        Identification and characterization of heat shock proteins in a parasitic wasp Chouioia cuneae (Hymenoptera: Eulophidae)

        Heat shock proteins (HSPs) are known to be induced in response to various stress factors. Although HSPs have been studied in a number of insects, not much is known about HSPs in the natural enemies of insects, especially parasitoids. In this study, we identified and characterized five full‐length HSP genes (Cchsp40, Cchsp60, Cchsp70, Cchsp83, and Cchsp90) from an endoparasitic chalcid wasp, Chouioia cunea, which parasitizes the fall webworm, Hyphantria cunea pupae, a worldwide pest. The expression of Cchsps in response to temperature, pesticide stresses and UV radiation were also investigated by quantitative real‐time polymerase chain reaction (RT‐qPCR). The results showed that all five Cchsps were induced in response to hot and cold temperatures. Four pesticides induced the abundant expression of Cchsp70, Cchsp83 and Cchsp90 while ultraviolet radiation up‐regulated Cchsp40, Cchsp70, Cchsp83 and Cchsp90. These results indicate the different transcriptional profiles of the five different Cchsps in response to various abiotic stresses. The findings of this study provide insights into the response of C. cunea to abiotic stresses and insight into the use of this parasitoid in biological control strategies.

        Improved lithium storage performance of CeO2-decorated SrLi2Ti6O14 material as an anode for Li-ion battery

        In this work, the CeO2-decorated SrLi2Ti6O14 anode was successfully prepared through a solid-state process. The space-charge effect induced by the internal adsorption of ions on the CeO2 surface can easilyresult in a formation of an excellent conductive interfacial layer between CeO2 and SrLi2Ti6O14. The goodelectrical contact between CeO2 and SrLi2Ti6O14 offers more active sites for the electrolyte storage andredox reaction, and promotes the intercalation/deintercalation of lithium ions, and thus improves therate performance and cycle stability. Due to its unique structure and composition, the CeO2-decoratedSrLi2Ti6O14 composites exhibit high reversible capacities, good cycle performance and outstanding rateperformance. Especially, the CeO2 (5 wt%)-decorated SrLi2Ti6O14 anode shows the most excellent electrochemicalperformance, which delivers a large charge capacity of 121.3 mAh g 1 and a capacity retentionof 94.48% after 100 cycles at 0.5 A g 1. However, the corresponding charge capacity and capacity retentionof pristine SrLi2Ti6O14 are 100.5 mAh g 1 and 86.77%, respectively. The CeO2(5 wt%)-decoratedSrLi2Ti6O14 with enhanced rate capacity, cycle stability and structural stability is a potential electrodematerial candidate for Li-ion battery.

        Microstructures and nano mechanical properties of the metal tungsten film

        The W film was prepared on 1045 steel by magnetron sputtering, with the thickness of 2 ㎛, its surface and cross-section morphologies were investigated with SEM, and the phase structure was analyzed with XRD. X-ray stress determinator was utilized to measure its residual stress, and the nano-hardness and elastic modulus of the film were surveyed by nano-indentation tester. The results show that the surface of W film is very compact and smooth; the particles arranged regularly, the granularity of the thin film is about 1 ㎛. The microcracks, cavities and desquamation were not found in the film and interface, and the bonding between the W film and substrate is well. The XRD results showed that the W film had a body-centered cubic structure, the lattice constant: ɑ = 0.316 nm, the growth preferred orientations are (110) and (220). The compressive stress (-169 MPa) was found on the surface. The average nano-hardness and elastic modulus of W film are 15.22 GPa, 176.64 GPa, respectively, and the mechanical properties of W film are well.         cDNA cloning, expression, and immunolocalization of gonadinhibiting hormone (GIH) in Litopenaeus vannamei

