Oxidative stress is well known to be implicated in the development of asthma. The mitochondrial respiratory chain is a major site of intracellular reactive oxygen species (ROS) generation and, at the same time, an important target for the damaging effects of ROS. Mito-Tempo is a specific mitochondrial ROS inhibitor and it is known to be associated with opening of mi-tochondrial permeability transition pore and inhibition of cell necroptosis or apoptosis. However, there is little information on the protective effects of Mito-Tempo on the inflammatory airway disorders including bronchial asthma and its acute exacerbation. We investigate the effects of Mito-tempo on the allergic airway inflammation and hyperresponsiveness using the mice sensitized with OVA and LPS and then challenged with OVA (OVALPS-OVA mice). The OVALPS-OVA mice showed the typical features of neutrophilic asthma; increased airway inflammatory cells, the pathologic changes, the increased levels of Th2 cytokines in lungs of OVALPS-OVA mice, increased mitochondrial ROS generation, and increased bronchial hyperresponsiveness. Interestingly, we found that in OVALPS-OVA mice, Mito-Tempo, a novel mitochondrial targeting agent significantly reduced the increases in inflammatory cytokines, mitochondrial ROS generation, airway inflammation, and bron-chial hyperresponsiveness. These findings indicate that mitochondrial dysfunction including oxidative damage may be im-plicated in the pathogenesis of bronchial asthma and provide the therapeutic potential of a mitochondrial targeting agent, Mito-Tempo, for bronchial asthma.
이청준의 창작론 연구 (1) -``진실의 증거``를 향한 도정(道程)

이청준은 소설가가 주인공이거나 서술자인 ``소설가 소설``을 통하여 자신의 창작론을 표명해 왔다. 그의 소설가 소설은 ``소설이란 무엇인가``에 대한 탐색의 노정이 담긴 소설들로서 창작론의 입장에서 볼 때 각별하게 논의할 점을 제시해 준다. 이 논문은 이청준의 창작론을 그의 대표적인 ``소설가 소설``인 소문의 벽, 『지배와 해방』, 「비화밀교」, 『자유의 문』, 『인문주의자 무소작 씨의 종생기』 등을 비롯하여 작가의 직접적인 어조가 드러난 산문, 대담 등 2차 텍스트를 적극적으로 활용하여 논의하였다. 이 논문에서는 창작원리로서 이청준 문학을 해석할 수 있는 틀과 사유의 밑 바탕을 해석하는 데에 초점을 두고, 이청준의 창작론이 진술 욕망과 좌절의 징후, 자유의 질서와 삶의 진실, 진실의 증거와 갱신 등으로 전개되어 나가는 과정을 읽었으며, 논의를 진행하면서 ``진실의 증거``라는 주요한 화두가 이청준의 초기부터 후기 소설에 이르는 동안 지속적인 고뇌의 깊이를 마련한 창작론의 대전제임을 확인할 수 있었다. 이청준은 개인과 사회, 현실과 역사, 문학과 종교 사이에서 끊임없이 소설의 길을 탐색해 왔다. 이청준에게 있어서 ``진실의 증거``는 곧 자유의 질서를 획득하는 것과 다르지 않다. 그리고 그 진실이란 작가와 독자, 그리고 사회의 삶과 유리되지 않은 ``자유의 질서``를 마련하기 위한 것이어야 한다. 즉 작가가 소설 속에 담아내는 진실은 ``인간의 삶``과 유리되지 않는 것으로서 삶의 자유를 얻기 위해 진실의 증거가 필요하다는 것이다. The purpose of this paper is to search through what the creative writing theory of Lee Cheong-jun is and how it is embodied in his works. For this, his major works of fiction in which novelists play a narrator or a main character, The Wall of Rumors, Rule & Liberation, Secret Fire Esoteric, Gate of Freedom, The Life Story of Humanist Mr. Musojak and secondary texts from his prose works, lectures, symposiums are analysed. In The Wall of Rumors, through the story of a novelist who has the dread of making a statement, Lee Cheong-jun embodies his creative writing theory in which the novelist has the desire to make a true statement and fiction must be comprised of novelist`s free statements as well as present symptoms of the age`s crises. In Rule & Liberation, by showing the novelist`s lecture as it is, he reveals his creative writing theory. There, taking a step further, he writes that the novelist has a desire to present ``order of freedom``; research for the brave ways against inhumane systems and repression, and fiction is a work of broadening or maintaining life`s freedom by presenting actively the order of freedom. In Secret Fire Esoteric, through the story of a ritual not wanted to be revealed, he shows that the novelist`s statements can be restricted by not only social systems and repression but also excessive spiritualism. Because the novelist narrator sympathized with the spiritualist who disclosed that any truths or facts, when revealed, can be compromised with the mainstream, he felt restriction being placed upon the novelist`s free statements. In Gate of Freedom, by showing the contrast between two main characters, the novelist and the spiritualist, Lee Cheong-jun reveales a matter not resolved in Secret Fire Esoteric. In other words, fiction is the novelist`s free statement on truth, specifically it is a work trying to broaden or maintain practical freedoms and love based on morality, and it is only possible through sustained self-reflection and change. In The Life Story of Humanist Mr. Musojak, through the allegorical life story of the storyteller, Musojak, Lee Cheong-jun presents that fiction should be a story not separated from the real world, and it is possible when fiction reveals the evidence of the truth. Lee Cheong-jun has undergone a process to complete his creative writing theory of ``A Journey to the Evidence of Truth`` through his novelist`s works of fiction.
VDUP1 Is Required for the Development of Natural Killer Cells

