We propose a spheroid chip that uses removable cell trapping barriers and that is capable of forming and extracting multicellular spheroids. By using a conventional well plate and flask, it is difficult to form small-sized spheroids, which resemble avascular 3D cell-cell interaction. It was difficult to extract spheroids using conventional microchips and fixed cell trapping barriers. The proposed chip, however, facilitates both formation and extraction of spheroids by using removable cell trapping barriers formed by membrane deflection. The cell trapping barriers, formed at the membrane pressure of 50 ㎪, hold the cells in the trapping region at a cell inlet pressure of 145?155 ㎩. After incubation for 24 h, the trapped cells form uniform spheroids. We successfully extract the spheroids at a cell inlet pressure of 5 ㎪ after removing the membrane pressure. The extracted spheroids have a diameter of 197.2 ± 11.7 Bm with a viability of 80.3 ± 7.7%. Using the proposed chip, uniform spheroids can be formed and these spheroids can be safely extracted for carrying out the post-processing of spheroids.

Sleep Problems and Daytime Sleepiness in Children with Nocturnal Enuresis

        Gu, Yun-Mo,Kwon, Jung Eun,Lee, Gimin,Lee, Su Jeong,Suh, Hyo Rim,Min, Soyoon,Roh, Da Eun,Jo, Tae Kyoung,Baek, Hee Sun,Hong, Suk Jin,Seo, Hyeeun,Cho, Min Hyun Korean Society of Pediatric Nephrology 2016 Childhood kidney diseases Vol.20 No.2

Purpose: Nocturnal enuresis (NE) is one of the most common problems in childhood. NE has a multifactorial etiology and is influenced by sleep and arousal mechanisms. The aim of the present study was to prospectively evaluate sleep problems and patterns in children with NE compared with normal healthy controls. Methods: Twenty-eight children with NE and 16 healthy controls were included in the study. To evaluate sleep habits and disturbances, parents and children filled out a questionnaire that included items about sleep patterns and sleep-related behaviors prior to treatment for NE. Demographic factors and other data were compared for the two groups based on the responses to the sleep questionnaire. Results: Night awakening, sleepwalking, and periodic limb movements were more prevalent in children with NE, but symptoms of sleep-disordered breathing were not increased in this group. There were statistically significant differences in periodic limb movements and daytime sleepiness between the two groups. Conclusion: Children with NE seemed to have more sleep problems such as night awakening, sleepwalking, and periodic limb movements. In addition, a higher level of daytime sleepiness and hyperactivity in patients with NE suggested a relationship between NE and sleep disorders.

Seven-Day Continuous Abstinence Rate from Smoking at 1, 2, or 3 Years after the Use of Varenicline

( Jin Se Kim ),( Ju Young Jang ),( Eun Hye Park ),( Joo Young Lee ),( Kang Mo Gu ),( Jae Woo Jung ),( Jae Chol Choi ),( Jong Wook Shin ),( In Won Park ),( Byoung Whui Choi ),( Jae Yeol Kim ) 대한결핵 및 호흡기학회 2015 Tuberculosis and Respiratory Diseases Vol.78 No.2

Background: Varenicline, a selective partial agonist/antagonist of the α4β2 nicotinic receptor, has proven effectiveness for smoking cessation by several randomized, controlled trials. Because few studies have evaluated the long-term efficacy of varenicline, we tried to evaluate the smoking status of varenicline users up to 3 years after the initial prescription of the drug. Methods: We interviewed varenicline users who were prescribed the drug from June 2007 to May 2010 by telephone, from June 2010 to May 2011. Results: One-hundred and thirty-three of 250 varenicline users (53.2%) were available for the survey. Seven-day continuous abstinence from smoking was adhered to by 17 of 39 respondents (43.6%) at 1 year, and 11 of 36 (30.6%) and 19 of 58 (32.8%) at 2 and 3 years since the first use of varenicline, respectively. Compared to current smokers, successful quitters were older (55.0 years vs. 49.9 years, p=0.01), had better compliance to the 12-week course (27.7 vs. 9.3%, p=0.01), and had taken varenicline longer (10.1 vs. 5.9 weeks, p=0.01). Fifty-four of 71 current smokers (76.1%) were willing to stop smoking in the near future. The preferred ways to cease smoking were will-power (48.1%), varenicline (25.9%), nicotine replacement therapy (11.1%), and others (14.9%). Conclusion: Smokers should be encouraged to stick to the proven way for recommended period of time for successful cessation of smoking.

Although primary small intestinal carcinoma (SIC) is morphologically similar to colorectal carcinoma and shares many of the genetic changes of carcinogenesis, little is known about the role of defective mismatch repair (MMR) genes involved in the SIC. The aim of this study is to investigate the role of defective MMR genes and correlation between clinicopathological factors and loss of MMR protein in SIC.

      • Novel Mediator Proteins of the Small Mediator Complex in Drosophila SL2 Cells

        Gu, Ji-Young,Park, Jin Mo,Song, Eun Joo,Gaku, Mizuguchi,Yoon, Jeong Ho,Kim-Ha, Jeongsil,Lee, Kong-Joo,Kim, Young-Joon 梨花女子大學校 藥學硏究所 2002 藥學硏究論文集 Vol.- No.11

The Mediator complex is generally required for tran-scriptional regulation In species ranging from yeast tohuman. Throughout evolution, the functional diversityof the Mediator complex has been enhanced to meet theincreasing requirements for sophisticated gene regula-tion. It is likely that greater structural complexity isthus required to accomplish these new, complex regula-tory functions. In this study, we took systematic steps toexamine various types of Mediator complexes inDro.sophila melanogaster. Such efforts led to the identifica-tion of three distinct forms of Mediator complexes. Inexploring their compositional and functional heteroge-neity, we found that the smallest complex (C1) is highlyenriched in a certain type of Drosophilacells and pos-sesses novel Mediator proteins. The subonits staredamong the three Mediator complexes (C1, C2, and C3)appear to lorn a stable modular structure that serves asa binding surface for transcriptional activator proteins. However, only C2 and C3 were able to support activatedtranscriptioa in vitro. These findings suggest that dif-ferent cell types may require distinct Mediator com-plexes, bene of which may participate in nuclear pro-cesses other than the previousIy identified functions.

Vol.11 No.2