        In this study, the full-length GIH cDNA sequence from Litopenaeus vannamei was cloned from the eyestalk by reverse transcriptase polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends. The fulllength GIH cDNA was 865 bp with a 288 bp open-reading frame, which encoded a 96 amino acid prepro-GIH with 17 amino acid signal peptide. L. vannamei GIH (LvGIH) can be classified as a member of type-II crustacean hyperglycemic hormone polypeptide family. LvGIH shares 93.8 and 66.7 % amino acid sequence identity with GIH from Penaeus monodon, and the molt-inhibiting hormone from Marsupenaeus japonicas, respectively. By quantitative real-time PCR (qPCR), LvGIH mRNA transcripts were detected in fertilized eggs, nauplius, zoea, mysis and juveniles of 25, 35 and 40 days old. LvGIH transcript levels increased significantly with the development from fertilized eggs to juveniles. LvGIH transcript levels were highest in juveniles at 35 days old. By RT-PCR, LvGIH mRNA transcripts were detected only in the eyestalks and brains but not in the muscles, intestines, gills, heart, hepatopancreas, ovaries and testes of adults, and there was no difference in the expression level of LvGIH between males and females. Using the P. monodon anti-GIH antibody, we showed that LvGIH was located mainly in the XO-SG and slightly in axon, with similar fluorescence intensity found in XO and SG. To summarize, we have cloned and characterized the GIH of the shrimp L. vannamei. In addition to the GIH properties described in other crustaceans, a peak of LvGIH expression was identified at the time of sexual differentiation (i.e., day 35 larvae) suggesting that LvGIH may also be involved in the control of this process.

      • Knockdown of HMGN5 Expression by RNA Interference Induces Cell Cycle Arrest in Human Lung Cancer Cells

        HMGN5 is a typical member of the HMGN (high mobility group nucleosome-binding protein) family which may function as a nucleosomal binding and transcriptional activating protein. Overexpression of HMGN5 has been observed in several human tumors but its role in tumorigenesis has not been fully clarified. To investigate its significance for human lung cancer progression, we successfully constructed a shRNA expression lentiviral vector in which sense and antisense sequences targeting the human HMGN5 were linked with a 9-nucleotide loop. Inhibitory effects of siRNA on endogenous HMGN5 gene expression and protein synthesis were demonstrated via real-time RT-PCR and western blotting. We found HMGN5 silencing to significantly inhibit A549 and H1299 cell proliferation assessed by MTT, BrdU incorporation and colony formation assays. Furthermore, flow cytometry analysis showed that specific knockdown of HMGN5 slowed down the cell cycle at the G0/G1 phase and decreased the populations of A549 and H1299 cells at the S and G2/M phases. Taken together, these results suggest that HMGN5 is directly involved in regulation cell proliferation in A549 and H1299 cells by influencing signaling pathways involved in cell cycle progression. Thus, our finding suggests that targeting HMGN5 may be an effective strategy for human lung cancer treatment.

        Control Design for Stochastic Nonlinear Systems with Full-state Constraints and Input Delay: A New Adaptive Approximation Method

        In this paper, the full state constraints and input delay of stochastic nonlinear systems are studied. A new adaptive control algorithm is proposed using backstepping approach and multi-dimensional Taylor network (MTN) method. Firstly, the input delay problem is dealt with by introducing a new variable using the Padé approximation with Laplace transform. Secondly, MTNs are employed to approximate unknown nonlinear functions, and the barrier Lyapunov functions (BLFs) are constructed to deal with the state constraints. Based on this, a new approximation-based adaptive controller is proposed. Thirdly, it is proved that the proposed control method can ensure that all signals in the closed-loop system are semi-global ultimately uniformly bounded (SGUUB) in probability and the tracking error converges to a small neighborhood of the origin. Finally, two simulation examples are given to illustrate the effectiveness of the proposed design method.

