Objective: To investigate the dosage of bovine type II collagen (BnCII) for the induction of oral tolerance in CIA animals, and to verify the changes of immune response and TGF-β production of mesenteric lymph node cells in tolerized CIA animals. Methods: Oral tolerance was induced by feeding of variable doses (5㎍, 10㎍, 20㎍ and 40㎍) of BnCII to DBA/1 mice 4 times per week during 2 weeks, and control mice were given ovalbumin (1000㎍), before immunization. We examed clinical assessment; incidence of arthritis, severity of arthritis, arthritic limb by visual analysis. IgG antibodies to BnCII were measured by ELISA, T cell responses to BnCII and PHA were quantified by antigen (CII)-induced 3H-thymidine incorporation into lymphocytes of mesenteric lymph node, draining lymph node, and spleen. TGF-β in supernatants obtained from lymph node culture medium was measured by ELISA. Results: Arthritis limbs were observed in 100% of control at 5 weeks after subcutaneous BnCII injection. The incidences of CIA in all tolerized group were significantly lower than that in control 5 weeks after immunization (control 100% vs. 5㎍ feeding group: 50%, 10㎍ feeding group: 50%, 20㎍ feeding group: 50%, 40㎍ feeding group: 55.5%, P<0.01). In comparison to control, mean articular indices were lower in all tolerized groups (control 5.13: 5㎍ feeding group 3.50, 10㎍ feeding group 2.75, 20㎍ feeding group 2.87, 40㎍ feeding group 2.63, P<0.05). Arthritic limbs were also significantly lower in tolerized groups (control 58.3: 5㎍ feeding group 20.8, 10㎍ feeding group 16.7, 20㎍ feeding group 20.8, 40㎍ feeding group 20.8, P<0.05). The titers of IgG antibody to CII were lower in tolerized group than that in control [tolerized group; median 10 (min. 0, max. 48), control; median 33 (min. 8.6, max. 101), P<0.05]. The proliferative responses to BnCII were significantly suppressed in tolerized (control 8010±2319cpm, tolerized group 4500±2060cpm, P<0.01). High TGF-β production was noted in tolerized group (control; 28pg/ml, BnCII feeding group; 73pg/ml). Conclusion: Oral tolerance in DBA/1 mice was successfully induced from low doses of BnCII (5㎍) and suppressed T and B cell function in conjunction with increased TGF-β production may play an important role for the induction of CII induced oral tolerance in DBA/1 mice.

목 적 : 현재 시중에 판매되고 있는 연무기 기종에 따라 그 성능이 다를 것으로 보고 제품 선택에 도움이 되고자 국내 시판중인 16가지 연무기 기종의 성능을 실험을 통하여 비교하였다. 방 법 : 16가지 연무기/압축기 제품(Pariboy Type 38/Long life, Pariboy Type N/Long life, Pariboy Type N/Salter 8900, Pariboy Type N/LC, Devilbiss pulmoaid-LT/Hudson, Devilbiss pulmoaid/Hudson, Mesmed neb-300/Own, San-up 3040/Hudson, Midas(Basic)/Own, AirJolie2/Hudson, Thomas 1127/Salter 8900, Noel NE-2000/Salter 8900, Omron CX3/Hudson, Chang Woo CWN-100/Salter 8900, Voyage/Mefar, ASI-Pro/Medel jet pluse)을 가지고 Mass output과 입자 크기를 측정하였다. Mass output은 생리식염수 4 mL를 2분간 분무 전, 후의 무게변화를 정밀 전자저울로 측정하였다. 동일한 방법으로 생리식염수 3 mL를 채워 실험하였다. 입자의 크기는 0.9% 생리식염수 4 mL를 채우고 5분간 작동하면서 laser diffraction법을 이용하여 측정하였다. 결 과 : 분당흡입량은 Pariboy type N/Long life이 0.184 mg/min로 가장 높은 값을 보였으며 0.019 mg/min로 가장 낮은 수치를 나타낸 Mesmed Neb-300/Own과는 약 9.6배의 차이를 보였다. Pariboy Type N/Long life이 Mass output이 0.68 mg/min로 가장 컸고 0.13 mg/min으로 가장 작은 수치를 보인 Mesmed-Neb/Own과는 5.2 배 차이가 있었다. 모든 제품은 5분간의 입자의 크기를 측정하였으나 검사시간 동안 입자크기의 차이는 보이지 않았다. $5{\mu}m$ 이하의 입자크기의 최대값은 64%였고 최소값의 연무기는 20%였다. MMAD는 Pariboy Type N/Long life이 $3.76{\mu}m$로 가장 작았으며 나머지 제품은 모두 $5{\mu}m$ 이상이었다. 결 론 : 시중에 판매되는 연무기들의 기능이 다양하므로 제품의 선택에 있어서 그 성능을 고려할 필요가 있다. Purpose : We assessed the dynamic characteristics of 16 nebulizer/compressor combinations currently available in Korea. Methods : The 16 nebulizer/compressor combinations(Pariboy Type 38/Long life, Pariboy Type N/Long life, Pariboy Type N/Salter 8900, Pariboy Type N/LC, Devilbiss pulmoaid-LT/Hudson, Devilbiss pulmoaid/Hudson, Mesmed neb-300/Own, San-up 3040/Hudson, Midas(Basic)/Own, AirJolie 2/Hudson, Thomas 1127/Salter 8900, Noel NE-2000/Salter 8900, Omron CX3/Hudson, Chang Woo CWN-100/Salter 8900, Voyage/Mefar, Chang Woo ASI-Pro/Medel jet pulse) were evaluated in terms of particle size and mass output. In addition, we determined the effects of nebulizer fill volume on mass output. Results : Pariboy Type N/Long life has the highest respirable mass of 0.184 mg/min and Mesmed Neb-300/Own has the lowest 0.019 mg/min. Pariboy Type N/Long life has the highest mass output of 0.68 mg/min and the shortest mass median aerodynamic diameter(MMAD) of $3.76{\mu}m$. All combinations other than Pariboy Type N/Long life produced a MMAD of over $5{\mu}m$. MMAD over a 5 min nebulization ranged 3.76 to $9.83{\mu}m$. There were no significant effects of fill volume on mass output. Conclusion : We concluded that there is a wide variation in performance of nebulizer/compressor combinations. The characteristics of nebulizer/compressor combinations should be considered in selecting products.

