        Hypoxia Mediates Runt-Related Transcription Factor 2 Expression via Induction of Vascular Endothelial Growth Factor in Periodontal Ligament Stem Cells

        Periodontitis is characterized by the loss of periodontal tissues, especially alveolar bone. Common therapies cannot satisfactorily recover lost alveolar bone. Periodontal ligament stem cells (PDLSCs) possess the capacity of self-renewal and multilineage differentiation and are likely to recover lost alveolar bone. In addition, periodontitis is accompanied by hypoxia, and hypoxia-inducible factor-1α (HIF-1α) is a master transcription factor in the response to hypoxia. Thus, we aimed to ascertain how hypoxia affects runt-related transcription factor 2 (RUNX2), a key osteogenic marker, in the osteogenesis of PDLSCs. In this study, we found that hypoxia enhanced the protein expression of HIF-1α, vascular endothelial growth factor (VEGF), and RUNX2 ex vivo and in situ. VEGF is a target gene of HIF-1α, and the increased expression of VEGF and RUNX2 proteins was enhanced by cobalt chloride (CoCl2, 100 μmol/L), an agonist of HIF-1α, and suppressed by 3-(5′-hydroxymethyl-2′-furyl)-1-benzyl indazole (YC-1, 10 μmol/L), an antagonist of HIF-1α. In addition, VEGF could regulate the expression of RUNX2, as RUNX2 expression was enhanced by human VEGF (hVEGF165) and suppressed by VEGF siRNA. In addition, knocking down VEGF could decrease the expression of osteogenesis-related genes, i.e., RUNX2, alkaline phosphatase (ALP), and type I collagen (COL1), and hypoxia could enhance the expression of ALP, COL1, and osteocalcin (OCN) in the early stage of osteogenesis of PDLSCs. Taken together, our results showed that hypoxia could mediate the expression of RUNX2 in PDLSCs via HIF-1α-induced VEGF and play a positive role in the early stage of osteogenesis of PDLSCs

        Mechanical evaluation of polymer microneedles for transdermal drug delivery: In vitro and in vivo

        In this study, we reported two types of PMNs based on polylactic acid (PLA) and polyvinyl alcohol (PVA),respectively. Parafilm M film, porcine skin, and rats’ models were operated to evaluate the mechanicalproperties in vitro and in vivo to find optimal parameters for efficient insertion. Insertion depth was measuredusing Digital Force Gauge by changing insertion force and speed, respectively. Results showed thatincreasing the insertion force and speed used for PMNs application led to a significant increase in thedepth of insertion. A force of 18 N under a speed of 330 mm/min was the optimal condition for insertingPMNs array into ParafilmM film and porcine skin. In addition, PLA-MNs exhibited higher robustness andenhanced homogeneity in insertion depth compared with PVA-MNs, but PVA-MNs were able to reachmuch deeper insertion depth. Moreover, Sprague Dawley (SD) rat experiments confirmed the effectivenessof optimal insertion parameters for transdermal drug delivery. This study illustrated not only thedevelopment of novel PMNs but also the mechanical evaluation for the design of PMNs.

        In Vitro Evaluation and Monitoring of the Expression Level and Localization of Aldose Reductase Using Functionalized Quantum Dots and EGFP

        The optimization of aldose reductase (AR) expression levels and tracking of the AR expression sites within the cell is an essential step in developing a platform for the effective production of aldose reductase inhibitors (ARIs). In this study, we have demonstrated the use of both immunocytochemistry and quantum dots-based immunofluorescence techniques for observing and detecting the expression level and localization of AR in the cytoplasm and cell membrane of a eukaryotic cell model with high levels of AR protein expression. Our results show that high expression levels of human AR can be achieved using the eukaryotic cell model that we have developed. The overexpressed AR can be used for translational studies of hAR and the screening of ARIs. More importantly, the use of the established quantum dots-based immunofluorescence technique in the intracellular labeling of AR allows the determination of the expression and distribution of the AR gene. Overall, the use of the interdisciplinary approach of both genetic engineering and quantum dot-based immunofluorescence allows not only the effective production of a desired protein, but also the determination of the cellular localization of such an expressed protein.