        Mycena subpiligera sp. nov., a Symbiotic Species from China Associated with the Seed Germination of Gastrodia elata

        Mycena subpiligera, a new taxon in sect. Fragilipedes that can strongly enhance the germin- ation efficiency of Gastrodia elata seeds, was discovered in subtropical areas of China. As revealed by a morphological comparison with related Mycena species as well as maximum likelihood (ML) and Bayesian phylogenetic analyses based on sequences of the internal tran- scribed spacer (ITS) and the large subunit (LSU) regions of nuclear ribosomal RNA, the new taxon can be distinguished from phenotypically similar and phylogenetically related species. Optimal cultural conditions for M. subpiligera basidiomata are reported, and the germination rate of the new species is compared with that of M. citrinomarginata.

        Optimization of Processing Conditions of Chinese Smoke-cured Bacon (Larou) with a New Natural Coating Solution during Storage Period

        The study aims to determine the optimum sterilization rate and water activity of Chinese traditional smoke-cured bacon product (Larou) in the preservation with natural coating solution. With the response surface methodology (RSM), we analyzed 3 factors of processing conditions (the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan) and 2 response factors (sterilization rate and water activity). Sterilization rate and water activity of Larou were largely affected by the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan. The final optimum concentrations of lysozyme, sodium alginate, and chitosan were 0.09, 1.40, and 1.60% and realized the high sterilization rate. Water activity of sliced Larou was significantly correlated with the sterilization rate. Low-field nuclear magnetic resonance analysis verified the optimum processing conditions. The coating resulted in 99.69% rate of reducing bacteria after 30-day storage. The data of the total number of colony, peroxidation value, moisture content, pH, and sensory evaluation provided the theoretical basis for extending the shelf life of Chinese traditional smoke-cured bacon product (Larou) with natural coating solution.

      • Intracellular Mechanistic Understanding of 2D MoS<sub>2</sub> Nanosheets for Anti-Exocytosis-Enhanced Synergistic Cancer Therapy

        <P>Emerging two-dimensional (2D) nanomaterials, such as transition-metal dichalcogenide (TMD) nanosheets (NSs), have shown tremendous potential for use in a wide variety of fields including cancer nanomedicine. The interaction of nanomaterials with biosystems is of critical importance for their safe and efficient application. However, a cellular-level understanding of the nano-bio interactions of these emerging 2D nanomaterials (<I>i</I>.<I>e</I>., intracellular mechanisms) remains elusive. Here we chose molybdenum disulfide (MoS<SUB>2</SUB>) NSs as representative 2D nanomaterials to gain a better understanding of their intracellular mechanisms of action in cancer cells, which play a significant role in both their fate and efficacy. MoS<SUB>2</SUB> NSs were found to be internalized through three pathways: clathrin → early endosomes → lysosomes, caveolae → early endosomes → lysosomes, and macropinocytosis → late endosomes → lysosomes. We also observed autophagy-mediated accumulation in the lysosomes and exocytosis-induced efflux of MoS<SUB>2</SUB> NSs. Based on these findings, we developed a strategy to achieve effective and synergistic <I>in vivo</I> cancer therapy with MoS<SUB>2</SUB> NSs loaded with low doses of drug through inhibiting exocytosis pathway-induced loss. To the best of our knowledge, this is the first systematic experimental report on the nano-bio interaction of 2D nanomaterials in cells and their application for anti-exocytosis-enhanced synergistic cancer therapy.</P> [FIG OMISSION]</BR>

        Neural Network-based Event-triggered Adaptive Asymptotic Tracking Control for Switched Nonlinear Systems

        In this paper, an adaptive event-triggered asymptotic tracking control problem is addressed for switched nonlinear systems with unknown control directions. In existing control schemes, the proposed controller is not directly aimed at the original system, which affects the control performance. Different from the existing control schemes, based on the original system, an event-triggered control law is constructed in this paper. The proposed event-triggered controller guarantees that the tracking error ς1 asymptotically converges to the origin. Finally, the effectiveness of the proposed controller design scheme is proved by simulation examples.
