        Limonoids from the roots of Trichilia sinensis and their cytotoxicities

        Six new compounds (1–4, 8, 10), along with six known limonoids (5–7, 9, 11, 12), were isolated from the roots of Trichilia sinensis. Their structures were elucidated on the basis of extensive spectroscopic methods including 1H NMR, 13C NMR, DEPT, HSQC, HMBC, 1H–1H COSY and ROESY experiments, as well as by comparison with the literature. All the compounds were evaluated for cytotoxicities against K562, SGC-7901 and BEL-7402 cell lines. Compounds 2, 7, 10, 11, and 12 showed weak inhibitory activity to the selected cell lines.

        Peripheral nerve defects repaired with autogenous vein grafts flled with platelet-rich plasma and active nerve microtissues and evaluated by novel multimodal ultrasound techniques

        Background: Developing biocompatible nerve conduits that accelerate peripheral nerve regeneration, lengthening and functional recovery remains a challenge. The combined application of nerve microtissues and platelet-rich plasma (PRP) provides abundant Schwann cells (SCs) and various natural growth factors and can compensate for the deficiency of SCs in the nerve bridge, as well as the limitations of applying a single type of growth factor. Multimodal ultrasound evaluation can provide additional information on the stiffness and microvascular flow perfusion of the tissue. This study was designed to investigate the effectiveness of a novel tissue-engineered nerve graft composed of an autogenous vein, nerve microtissues and PRP in reconstructing a 12-mm tibial nerve defect and to explore the value of multimodal ultrasound techniques in evaluating the prognosis of nerve repair. Methods: In vitro, nerve microtissue activity was first investigated, and the effects on SC proliferation, migration, factor secretion, and axonal regeneration of dorsal root ganglia (DRG) were evaluated by coculture with nerve microtissues and PRP. In vivo, seventy-five rabbits were equally and randomly divided into Hollow, PRP, Micro-T (Microtissues), Micro-T + PRP and Autograft groups. By analysing the neurological function, electrophysiological recovery, and the comparative results of multimodal ultrasound and histological evaluation, we investigated the effect of these new nerve grafts in repairing tibial nerve defects. Results: Our results showed that the combined application of nerve microtissues and PRP could significantly promote the proliferation, secretion and migration of SCs and the regeneration of axons in the early stage. The Micro-T + PRP group and Autograft groups exhibited the best nerve repair 12 weeks postoperatively. In addition, the changes in target tissue stiffness and microvascular perfusion on multimodal ultrasound (shear wave elastography; contrast-enhanced ultrasonography; Angio PlaneWave UltrasenSitive, AngioPLUS) were significantly correlated with the histological results, such as collagen area percentage and VEGF expression, respectively. Conclusion: Our novel tissue-engineered nerve graft shows excellent efficacy in repairing 12-mm defects of the tibial nerve in rabbits. Moreover, multimodal ultrasound may provide a clinical reference for prognosis by quantitatively evaluating the stiffness and microvescular flow of nerve grafts and targeted muscles

        MiR-200c-3p aggravates gastric cell carcinoma via KLF6

        Background Gastric cell carcinoma (GCC) is a common and high-incidence malignant gastrointestinal cancer that seriously threatens human life and safety. Evidences suggest that microRNAs (miRNAs) exhibit an essential role in regulating the occurrence and development of GCC, while the efects and possible mechanisms remain to be further explored. Objective This study was designed to explore whether miR-200c-3p exerted its functional role in the growth and metastasis of GCC, and investigate the possible mechanisms. Methods The expression levels of miR-200c-3p in GCC tissues and cell lines were detected by qRT-PCR analysis. The functional role of miR-200c-3p in the viability, proliferation, migration and invasion of GCC cells were evaluated by CCK-8, EdU, wound healing and Transwell assays. In addition, the candidate targets of miR-200c-3p was predicted and confrmed by dual-luciferase reporter assay. Moreover, the relationship between miR-200c-3p and target (Krüppel like factor 6, KLF6) was assessed by qRT-PCR and western blot assays. Besides, the expression levels of KLF6 in GCC cells were determined by qRT-PCR and western blot assays. Furthermore, the role of KLF6 in the viability, proliferation, migration and invasion of GCC cells mediated with miR-200c-3p mimics was evaluated by CCK-8, EdU, wound healing and Transwell assays. Results In the present study, a new tumor promoting function of miR-200c-3p was disclosed in GCC. We found that the expression of miR-200c-3p was obviously increased in clinic GCC tissues and cell lines. In addition, down-regulation of miR-200c-3p suppressed cell viability, proliferation, migration, and invasion in GCC cells. Moreover, KLF6 was verifed as a direct target of miR-200c-3p by binding its 3’-UTR. Additionally, KLF6 was remarkably decreased and was negatively associated with the miR-200c-3p expression in GCC cell lines. Furthermore, over-expression of KLF6 retarded the efects of miR-200c-3p on the growth and metastasis of GCC cell lines. Conclusions MiR-200c-3p potentially played a tumor-promoting role in the occurrence and development of GCC, which may be achieved by targeting KLF6.

