Thrap3 promotes R-loop resolution via interaction with methylated DDX5

Transcription-replication conflicts lead to DNA damage and genomic instability, which are closely related to human diseases. A major source of these conflicts is the formation of R-loops, which consist of an RNA-DNA hybrid and a displaced single-stranded DNA. Although these structures have been studied, many aspects of R-loop biology and R-loop-mediated genome instability remain unclear. Here, we demonstrate that thyroid hormone receptor-associated protein 3 (Thrap3) plays a critical role in regulating R-loop resolution. In cancer cells, Thrap3 interacts with DEAD-box helicase 5 (DDX5) and localizes to R-loops. Arginine-mediated methylation of DDX5 is required for its interaction with Thrap3, and the Thrap3-DDX5 axis induces the recruitment of 5’-3’ exoribonuclease 2 (XRN2) into R-loops. Loss of Thrap3 increases R-loop accumulation and DNA damage. These findings suggest that Thrap3 mediates resistance to cell death by preventing R-loop accumulation in cancer cells.
Two Polymorphisms of Interleukin-4 Gene in Korean Adult Periodontitis

Adult periodontitis is a multifactorial disease characterized by multple genetic and environmental factors. In view of the importance of interleukin-4 (IL-4) gene as a genetic factor for adult periodontitis, we investigated the relationship between two polymorphisms (-590 C $\rightarrow$ T polymorphism and 70 bp repeat polymorphism) of the human IL-4 gene and adult periodontitis in the Korean population. Genomic DNA was extracted from white blood cells of 32 adult periodontitis patients and 150 normal controls, respectively. There were no significant differences in the allele, genotype and haplotype distributions of two polymorph isms between normal controls and adult periodontitis group. Therefore, our results suggest that IL-4 gene locus contributes little to the interindividual susceptibility for adult periodontitis in Korean population.

These days, robust autonomous navigation incorporates the information on the digital map including variety of road features such as position of lane markings, road boundaries and etc. In this study, a method of extracting road features are developed using two dimensional laser range finder (LRF). In particular, range measurements are employed to identify the road surface using split and merge method. In addition, the reflectivity measurements are used to identify lane markings on the road surface. Finally a digital map of road surface is constructed which is useful for robust autonomous navigation. The proposed method is verified using experimental LRF data measured on the road environments.

In this study, two-stage hypersonic scramjet vehicle was designed for the flight condition of Mach number 6. In order to launch at sea level, two stage concept was applied. The first stage of the vehicle is solid rocket-powered and is mounted under the second stage. The second stage is powered by scramjet propulsion system and gas wings. The suggested mission scenario is to deliver 0.2 ton payload to the range of 2,000 km. For the first step of conceptual design, trajectory of air vehicle was calculated by 3-DOF trajectory code. Based on the result of trajectory code, scramjet engine design and mass estimation were performed by non-equilibrium nozzle flow code and NASAs HASA model, respectively. In order to find best solution, all steps of designing process was iterated until they was reached.
초음속 연소에서 연료 분사구 형상에 따른 연소성능 변화에 대한 실험적 연구

초음속 연소에서 연료를 분사하는 분사구의 형상에 따른 연소 성능의 변화를 파악하기 위하여 실험과 준일차 해석(Quasi-One Dimensional Analysis)을 수행하였으며, 결과 값을 본 팀에서 동일 설비 및 조건으로 2008년에 시험을 수행한 데이터와 비교해 보았다. 연료 분사구 형상은 2008년 수행하였던 시험모델과 비교하여 연료 분사구의 크기를 줄이고 개수를 증가시켰으며, 연료 분사구의 크기 및 개수는 동일한 연료 압력에서 연료의 분사량과 침투깊이가 동일하도록 결정되었다. 실험결과 공동이 없을 시에는 연료 분사구의 크기를 줄이고 개수를 늘렸을 때 연소성능이 큰 폭으로 증가하였지만, 일자형 공동에서는 그 영향이 미미하였다. 하지만 지그재그형 공동에서는 연료 분사구의 크기를 줄이고 개수를 늘렸을 때 오히려 연소성능이 저하되었다. In order to investigate the effect of fuel injection hole configuration within the scramjet combustor, experiment and quasi-one-dimensional analysis was performed. And the results were compared with experiment and analysis result which were performed in 2008 with same facility and test condition. Fuel injection hole size was decreased and quantity was increased. However the depth of fuel penetration and fuel flow were maintained. As a test result, combustion performance was increased significantly with no-cavity injector and slightly with plain-cavity. However, combustion performance with zigzag-cavity was decreased.

Paraffin-embedded tissue samples from 30 cases of non-Hodgkin's lymphoma(NHL) and 10 of reactive hyperplasia, were processed for interphase cytogenetic chromosomal study. We performed non-fluorescent in situ hybridization(NFISH) using enzymatic method with digoxigenin-labeled DNA centromeric probes for chromosome 7, 12, 18 and X, and painting probe for chromosome 18. Chromosomal aberrations were observed in 27(90%) out of 30 cases of NHL. The most commonly observed numerical aberratiort was extracopy of X chromosome. There were some characteristic aberrations corresponding to each grades and groups of NHL by International Working Formulation: In low grade NH]L(9 cases), a third was associated with extracopy of chromosome 12, and disomy X was frequently found in small Iymphocytic Iymphoma(75%). With intermediate grade(16 cases), tetraploidy(25%), translocation of chromosome 18(25%), and extracopy of chromosome 18(19%) were chararteristically associated. These result suggest that interphase NFISH is a easily performable method in retrograde cytogenetic study of archival materials. Some specifically correlated chromosomal aberrations corresponding to the histopathologic grades and groups could provide us more valuable information for determining pathologic diagnosis and assessing the clinical outcome of NHL.

비행 마하수 6으로 운용되며, 지상 정지 추력으로부터 사용이 가능하도록 2단 추진체 개념이 적용된 스크램제트 엔진 비행체에 대한 개념설계를 수행하였다. 1단은 로켓을 적용되었으며, 2단은 탄화수소 계열의 연료를 사용하는 스크램제트 엔진이 적용되었다. 개념설계를 위하여 반경 2,000km의 운용거리와 0.2톤의 탑재체 무게를 가정하였다. 개념설계의 첫 번째 단계로 3-DOF 코드를 이용하여 비행궤도를 계산하였으며, 계산된 비행계도를 바탕으로 일차원-비평형 유동 코드와 NASA의 HASA 데이터베이스를 이용하여 스크램제트 엔진에 대한 개념설계를 수행하였다.
