      • Cross-talk between STAT6 and Ras/MAPK Pathway for the IL-4-mediated T Cell Survival

        As a prototypic Thl vs Th2 cytokine, IFN-$\gamma$ and IL-4 activate distinct STAT proteins, STAT1 and STATE, respectively. In cytokine-producing Jurkat T cells, IL-4 is effectively rescued from cell death that is induced by dexamethasone, but IFN-$\gamma$ failed to do so. Since the Ras/MAPK pathway is known to play an important role in cytokine-induced cell survival, we investigated the mechanism of T cell survival through the analysis of functional cross-talk between Ras/MAPK and distinct STAT proteins that are activated by IL-4 and IFN-$\gamma$. Although IL-4 and IFN-$\gamma$ each induced the activation of STATE and STATI. in Jurkat T cells, respectively, only IL-4 was capable of inducing MAPK. Along with tyrosine kinase inhibitors, MEK/MAPK inhibitors also caused a significant suppression of the IL-4-induced STATE activity. This suggests a positive regulation of STATE by MAPK during IL-4 signal transduction. Furthermore, transfection studies with dominant active (da) vs dominant negative (dn) Ras revealed that daRas, but not dnRas, selectively up-regulated the expression and activity of STATE with a concomitant increase in MAPK activity. These results, therefore, suggest that there is a functional cross-talk between the Ras/MAPK and Jak/STAT6 pathways, which may have a role in the IL-4-induced T cell survival.

        Interleukin-4(IL-4) specifically induces the expression of its own receptor, IL-4R and type II IgE receptor, FceR1l in a dose-dependent manner in human B lymphocytes. We observed that the IL-4-induced expression of IL-4R and Fce Rll were both down-regulated by interferon-T(IFN-T) at mRNA as well as surface protein levels in normal B lymphocytes. Such co-ordinated regulation of the IL-4-induced IL-4R and FceRlI also occurred by anti-CD40 and steroid, each regulating the IL-4-induced response in an opposite direction. While anti-CD40, which is re-ported to agument the IL-4-induced B cell activation, further enhanced the IL-4-induced IL-4R and Fee RU expression, methylprednisolon(MPD) strongly inhibited the level of these receptors. The anti-CD40 or MPD-induced modulation of the IL-4-induced IL-4R and FceRII was further subject to the down-regulation by lFN-T. As compared to normal B lymphocytes, a differential regulation of IL-4R and FceRII was noted in a transformed B cell line, where IFN- Tup-regulated the IL-4-induced responses. Still, there was a co-ordinated regulation of the IL-4R and FceRII in these cells. Taken together, these results suggest that modulation of IL-4R expression and FccRII induction are closely correlated events during the IL-4-induced cellular activation

        저칼슘식이 투여와 방사선조사가 임프란트의 골유착에 미치는 영향에 관한 실험적 연구

        Purpose : To investigate osseointergration of titanium implants into the tibia of rabbits, which were fed a low calcium diet and irradiated. Materials and Methods : To prepare the experimental model, control group was fed a normal diet and experimental group was fed a low calcium diet for 4 weeks. And then, titanium implants were inserted into the tibia of each rabbit. Experimental group was subdivided into two groups; low calcium diet/non-irradiation group and low calcium dietfirradiation group. The low calcium diet/irradiation group was irradiated with a single absorbed dose of 15 Gy at the 5th postoperative day. At 12, 19, 33, 47, and 61 days after implantation (1, 2, 4, 6, and 8 weeks after irradiation), the bone formation in the bone-implant interface area was examined by light microscopy and fluorescent microscopy. Result and Conclusions : 1. In the control group, there began to form woven bone in the bone-implant interface area at 12 days after implantation. As the experimental time was going on, the amount of bone which was in contact with the implant was increased. 2. In the low calcium diet/non-irradiation group, there began to form woven bone in the bone-implant interface area at 19 days after implantation. Although the amount of bone which was in contact with the implant was increased as the experimental time was going on, the extent of increased bone was weak as compared with control group. 3. In the low calcium diet/irradiation group, there began to form woven bone incompletely in the bone-implant interface area at 19 days after implantation, but there were vascular connective tissues in the bone-implant interface area over the entire experimental period. 4. In the control group and low calcium diet/non-irradiation group, bone labeling bands were observed at 33 days after implantation, which suggests that the bone formation and remodeling was in process, but interstitial bone remodeling was not observed in the low calcium diet/irradiation group.         치성 및 비치성 조직이 악골 창상치유에 미치는 영향에 관한 실험적 연구

