 대하여 검토하였다. 이화학적 특성으로 총 폴리페놀 화합물과 flavonoid 함량을 측정하였고, 생리활성 및 항산화작용은 DPPH 자유라디칼 소거활성, Fe/Cu 환원력, 간장 microsome을 이용한 지질 과산화 억제활성, linoleic acid 산화 실험계를 이용하여 측정하였다. 쌀겨발효 추출물의 총 폴리페놀과 flavonoids 함량은 각각 19.92 ㎎/g과 11.56 ㎎/g으로 미배아대두발효 추출물보다 높은 수치를 보였다. 쌀겨발효 추출물은 DPPH free radical 소거 활성능에서도 69.8%로 상당히 높은 소거율을 나타내었으며 환원력 또한, 처리농도 증가와 함께 높아지는 경향을 보였다. 간장 microsome을 이용한 지질 과산화 억제활성, linoleic acid 산화 실험계를 통한 지질과산화 억제활성을 확인한 결과, 쌀겨발효 추출물이 미배아대두발효 추출물보다 우수함을 확인하였다. 따라서, 본 연구는 건강식품소재 개발을 위해 쌀겨발효 추출물의 생리활성물질과 항산화 활성을 검토하는 기초 자료로 활용될 수 있을 것이라 사료된다. This study suggests that fermented rice bran extract contains natural antioxidants. The contents of bioactive materials (e.g., polyphenolic compounds and flavonoids), antioxidative properties (DPPH (α,α’-diphenyl-β-picrylhydrazyl) free radical scavenging activity, Fe reducing, Cu reducing power, peroxidation of linoleic acid and rat hepatocyte microsome) were tested by in vitro experimental models using fermented rice bran (FRB) extract. The concentrations of phenolic compound and flavonoid were 19.92 ㎎/g and 11.56 ㎎/g, respectively. In oxidation in vitro models using DPPH free radical scavenging activity, (free radical scavenging activity 69.8%) Fe reducing power and Cu reducing power (effect of dose-dependent manner), Fe<SUP>2+</SUP>/ascorbate induced linolenic acid peroxidation by ferric thiocyanate and thiobarbituric acid (TBA) methods (inhibition activity 81%), and autooxidation of rat hepatic microsomes membrane (lipid peroxidation inhibition activity 38%), antioxidative activities were stronger in FRB extract than FRS (Fermented Rice and Soybean, positive control) extract and, these effects were dose-dependent manner. From these results, FRB extract was shown to have the most potent antioxidative properties and contain the highest amounts of antioxidative compounds such as phenolic compounds and flavonoids. Overall, these results may provide the basic data to understand the antioxidative properties of fermented rice bran for development of functional foods.

        A Study on DPPH and ABTS Antioxidant Activity and Sensory Evaluation of Seolgitteok with Walnut and Health CRISIS(Juglans Regia)

        Purpose: Life expectancy of human beings has increased due to marked advancement of medical science. Along with this increase, people are becoming more interested in living longer in good health, which in turn has imparted significant effects on dietary life. As such, the advantageous physiological characteristics of food items are being highlighted and gaining in popularity. Therefore, this study is concerned with production of “tteok”(rice cake) with the addition of walnut, a functional ingredient capable of suppressing oxidation and thus having significant effects with regard to diseases. Method: Based on various research data, it is known that continuous walnut consumption is effective for inducing antioxidation in the human body. Moreover, from experimental results of studies on cancer and stress-mediated diseases, it is known that phenols and flavonoids are effective in inducing antioxidation. Accordingly, this study is aimed at examining the extent of the antioxidation effect from the addition of walnut when making Seolgitteok with non-glutinous rice, as well as the commercial value of that effect. Results: For this purpose, the correlation between walnut contents of 0%, 1%, 3%, 6% and 9%, as the control group, phenol and flavonoid The content was correlated with the dose through DPPH and ABTS elimination. The chromaticity and other properties of steamed Seolgitteok were quantified through sensory evaluation to identify the optimal walnut content for commercialization. Conclusion: Accordingly, the results of this study were deemed to be helpful for purposes of qualitative improvement of the tteok industry by disclosing the physiological characteristics of Seolgitteok containing walnut.

      • Monodisperse Hydrogel Microparticles with Transition Metal Dichalcogenides for Dermal Antioxidation

        Two-dimensional transition metal dichalcogenides (TMDs) have provoked an extensive interest in the past few years due to their layer-dependent properties, such as radical scavenge and antioxidation. In this work, we fabricate a monodisperse poly (ethylene glycol) diacrylate hydrogel microparticles in which TMDs are physically immobilized in their meshes. Then, we incorporate quenchers to investigate how they affect the quenching efficiency through the hydrogel phase. Using them, we analyze their optical properties and durability of antioxidation activity. Finally, we propose that the TMDs-immobilized hydrogel microparticles are truly useful for exploring new biomolecular applications in the fields of dermatology and cosmetics.

      • Novel transition metal dichalcogenide antioxidants for dermatological applications

        Recently, two-dimensional transition metal dichalcogenides (TMDs) have been attracted wide attention in the past few years due to their layer-dependent properties, such as radical scavenge and antioxidation. In this work, we fabricate extremely stable nanoscale dispersions of TMDs which are encapsulated in the hydrophobic core of amphiphilic block polymeric micelles. We especially focus on figuring out how the type of TMDs affects the dispersion stability and antioxidation power in polymeric micelle system. We also analyze their quenching effect as well as optical properties. Finally, we demonstrate that the approach proposed in this study has great potential in exploring new biomolecular applications in fields of dermatology and cosmetics.
