<목적> H. pylori 제균 치료성적을 좌우하는 요소에는 약제와 대상 환자군의 선정, 균검사방법의 차이, 항생제 저항성 등이 중요시되고 있다. 이 중에서도 항생제 저항성은 나라간의 H. pylori 제균 성적을 다르게 하는 대표적인 원인이다. 우리나라는 제균률이 외국보다 저조하여 85%내외로 보고되고 있다. 본 연구의 목적은 H. pylori 제균 실패율과 clarithromycin 내성률을 조사하여 제균 실패 원인으로서의 clarithromycin

There are multiple experiments conducted with male Korean native ducks (KND) to evaluate the optimal levels of limiting amino acids (AA). In Exp. 1, a total of 450 one-d-old male KNDs were divided into five groups with six replicates and fed experimental diets with varying levels of lysine, total sulfur amino acids (TSAA) and threonine (T1, 0.90/0.74/0.70%; T2, 1.00/0.82/0.77%; T3, 1.10/0.90/0.85%; T4, 1.20/0.98/0.93%; T5, 1.30/1.07/1.01%) to 21 d of age. In Exp. 2, one-d-old male KND were received and fed commercial starter diet from hatching to 21 d of age, and then divided into five groups with six replicates and fed one of five diets varying levels of lysine, TSAA, and threonine (T1, 0.73/0.62/0.54%; T2, 0.80/0.68/0.60%; T3, 0.87/0.74/0.65%; T4, 0.94/0.80/0.70%; T5, 1.01/0.86/0.75%) during 22 to 56 d of age, respectively. The BW gain was linearly increased as dietary limiting AA levels increased to 1.20% lysine, 0.98% TSAA and 0.93% threonine. There were no significant differences in feed intake, gain:feed and uniformity among groups. In Exp. 2, the BW gain and gain:feed were not affected by dietary limiting AA levels. There were no significant differences in carcass characteristics and meat quality among groups. The growth performance and carcass characteristics did not show the significant response to increasing dietary limiting AA levels in KND during 22 to 56 d of age. In conclusion, the levels of lysine, TSAA and threonine necessary to maximize growth for starter phase were at least 1.20%, 0.98%, and 0.93%, respectively. On the other hands, KND require relatively low levels of limiting AA for late growth and carcass yield. The dietary levels of 0.73% lysine, 0.62% TSAA and 0.54% threonine appear to be adequate during growing phase.
Rapid and label-free bioanalytical method of alpha fetoprotein detection using LSPR chip

Alpha fetoprotein (AFP) is a cancer marker, particularly for hepatocellular carcinoma. Normal levels of AFP are less than 20ng/mL; however, its levels can reach more than 400ng/mL in patients with HCC. Enzyme linked immunosorbent assay (ELISA) and radioimmunoassay (RIA) have been employed for clinical diagnosis of AFP; however, these methods are time consuming and labor intensive. In this study, we developed a localized surface plasmon resonance (LSPR) based biosensor for simple and rapid detection of AFP. This biosensor consists of a UV-Vis spectrometer, a cuvette cell, and a biosensor chip nanopatterned with gold nanoparticles (AuNPs). In our LSPR biosensor, binding of AFP to the surface of the sensor chip led to an increasing magnitude of the LSPR signals, which was measured by an ultraviolet-visible (UV-Vis) spectrometer. Our LSPR biosensor showed sufficient detectability of AFP at concentrations of 1ng/mL to 1μg/mL. Moreover, the overall procedure for detection of AFP was completed within 20min. This biosensor could also be utilized for a point of care test (POCT) by employing a portable UV-Vis spectrometer. Owing to the simplicity and rapidity of the detection process, our LSPR biosensor is expected to replace traditional diagnostic methods for the early detection of diseases.
Cooperative Activation of PI3K by Ras and Rho Family Small GTPases

