        가압가열 및 microwave에 의한 중력분 반죽 gliadin의 항원성 변화

        본 연구에서는 가압가열 및 microwave 처리에 의한 중력분 반죽 추출물 내의 gliadin 단백질의 항원성 변화에 대해 살펴보았다. 중력분 반죽에 가압가열과 microwave를 단독 또는 병행으로 처리하여 ci-ELISA, SDS-PAGE 및 immunoblotting을 실시하였으며, 가압가열 처리에 의해서 anti-gliadin IgG 항체와 gliadin과의 결합력이 다소 감소한 것을 확인하였다. 특히 30 min 이상 처리시 더욱 감소한 것으로 나타났으며, SDS-PAGE와 immunoblotting 결과에서도 gliadin band의 강도가 약해지고 항체와의 반응도 나타나지 않았다. Microwave 처리의 경우, 5 min 이상 처리시 일부 gliadin 단백질의 소실이 관찰되었으나, 항원성에는 큰 변화가 없었다. 또한 가압가열 및 microwave 병행 처리에 의해 항원-항체 결합력이 더욱 감소되었으며, 특히 가압가열 50 min, microwave 10 min 처리시 약 35.0%로 감소되었다. 이상의 결과를 통해 가압가열 처리에 의해 중력분 반죽 추출물 내 gliadin의 항원성이 감소되는 것을 확인하였으며, microwave와 병행 처리하는 경우, 더욱 감소하는 것을 확인하였다. The aim of this study was to determine the optimal physical treatment to reduce the antigenicity of gliadin in wheat dough. Medium wheat dough was treated with an autoclave (5, 10, 30, and 50 min at 121℃, 1 atm), a microwave (1, 5, and 10 min) or both (10, 30, and 50 min/5, 10 min). The proteins in the dough extracts were analyzed by SDS-PAGE and the binding ability of anti-gliadin IgG to gliadin was examined by ci-ELISA and immunoblotting. Results showed that the ability of anti-gliadin IgG to bind to gliadin in wheat dough treated with an autoclave alone or in combination with a microwave was decreased. Especially, it declined to ~77% after autoclaving for 30 min and 35% after both autoclaving for 50 min and microwaving for 5 min. In addition, the intensity of gliadin bands in SDS-PAGE were weakened and anti-gliadin IgG did not recognize gliadin in immunoblotting. However, microwaving alone did not affect the antigenicity of gliadin in wheat dough. These results indicate that autoclaving may affect the reduction of the antigenicity of gliadin in medium wheat dough. Moreover, autoclaving in combination with microwaving is more effective for reducing the antigenicity of wheat dough.

      • 동충하초 투여가 최대하 운동 시 지방연소에 미치는 영향

        The purpose of this study was to evaluate effects of Cordyceps militaris on fat oxidation during submaximal exercise. Fifteen healthy male collegiate students(ingestion group, N=7; control group, N=8) participated as subjects after signing an informed consent following overnight, subjects completed maximal graded execise test on treadmill to determine exercise intensity(50%VO2max). To evaluate the fat oxidation using indirect calorimeter, it was performed submaximal treadmill to determine exercise lasting. 40min at 50%VO2max after 5day and consumed Cordyceps militaris on separate days. This study was used 2×4 two way repeated ANOVA to analyze physiological and metabolic variables between two groups and metabolic variables between two groups and four exercise periods(l0min, 20min, 30min 40min), and used Scheffe to post-hoc test. Significance was set at α=.05. The results of this study were as followed: First, there were significantly highest in ingestion group for V02, percentage of fat oxidetion(%fat), fat per kilocarolies(Fkcal), and total per kilocarolies(Tkcal)(p<.05), but respiratory exchange ration(RER) and heart rete(HR) were significantly lower in ingestion group(p<.05). Second, RER, %fat, Fkcal were significantly difference in 10, 20, 30, 40min of exercise period(p<.05). This results suggest that there is the effect of Cordyceps militaris on fat oxidation submaximal exercise. Additionally, to evaluate effect of Cordyceps militaris as ergogenic aid, it should be required the future study that included extend subjects, various dosage, and chronic consumption.

