      • Pharmacological mechanism responsible for the <i>Atractylodes japonica</i>-induced distal colonic contraction in rats

        <P><B>Abstract</B></P><P><B>Background and aim</B></P><P><I>Atractylodes japonica</I> Koidz (Compositae) has been commonly used to treat the gastrointestinal (GI) disorders in Korean traditional medicine, but its pharmacological roles in the regulation of GI motility have not been clarified yet.</P><P><B>Methods</B></P><P><I>Atractylodes japonica</I> was sequentially partitioned with MeOH, <I>n</I>-hexane, CHCl<SUB>3</SUB>, EtOAc and <I>n</I>-BuOH saturated with H<SUB>2</SUB>O, and the effects of <I>Atractylodes japonica</I> extracts on the spontaneous contractility of GI muscle strips prepared from rats were measured.</P><P><B>Results</B></P><P>Among five different fractionations, EtOAc extracts of <I>Atractylodes japonica</I> (AJEA) dose-dependently increased the low frequency contraction of distal colon longitudinal muscles (DCLM), and the ED<SUB>50</SUB> values were revealed to be 1.71×10<SUP>−9</SUP>g/ml. Among GI tracts, a prominent contractile response to AJEA was observed only in the DCLM. The contractile patterns produced by AJEA remarkably differed from those caused by acetylcholine and 5-HT. 4-DAMP and methoctramine at 0.5μM significantly blocked the AJEA (1.0μg/ml)-induced contraction of DCLM, but ondansetron, GR113808 and methysergide at 1.0μM in combination did not change the AJEA-induced DCLM contractions. Acetylethylcholine mustard (5.0μM) significantly diminished the AJEA-induced DCLM contractions, whereas <I>p</I>-chlorophenyl alanine (1.0μM) did not affect the stimulatory effects of AJEA on the DCLM contractions.</P><P><B>Conclusion</B></P><P>The present results suggest that AJEA may specifically act on the DCLM among GI smooth muscles, and AJEA-induced DCLM contraction is likely mediated, at least, by activation of ChAT and acetylcholinergic muscarinic receptors.</P>

        풍륭(風隆)·음릉천(陰陵泉)에 대한 약침(藥鍼)(창출(蒼朮), 의이인(薏苡仁))이 고지방식이(高脂肪食餌)로 유발(誘發)된 비만백서(肥滿白鼠)에 미치는 영향(影響)

        Objectives : This research was performed to investigate the effect of herbal acupuncture (Atratylodes japonica Koizumi and Coix lachrymajobi var. mayeun Stapf) at Pungnyung(ST40) and Umnungchon(SP9) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into normal group(Sham), high fat diet group(Control), high fat diet and Atractylodes japonica-herbal acupuncture in ST40 SP9 treated group (AJ), high fat diet and Coix lachrymajobi var. mayeun Stapf-herbal acupuncture in ST40 SP9 treated group(CL), high fat diet and Atractylodes japonica+Coix lachrymajobi var. mayeun Stapf-herbal acupuncture in ST40 SP9 treated group(AJ+CL). Herbal acupuncture was bilaterally treated at the level of 132.5mg/kg body weight per day. Results : Body weight and food efficiency were decreased in AJ, AJ+CL. Feed intake was decreased in AJ, CL. The level of serum HDL-cholesterol was increased in AJ, CL. The level of serum total cholesterol, LDL-cholesterol and phospholipid were not significant. The level of serum triglyceride, free fatty acid were increased in AJ. The level of serum AST was decreased in AJ, AJ+CL. The level of serum ALT was decreased in AJ, CL, AJ+CL. The level of serum ALP was decreased in AJ+CL. The HDL to Total cholesterol ratio was increased in AJ. Conclusions : Atractylodes japonica-herbal acupuncture in ST40 SP9 can control the body weight, feed intake and feed efficiency ratio, HDL-cholesterol and herbal acupuncture combined Atractylodes japonica with Coix lachrymajobi var. mayeun Stapf is effective on Body weight, feed intake, feed efficiency ratio, protection of liver function and prevention cardiovascular risk by obesity.