<P><B>Summary</B></P><P>Vitamin D<SUB>3</SUB> upregulated protein 1 (VDUP1) is a stress-response gene that is upregulated by 1,25(OH)<SUB>2</SUB>D<SUB>3</SUB> in tumor cells. The in vivo roles of VDUP1 were investigated by producing mice lacking VDUP1 (VDUP1<SUP>−/−</SUP> mice). VDUP1<SUP>−/−</SUP> mice showed minimal changes in the development of T and B cells, but there was a profound reduction in the numbers of natural killer (NK) cells. As well, these mice showed decreased NK activity. In the VDUP1<SUP>−/−</SUP> mice, the expression of CD122 was reduced, demonstrating that VDUP1 is required for CD122 expression and NK maturation. In addition, severe lymphoid hyperplasia in the small intestine was observed in VDUP1<SUP>−/−</SUP> mice. Taken together, these results suggest that VDUP1 is a critical factor for the development and function of NK cells in vivo.</P>
Six Taxa of Newly Recorded Species of Charophytes (Charophyta) in Korea

Six taxa of unrecorded Korean species of charophytes are described. Freshwater and aerial charophytes were collected from the bark of trees, reservoirs, small ponds and swamps from April 2017 to September 2017. The newly recorded species of Korea were Cosmarium quadrifarium f. octastichum, Euastrum turneri, Spondylosium nitens var. triangulare, Staurastrum kouwetsii, Staurodesmus dickiei var. latus, and Klebsormidium subtile. K. subtile was transferred from Stichococcus subtilis.

Low-abundance cellular proteins normally invisible on the standard two-dimensional SDS-polyacrylamide gel electrophoresis (2-DE SDS-PAGE) map must be enriched appropriately in order to be visualized and identified in cells or tissues. We applied proteins of H. pylori strain 26695 to a immobilized heparin-affinity resin, which has an affinity for nucleic acid-binding proteins, protein biosynthesis factors, and growth factors. The whole cell extract of H. pylori strain 26695 was fractionated by the heparin-agarose chromatography, and was analyzed by 2-DE. The 2-DE SDS-PAGE displayed spots after silver staining, which were identified by matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS). Among the ca. 150 spots that were processed, 79 proteins representing 57 genes were identified. Eleven proteins were determined to be nucleic acid-associated. Eighteen proteins were newly identified in this study, including DNA topoisomerase I. These results may provide guidance for enriching low abundance proteins of H. pylori and contribute to the construction of a master protein map of H. pylori.

Objective: The aim of this study was to investigate differentially expressed genes and their functions in the hippocampus and striatum after heroin administration in cynomolgus macaques of different ages. Methods: Cynomolgus monkeys were divided by age as follows: 1 year (A1, n = 2); 3 to 4 years (A2, n = 2); 6 to 8 years (A3, n = 2); and older than 11 years (A4, n = 2). After heroin was injected intramuscularly into the monkeys (0.6 mg/kg), we performed large-scale transcriptome profiling in the hippocampus (H) and striatum (S) using RNA sequencing technology. Some genes were validated with real-time quantitative PCR. Results: In the hippocampus, the gene expression of A1H was similar to that of A4H, while the gene expression of A2H was similar to that of A3H. Genes associated with the mitogen-activated protein kinase signaling pathway (STMN1, FGF14, and MAPT) and -aminobutyric acid-ergic synapses (GABBR2 and GAD1) were differentially expressed among control and heroin-treated animals. Differential gene expression between A1S and A4S was the least significant, while differential gene expression between A3S and A2S was the most significant. Genes associated with the neurotrophin signaling pathway (NTRK1 and NGFR), autophagy (ATG5), and dopaminergic synapses (AKT1) in the striatum were differentially expressed among control and heroin-treated animals. Conclusion: These results suggest that even a single heroin exposure can cause differential gene expression in the hippocampus and striatum of nonhuman primates at different ages.