      • Ether-free polymeric anion exchange materials with extremely low vanadium ion permeability and outstanding cell performance for vanadium redox flow battery (VRFB) application

        <P><B>Abstract</B></P> <P>Ether-free poly(<I>p</I>-phenylene)-based anion exchange membrane materials with outstanding vanadium ion blocking properties are reported. The synthesized ionomers with a rigid backbone structure show small dimensional variations despite the high ion exchange capacities. Among the synthesized membranes, the QPPP-2 membrane with ion exchange capacity of 2.1 meq·g<SUP>−1</SUP> exhibits high anion (OH<SUP>−</SUP>, SO<SUB>4</SUB> <SUP>2−</SUP>, and Cl<SUP>−</SUP>) conductivity. The QPPP-2 membrane shows an extremely low diffusion coefficient (2.12 × 10<SUP>−9</SUP> cm<SUP>2</SUP> min<SUP>−1</SUP>) for VO<SUP>2+</SUP>, which is much lower than that of Nafion 115 (2.88 × 10<SUP>−6</SUP> cm<SUP>2</SUP> min<SUP>−1</SUP>), indicating the vanadium ion blocking property of QPPP-2. Moreover, the chemical and operational stabilities of the membranes are investigated <I>via</I> ex situ soaking tests in 0.1 M VO<SUB>2</SUB> <SUP>+</SUP> solution and in situ operation tests for 100 cycles, respectively. The vanadium redox flow battery single cell assembled with the QPPP-2 membrane exhibits a coulombic efficiency of 99%, voltage efficiency of 87%, and energy efficiency of 86% during 100 cycles at 80 mA cm<SUP>−2</SUP>.         총경동맥 폐쇄시간에 따르는 국소 뇌혈류 변화 : 실험적 연구 An Experimental Cat Model

        The microvasculature of the brain is also quite susceptible to ischemic insult, and substantial portions of the brain are not reperfused after restoration of the blood supply following overtime of critical ischemic periods. The purpose of this series of experiments was to determine the effects of ischemia on subsequential regional cerebral blood flow measurements and cortical electric activities following reperfusion after ischemia and also to define the proper time of vascular occlusion without irreversible neural damage. Cerebral ischemia was induced in cat by bilateral common carotid occlusions for periods of 10, 30, to 60 minutes, and the blood supply was reperfused for 3 hours after clamp-off. Regional cerebral blood flow(rCBF) was measured by hydrogen clearance technique following ischemia, restoration of blood supply and electroencephalogram recovery could be predicted according to the rCBF. Forty adult cats weighing 2.7 to 4.0㎏ were used in this study. The animals were divided into 4 groups of 10 cats each : normal control, 10 min-clamped, 30 min-clamped, and 60 min-clamped groups. The results obtained were as follows : 1) The mean rCBF was 24.6±7.0㎖/100g/min in control group. 2) Bilateral carotid occlusions resulted in a reduction of the rCBF(12.4±4.1㎖/100g/min) to 50% of control flow on both hemispheres. 3) Sequential changes of the rCBF after reperfusion : (1) There was restored the rCBF(21.3±5.1㎖/100g/min) to control flow in the 10 minutes-clamped group. (2) There was a 85% recovery of control flow in the 30 minutes-clamped group. (3) There was a only 25% recovery of control flow in the 60 minutes-clamped group. 4) A close correlation was found between cortical electrical activity and rCBF suggesting a threshold relationship. (1) The changes of cortical electric activity began to notice at rCBF less than 17.4±4.7㎖/100g/min. (2) The recovery of cortical electric activity noted at rCBF more than 10.2±2.3㎖/100g/min. 5) There was no evidence of ischemic involvement at the cortex, white matter and basal ganglia in the 10 minutes clamped group, but demonstrated a dense wedge shaped infarct at the cortex and uncus herniation in the 60 minutes clamped group. The rCBF and cortical electric activity restored to normal values in reperfusion within 10 minutes after occlusion of both common carotid arteries.