      • Zinc Oxide-Enhanced Piezoelectret Polypropylene Microfiber for Mechanical Energy Harvesting

        <P>This paper reports zinc oxide (ZnO)-coated piezoelectret polypropylene (PP) microfibers with a structure of two opposite arc-shaped braces for enhanced mechanical energy harvesting. The ZnO film was coated onto PP microfibers via magnetron sputtering to form a ZnO/PP compound structure. Triboelectric Nanogenerator (TENG) based on ZnO/PP microfiber compound film was carefully designed with two opposite arc-shaped braces. The results of this study demonstrated that the mechanical energy collection efficiency of TENG based on piezoelectret PP microfiber was greatly enhanced by the coated ZnO and high-voltage corona charging method. We found that, with the step-increased distance of traveling for the movable carbon black electrode, an electrical power with an approximately quadratic function of distance was generated by this mechanical-electrical energy conversion, because more PP microfibers were connected to the electrode. Further, with a full contact condition, the peak of the generated voltage, current, and charges based on the ZnO/PP microfibers by this mechanical-electrical energy conversion with 1 m/s<SUP>2</SUP> reached 120 V, 3 μA, and 49 nC, respectively. Moreover, a finger-tapping test was used to demonstrate that the ZnO/PP microfiber TENG is capable of lighting eight light-emitting diodes.</P> [FIG OMISSION]</BR>

        Distributed Power and Rate Control for Cognitive Radio Networks

        In this paper, a distributed power and end-to-end rate control algorithm is proposed in the presence of licensed users. By Lagrangian duality theory, the optimal power and rate control solution is given for the unlicensed users while satisfying the interference temperature limits to licensed users. It is obtained that transmitting with either 0 or the maximum node power is the optimal scheme. The synchronous and asynchronous distributed algorithms are proposed to be implemented at the nodes and links. The convergence of the proposed algorithms are proved. Finally, further discussion on the utility-based fairness is provided for the proposed algorithms. Numerical results show that the proposed algorithm can limit the interference to licensed user under a predefined threshold.

        The natural product salicin alleviates osteoarthritis progression by binding to IRE1α and inhibiting endoplasmic reticulum stress through the IRE1α-IκBα-p65 signaling pathway

        Despite the high prevalence of osteoarthritis (OA) in older populations, disease-modifying OA drugs (DMOADs) are still lacking. This study was performed to investigate the effects and mechanisms of the small molecular drug salicin (SA) on OA progression. Primary rat chondrocytes were stimulated with TNF-α and treated with or without SA. Inflammatory factors, cartilage matrix degeneration markers, and cell proliferation and apoptosis markers were detected at the mRNA and protein levels. Cell proliferation and apoptosis were evaluated by EdU assays or flow cytometric analysis. RNA sequencing, molecular docking and drug affinity-responsive target stability analyses were used to clarify the mechanisms. The rat OA model was used to evaluate the effect of intra-articular injection of SA on OA progression. We found that SA rescued TNF-α-induced degeneration of the cartilage matrix, inhibition of chondrocyte proliferation, and promotion of chondrocyte apoptosis. Mechanistically, SA directly binds to IRE1α and occupies the IRE1α phosphorylation site, preventing IRE1α phosphorylation and regulating IRE1α-mediated endoplasmic reticulum (ER) stress by IRE1α-IκBα-p65 signaling. Finally, intra-articular injection of SA-loaded lactic-co-glycolic acid (PLGA) ameliorated OA progression by inhibiting IRE1α-mediated ER stress in the OA model. In conclusion, SA alleviates OA by directly binding to the ER stress regulator IRE1α and inhibits IRE1α-mediated ER stress via IRE1α-IκBα-p65 signaling. Topical use of the small molecular drug SA shows potential to modify OA progression.