        해부형태적 특징과 SCAR Marker를 이용한 백출의 기원식물 판별

        국내 한약재 시장에서 유통되고 있는 백출 건재약재의 기원을 판별하고자 해부형태적 특징과 종 특이적인 SCAR 마커 (SAjR2, SAmR1)를 적용한 실험 결과는 다음과 같다. 해부형태적 특징으로 A. japonica에서는 주피에 코르크층이 매우 발달하여 있었고, 코르크층 내부 또는 아래에 석세포대가 연속적으로 발달하고 있었다. 유실은 A. japonica에서 뿌리횡단면에 전체적으로 다수 분포하여 잘 발달하고 있었으며 피충부분과 잘 발달된 방사조직 내에서도 목섬유다발이 다수 관찰됐다. 또한 유세포 내의 침상 결정물도 A. macrocephala보다 많이 함유하고 있었다. 이러한 특징들을 종합한 결과, 국내 유통 국산 및 수입 백출은 모두 A. japonica의 해부형태적 특징을 따르고 있어 A. japonica로 동정되었다. 한편 RAPD clone으로부터 개발된 A. japonica 특이적인 SAjR2를 이용하여 PCR 한 결과 모든 샘플에서 600bp 크기의 한국백출 특이적인 밴드가 형성되었으며, A. macrocephala 특이적인 SAmR1을 이 용한 결과에서는 작물시험장에서 재배중인 A. macrocephala에서만 1200 bp크기의 특이적인 밴드가 형성되어, 시중에서 유통되고 있는 한국백출과 중국백출은 모두 A. japonica로 동정되었다. 결론적으로 해부형태적 특성과 SCAR 마커는 A. japonica와 A. macrocephala 기원식물의 동정과 유통 건재 약재를 판별하기에 유용한 방법이었다. Finding a means to discriminate the commercial herb medicines when they were dried and sliced is a very important and imminent project in Korea. To differentiate plant origins and the commercial herb medicines of Atractylodes japonica and A. macrocephala, two discriminative methods using anatomical characteristics and SCAR marker were applied. It was possible to discriminate plant origins and the commercial herb medicines between A. japonica and A. macrocephala by anatomical characteristics: development of periderm, layer of stone cell, distribution of laticiferous vessels, development of xylem fiber in xylem ray, contained quantity of clustered crystals and others. While, two SCAR markers were developed from RAPD clones: SAjR2 (600 bp) from AjR2 and SAmR1 (1,200 bp) from AmR1. These two markers were enough for discrimination plant origins and the commercial herb medicines between A. japonica and A. macrocephala. The result of application of anatomical characteristics and SCAR markers to investigate current status in domestic herb market, Daegu and Kumsan herb market, it was identified to be current herb medicines of A japonica.

        Objective : This research was performed to investigate the effect of herbal acupuncture(Atratylodes japonica, Coix lachrymajobi, Ephedra sinica, Atratylodes japonica mixed with Coix lachrymajobi and Ephedra sinica mixed with Green tea) at Pungnyung(ST40) and Umnungchon(SP9) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Method : Experimental groups were divided into normal group(Normal), high fat diet group(Control), high fat diet and Atractylodes japonica-herbal acupuncture group(AJ), high fat diet and Coix lachrymajobi-herbal acupuncture group(CL), high fat diet and Ephedra sinica-herbaI acupuncture group(ES), high fat diet and Atractylodes japonica+Coix lachrymajobi-herbal acupuncture group(AJ+CL), Ephedra sinica+Green tea-herbal acupuncture group(ES+GT). Herbal acupuncture was bilaterally treated at the level of 132.5mg/kg body weight per 2day. Results : Body weight and food efficiency were decreased in AJ, ES, AJ+CL, ES+GT. The level of serum total cholesterol, triglyceride and free fatty acid were increased in AJ, ES, ES+GT. That of serum HDL-cholesterol was increased in AJ. The change of food intake, the level of serum phospholipid and ALP were not significant. The HDL to Total cholesterol ratio was increased in AJ and ES. Conclusion : Atractylodes japonica-herbal acupuncture in ST40 SP9 is effective on Body weight, food efficiency ratio, the level of serum lipid, protection of liver function and prevention cardiovascular risk by obesity induced by high fat diet. Herbal acupuncture mixed Ephedra sinica with Green tea can control the body weight, food efficiency ratio and the level of serum lipid.