        가압가열 및 Microwave 처리에 의한 중력분 Gliadin의 항원성 변화

        본 연구에서는 가압가열 및 microwave 처리가 gliadin의 항원성에 미치는 영향을 살펴보기 위해 중력분에 가압가열과 microwave를 단독 또는 병행으로 처리하여 Ci-ELISA, SDS-PAGE 및 immunoblotting을 실시하였다. 가압가열 처리의 경우, 처리 시간이 길어질수록 IgG와의 결합력이 감소하였으며, 특히 50분 처리구에서 약 69%로 가장 낮은 결합력을 보였다. 또한 SDS-PAGE와 immunoblotting 결과에서도 무처리구에서 강하게 보였던 gliadin band가 처리에 의해 거의 소실되고 항체와 반응하지 않았다. 가압가열 및 microwave를 병행 처리한 경우도 마찬가지로 gliadin의 결합력이 다소 감소하였으며, 처리구 중에서는 가압가열 50분, microwave 5분 처리구에서 약 73%로 가장 낮은 결합력을 보였다. 반면 microwave를 단독으로 처리하였을 때에는 일부 단백질의 변화는 관찰되었으나 항원성 감소에는 큰 영향을 미치지 않았다. 이상의 결과를 통해 가압가열을 단독 처리에 의해 gliadin의 항원성이 다소 감소되었으며, microwave 병행 처리에 의한 차이는 크게 나타나지 않은 것을 확인하였다. This study was conducted to evaluate the effect of physical treatments on the antigenicity of gliadin in medium wheat flour. The wheat flour was treated with an autoclave (5, 10, 30, 50 min), a microwave (1, 5, 10 min), and both (10, 30, 50 min/ 5, 10 min), and investigated by SDS-PAGE, immunoblotting and Ci-ELISA using anti-gliadin IgG. The results showed that the binding ability of anti-gliadin IgG to gliadin in wheat flour was slightly decreased when autoclaved or when autoclaved and microwaved. Especially, it was reduced to about 69% after autoclaving for 50 min and 73% after autoclaving for 50 min and microwaving for 5 min. In addition, gliadin bands in the 50 min autoclaved group disappeared in both SDS-PAGE and immunoblotting. On the other hand, the antigenicity of gliadin was unaffected by microwaving alone. Consequently, there were no considerable changes in using an autoclave alone or in combination with a microwave. These results suggest that autoclaving may affect the reduction of the antigenicity of gliadin in medium wheat flour.

        도심 수목원에서의 경관감상과 산책이 인체의 생리적 안정에 미치는 영향

        This research was conducted to investigate the impact of viewing scenery and walking in the urban forest onphysiological relaxation of humans. The experiment was conducted in Hanbat Arboretum located in Daejeon, and thecontrol experiment was conducted in front of Daejeon City Hall. The subjects that participated in the experimentcomprised 24 Korean male university students in their 20s (average age, 21.1±2.5 years), participated in the experimentas the subject, and these subjects were classified into three groups divided into three locations such as the pine forest inArboretum, the pond in Arboretum, and the city. The subjects sat down and viewed scenery for 10 min, and they thenwalked for 15 min. Further, physiological changes were measured using indicators such as heart rate variability (HRV),blood pressure, and pulse rate. As a result, when subjects viewed scenery and walked in the urban forest, a statisticallysignificant increase in the high frequency (HF) power of HRV and decrease in systolic pressure was observed comparedwith when subjects viewed scenery and walked in the city. Through this research, it was found that activities like viewingscenery or walking in Arboretum of the city are effective in increasing the physiological relaxation of the city residents.

