        Growth Performance and Carcass Characteristics of Korean Native Ducks Fed Diets with Varying Levels of Limiting Amino Acids

        There are multiple experiments conducted with male Korean native ducks (KND) to evaluate the optimal levels of limiting amino acids (AA). In Exp. 1, a total of 450 one-d-old male KNDs were divided into five groups with six replicates and fed experimental diets with varying levels of lysine, total sulfur amino acids (TSAA) and threonine (T1, 0.90/0.74/0.70%; T2, 1.00/0.82/0.77%; T3, 1.10/0.90/0.85%; T4, 1.20/0.98/0.93%; T5, 1.30/1.07/1.01%) to 21 d of age. In Exp. 2, one-d-old male KND were received and fed commercial starter diet from hatching to 21 d of age, and then divided into five groups with six replicates and fed one of five diets varying levels of lysine, TSAA, and threonine (T1, 0.73/0.62/0.54%; T2, 0.80/0.68/0.60%; T3, 0.87/0.74/0.65%; T4, 0.94/0.80/0.70%; T5, 1.01/0.86/0.75%) during 22 to 56 d of age, respectively. The BW gain was linearly increased as dietary limiting AA levels increased to 1.20% lysine, 0.98% TSAA and 0.93% threonine. There were no significant differences in feed intake, gain:feed and uniformity among groups. In Exp. 2, the BW gain and gain:feed were not affected by dietary limiting AA levels. There were no significant differences in carcass characteristics and meat quality among groups. The growth performance and carcass characteristics did not show the significant response to increasing dietary limiting AA levels in KND during 22 to 56 d of age. In conclusion, the levels of lysine, TSAA and threonine necessary to maximize growth for starter phase were at least 1.20%, 0.98%, and 0.93%, respectively. On the other hands, KND require relatively low levels of limiting AA for late growth and carcass yield. The dietary levels of 0.73% lysine, 0.62% TSAA and 0.54% threonine appear to be adequate during growing phase.

      • Characterization of a family B DNA polymerase from Thermococcus barophilus Ch5 and its application for long and accurate PCR

        <P>The family B DNA polymerase gene from the euryarchaeon Thermococcus barophilus Ch5 (Tba5) contains an open reading frame of 6198 base pairs that encodes 2065 amino acid residues. The gene is split by three inteins that must be spliced out to form the mature DNA polymerase. A Tba5 DNA polymerase gene without inteins (genetically intein-spliced) was expressed under the control of the pET-28b(+)T7lac promoter in E. coli Rosetta 2(DE3)pLysS cells. The molecular mass of the purified Tba5 DNA polymerase was about 90 kDa consistent with the 90,470 Da molecular mass calculated based on the 776 amino acid sequence. The optimal pH for Tba5 DNA polymerase activity was 7.5 and the optimal temperature was 70-75 degrees C. The enzyme possessed 3' -> 5' exonuclease activity and was activated by magnesium ions. PCR amplification using Tba5 DNA polymerase enables high-yield for 1- to 6-kb target DNA products, while 8- to 10-kb target DNA products were amplified at low or inefficient levels. To simultaneously improve product yield and amplification fidelity, Tba5 plus DNA polymerase mixtures were constituted with various amounts of Tba5 DNA polymerase mixed with Taq DNA polymerase. The Tba5 plus DNA polymerase mixtures robustly amplified up to 25-kb lambda DNA fragments. In addition, the PCR error rate of Tba5 plus3 and Tba5 plus4 mixtures were much lower than those of wild-type Tba5 DNA polymerase, Pfu DNA polymerase, Taq DNA polymerase, and Pfu plus DNA polymerase.       • Trichloroethylene 취급 작업장의 공기 중 농도와 근로자의 요중 Trichloroacetic Acid 농도의 상관관계에 관한 조사연구

        부산ㆍ경남지역의 Trichloroethylene 취급 사업장 중 4개업종 20개소를 대상으로 기중 Trichloroethylene 농도를 측정하고, 이들 사업장 근로자 중 101명을 대상으로 요중 Trichloroaceticacid 농도를 분석하여 Trichloroethlene 폭로 정도를 평가하였다. Trichloroethylene, which is a chlorinated unsaturated hydrocarbon, is widely used as a solvent in degreasing operations. In the present study, the time-weighted average intensity of trichloroethylene exposure at work environments in 20 manufactures of 4 industries was assessed, at the same time trichloroacetic acid excretion in urine among 101 workers exposed to trichloroethylene was measured, and the exposure-excretion relationship was investigated. The results were summarized as follows; 1.The environmental trichloroethylene level was highest in metal machine industry(121.3ppm), followed by fabricated metal industry (106.0ppm), precision machine industry(65.0 ppm) and electrical machine industry(48.0 ppm). 2.The rate of urinary trichloroacetic acid concentration over 75mg/l among 101 workers was 42.6%(43 workers). 3.There is a linear correlation between the environmental exposure and urinary excretion(Y=1.05584X+35.65591, r=0.52401, p=0.0000).

