DMSO‐ and Serum‐Free Cryopreservation of Wharton's Jelly Tissue Isolated From Human Umbilical Cord

<P><B>ABSTRACT</B></P><P>The facile nature of mesenchymal stem cell (MSC) acquisition in relatively large numbers has made Wharton's jelly (WJ) tissue an alternative source of MSCs for regenerative medicine. However, freezing of such tissue using dimethyl sulfoxide (DMSO) for future use impedes its clinical utility. In this study, we compared the effect of two different cryoprotectants (DMSO and cocktail solution) on post‐thaw cell behavior upon freezing of WJ tissue following two different freezing protocols (Conventional [−1°C/min] and programmed). The programmed method showed higher cell survival rate compared to conventional method of freezing. Further, cocktail solution showed better cryoprotection than DMSO. Post‐thaw growth characteristics and stem cell behavior of Wharton's jelly mesenchymal stem cells (WJMSCs) from WJ tissue cryopreserved with a cocktail solution in conjunction with programmed method (Prog‐Cock) were comparable with WJMSCs from fresh WJ tissue. They preserved their expression of surface markers, pluripotent factors, and successfully differentiated in vitro into osteocytes, adipocytes, chondrocytes, and hepatocytes. They also produced lesser annexin‐V‐positive cells compared to cells from WJ tissue stored using cocktail solution in conjunction with the conventional method (Conv‐Cock). Real‐time PCR and Western blot analysis of post‐thaw WJMSCs from Conv‐Cock group showed significantly increased expression of pro‐apoptotic factors (BAX, p53, and p21) and reduced expression of anti‐apoptotic factor (BCL2) compared to WJMSCs from the fresh and Prog‐Cock group. Therefore, we conclude that freezing of fresh WJ tissue using cocktail solution in conjunction with programmed freezing method allows for an efficient WJ tissue banking for future MSC‐based regenerative therapies. <P>Electrically driven hybrid light‐emitting diodes (LEDs) consisting of quantum dots, wires, and wells based on the nanometer‐sized pyramid GaN structure are reported by Taek Kim, Yong‐Hoon Cho, and co‐workers on page 5364. The LEDs exhibit mixed emissions from InGaN quantum dots, wires, and wells formed at the tops, edges, and sidewalls of the pyramids, respectively. The hybrid LEDs containing low‐dimensional quantum structures provide a broad‐band, highly efficient visible lighting source. </P>
Highly Efficient Organic THz Generator Pumped at Near‐Infrared: Quinolinium Single Crystals

<P><B>Abstract</B></P><P>A novel highly efficient ionic electro‐optic quinolinium single crystals for THz wave applications is reported. Acentric quinolinium derivatives, HMQ‐T (2‐(4‐hydroxy‐3‐methoxystyryl)‐1‐methylquinolinium 4‐methylbenzenesulfonate) and HMQ‐MBS (2‐(4‐hydroxy‐3‐methoxystyryl)‐1‐methylquinolinium 4‐methoxybenzenesulfonate) exhibit high order parameters cos<SUP>3</SUP><I>θ</I><SUB>p</SUB> = 0.92 and cos<SUP>3</SUP><I>θ</I><SUB>p</SUB> = 1.0, respectively, as well as a large macroscopic optical nonlinearity, which is in the range of the benchmark stilbazolium DAST (<I>N,N</I>‐dimethylamino‐<I>N</I>’‐methylstilbazolium 4‐methylbenzenesulfonate) and phenolic polyene OH1 (2‐(3‐(4‐hydroxystyryl)‐5,5‐dimethylcyclohex‐2‐enylidene)malononitrile) crystals. As‐grown unpolished bulk HMQ‐T crystals with a side length of about 6 mm and thickness of 0.56 mm exhibit 3.1 times higher THz generation efficiency than 0.37 mm thick OH1 crystals and about 8.4 times higher than 1 mm thick inorganic standard ZnTe crystals at the near‐infrared fundamental wavelength of 836 nm. Therefore, HMQ crystals with high order parameter obviously have a very high potential for high power THz‐wave generation and its applications.</P>

<P><B>Abstract</B></P><P>In the present study, we used TaqMan real‐time polymerase chain reaction to quantify and compare infection of white spot syndrome virus (WSSV) with shrimp production of <I>Fenneropenaeus chinensis</I> cultured in outdoor ponds along the west coast of the South Korea. In 2007, a total of 60 specimens in summer and 116 specimens in autumn were collected from 12 growing‐out ponds and 12 harvest ponds respectively. Pond harvest data were obtained from farmers. Of the summer samples, all specimens were WSSV positive, with a wide range of 12.4–7.0 × 10<SUP>7</SUP> (mean 7.5 × 10<SUP>6</SUP>) copies ng<SUP>−1</SUP> DNA; shrimp production was 1.7 metric tonnes per hectare (mt ha<SUP>−1</SUP>). Of the 116 autumn‐sample specimens, 81 (69.8%) were WSSV positive; WSSV infection had been decreased dramatically, to 0–7.2 (mean 3.5) copies ng<SUP>−1</SUP> DNA. Shrimp production of autumn ponds was 2.1 mt ha<SUP>−1</SUP>. Statistical analysis indicated that the difference in WSSV infections detected in summer and autumn was highly significant (<I>P</I><0.01). In summer, seven ponds (58.3%) with low‐WSSV infection loads (0–1000 WSSV copies ng<SUP>−1</SUP> DNA) had shrimp production of 2.7 mt ha<SUP>−1</SUP>; the others had shrimp production of only 0.2 mt ha<SUP>−1</SUP>. The mean shrimp production between the two infection levels showed a highly statistically significant difference (<I>P</I><0.01).</P>