        This study was for comparing healing patterns and effects between with odontogenic and nonodontogenic tissues on the defected mandible. Experimental bone defects that measured 3 mm in diameter were created on the mandibular body of guinea pig by removal of bone with the use of trephine burs and bone defects were grafted with Biogran(Orthovita Co., U.S.A.)and covered with Dura Mata(Pfrimmer-Viggo GmbH Co., Germany). Guinea pigs were serially terminated by fours on the 3 day, the 1 week, the 2 weeks. the 3 weeks, the 4 weeks, and the 5 weeks after experiment, and the mandibular body was removed and fixed with 10% neutral formalin. They were decalcified adnembedded in paraffin as using the usual methods. The specimen sectioned and stained with hematoxylin and eosin and toluidine blue. They were observed with a light microscope and a polarizing microscope. The obtained results were as follows: 1. Defected bone was healed fast from the odontogenic tissues in early stage of the experiment. 2. The arrangement of the bone matrix was relatively regular in the bone from the nonodontogenic tissues, but irregular in the bone from the odontogenic tissues. 3. Compact bone has started to be resorbed and changed to the pattern of matrix bone tissue from 3 weeks after experiment.

        가정 호흡재활 프로그램 관련 연구 논문 분석

        Purpose: The purpose of this study was to analyze the trend of research on the use of home-based pulmonary rehabilitation. Method: Using Medline and CINAHL search engine, experiemental research, titled as 'home-based pulmonary rehabilitation' and 'chronic lung disease', statistics were collected. The total 29 experimental studies published between 1981 to 2000 were selected and analyzed according to the ATS guidelines. Result: 1) The study samples in research were mostly of men, whose lung function was moderate to severe. 2) The total period of the intervention program varied from 5 to 12 week. The program included excercise intervention and educational intervention. Psychosocial intervention was minimal. 3) The outcome of the home-based pulmonary rehabilitation had been measured mostly by physical aspects. Quality of life and cost were less frequently measured. 4) In terms of effectiveness of the home based pulmonary rehabilitation program, physiologic outcome, such as lung function and hematologic markers, there was more 'no effect' than 'positive effect', whereas there was more 'positive effect' in decreasing dyspnea, improving excercise capacity, and improving quality of life. Conclusion: Based on these findings, future research on home-based pulmonary rehabilitation should be emphasized and provide standardized protocol, including psychosocial intervention, and analyses on cost and quality of life.

        Background: With reports on the relation between smoking and depression appearing since 1980, it has become clear that depression is one of the psychodynamics of smoking, though there has been little or no progress made in the study of whether or not there is indeed a relation between depression and nicotine dependence. Purpose: The purpose of the study was to investigate the effects of depression on the amount and period of smoking and nicotine dependence among university man student smokers. Method: In periodical examination between 14 and 21, May, 2003, using 170 university male. The CES-D(Center for Epidemiologic Studies Depression) Scale and the FTQ(Fagerstrom Tolerance Qustionnaire) were utilized to measure the level of depression and nicotine dependency. The amount of smoking was calculated based on the number of cigarette used. Result: Results indicated that depression had something to do with amount of smoking. Nicotine dependence also was related to depression but total value was relatively low. finally the period of smoking had nothing to do with depression. Conclusion: The results of this study revealed that the amount of smoking of university man student smokers was very high. Both the amount of smoking and nicotine dependence were affected by levels of depression. Accordingly psychiatric nursing access such as emotional support is needed to manage depression and decrease smoking.
