<P>Abstract</P><P>Recent studies have suggested that virulent strains of <I>Mycobacterium tuberculosis</I> induce apoptosis in macrophages less often than do attenuated strains. K-strain, which belongs to the Beijing family, is the most frequently isolated clinical strain of <I>M. tuberculosis</I> in Korea. In this study, we investigated the differential induction of cell death in human monocytic THP-1 cells by K-strain and H37Rv, a virulent but laboratory-adapted strain of <I>M. tuberculosis</I>. Although no significant difference in growth rate was observed between the cells exposed to K-strain and those exposed to H37Rv, the levels of protective cytokines such as tumour necrosis factor (TNF)-&agr;, interleukin (IL)-6 and IL-12p40 were lower in K-strain-infected cells than in H37Rv-infected cells. Cell viability assays showed that both K-strain and H37Rv, but not heat- or streptomycin-killed bacteria, induced THP-1 cell death in a TNF-independent manner. In contrast, double staining with fluorochrome-labelled inhibitors of caspase and propidium iodide and lactate dehydrogenase release assays revealed that K-strain induced significantly higher levels of necrotic cell death, rather than apoptosis, in THP-1 cells than did H37Rv. Anti-apoptotic <I>Bcl-2</I>, <I>Mcl-1</I>, <I>Bfl-1</I> and <I>Bcl-xL</I> in the cells were significantly upregulated following infection with K-strain compared with H37Rv, whereas <I>Bax</I> was slightly upregulated in response to infection with both H37Rv and K-strain. These results suggest that the highly virulent K-strain keeps cellular apoptosis as a host defense mechanism to a minimum and induces necrosis in macrophages.</P>

N/A Background: Potassium channel opener (K-opener) opens ATP-sensitive K+-channel located at membrane and induces potassium efflux from cytosol, resulting in intracellular hyperpolarization. Newly synthesized K-opener is currently examined for pharmacologic potency by means of rubidium release test from smooth muscle strip pre-incubated with Rb-86. Since in-vive behavior of thallium is similar to that of rubidium, we hypothesized that K-opener can alter T1-201 kinetics in vivo. Purpose: This study was prepared to investigate the effects of pinacidil (one of potent K-openers) on the T1-201 uptake and clearance in cultured myocyte, and in-vivo biodi- stribution in mice. Methods: Spontaneous contracting myocytes were prepared to imitate in-vivo condition from 20 hearts of 3-5 days old Sprague-Dawley rat and cultured for 3-5 days before use (5×105 cells/ml). Pinacidil was dissolved in 10% DMSO solution at a final concentration of 100nM or 10uM and was co-incubated with T1-201 in HBSS buffer for 20-min to evaluate its effect on cellular T1-uptake, or challenged to cell preparation pre-incubated with T1-201 for washout study. Two, 40 or 100μg of pinacidil was injected intravenously into ICR mice at 10 min after 5μCi T1-201 injection, and organ uptake and whole body retention rate were measured at different time points. Results: Co-incubation of pinacidil with T1-201 resulted in a decrease in T1-201 uptake into cultured myocyte by 1.6 to 2.5 times, depending on pinacidil concentration and activity of T1-201 used. Pinacidil enhanced T1-201 washout by 1.6-3.1 times from myocyte preparations pre-incubated with T1-201. Pinacidil treatment appears to be resulted in mild decreases in blood and liver activity in normal mice, in contrast, renal and cardiac uptake were mildly decreased in a dose dependent manner. Whole body retention ratios of T1-201 were lower at 24 hour after injection with 100μg of pinacidil than control. Conclusion: These results suggest that treatment with K-opener may affect the interpretation of T1-201myocardial images, due to decreasing thallium accumulation and enhancing washout from myocardium.

Advanced glycation end products (AGEs) are involved in the development of diabetic complications such as diabetic retinopathy. 5'-methoxybiphenyl-3,4,3'-triol (referred to as K24) was isolated using bioactivity-guided fractionation of Osteomeles schwerinae C. K. Schneid. and identified as a potent AGE inhibitor. To identify the protective effect of K24 on disruption of the blood-retinal barrier, AGE-RSA was intravitreally injected into rat eyes. K24 had an inhibitory effect on AGE-RSA-induced retinal vascular leakage by suppressing the expression of vascular endothelial growth factor (VEGF) and decreasing the loss of occludin. In addition, we examined whether K24 has a preventive effect against retinal pathogenic angiogenesis in an oxygen-induced retinopathy (OIR) mouse model. K24 significantly reduced the retinal non-perfused area and neovascular tufts in the OIR mice. These data indicate that K24 could serve as an innovative pharmaceutical agent to prevent blood-retinal barrier breakage and retinal pathogenic angiogenesis through an anti-VEGF mechanism.

