        Expression of three neuropeptide genes (SfEH, SfETH, and SfCCAP) in response to insecticide stress on the white-backed planthopper Sogatella furcifera (Hemiptera: Delphacidae)

        Insect neuropeptides are involved in many important developmental processes. Here, we cloned three neuro peptide genes (SfEH, SfETH, and SfCCAP) from Sogatella furcifera (Horváth) and analyzed the relative expression levels of these genes through Real-time Quantitative polymerase chain reaction (RT-qPCR) during different developmental stages and in different wing types of S. furcifera following exposure to sublethal concentrations (LC 10 and LC 25 ) of methylaminoavermectin benzoate. The results revealed that the three neuropeptide genes (SfEH, SfETH, and SfCCAP) of S. furcifera contained open reading frame sequences of 222, 636, and 408 bp, encoding 73, 211, and 135 amino acids, respectively. Moreover, the expression of SfEH and SfETH reached its peak on day 3 of the 5 th instar nymph stage, while the SfCCAP expressed pinnacle on 40 min after eclosion. Furthermore, the relative expression levels of the three genes were in the following order: SfEH, short-winged female > long-winged female > long-winged male; SfETH, long-winged female > short-winged female > longwinged male; and SfCCAP, short-winged female > long-winged male > short-winged female. Upon calculating the relative expression of the three peptide genes after 24 and 48 h of treatment with sublethal concentrations of methylaminoavermectin benzoate, it was found that the relative mRNA expression levels of SfEH and SfETH declined upon LC 10 treatment but increased upon LC 25 treatment. Meanwhile, the relative mRNA expression level of SfCCAP declined upon both LC 10 and LC 25 treatments. This result will help us understand the effects of pesticides on insect growth and development at the molecular level.

        Assessment of nephrotoxicity of herbal medicine containing aristolochic acid in mice

        Background/Aims: It is undetermined if herbal medicines (HM) containing aristolochic acid (AA)-containing have similar nephrotoxicity to AA itself. Methods: We administered HM containing a high concentration of AA for 5 days (short-term study) or a low concentration of AA for 30 days (long-term study) to C57BL/6 mice; for comparison, same dose of AA compound was used as controls. Results: The nephrotoxicity in the HM- and AA-treated mice was compared in terms of renal function, histopathology, oxidative stress, apoptotic cell death, and mitochondrial damage. Short-term HM treatment resulted in acute kidney injury (marked renal dysfunction, acute tubular necrosis, and neutrophil gelatinase-associated lipocalin [NGAL] expression) in which the severity of renal dysfunction and histopathology was comparable with that induced by the administration of AA alone. Long-term HM treatment resulted in features of chronic kidney disease (CKD, mild renal dysfunction and tubular atrophy and dilatation). No significant differences in these parameters were observed between the HM- and AA-treated mice. HM-induced oxidative stress (8-hydroxy-2’-deoxyguanosine and manganese- dependent superoxide dismutase expression) and apoptotic cell death (terminal deoxynucleotidyl transferase dUTP nick end labelling [TUNEL]-positive cells and active caspase-3 expression) were similar in HM- and AA-treated mice in the short-term and long-term studies. Mitochondrial injury, evaluated by electron microscopy, was also similar in HM- and AA-treated mice in the short-term and long-term studies. Conclusions: The nephrotoxic potential of HM containing AA was similar to that of AA itself.

        Thermal and Mechanical Response of Soil and Tunnel during Replacement of Shield Tail Brush by Freezing Method

        In the long-distance shield tunneling process, the shield tail brush needs to be replaced after wear failure, especially in cross-river and cross-sea tunnel projects. Taking the shield tail brush replacement project of Qingchun Road tunnel in Hangzhou, China as the research background, a three-dimensional sequential thermo-mechanical coupled numerical simulation was carried out to study the shield tail brush replacement with liquid nitrogen freezing by ABAQUS software. The development of temperature fields and displacement fields of soil, and the deformations of the tunnel sections along the longitudinal direction induced by freezing were obtained. The numerical model was further validated by site measurement. In addition, the effects of the frozen wall thickness, buried depth, excavation radius, and coefficients of linear thermal expansion of soils on the frost heave were studied.