        물리ㆍ화학적 처리에 의한 멸균 초콜릿 우유 오염균의 생육억제 효과

        멸균 초콜릿 우유로부터 분리한 내열성 균주에 대해 열, pH, 전해수, 오존처리, microwave 및 감마선 처리를 하여 균주의 사멸효과에 대해 알아보았다. 균주의 지방산 분석과 API kit를 통하여 균주를 동정한 결과, Bacillus lentus로 동정되었으며, 잠정적으로 Bacillus lentus M1으로 명명하였다. B. lentus M1에 110℃, 15분간 열처리하였을 경우 생육이 억제되었으며, pH 처리 시 pH 5 이하, 10 이상에서 생육이 억제된 것으로 나타났다. B. lentus M1에 대한 전해수의 항균활성을 paper disc법으로 측정한 결과, 높은 생육억제를 보였으며, 오존 처리의 경우 초기 균수가 10² CFU가량의 균을 10분 동안, 10³ CFU가량의 균을 30분 동안 처리 시 균의 생육이 억제되는 것으로 나타났다. Microwave를 1분간 처리 시 B. lentus M1이 모두 사멸한 것으로 나타났다. 감마선 조사의 경우, 1 kGy 조사 시 생균수가 1.61×10³ CFU로 초기 균수에 비해 4 log cycle 가량 균수가 감소하였으며 7 kGy에서 완전히 사멸하였다. 이상의 결과를 통해 열, pH, 전해수, 오존 처리 및 방사선 처리 방법이 멸균 초콜릿 우유의 생존 오염균인 B. lentus M1을 효과적으로 사멸시킬 수 있을 것으로 사료된다. This study was conducted to investigate the cause of microbiological contaminants in aseptic chocolate milk and evaluate the effect of a physicochemical treatment on the growth inhibition of isolated bacterial strains. The bacterium isolated from aseptic chocolate milk was identified as Bacillus lentus and was named B. lentus M1. In the heat and pH treatment, the growth of B. lentus was inhibited at 110℃ for >15 min and at pH’s <5 and >10. An electrolyzed water treatment against B. lentus M1, revealed 5 ㎜ growth past the inhibition zone. The effect of ozone gas on B. lentus M1 growth was evaluated using viable cell counts. When the initial number of B. lentus M1 was 10² and 10³ CFU, the bacteria were completely suppressed by ozone gas treatment for 10 and 30 min, respectively. In a microwave treatment, B. lentus M1 was sterilized following microwave treatment for 1 min. As the result of γ-irradiation against B. lentus M1, numbers decreased as the γ-irradiation dosage increased. These results show the growth inhibition effects against contaminants in aseptic chocolate milk using physicochemical treatments.

        물리․화학적 처리에 의한 요구르트 오염균의 생육 억제효과

        물리?화학적 처리에 의한 요구르트 오염균의 생육억제 효과를 알아보기 위해, 요구르트의 주요 오염균을 분리?동정하고, 열, pH, 전해수, 오존가스, microwave 처리 및 감마선을 조사하여 오염균주에 대한 사멸효과를 알아보았다. 오염된 요구르트로부터 분리한 효모의 지방산 조성 분석과 API(Analytic Profile Index) kit 분석을 실시한 결과, Hanseniaspora uvarum으로 동정되었으며 잠정적으로 Hanseniaspora uvarum Y1으로 명명하였다. H. uvarum Y1의 열 및 pH 처리에 의한 생육억제 효과를 측정한 결과, 70℃ 및 80℃에서 15분 가열처리로 균이 사멸되었으며, pH 처리 시 pH 2, 3 및 9에서 생육이 다소 억제되었으며, pH 1 및 10에서 완전히 억제되었다. 전해수 처리의 경우, clear zone이 5 mm 이상으로 H. uvarum Y1이 전해수에 높은 감수성을 가지는 것으로 나타났다. 오존가스 처리에 의한 H. uvarum Y1의 사멸효과를 측정한 결과, 102 CFU의 균은 10분, 103 CFU의 균은 20분 처리 시 모두 사멸한 것으로 나타났으며, microwave 처리의 경우, 106 CFU 가량의 균이 1분 처리 시 모두 사멸되었다. 방사선 조사의 경우, 균수를 90% 이상 감소시키는데 필요한 조사선량이 20 kGy 이상으로 H. uvarum Y1은 감마선에 저항성이 있는 균임을 알 수 있었다. 이상의 결과를 종합해볼 때 열, pH, 전해수, 오존가스, microwave 처리를 통해 요구르트 오염균주인 H. uvarum Y1의 생육을 효과적으로 억제할 수 있을 것으로 생각된다. This study was conducted to investigate the cause of microbiological contamination in yogurt and evaluate the effect of physicochemical treatment on the growth inhibition of Hanseniaspora uvarum isolated from yogurt. The yeast strain Hanseniaspora uvarum Y1 was subjected to heat and pH treatments. H. uvarum Y1 was killed at 70oC and 80oC after 15 min and survived in a wide pH range from pH 2 to 9. However, it did not survive under pH 1 and over pH 10. In a disk diffusion susceptibility test on H. uvarum Y1, a clear zone (5 mm) of growth inhibition was observed upon treatment with electrolyzed water. The effect of ozone gas on the growth of H. uvarum Y1 was evaluated by viable cell count. Initial cell numbers of 102 and 103 CFU/mL of H. uvarum Y1 were completely killed by treatment for 10 and 30 min, respectively. H. uvarum Y1 was also sterilized by microwave treatment for 1 min. When treated with gamma-irradiation, the rate of killing of H. uvarum Y1 was proportional to the irradiation dose. and complete killing occurred at a dose of 50 kGy.