<P>We had developed four years ago a superconducting property measurement system (SPMS) that can be used to acquire electrical and thermal properties of high-temperature superconductor (HTS) tape samples. The SPMS was composed of the sample holder for mounting an HTS tape and 3 T HTS magnet, both cooled individually by conduction using a cryocooler. The maximum dimension of the HTS tape sample that could be loaded on the sample holder was 15 mm in width and 250 mm in length. The conduction-cooled sample holder was again fabricated to increase the maximum test current up to 800 A. Maximum transport current was 763 A in magnetic field intensity 3 T. The measuring temperature range was 15-50 K.</P>
풍압력하에서 피예항중인 손상선박의 침로안정성에 관한 연구

본 논문은 손상선박의 안전대책에 관한 연구의 일환으로, 황천항행중인 선박이 충돌, 좌초 등 원인에 의해 손상을 받았을 때를 가상하고, 손상선박을 다른 안전한 장소로 예항하고자 할 때의 침로안정성 문제를 다루고 있다. 외력으로는 바람의 영향만을 고려할 때 예선·피예선계의 침로안정성 평가를 위한 특성방정식을 도술하고, 피예선의 각 손상상태에 따른 침로안정성을 수치계산하였다. 그 결과 손상상태, 풍속, 풍향 및 예항삭의 길이 등이 침로안정성에 미치는 영향을 평가할 수 있었다. This paper is related with one of safety plans to rescue a damaged-ship whether by collision, grounding or internal accident. We discuss the problem on course stability of damaged-ship while towed under severe wind pressure. The characteristic equation to assess the stability on course, is derived from sway and yaw coupled motion of towing and towed vessels with wind effect. Through the numerical calculation on course stability of towing and towed vessels system, the relationship between the course stability of a towed damaged-ship and wind direction or towrope length, is clarified with the parameters of weather and damage conditions.
마이크로 센서용 다채널 전해질 분석기의 샘플챔버 개선 및 샘플상호간의 간섭현상 보정

In this paper, the sample chamber for multichannel electrolyte analyzing system was designed and the correction algorithm for interference among samples was proposed. The sample chamber was designed to replace the ISFET sensor more easier. The principles of design were the structural independency of each channel and the shield of sample solution from air contact. The correction algorithm could reject the interference effect of mixed sample solution and compensate the voltage difference between single electrolyte solution and mixed electrolyte solution. To approve the proposed algorithm, the several experiments were carried out under the condition of mixed samples. We acknowledged that with our proposed algorithm, the sensor output influenced by mixed electrolyte solution was improved with measurement accuracy.

A new potassium sensitive field effect transistor modified with 4`-aminobenzo-15-crown-5 was prepared and its response characteristics were evaluated. The response slope of K^+-ISFET for pH was 30.0 mV/decade and the response time was more than 3 minutes. And the response slope and time of the K^+-ISFET for potassium ion was 19.5±0.2 mV/decade and about 3 minutes, respectively. The linear response range of the sensor for potassium ion was 2.0X10^(-4)∼1.0X10^(-2)M. The selectivity coefficients of the K^+-ISFET for the alkali and alkaline earth metal ions were also evaluated. Sodium, ammonium and calcium ions exhibited relatively significant interference. The long term stability of the sensor was remarkably improved and it could be used for more than 50 days.

A cartridge for FET type sensors was designed and fabricated by using micromachining techniques such as silicon etching and bonding techniques. The photosensitive polymer(THB 30) is used to define an uniform micropool and to encapsulate the sensor surface. A REFET is fabricated to replace the conventional reference electrode using photosensitive polymer(OMR 83). The fabricated cartridge includes four electrolyte sensors (H^(+), K^(+), Ca^(2+), Na^(+)), and the dead volume of intergrated flow channel is about 8μl.

The selective chemical etching of Si_(1)-_(x)Ge_(x). heteroepitaxial layer grown by rapid thermal CVD (RTCVD) method has been studied with respect to Ge content ( 0.15 ≤ x ≤ 0.30 ). The properties of HF : H_(2)O_(2) : CH_(3)COOH (1:2:3) chemical etchant is highly selective in the Si_(1)-_(x)Ge_(x) / Si heteroepitaxial structure. The selectivity is presented better than 100 for Ge content (x≥0.20) and increased with Ge content in Si_(1)-_(x)Ge_(x) heteroepitaxial layer.