      • Rapid and label-free bioanalytical method of alpha fetoprotein detection using LSPR chip

        Alpha fetoprotein (AFP) is a cancer marker, particularly for hepatocellular carcinoma. Normal levels of AFP are less than 20ng/mL; however, its levels can reach more than 400ng/mL in patients with HCC. Enzyme linked immunosorbent assay (ELISA) and radioimmunoassay (RIA) have been employed for clinical diagnosis of AFP; however, these methods are time consuming and labor intensive. In this study, we developed a localized surface plasmon resonance (LSPR) based biosensor for simple and rapid detection of AFP. This biosensor consists of a UV-Vis spectrometer, a cuvette cell, and a biosensor chip nanopatterned with gold nanoparticles (AuNPs). In our LSPR biosensor, binding of AFP to the surface of the sensor chip led to an increasing magnitude of the LSPR signals, which was measured by an ultraviolet-visible (UV-Vis) spectrometer. Our LSPR biosensor showed sufficient detectability of AFP at concentrations of 1ng/mL to 1μg/mL. Moreover, the overall procedure for detection of AFP was completed within 20min. This biosensor could also be utilized for a point of care test (POCT) by employing a portable UV-Vis spectrometer. Owing to the simplicity and rapidity of the detection process, our LSPR biosensor is expected to replace traditional diagnostic methods for the early detection of diseases.

        사료내 식물성 Phytase 첨가가 산란계의 생산성 및 인 이용성에 미치는 영향

        본 연구는 식물성 phytase 처리수준이 산란계의 생산성에 미치는 영향 및 인 이용률에 미치는 영향을 검토하기 위하여 실시하였다. NRC 사양표준의 유효 인 요구량을 충족시킨 대조구와 무기태인 사용량을 50%로 제한한 처리구로 나누고, 각각의 처리구에 식물성 phytase를 각각 0.05%, 0.1% 및 0.2% 첨가하였다. 58주령의 갈색 산란계(ISA-Brown) 240수를 공시하여 총 6주간 사양실험을 실시하였고 산란종계를 이용하여 인 및 질소의 이용성을 측정하였다. 그 결과를 요약하면 다음과 같다. 1. 50% 무기태인 수준에서 식물성 phytase의 첨가에 따른 일당증체량, 산란율의 저하는 나타나지 않았다. 2. 50% 무기태인 수준에서 식물성 phytase 0.05% 첨가수준에서 일시적으로 난각질 저하 현상이 나타났으나 0.1% 이상 첨가수준에서는 난각질의 변화가 관찰되지 않았다. 3. 효소의 첨가수준이 증가함에 따라 P 배설량의 감소와 P 이용률이 개선되었다(P<0.05). 4. 식물성 phytase 첨가는 산란계의 성적 저하없이 사료내 무기태인의 사용량을 줄일 수 있음이 시사되었다. Two experiments were conducted to investigate the effect of dietary supplementation of plant phytase(Phytazyme^R) in corn-soybean meal based diets on utilization of phytase-bound phosphorus in laying hens and evaluate nitrogen(N) digestibility and phosphorus(P) availability in breeders. In he experiment one, three levels of the Phytazyme^R(0.05, 0.1, and 0.2% of diet) were added to diets containing a half of control TCP level(0.96%) for 4 wks. Feed consumption, egg production rate, egg weight and eggshell quality were recorded weekly. At the end of experiment, 8 birds per treatment were sacrificed, liver weight were weighed and right tibiae were removed for determination of P content. The second experiment was conducted to evaluate the P availability and nitrogen digestibility in breeders fed same diets for 2 wks. Feed and excreta were collected to determine the P and N contents for the last three days of experiment two. Addition of Phytazyme^R resulted in no effects on feed intake, egg product rate, egg weight and egg shell quality. P excretion decreased and its availability enhanced as phytase supplementation increased in diets. Dietary supplementation of Phytayme ^R above 0.1% level in corn-soybean meal based diets did not have an adverse effect on production and decreased level of phosphorus in excreta.