        The three-point crossover technique for immediate reconstruction of the umbilicus

        The umbilicus is an important aesthetic component of the abdomen; therefore, its absence is both cosmetically and psychologically distressing to the patient. However, loss of the umbilicus during abdominal surgical procedures is often unavoidable. Umbilical reconstruction is aimed at obtaining a natural, three-dimensional appearance. We propose a simple method for immediate umbilical reconstruction with good long-term results. This technique was used successfully on a patient who underwent tumor excision. A 49-year-old woman presented with a large mass, measuring 5.8×4.0 cm, on her umbilicus. The mass, an epidermal cyst, developed after laparoscopic uterine myomectomy 5 years earlier. Complete excision of the mass resulted in a large defect, and immediate umbilical reconstruction was planned. Our procedure involved apposing and anchoring two opposing flaps onto the abdominal wall, so that the umbilicus would retain its depth over a long period of time. Negative-pressure wound therapy was applied for 72 hours as a mild compressive dressing. No complications were encountered. The healing process was uneventful and the aesthetic outcome was pleasing; a natural-appearing navel was created. The patient was satisfied with the end result. This technique provides a permanent and sufficient depression for the umbilicus.

      • Ginseng extract reduces tacrolimus-induced oxidative stress by modulating autophagy in pancreatic beta cells

        <P>We previously reported that long-term treatment with a calcineurin inhibitor impairs autophagy process in pancreatic beta cells. This study investigated the effect of Korean red ginseng extract (KRGE) on autophagy modulated by oxidative stress. In mice with tacrolimus (Tac)-induced diabetes mellitus, KRGE alleviated islet dysfunction and decreased oxidative stress and autophagic vacuoles. In vitro, KRGE decreased autophagosome formation and attenuated lysosomal degradation, accompanied by improved beta cell viability and insulin secretion. Addition of 3-methyladenine (3-MA), an inhibitor of autophagosomes, to KRGE further improved cell viability and insulin secretion, and bafilomycin A (BA), an inhibitor of lysosomal function, reduced the effects of KRGE. At the subcellular level, Tac caused mitochondrial dysfunction (impaired mitochondrial oxygen consumption, ATP production, and increased reactive oxygen species production). But KRGE improved these parameters. The effect of KRGE on mitochondrial function enhanced by 3-MA but decreased by BA, suggesting a causal relationship between KRGE effect and autophagy modulation in Tac-induced mitochondrial dysfunction. These findings indicate that KRGE modulates autophagy favorably by reducing Tac-induced oxidative stress, and this effect is closely associated with improvement of mitochondrial function.</P>

        The effect of the modification methods on the catalytic performance of activated carbon supported CuO-ZnO catalysts

        Activated carbon (AC) was modified by ammonium persulphate or nitric acid, respectively. AC and the modified materials were used as catalyst supports. The oxygen groups were introduced in the supports during the modifications. All the supports were characterized by N2-physisorption, Raman, X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM), and thermogravimetric analysis. Methanol synthesis catalysts were prepared through wet impregnation of copper nitrate and zinc nitrate on the supports followed by thermal decomposition. These catalysts were measured by the means of N2-physisorption, X-ray diffraction, XPS, temperature programmed reduction and TEM tests. The catalytic performances of the prepared catalysts were compared with a commercial catalyst (CZA) in this work. The results showed that the methanol production rate of AC-CZ (23 mmol-CH3OH/(g-Cu·h)) was higher, on Cu loading basis, than that of CZA (9 mmol-CH3OH/(g-Cu·h)). We also found that the modification methods produced strong metal-support interactions leading to poor catalytic performance. AC without any modification can prompt the catalytic performance of the resulted catalyst.