        Effects of the Rhizomes of Atractylodes japonica and Atractylenolide I on Allergic Response and Experimental Atopic Dermatitis

        Although some anti-allergic activities of the rhizome of Atractylodes japonica have been previously reported, the active principle(s) for anti-allergic action is not fully elucidated and the effect of this plant material on atopic dermatitis (AD) is not known. In this study, the 70% ethanol extract of the rhizome of A. japonica was found to significantly inhibit 5-lipoxygenase (5-LOX)-catalyzed leukotrienes (LT) production from rat basophilic leukemia (RBL)-1 cells. From the extract of A. japonica, three major sesquiterpene derivatives including atractylenolide I, atractylenolide III and eudesma-4,7-dien-8-one were successfully isolated. Among these compounds, only atractylenolide I was shown to strongly inhibit 5-LOX from RBL-1 cells (IC50 = 18.6 μM). To evaluate the effects of experimental AD, the ethanol extract of A. japonica (200 mg/day) was administered orally to hapten-treated NC/Nga mice which is an animal model of AD. It was firstly found that the extract significantly inhibited AD-like symptoms in mice, as judged by severity score and scratching behavior. Taken together, it is concluded that A. japonica possesses the inhibitory activity on 5-LOX and an animal model of AD, and atractylenolide I may contribute, at least in part, to these anti-allergic actions of A. japonica.

        백출의 Atractylenolide I과 Atractylenolide III의 함량분석

        This study was conducted to determine the content of atractylenolides I and III in Atractylodes japonica. An established HPLC-DAD method was used to monitor contents of atractylenolides I and III in A. japonica obtained from Korea and China and compared with contents of A. macrocephala, A. chinensis, and A. lancea. Quantitative analysis of atractylenolides I and III was carried out on a Shiseido C18 column (S-5 µm, 4.6 mm I.D. × 250 mm) with gradient elution composed of acetonitile-water. The results show that the average content of atractylenolides I and III in A. japonica were 0.0954 and 0.1963%,respectively, and contents of A. lancea were higher than A. macrocephala, A. chinensis. In this study, we identified the differentiation of the quality of A. japonica from different species and collected locations and established content standard of atractylenolides I and III in A. japonica and this content standard was helpful to quality control of A. japonica.

        임간재배 시 광조절과 부엽토 처리에 따른 삽주의 생육 및 광합성 특성

        This study was carried out to determine the effects of light controls and leaf mold on root growth and physiological responses of Atractylodes japonica growing in forest farming. The experiment was performed by light controls (100%, 62.5%, 40.3% and 19.7% of full sunlight) and application of leaf mold to soil. Height, stem diameter, number of flower buds and root collar diameter were the highest in leaf mold within 62.5% of full sunlight (relative light intensity 62.5%). And these were the higher in leaf mold within each light level. As the shading level increased, light saturation point and maximum photosynthesis rate decreased. As the light level decreased, SPAD value increased in control and leaf mold. As a result of surveying the whole experiment, A. japonica was judged worse root growth under the lower light level. It was concluded that the light level was one of the most important factors to produce A. japonica. Also, producing high-quality of A. japonica with the price competitiveness by using leaf mold like the experiment can be an effective way to increase incomes for farmers.