Phosphoinositide 3-kinases (PI3Ks) and Ras and Rho family small GTPases are key regulators of cell polarization, motility, and chemotaxis. They influence each other's activities by direct and indirect feedback processes that are only partially understood. Here, we show that 21 small GTPase homologs activate PI3K. Using a microscopy-based binding assay, we show that K-Ras, H-Ras, and five homologous Ras family small GTPases function upstream of PI3K by directly binding the PI3K catalytic subunit, p110. In contrast, several Rho family small GTPases activated PI3K by an indirect cooperative positive feedback that required a combination of Rac, CDC42, and RhoG small GTPase activities. Thus, a distributed network of Ras and Rho family small GTPases induces and reinforces PI3K activity, explaining past challenges to elucidate the specific relevance of different small GTPases in regulating PI3K and controlling cell polarization and chemotaxis.
p21-Activated kinase 4 promotes prostate cancer progression through CREB

Prostate cancer is initially androgen-dependent but, over time, usually develops hormone- and chemo-resistance. The present study investigated a role for p21-activated kinase 4 (PAK4) in prostate cancer progression. PAK4 activation was markedly inhibited by H89, a specific protein kinase A (PKA) inhibitor, and PAK4 was activated by the elevation of cAMP. The catalytic subunit of PKA interacted with the regulatory domain of PAK4, and directly phosphorylated PAK4 at serine 474 (S474). Catalytically active PAK4 enhanced the transcriptional activity of CREB independent of S133 phosphorylation. Stable knockdown of PAK4 in PC-3 and DU145 prostate cancer cells inhibited tumor formation in nude mice. Decreased tumorigenicity correlated with decreased expression of CREB and its targets, including Bcl-2 and cyclin A1. Additionally, in androgen-dependent LNCap-FGC cells, PAK4 regulated cAMP-induced neuroendocrine differentiation, which is known to promote tumor progression. Finally, PAK4 enhanced survival and decreased apoptosis following chemotherapy. These results suggested that PAK4 regulates progression toward hormone- and chemo-resistance in prostate cancer, and this study identified both a novel activation mechanism and potential downstream effector pathways. Therefore, PAK4 may be a promising therapeutic target in prostate cancer.

To date, genome-wide population-based association studies have been predominantly conducted on genetic predispositions in adult individuals. In uncovering childhood FPG susceptibility, a combined meta-analysis identified a novel LEPR locus (rs17407594) reaching genome-wide significance (n=1,260, P<SUB>add</SUB>=4.98x10<SUP>-8</SUP>). We also observed an association with type 2 diabetes (T2D) risk in the Asia Genetic Epidemiology Network (AGEN) consortium (n=18,817, P<SUB>add</SUB>=2.06x10<SUP>-2</SUP>). In conclusion, our findings might expand understanding of genetic architecture contributing to glucose regulation and T2D risk.

<P>Gelatin derivative was prepared to coating titanium surface and photo-immobilize rhBMP-2 for improved osteoinduction and function. The conjugation was confirmed by P-31 and H-1 NMR. Photo immobilization of the azide-phosphonate-gelatin (Az-P-Gel) was evaluated by micropatterning. The rate of recombinant human bone morphogenetic protein (rhBMP-2) release was evaluated for performance of Az-P-Gel as a carrier. The water contact angle was determined to characterize Ti surface coated with Az-P-Gel. Ability to induce osteogenesis of Az-P-Gel was evaluated using alkaline phosphatase (ALP) activity assay, Alizarin Red S staining, and calcium colorimetric assay. 박판 A3003 Al합금의 Nd : YAG 레이저빔 용접에 관한 연구

This work was carried out to investigate the welding behavior of thin A3003 Al alloy sheets by Nd:YAG laser beam. Considering bead shape and mechanical properties, the laser pulse shapes selected were two kinds of 2-division and 3-division by varying power level and pulse duration. In order to obtain optimum conditions, the factorial design method and central composite design method were applied. Tensile test, optical microscopy, micro hardness test and TEM analysis were performed. Due to the annealing caused by thermal effect during laser welding, precipitates were coarsended. The HAZ was softened and failed during tensile test. The hardness of HAZ was lower than that of base metal, since the heat input relieved the work hardening effect and caused grain growth.

Generally, a thermal deformation is generated in the structure used at high temperature and this thermal deformation causes trouble such as the strength decline or the breakage of the structure. Especially, in the case of the precision mold such as a press type glass lens mold, the thermal deformation of a mold affects in the quality of products. In the present study, to obtain the thermal deformation at forming step and calculate the modified height of a glass lens mold at a design step, thermal stress analysis was carried out for a glass lens mold.