      • K-band frequency-selective limiter using an injection locked oscillator for blocker cancellation

        <P>A frequency-selective limiter (FSL) using an injection locked oscillator (ILO) is proposed for a blocker cancellation, which employs an ILO in the feedforward path as a high-Q notch filter to select and amplify the blocker from the input signal. The ILO also functions as a phase shifter providing a proper phase delay so that two blockers in the feedforward and thru paths cancel each other out in the combining node. The proposed FSL is fabricated in a commercial 0.15 mu m GaAs pHEMT process with a maximum blocker suppression of 27 dB in the 50 MHz spacing two-tone measurement at the K-band. Thus, it provides a blocker suppression higher than 20 dB over the 250 MHz blocker bandwidth.</P>

      • 肛門 閉塞을 동반한 鎖骨 頭蓋 異骨症 一例

        Cleidocranial dysostosis is rare congenital syndrome which was originally described by Marie and Sainton in 1897. According to the classic discription, the syndrome consist in 1) Various degree of hypoplastic change of the clavicles 2) Delayed ossification of the cranial sutures and incresed width of the skull 3) Defects in the teeth, and 4) Hereditary characteristics. And these authors used the term Cleidocranial Dysostosis. But Rhinehart called attention to the mutational character of the disease and accordingly recommended that its name be changed to Mutational Dysostosis. Subsequent to the discovery of cleidocranial dysostosis, new anomalies extending practically throughout the entire osseous system have been described. We experienced a new case of Cleidocranial Dysostosis patient with absence of distal portion of both clavicles, unusual scapular and defect of ossification on the skull. Also this patient has high type of obstruction of imperforated anus. And we present this case based on clinical and radiological findings with review of some literatures.

        Thermodynamic properties of five-layered HgBa<sub>2</sub>Ca<sub>4</sub>Cu<sub>5</sub>O<sub>12+y</sub> from equilibrium magnetization

        By using the Hao-Clem model, we analyzed the equilibrium magnetization of the grain-aligned HgBa<SUB>2</SUB>Ca<SUB>4</SUB>Cu<SUB>5</SUB>O<SUB>12+y</SUB> (Hg1245) with T<SUB>c</SUB>~108K. We obtained thermodynamic parameters, such as the penetration depth [λ<SUB>ab</SUB>(0)] and the coherence length [ξ<SUB>ab</SUB>(0)] from the thermodynamic critical field [H<SUB>c</SUB>] and the Ginzburg-Landau parameter [κ]. Compared to the four layered superconductor HgBa<SUB>2</SUB>Ca<SUB>3</SUB>Cu<SUB>4</SUB>O<SUB>10+y</SUB> (Hg1234), the obtained penetration depth was slightly increased indicating that the Cooper pair density of Hg1245 was decreased, which was one of the reasons why the transition temperature (T<SUB>c</SUB>) of the Hg1245 did not increase as compared to the Hg1234. The expected increase of T<SUB>c</SUB> for the material with the more CuO<SUB>2</SUB> planes was not achieved because of the insufficient doping of the Cooper pairs while adding CuO<SUB>2</SUB> planes.

        <P>Conclusion: Our findings indicate that Nox2 deficiency results in exaggerated experimental asthma, which is caused by enhanced Th2 effector function in a T-cell-intrinsic manner.</P>

        Since the College of Industrial Education of Chungnam National University took charge of technical teacher preparation in 1977, much efforts have been concentrated on the activities for creating favorable educational environments in terms of curricula, pedagogic affairs, inner and outer facilities, etc. In this study some imposed barriers which are likely to hinder this college from achieving better education were identified and analysed. Considerably large scale of the student enrollment in comparison with the market demand is a prevailing issue which causes an adverse effect for clarification of the educational goal of the college and another problem is concerned with the student entry criteria which do not require the pre-experiences in the field. Under this circumstances, a guide line to improve the teacher education is recommended as follow; 1) The number of the student enrollment should be adjusted to meet the minimum range in the wave of the technical teacher demand projection and the shortage of teacher could be covered by the graduates from the shortterm courses in this college. 2) The current curriculum is accordingly recommended to be diversified taking into consideration the qualification of the student. The suggested consists of four-year regular course, one-year course, and six-month course and the short-term courses are provided to induce the experienced people from industries. 3) In order to have teachers adjust themselves to changing technology, education on sound and fundamental knowledges should be provided in the pre-service education course and education on advanced knowledges and applied theories deferred to the in-service training course. 4) In the four-year pre-service education, the general education and pedagogic subjects are to be drastically intensified from 18 credits and 22 credits up to 40 credits and 30 credits respectively.
