유산양 유방염에 대한 Chitosan 약침의 치료 효과

To improve the therapeutic method for mastitis, treatment effect of chitosan pharmacopuncture was examined in three milking goats(A,B and C). Mastitis was artificially induced in two milking goats(A and B) and one goat(C) was naturally infected case with mastitis. The acupoint used was Yang-Ming and chitosan suspensions(provided from Tottory University, Japan: 30㎎ of chitosan/㎖)were injected to the acupoint with 5㎖/time for 3 days, respectively. As for the change of somatic cells in milk along with chitosan pharmacopuncture, they showed a decreasing tendency after increase on the 3rd day in all udders of B. In addition, as for the change of total leukocyte counts of peripheral blood with chitosan pharmacopuncture, they showed a decreasing tendency after increase on the 3rd day in B and C except A which showed a increasing tendency until on the 7th day. In the change of neutrophil/lymphocyte(N/L) ratio, increasing tendency was seen in A and C but increasing tendency after slight decrease was observed in B. Further, in the change of serum total protein and A/G ratio, serum total protein content was decreased after slightly increase on the 3rd day in A, and they were decreased after increase until on the 7th day in B and C, respectively. As for the change of A/G ratio, different pattern increasing tendency was shown in A, B and C.

A 3-month-old, 3.85kg female Jin-do dog with chief complaint of bloody diarrhea and depression was referred to the Veterinary Teaching Hospital, Chungnam National University. In fecal examination, trophozoites of giardia were observed and It was diagnosed as canine giardiasis. For treatment, metronidazole was administered orally for 5 days at a dose of 22mg/kg q12h. Diarrhea was not observed on 3 days after medication and most of clinical signs were disappeared on 7 days after medication with metronidazole. General conditions and activitivities became good on 13 days after medication. In addition, cysts or trophozoites of giardia were not detected any more in fecal examination using floating method with zinc sulphate solution. The present patient was a case of canine giardiasis that showed favorable response to metronidazole.

Investigation of the melanogenesis inhibitory effect of barnyard millet grain extracts (BME) led to isolation of compounds against mushroom tyrosinase and B16 melanoma cells. Chromatography yielded five phenolic compounds, among which n-p-coumaroyl serotonin (1, CS) exhibited potent non-competitive inhibition (IC50 =8.8 μM) compared with that of kojic acid (IC50=14.6 μM) against mushroom tyrosinase. More importantly,CS decreased melanin content to eight times lower (IC50 =11.6μM) in B16 melanocytes than that of feruloyl serotonin (2, FS,IC50 =86.5 μM) without cytotoxicity. CS significantly inhibited the protein expression of tyrosinase, tyrosinase-related protein-1(TRP-1), TRP-2, and microphthalmia-associated transcription factor using immunoblotting. Furthermore, molecular docking simulations were also carried out to investigate the structure related to the inhibitory activities. Therefore, BME can be used as a natural source of depigmentation to prevent melanogenesis.

To handle the development of antifungal drug resistance, the development of new structural modules and new modes of action for antifungals have been highlighted recently. Here, the antifungal activity of quinonemethidal triterpenoids such as celastrol, dihydrocelastrol, iguestein, pristimerin, and tingenone isolated from Tripterygium regelii were identified (MIC 0.269- 19.02 μM). C. glabrata was the most susceptible to quinonemethide among the tested fungi. Furthermore, quinonemethide suppressed cyctochrome c peroxidase expression dramatically, decreasing fungal viability caused by the accumulation of hydrogen peroxide. Thus, cyctochrome c peroxidase downregulation of quinonemethide may be a key mode of action for antifungals.