      • Using Automated Container Type Culture System Hericium erinaceum Mushroom Production Technology

        Time, place, and the high-tech industry to overcome barriers to the fusion of the so-called "smart agriculture" is changing the landscape of agriculture. Precision Agriculture's core temperature, humidity, location information, and summary information in real time, all significant ICT controlling technique. The system is a four-season container production for the mushroom cultivation temperature, humidity, illumination, self-regulation, such as carbon dioxide, the optimal environment for mushroom cultivation was implemented. Hericium erinaceum (also called Lion's Mane Mushroom, Bearded Tooth Mushroom, or Bearded Tooth Fungus) is an edible mushroom and medicinal mushroom in the tooth fungus group. Native to North America, Europe and Asia it can be identified by its tendency to grow all the spines out from one group (rather than branches), long spines (greater than 1 cm length) and its appearance on hardwoods. About 40 days of incubation 1.5kg oak sawdust was used as the test medium bag. The humidity was maintained more than 85~95% and temperature was 18~20°C, and CO2 density was 500~1,000ppm. The days for primordium formation showed us 6~12 days. The amount of fruitbody production was 110~260g per bag. This technique to establish new community support program in conjunction with the international movement, domestic sales and exports are also planned.

        배경 : 폐경후 골다공증은 심각한 문제로 대두되고 있다. 현재 Alendronate는 폐경후 골다공증의 치료제로서 널리 쓰이고 있다. 저자는 Alendronate의 골밀도에 효과를 알아보고 골다공증의 위험인자에 따른 Alendronate의 반응 정도를 평가하기 위하여 본 연구를 수행하였다. 방법 : Alendronate를 평균 12개월간 복용한 35명의 폐경후 골다공증환자에서 체질량지수와 폐경후 기간에 따른 치료효과를 조사하였다. 결과 : 전체 대상군에서 Alendronate 10㎎을 12개월간 투여한후 골밀도의 평균 변화율은 요추와 대퇴골 경부에서 각각 7.83±7.39%와 2.99±9.22%이었다. 과체중군에서 평균변화율은 요추와 대퇴골 경부에서 각각 7.15±4.5%와 3.05±2.86%였다. 양군간에 통계적 유의성이 있는 차이는 없었다. 폐경후 7년이 경과되지 않은 군에서의 평균변화율은 요추와 대퇴골 경부에서 각각 6.9±5.3%와 5.71±0.08%였다. 양군간에 통계적인 유의성이 있는 차이는 없었다. 결론 : 마른체형이 골다공증의 발병에는 영향이 있으나, Alendronate 치료효과의 차이는 없는 것을 확인할 수 있었다. 더불어 폐경이 있고 7년이 초과된 군과 년이 경과되지 않은 군에서의 Alendronate 치료효과의 차이는 없는 것을 확인할수 있었다. 이는 골흡수가 상당히 진행된 상태에서도 Alendronate가 치료효과를 가지고 있음을 반영한다고 할 수 있겠다. Postmenopausal osteoporosis is a serious health problem. The aminophosphonate Alendronate is widely used for treatment of postmenopausal osteoporosis. We studied the effect of l0㎎ of alendronate given daily for mean 12 months on bone mineral density in 35 women(mean age 61yrs. mean BMI 23.24㎏/m^(2)) with osteoporosis. An over weight group is defined such that BMI was above 23㎏/m^(2). All the women received 1500㎎ calcium carbonate daily. The bone mineral density of L-spine and femoral neck was measured by dual-energy X-ray absorptiometry. Alendronate increased BMD at mean 12 months at the L-spine (7.15±4.5%) and femoral neck(3.05±2.86%) in normal women. Alendronate increased bone mineral density at 12 months at the L-spine(6.00±5.3%) and femoral neck(4.17±4.87%) in overweight women. There was no difference between two groups. In the less than 7 years group of postmenopausal duration. alendronate increased bone mineral density at 12 months at the L-spine(6.9±4.9%) and femoral neck(3.18±7.1%). In the more than 7 years group of postmenopausal duration, alendronate increased hone mineral density at 1% months at the L-spine(6.5±5.3%) and femoral neck(5.71±0.08%). There was no difference between two groups. In postmenopausal women with osteoporosis, therapy with alendronate produced significant increases in BMD of the L-spine and femoral neck in both group. There was no difference between normal and over-weight groups. and between less than 7 years group and more than 7 years group of postmenopausal duration.
