Saussurea pulchella (Asteraceae) is widely distributed in Korea and has been used in Korean folk medicine for the treatment of inflammation, hypertension, hepatitis, and arthritis. Pulchellamin G is an amino acid-sesquiterpene lactone conjugate isolated from S. pulchella. In the present study, we focused on the anti-inflammatory effect of pulchellamin G, which acts by inducing the expression of heme oxygenase (HO)-1. HO-1 plays important roles in cytoprotection since it has antioxidant, anti-inflammatory, antiproliferative, and antiapoptotic properties. Pulchellamin G inhibited the mRNA and protein expression of inducible nitric oxide synthase (iNOS), iNOS-derived nitric oxide (NO), and cyclooxygenase (COX)-2 and COX-derived prostaglandin E2 (PGE<SUB>2</SUB>) production in lipopolysaccharide (LPS)-stimulated murine peritoneal macrophages. The compound also reduced tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) production and suppressed the phosphorylation and degradation of IκB-α and nuclear translocation of p65 in murine peritoneal macrophages in response to LPS stimulus. The inhibitory effects of pulchellamin G on nuclear factor kappa B (NF-κB) translocation was impaired by co-treatment of the cells with HO activity inhibitor tin protoporphyrin (SnPP). By using SnPP, we verified that the inhibitory effects of pulchellamin G on the pro-inflammatory mediators NO, PGE<SUB>2</SUB>, TNF-α, and IL-1β are associated with induction of HO-1 expression. Our data suggest that pulchellamin G might have potent therapeutic effects and it should be considered in the development of treatments for various inflammatory diseases.

<P>We performed mass spectrometry (MS)-based metabolite profiling of Cheonggukjang (CGJ) and three types of garlic (normal, sprouted, and fermented) added CGJ (G-CGJ). Metabolite profiling suggested that the major distinguishing factor between the four types of CGJ lay in whether garlic was added, although different metabolic states were observed in G-CGJ. Among the discriminant metabolites between CGJ and G-CGJ, the levels of four amino acids such as L-valine, L-isoleucine, L-leucine, and glycine were decreased in G-CGJ compared to CGJ because garlic inhibited the protease activity of Bacillus subtilis during fermentation. In addition, the relative contents of L-cysteine, s-allyl-cysteine, s-allyl-cysteine sulfoxide, soyasaponin beta g, and soyasaponin gamma g, which showed positive correlation with antioxidant activities, were high in G-CGJ. These results suggest that MS based metabolite profiling could be a useful tool for understanding the metabolic differences of fermented foods according to the additives, and their relationship with antioxidant activity. <P><B>Abstract</B></P> <P>Single grid-based phase-contrast x-ray imaging (g-PCXI) technique, which was recently proposed by Wen et al. to retrieve absorption, scattering, and phase-gradient images from the raw image of the examined object, seems a practical method for phase-contrast imaging with great simplicity and minimal requirements on the setup alignment. In this work, we developed a useful simulation platform for g-PCXI and performed a simulation to demonstrate its viability. We also established a table-top setup for g-PCXI which consists of a focused-linear grid (200-lines/in strip density), an x-ray tube (100-μm focal spot size), and a flat-panel detector (48-μm pixel size) and performed a preliminary experiment with some samples to show the performance of the simulation platform. We successfully obtained phase-contrast x-ray images of much enhanced contrast from both the simulation and experiment and the simulated contract seemed similar to the experimental contrast, which shows the performance of the developed simulation platform. Purpose: This study aims to identify the effects of self-esteem, relationship with children, Social Networking Services(SNS) use intensity on depression in middle-aged women with college-age children, and to provide basic data for the health promotion of middle-aged women. Method: A sample of 133 middle-aged people was purposely selected from parents of university students. Questionnaires used for this study were CES-D(The Center for Epidemiological Studies-Depression Scale), Self-esteem Scale, Relationship with Children Scale, SNS Intensity Scale. The collected data was analyzed by SPSS 21 with descriptive statistics, independent t-test, ANOVAs, Pearson correlation, multiple regression. Result: Self-esteem was correlated with relationship with children (r=0.385, p<0.01) and depression (r=-0.440, p<0.01). Relationship with children was correlated with depression(r=-0.402, p<0.01). As a result, a factor having the greatest influence on the subject's depression degree was self-esteem(β=-0.335), followed by relationship with children(β=-0.274). Conclusion: The result of this study suggested that depression in middle-aged women is closely related to self-esteem and relationship with children. This result can be used in Nursing by developing the theoretical basis for education program related to enhancing self-esteem and relationship with children.

We fabricated the amorphous chalcogenide As_2S_3 thin film by thermal evaporation. We also measured and analysed the optical constants of refractive index, absorption coefficient, thickness of deposited amorphous As_2S_3 thin film and optical energy gap by simple measurement of transmittance spectrum.

We fabricate a Cu<SUB>2</SUB>ZnSnS<SUB>4</SUB> (CZTS) absorber layer, by using single step electrodeposition of CZTS precursor, deposited at -1.05 V, followed by high temperature annealing in a sulfur atmosphere. X-ray diffraction pattern indicates that the as-grown sample is amorphous in nature, and polycrystalline CZTS thin films with kesterite crystal structure have been obtained by sulfurization from 450 to 580 <SUP>o</SUP>C. Surface morphologies of the as-grown sample show some voids with agglomerated particles. After sulfurization, the morphologies of the annealed samples become more uniform, and dense. EDAX study reveals that the sulfurized samples are nearly stoichiometric, being Cu-rich and S-deficient in composition. The band gaps of the annealed samples are found to be in the range from 1.9 to 1.5 eV.

Peroxiredoxin (Prx) is a family of bifunctional proteins that exhibit peroxidase and chaperone activities. Prx proteins contain a conserved Cys residue that undergoes a redox change between thiol and disulfide states. 2-Cys Prx enzymes, a subgroup of Prx family, are intrinsically susceptible to reversible hyperoxidation to cysteine sulfinic acid during catalysis. Cysteine hyperoxidation of Prx was shown to result in loss of peroxidase activity and a concomitant gain of chaperone activity. Reduction of sulfinic Prx enzymes, the first known biological example of such a reaction, is catalyzed by sulfiredoxin (Srx) in the presence of ATP. Srx appears to exist solely to support the reversible sulfinic modification of 2-Cys Prx enzymes. Srx specifically binds to 2-Cys Prx enzymes by recognizing several critical surface-exposed residues of the Prxs, and transfer the γ-phosphate of ATP to their sulfinic moiety, using its conserved cysteine as the phosphate carrier. Effects of culture medium supplements and osmotic stress treatment on embryogenic callus induction and somatic embryogenesis were investigated in order to optimize tissue culture conditions of alfalfa(Medicago sativa L.). SH medium containing 5 ㎎/L 2,4-D and 0.2 ㎎/L kinetin was optimal for embryogenic callus induction from cotyledon tissue of alfalfa. Somatic embryos were formed when the embryogenic callus was cultured on SH medium supplemented with 1 ㎎/L 2,4-D and 2 ㎎/L BA. Supplementation of 5 mM L-pro-line and 1g/L casein hydrolysate into the regeneration medium further increased plant regeneration frequency. Osmotic stress treatment of callus appeared to improve the frequency of somatic embryo formation, but the frequency of somatic embryo formation differed by the osmotic stress treatment using different osmotic stressors. The highest plant regeneration frequency of 30.7% was observed when embryogenic callus was treated with 0.7M sucrose for 18h. Efficient regeneration system established in this study will be useful for molecular breeding of alfalfa through genetic transformation.