In this study, a high-performance vertical gallium nitride (GaN) power transistor was designed by using two-dimensional technology computer-aided design simulator. In addition, the vertical GaN transistor is used to analyze the DC/DC boost converter. Systems requiring high voltages of 1000 V or more, such as electric vehicles, need wide devices to achieve a high breakdown voltage when using conventional power devices. However, vertical GaN transistors can be fabricated with a small device area and a high breakdown voltage. The proposed device has an off-current of 413 pA/㎠, an on-current of 22 kA/㎠, and a high breakdown voltage of 1693 V due to good gate controllability and the undoped-GaN layer. The designed device was used to construct a boost converter that doubled the input voltage and its characteristics were examined. The boost converter produced an output voltage of 1955 V and the voltage conversion efficiency was high at 97.75 %.

Ficin isolated from fig tree latex is a unique enzyme having both protease and peroxidase activities. Recently, it has been revealed that peroxidase activity of ficin is about 60% or more at pH 4.5 ~ 5.0 which is the pH of normal skin. However, other efficacy studies and mechanisms of action about ficin are not well known. Thus, the objective of the present study was to investigate the anti-oxidation, anti-inflammatory, and whitening effects of ficin through in vitro studies. First, the antioxidant activity of ficin was confirmed by DPPH and H2O2 assays. The DPPH radical scavenging activity was about 78.7% level of L-ascorbic acid (vitamin C) at the same concentration of 1,000 μg/mL and the H2O2 scavenging activity was about 72.3 ± 4.1% of L-ascorbic acid. Second, ficin suppressed NO production in LPS-stimulated RAW264.7 cells in a concentration-dependent manner (at concentration of 10, 25, 50, and 100 μg/mL, NO production was 90.5 ± 0.5%, 77.3 ± 0.5%, 67.9 ± 1.1%, and 59.6 ± 0.5%, respectively, compared to the control). Ficin could inhibit mRNA and protein expression levels of inducible NO synthase (iNOS), implying its anti-inflammatory activity. To evaluate inhibitory effect of ficin on inflammation signaling pathway, we determined whether ficin could inhibit phosphorylation of inflammatory signaling proteins such as IκB and NF-κB as well as MAPKs (ERK, JNK, p38). Third, we verified the whitening effect of ficin. After B16F10 cells (mouse melanoma cells) were treated with α-MSH (200 nM) along with ficin at concentrations of 0, 5, 10, and 20 μg/mL for 96 h, melanin contents were increased to 227.1 ± 5.9% by α-MSH alone, but decreased to 197.9 ± 2.4, 107.3 ± 0.9, and 105.2 ± 0.9% in the presence of ficin at 5, 10, and 20 μg/mL, respectively. These results suggest that ficin could be used as a good cosmetic material with enzyme stabilization and cell penetration technology in the future.
