Dopamine transporter (DAT) is a highly specialized membrane-spanning protein that aids in terminating dopaminergic neurotransmission by sodium-dependent reuptake of released dopamine into presynaptic neurons. In present study, the cells which stablyexpressed DAT were prepared y transfection of CV-1 cells (monkey kideny cell line) with plasmid pRc/CMV-DAT which is the expression vector pRc/CMV-based plasmid containing bovine DAT cDNA. CV-1 cells, transfected with pRc/CMV-DAT, stably expresses a high affinity DAT: in dopamine uptake assay, the cpm value uptaken by transfected cells was 30-50 folds higher than those of nontransfectants. The transfected cells accumulated [³H]dopamine in a dose-dependent manner with a Km of 991.6nM. Even though high doses of norpinephrine, epinephrine, serotonin, and choline neurotransmitters inhibited the uptake of [³H]dopamine, DAT espressed in transfected cells was proven to be much more specific to dopamine. The psychotropic drugs such as GBR12909, DFT(cocaine congener), normifensine, clomipramine, desipramine, and impramine inhibited significantly the dopamin uptake in tissue culture cells stably transfected with DAT cDNA. It has been known that rat and human DAT generally have the activities of Parkinsonism-inducing neurotoxin, MPPㆍ-uptake and cocaine-binding. However, MPPㆍ uptake activity of expressed bovine DAT was significantly lesser than that of expressed rat DAT< and no CFT binding activity was seen. Radioactive in situ hybridization was done to map the cellular localization of DAT mRna containing cells inthe adult rat central nervous system. The strong hybridization signals were detected only in the substantia nigra pars compacta and ventral tegmental area. The restricted anatomical localization of DAT mRNA-containing cells confirms the DAT as a presynaptic marker of dopamine containing cells confirms the DAT as a presynaptic marker of dopamine containing cells in the rat brain. All of ? species homologuses of the DAt(rat, bovine, and human) possess consensus sites for phosphorylation by cAMP-dependent protein kinase A(PKA), Ca?-calmodulin-dependent kinase Ⅱ(Cam kinase Ⅱ), and protein kinase C(PKC). The existence of putative phosphorylation sites indicates that the functions of DAT may be regulated by protein kinases or phosphatases. Concentration-and time-course preincubations of DAT expressing cells with protein kinase activators or inhibitors such as phorbol 12-myristrate 13-acetate (PMA, a protein kinase C(PKC) activator), staurosphorin(a nonspecific protein kinase inhibitor), calphostin C(a specific PKC inhibitor), forskolin (a cAMP-dependent protein kinase A activator), and EGTA (Ca?-chelating agent, a effector of Ca?-calmodulin-dependent kinase Ⅱ) at 37℃ were followed by DAT functional assays. Dopamine uptake into DAT expressing cells was severly decreased by PMA pretreatment, and increased by staurosphorin or calphostin C. PKC activator and inhibitors showed similar effects on MPPㆍ uptake of DAT. However, DAT functions were not altered by forskolin or EGTA. These results implies that the functions of DAT modulated by protein kinase C.

This study was conducted to determine the effect of treatment with intravenous human immunoglobulin G (hIVIgG) on outcome in dogs with idiopathic immune-mediated hemolytic anemia (IMHA), and to identify prognostic variables that determine outcome in affected dogs. Thirty-seven dogs that met the inclusion criteria were enrolled in a retrospective study. The dogs were categorized into two groups based on their having received hIVIgG. There was no significant difference in survival between the hIVIgG group and the non-hIVIgG group. Mortality during hospitalization and at 1 month, 1 year, or 2 years after discharge was not significantly different between the hIVIgG and the non-hIVIgG groups. Hemoglobinuria was significantly less prevalent in dogs that lived more than 1 year than in those who lived less than 1 year, and was less prevalent in dogs that lived more than 2 years than in those who lived less than 2 years. However, there was no difference in the presence of hemoglobinuria between dogs that lived less than 1 month and those that lived more than 1 month. Overall, there was no evidence of a beneficial effect of hIVIgG in dogs with idiopathic IMHA.