      • A Lesson to Learn from Conflicts between Chinese Tradition and Modernity

        Traditional Chinese culture is composed mainly of Confucianism, Buddhism. Those thought systems in evolution have greatly shaped Chinese society and have been affected by the changes of the times in return. Thanks to their positive qualities, chinese culture based on those three "isms" is embraced by world culture, as a part of the counterpart of Western culture and important compensation for Western loss and miss. On the other side its shortcomings and faults make Chinese tradition an awkward disfigurement in the social progress of modernity in the vital fields of Chinese life. It is observed that the conflicts between tradition and modernity are highly reflected in Chinese political life, economic development and the philosophical discipline. Modernity once suggested Western threat and now provides a bright vision for opening China. In traditional and anti-traditional China at a time it met strong resistance and is now warmly embraced with littler doubt about its negative effects. The good and bad qualities of tradition are combined to have produced confusion for modern China in its national modernization program starting from the end of the 19th century. Therefore it is not an easy job, moreover a constructive task to evaluate the positive and negative influences of Chinese tradition on Chinese life and Chinese psyche in the modern-world context right into the new century. My thesis will focus on China's modernization process during which conflicts arose between Chinese tradition and modernity by asking questions more than I am able to answer them. Making clear and sure of the function and influence of traditional Chinese values in modern Chinese life is for the purpose of overcoming the shortcomings of Chinese tradition and making the better part of Chinese culture true national legacy.

        Full Length cDNA, Genomic Organizations and Expression Profiles of the Porcine Proteasomal ATPases PSMC5 Gene

        PSMC5 subunit, which belongs to the 26S proteasomal subunit family, plays an important role in the antigen presentation mediated by MHC class I molecular. Full-length cDNA of porcine PSMC5 was isolated using the in silico cloning and rapid amplification of cDNA ends (RACE). Amino acid was deduced and the primary structure was analyzed. Results revealed that the porcine PSMC5 gene shares the high degree of sequence similarity with its mammalian counterparts at both the nucleotide level and the amino acid level. The RT-PCR was performed to detect the porcine PSMC5 expression pattern in seven tissues and the result showed that high express level was observed in spleen, lung, marrow and liver while the low express level was in muscle. The full-length genomic DNA sequence of porcine PSMC5 gene was amplified by PCR and the genomic structure revealed that this gene was comprised by 12 exons and 11 introns. Best alignment of the cDNA and genomic exon DNA sequence presents 4 mismatches and this information potentially bears further study in gene polymorphisms.

      • Structural Directed Growth of Ultrathin Parallel Birnessite on β-MnO<sub>2</sub> for High-Performance Asymmetric Supercapacitors

        <P>Two-dimensional birnessite has attracted attention for electrochemical energy storage because of the presence of redox active Mn<SUP>4+</SUP>/Mn<SUP>3+</SUP> ions and spacious interlayer channels available for ions diffusion. However, current strategies are largely limited to enhancing the electrical conductivity of birnessite. One key limitation affecting the electrochemical properties of birnessite is the poor utilization of the MnO<SUB>6</SUB> unit. Here, we assemble β-MnO<SUB>2</SUB>/birnessite core–shell structure that exploits the exposed crystal face of β-MnO<SUB>2</SUB> as the core and ultrathin birnessite sheets that have the structure advantage to enhance the utilization efficiency of the Mn from the bulk. Our birnessite that has sheets parallel to each other is found to have unusual crystal structure with interlayer spacing, Mn(III)/Mn(IV) ratio and the content of the balancing cations differing from that of the common birnessite. The substrate directed growth mechanism is carefully investigated. The as-prepared core–shell nanostructures enhance the exposed surface area of birnessite and achieve high electrochemical performances (for example, 657 F g<SUP>–1</SUP> in 1 M Na<SUB>2</SUB>SO<SUB>4</SUB> electrolyte based on the weight of parallel birnessite) and excellent rate capability over a potential window of up to 1.2 V. This strategy opens avenues for fundamental studies of birnessite and its properties and suggests the possibility of its use in energy storage and other applications. The potential window of an asymmetric supercapacitor that was assembled with this material can be enlarged to 2.2 V (in aqueous electrolyte) with a good cycling ability.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/ancac3/2018/ancac3.2018.12.issue-2/acsnano.7b03431/production/images/medium/nn-2017-03431b_0007.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nn7b03431'>ACS Electronic Supporting Info</A></P>