        삽주의 어린순 채취(採取)회수 및 질소분시(窒素分施)가 생육(生育)과 수양(收量)에 미치는 영향

        자생(自生) 상태(狀態)로 채취(採取) 이용되거나 수입(輸入)되어 사용되는 창출(蒼朮), 백출(白朮)의 기원식물인 삽주(Atractylodes japonica)의 효과적인 재배기술을 확립하여 생약(生藥)의 수입(輸入)대체 기술(技術)을 개발하고자 산채용 삽주싹을 무채취(無採取), 1회(回) 채취(採取), 2회(回) 채취(採取)후 질소질 비료를 전양기복(全量基服), 50-30-20%, 40-30-30% 분시(分施) 처리(處理)하여 채험(採驗)한 결과를 요약하면 다음과 같다. 1. 초장생육(草長生育)은 어린싹을 채취(採取)함에 따라 철저히 억제되었으나 삽주 생육(生育)에는 1회 채취시(採取時)가 적절할 것으로 보였다. 2. 지상부(地上部) 건물중(乾物重)은 생육후기 질소 분시율이 많을수록 무채취 및 1회 채취에서는 증가하였으나 2회 채취에서는 차이가 없었다. 3. 어린싹 수양(收量)은 1회(回) 채취시(採取時) 평균(平均) 269~288kg/10a 수준이었으며, 2회(回) 채취시(採取時)는 싹의 수양(收量)이 적고 거칠어 이용성이 떨어졌다. 4. 근경(根莖) 수양(收量)은 무채취(無埰取)(전양기비(全量基肥)) 대비 어린싹 2회(回) 채취시(採取時)는 70% 수준으로 감소하였으나, 1회(回) 채취(採取) 및 질소(窒素), 50-30-20%로 분시(分施)한 처리(處理) 수양(收量)은 100% (471kg/10a) 증수되었다. 5. 삽주는 5월말~6월초 지상부(地上部) 생육(生育)이 완료되고 6~7월경 잠아로부터 신초가 재형성되어지므로 5월상 - 하순경에 절단자극에 의해 일찍 신초를 형성시키는 것이 근경수량(根莖收量)의 증대에 효과적인 기술로 사료되었다. This Experiment was conducted to know effect of cutting times and nitrogen split application rates on shoot and root-related characters of Atractylodes japonica. The cutting time was forced on none, once and twice along with three levels of nitrogen split application, all basic 50 -30 -20% and 40 -30 -30%. The plant height declined with increased cutting time and dry matter weight was increased with nitrogen split application under none and once cutting. Average yield of cutted shoot as edible wild plant was 257kg/10a at once cutting condition, but twice cutting condition was little and unfavorable to use edible wild plant because shoot was coarse. Yield 0 dry rhizome was the highest at once shoot cutting und­er 50 -30 -20% nitrogen split application by 471kg/10a.Especially, culture of Atractylodes japonka needed once shoot cutting one the 10th-30th of May.

        Prevention of Arthritic Inflammation Using an Oriental Herbal Combination BDX-1 Isolated from Achyranthes bidentata and Atractylodes japonica

        An oriental herbal combination (BDX-1) was isolated from Achyranthes bidentata and Atractylodes japonica. We previously tested the clinical effectiveness of BDX-1 in rheumatoid arthritis (RA) patients and found that it has a beneficial therapeutic effect. Here, we provide experimental evidence for the effectiveness of BDX-1 on RA in murine models. The oral administration of BDX-1 was found to markedly inhibit collagen-induced arthritis, adjuvant-induced arthritis, and zymosan-induced inflammation. It also inhibited carrageenan-induced acute edema and acetic acid-induced writhing response. In addition, the biological activity of BDX-1 was found to be strongly increased by fermentation. Our results suggest that BDX-1 could be useful for the treatment of rheumatoid arthritis.