N/A The epidemiology and clinical aspect of peptic ulcer is ever-changing along with changes in environment. Therefore, we had a clinical analysis in 1501 patients, who were diagnosed as peptic ulcer with gastroscopy in Chungnam National University Hospital from January 1984 to June 1988. The results were as follows: 1) Of the total 8912 cases of endoscopy, the numbers of patient with gastric ulcer were 784(8.8%), with duodenal ulcer, 591(6.6%) and with combined ulcer, 126(1.4%) 2) The mean age of patients with gastric ulcer was 50.9+14.9, and that with duodenal ulcer was 41.76+13.8. 3) The ratio of males to females was 3.7: 1 in gastric ulcer group and 2.4: 1 in duodenal ulcer group. 4) The most common stympom was epiagastric pain, 48.3% of gastric ulcer patients and 47.2% of duodenal ulcer patients, And the patients with hematemesis and/or melena were 19.5% of gastric ulcer patients and 12.9% of duodenal ulcer patients 5) During the period 1984-1988, the annual proportion of gastric ulcer were 58.8%, 51.4%, 53.3%, 50%, and 50.7%. And that of duodenal ulcer were 36.5%, 39.4%, 38.2%, 40.5%, and 41.1%. 6) The patients with multiple ulcers were 26.5% of gastric ulcer group and 15.7% of duodenal ulcer group. 7) The patients of 78.1% of total peptic ulcer was in active stage, 11% in healing stage, and 10.9% in scarring stage. 8) In 30.8% of gastric ulcers, the size of ulcer was within 1.0 to 1.4 cm and in 31.6% of duodenal ulcers the size of ulcer was less than 0.4 cm. 9) The most common site of gastric ulcer was angle of stomach (42.7%), and that of duodenal ulcer was bulb (98.6%). 10) The proportion of patients with combined ulcer of stomach and duodenum was 8.3% of total patients and that of channel ulcer was 3.1%.

저자들은 전신성 강직경련 및 발육지경, 호흡곤란으로 입원하여 추적관찰 중이던 15개월 남아에서 낮은 혈청 구리농도와 ceruloplasmin 농도를 보이며 전형적인 임상증상과 ATP7A의 Exon 19에서 점돌연변이를 보인 특징적인 Kinky hair 질환 1례를 경험하였기에 보고하는 바이다. Kinky hair disease is X-lined recessive neurodegenerative disorder produced by defects in a gene(ATP7A) that encodes an intracellular copper-transporting ATPase. About 90-95% of the patients have a severe clinical course leading to death in early childhood. ATP7A mutations associated with Menkes disease show great variety from cytogenetic abnormalities to partial gene deletions to single base-pair changes. We experienced a 15 month old boy with loss of developmental milestones, hypotonia, seizures and failure to thrive. On laboratory findings, the levels of serum copper and ceruloplasmin were low. Electron microscopy of hair illustrated pathognomic pili torti and other abnormalities such as trichorrhexis nodosa and trichoptilosis(longitudinal splitting of the shaft). Brain magnetic resonance image showed diffuse cerebral and cerebellar atrophy with tortousity of cerebral blood vessels. Genetic defect was evaluated. Our sequencing data on the amplified exon 19 of ATP7ase genomic DNA confirmed point mutation, G1255A, resulting in a glycine-to-arginine conversion. So, we report a brief view with the related literatures.
신장이식 환자들의 면역억제제 복약에 대한 이해도 및 이행도에 관한 조사

Survey on comprehension and compliance of immunosuppressant medication was conducted in 97 kidney transplant recipients at the organ transplant clinic in Seoul National University Hospital from Septemer 23 to October 25, 1996. Medication teaching and patient counseling pharmacist consulted with patients by direct or telephone communications and checked medical records of them to find whether the patients understand about their medications, such as drug name, the reasones of medications, side effects, etc. From the present survey, it was noticed that noncompliance was 9.7%, which was lower percent than that of other reports or other diseases. Also the compliance was not effected significantly by ages(p=0.46) or duration of post-transplantation(p=0.148). The most of patients understood well about immunosuppresants which were administered to them, such as the drug name(98%), dosage form and content(96%) and variable adverse effects, but they didn't understand about the importance of therapeutic drug monitoring(TDM) of cyclosporine and blood sampling condition (fasted). Also they understood well about the risk of the co-administration of other drugs(98%) and 47% of them knew that the reasons of risks were resulted from drug interaction. From the results of present study, it was found that noncompliance of immunosuppresants was low percent, but in order to maximize long-term renal allograft survival, the pharmacists must instruct the organ transplant patients about various informations which they didn't understand, such as TDM and drug interactions.
난소적축술로 인한 폐경기 여성에서 호르몬 대체요법이 우울증에 미치는 효과

Objective: To determine the quantitative and contents of depressive trends in premenopausal and postmenopausal group performed with total abdominal hysterectomy with both adnexectomy and HRT group. Method: The depressive trends were measured by SDS scoring keys in 125 premenopausal women and 68 postmenopausal women. And they are reexamed with SDS scoring after HRT for 3 months (HRT group). Result: The depression score, SDS were increased in postmenopausal group compared to control group (P<0.05) and decreased in HRT group (P<0.02). In the severity of depression, in decisiveness ranked first in control group but, in postmenopausal and HRT group decreased libido ranked first. Conclusion: These results suggests that HRT in oophorectomized postmenopausal women depressed mood but, unfortunately decreased libido seems not to be improved with HRT for 3 months.