        Polymorphisms in Epigenetic and Meat Quality Related Genes in Fourteen Cattle Breeds and Association with Beef Quality and Carcass Traits

        Improvement for carcass traits related to beef quality is the key concern in beef production. Recent reports found that epigenetics mediates the interaction of individuals with environment and nutrition. The present study was designed to analyze the genetic effect of single nucleotide polymorphisms (SNPs) in seven epigenetic-related genes (DNMT1, DNMT3a, DNMT3b, DNMT3L, Ago1, Ago2, and HDAC5) and two meat quality candidate genes (CAPN1 and PRKAG3) on fourteen carcass traits related to beef quality in a Snow Dragon beef population, and also to identify SNPs in a total of fourteen cattle populations. Sixteen SNPs were identified and genotyped in 383 individuals sampled from the 14 cattle breeds, which included 147 samples from the Snow Dragon beef population. Data analysis showed significant association of 8 SNPs within 4 genes related to carcass and/or meat quality traits in the beef populations. SNP1 (13154420A>G) in exon 17 of DNMT1 was significantly associated with rib-eye width and lean meat color score (p<0.05). A novel SNP (SNP4, 76198537A>G) of DNMT3a was significantly associated with six beef quality traits. Those individuals with the wild-type genotype AA of DNMT3a showed an increase in carcass weight, chilled carcass weight, flank thicknesses, chuck short rib thickness, chuck short rib score and in chuck flap weight in contrast to the GG genotype. Five out of six SNPs in DNMT3b gene were significantly associated with three beef quality traits. SNP15 (45219258C>T) in CAPN1 was significantly associated with chuck short rib thickness and lean meat color score (p<0.05). The significant effect of SNP15 on lean meat color score individually and in combination with each of other 14 SNPs qualify this SNP to be used as potential marker for improving the trait. In addition, the frequencies of most wild-type alleles were higher than those of the mutant alleles in the native and foreign cattle breeds. Seven SNPs were identified in the epigenetic-related genes. The SNP15 in CAPN1 could be used as a powerful genetic marker in selection programs for beef quality improvement in the Snow Dragon Beef population.

        中国经济奇迹的标志性与持续性 -纪念中国改革开放政策实行30周年-

        일반적으로 사람들이 예상하는 성공범위를 초과하는 1개 국가 또는 지역의 경제발전에 대해 ‘경제 기적’을 이룩하였다고 말한다. 사회주의경제체제 하에서 시작된 중국경제는 그 후 전혀 다른 2단계, 즉 계획경제체제와 시장경제체제를 거쳐 지금에 이르렀다. 비록 중국이 1단계인 계획경제체제하에서 연평균 7%의 고속성장을 유지해왔지만 이에 대해 어느 누구도 중국경제의 기적이라고 표현하지는 않았는데, 그 이유는 다음과 같은 원인에서 비롯되었다. 첫째, 기존에 존재하던 계획경제제도하에서 이룩된 것으로서, 이미 전례가 있었기 때문에 ‘기적’이라는 표현을 사용하기에는 부족함이 있었고 그에 따른 문제점들도 선명하게 드러났기 때문이다. 둘째, 그 당시 고속성장을 지속하기 위해 지불한 사회․경제적인 비용이 너무 커서 계속 유지하기 어려웠기 때문이다. 셋째, 비(非)경제적 요인들에 의한 경제의 안정적인 성장을 지속할 수 있는 환경조성이 어려웠기 때문이다. 그러나 2단계, 즉1978년 시장경제방식으로 전환한 후 30년간 이룩한 경제성장 및 발전을 지켜본 후, 전세계는 점차 중국경제의 성공을 깨닫기 시작했다. 이러한 중국의 경제발전은 20세기후반 20년부터 21세기전반 10년간에 걸쳐 보기 드문 ‘경제적인 기적’이라고 할 수 있다. 중국이 개혁ㆍ개방정책을 추진한 30년 동안 이룩한 ‘경제기적’의 상징성은 기본 경제체제가 공유제 중심의 계획경제체제에서 시장경제체제로 전환하였다는 점에서 찾을 수 있다. 특히 이러한 전환과정에도 불구하고 중국경제의 고속성장은 인플레이션현상 없이 이루어졌다는 점이다. 그러나 경제적 기적의 이면에는 개인간 소득격차의 확대, 에너지 과소비에 따른 환경오염의 유발 등 문제점들도 동시에 나타났다. 따라서 앞으로 중국이 ‘경제 기적’을 지속하기 위해서는 첫째, 과거의 성장방식에서 벗어나 새로운 성장방식으로의 전환, 둘째, 원초기술의 확보, 혁신의 중시, 집중적인 기술개발 및 기술수입 후 재창조 등 혁신과정의 조합, 셋째, 기술의 보호, 기술개발의 촉진 및 효율적인 제도적 혁신, 넷째, 지속적으로 조화로운 거시경제 조정정책의 추진 등 4가지 조건을 유지하고 개선시켜 나가야 할 것이다.