      • Self-Rearrangement of Silicon Nanoparticles Embedded in Micro-Carbon Sphere Framework for High-Energy and Long-Life Lithium-Ion Batteries

        <P>Despite its highest theoretical capacity, the practical applications of the silicon anode are still limited by severe capacity fading, which is due to pulverization of the Si particles through volume change during charge and discharge. In this study, silicon nanoparticles are embedded in micron-sized porous carbon spheres (Si-MCS) via a facile hydrothermal process in order to provide a stiff carbon framework that functions as a cage to hold the pulverized silicon pieces. The carbon framework subsequently allows these silicon pieces to rearrange themselves in restricted domains within the sphere. Unlike current carbon coating methods, the Si-MCS electrode is immune to delamination. Hence, it demonstrates unprecedented excellent cyclability (capacity retention: 93.5% after 500 cycles at 0.8 A g(-1)), high rate capability (with a specific capacity of 880 mAh g(-1) at the high discharge current density of 40 A g(-1)), and high volumetric capacity (814.8 mAh cm(-3)) on account of increased tap density. The lithium-ion battery using the new Si-MCS anode and commercial LiNi0.6Co0.2Mn0.2O2 cathode shows a high specific energy density above 300 Wh kg(-1), which is considerably higher than that of commercial graphite anodes.</P>

        Temporal expression profiling of long noncoding RNA and mRNA in the peripheral blood during porcine development

        Objective: We investigated the temporal expression profiles of long noncoding RNA (lncRNA) and mRNA in the peripheral blood of pigs during development and identified the lncRNAs that are related to the blood-based immune system. Methods: Peripheral blood samples were obtained from the pigs at 0, 7, 28, and 180 days and 2 years of age. RNA sequencing was performed to survey the lncRNA and mRNA transcriptomes in the samples. Short time-series expression miner (STEM) was used to show temporal expression patterns in the mRNAs and lncRNAs. Gene ontology and Kyoto encyclopedia of genes and genomes analyses were performed to assess the genes' biological relevance. To predict the functions of the identified lncRNAs, we extracted mRNAs that were nearby loci and highly correlated with the lncRNAs. Results: In total of 5,946 lncRNA and 12,354 mRNA transcripts were identified among the samples. STEM showed that most lncRNAs and mRNAs had similar temporal expression patterns during development, indicating the expressional correlation and functional relatedness between them. The five stages were divided into two classes: the suckling period and the late developmental stage. Most genes were expressed at low level during the suckling period, but at higher level during the late stages. Expression of several T-cell-related genes increased continuously during the suckling period, indicating that these genes are crucial for establishing the adaptive immune system in piglets at this stage. Notably, lncRNA TCONS-00086451 may promote blood-based immune system development by upregulating nuclear factor of activated T-cells cytoplasmic 2 expression. Conclusion: This study provides a catalog of porcine peripheral blood-related lncRNAs and mRNAs and reveals the characteristics and temporal expression profiles of these lncRNAs and mRNAs during peripheral blood development from the newborn to adult stages in pigs.

        Bus voltage control of residential PV-HESS-grids using multimodal simplification

        This paper presents a residential power system that mainly includes photovoltaic (PV) panels, a hybrid energy storage system (HESS), a grid, and converters. The multimodal operation caused by the large number of states for each of the units increases the complexity of the system operation. It is difficult to adjust the operation state in the face of emergencies. A multimodal simplification method is proposed to simplify multimodal operation into three dominant modes (PV, HESS, and load) according to an energy management flowchart. A novel control strategy is proposed to realize the internal power distribution of the HESS, as well as the power allocation between the HESS and a single-phase full-bridge converter (SFC), while smoothing the double fluctuation of the bus voltage caused by the SFC. The effectiveness of the proposed multimodal simplification and control strategy is verified through the simulation and experimental results from a PV-HESS-grid.