        Potent in Vitro Anticancer Activity of Metacycloprodigiosin and Undecylprodigiosin from a Sponge-Derived Actinomycete Saccharopolyspora sp. nov.

        Bioassay-guided fractionation of $CHCl_{3}$ extract from the fermentation broth of a sponge Mycale plumose-derived actinomycete Saccharopolyspora sp. nov., led to the isolation of two known prodigiosin analogs - metacycloprodigiosin (1) and undecylprodigiosin (2). These compounds exhibited significant cytotoxic activities against five cancer cell lines: P388, HL60, A-549, BEL­7402, and SPCA4. This is the first report on the significant cytotoxicity of metacycloprodigiosin (1) against human cancer cell lines.

        Symmetries and new exact solutions of the novel (3+1)-dimensional sinh-Gorden equation

        In the present paper, some new exact solutions of the (3+1)-dimensional sinh-Gorden equation are displayed. First, based on the Lie point symmetry, basic infinitesimal generators given; also, and the Lie algebra and commutative relations are shown. Second, some exact solutions are obtained using the F-expansion method, and a great many Jacobian-elliptic function solutions are presented. Finally, other forms of solutions are derived using the traveling wave transform.

        Synthesis and characterization of core–shell associative polymer that prepared by oilfield formation water for chemical flooding

        A novel core–shell polymer (HBAPAM) with functionalized nano-SiO2 as core, multibranched associativepolymeric chains as shell was synthesized through in-situ free radical polymerization strategy. TGA, IR,1H NMR, and SEM proved the notion and method of preparing of HBAPAM was applicable. The core–shellstructure and multibranched morphology enhanced the supramolecular interaction of HBAPAM, whichdisplayed the desirable viscosified efficiency and viscoelasticity, especially, long-term stability in harshtemperature and salinity condition. HBAPAM featured the rheology in multiscale consistently indeionized water and formation water. Coreflooding experiments confirmed that HBAPAM had high costperformance in the variable permeability of oilfields.

        Quality improvement effects of electrolyzed water on rice noodles prepared with semidry-milled rice flours

        To investigate the effects of electrolyzed watertreatment on the qualities of rice noodles prepared withsemidry- milled rice flour, pasting properties and thermalproperties of rice flour, and the cooking and texturalproperties of rice noodles were determined. Higher peakviscosity and lower melting enthalpy were observed inelectrolyzed water (EW) treated rice flour. The hardness,gumminess and chewiness of rice noodle in slightly acidicelectrolyzed water treated rice noodles with availablechlorine concentration (ACC) 20.32 mg/L were increasedsignificantly (p\0.05). The cooking loss decreased significantlyin strong acidic electrolyzed water treated noodleswith ACC 10.09 mg/L treatment (p\0.05). Theresults indicated that EW could promote the gelatinizationof rice flour, and improve the textural qualities of ricenoodles. Therefore EW was appropriate to be used in ricenoodle production.
