        The LIM-only transcription factor LMO2 determines tumorigenic and angiogenic traits in glioma stem cells

        Glioblastomas (GBMs) maintain their cellular heterogeneity with glioma stem cells (GSCs) producing a variety of tumor cell types. Here we interrogated the oncogenic roles of Lim domain only 2 (LMO2) in GBM and GSCs in mice and human. High expression of LMO2 was found in human patient-derived GSCs compared with the differentiated progeny cells. LMO2 is required for GSC proliferation both in vitro and in vivo, as shRNA-mediated LMO2 silencing attenuated tumor growth derived from human GSCs. Further, LMO2 is sufficient to induce stem cell characteristics (stemness) in mouse premalignant astrocytes, as forced LMO2 expression facilitated in vitro and in vivo growth of astrocytes derived from Ink4a/Arf null mice and acquisition of GSC phenotypes. A subset of mouse and human GSCs converted into vascular endothelial-like tumor cells both in vitro and in vivo, which phenotype was attenuated by LMO2 silencing and promoted by LMO2 overexpression. Mechanistically, the action of LMO2 for induction of glioma stemness is mediated by transcriptional regulation of Jagged1 resulting in activation of the Notch pathway, whereas LMO2 directly occupies the promoter regions of the VE-cadherin gene for a gain of endothelial cellular phenotype. Subsequently, selective ablation of human GSC-derived VE-cadherin-expressing cells attenuated vascular formation in mouse intracranial tumors, thereby significantly prolonging mouse survival. Clinically, LMO2 expression was elevated in GBM tissues and inversely correlated with prognosis of GBM patients. Taken together, our findings describe novel dual roles of LMO2 to induce tumorigenesis and angiogenesis, and provide potential therapeutic targets in GBMs.

        A Combinatorial Chemistry Method for Fast Screening of Perovskite-Based NO Oxidation Catalyst

        <P>A fast parallel screening method based on combinatorial chemistry (combichem) has been developed and applied in the screening tests of perovskite-based oxide (PBO) catalysts for NO oxidation to hit a promising PBO formulation for the oxidation of NO to NO<SUB>2</SUB>. This new method involves three consecutive steps: oxidation of NO to NO<SUB>2</SUB> over a PBO catalyst, adsorption of NOx onto the PBO and K<SUB>2</SUB>O/Al<SUB>2</SUB>O<SUB>3</SUB>, and colorimetric assay of the NOx adsorbed thereon. The combichem experimental data have been used for determining the oxidation activity of NO over PBO catalysts as well as three critical parameters, such as the adsorption efficiency of K<SUB>2</SUB>O/Al<SUB>2</SUB>O<SUB>3</SUB> for NO<SUB>2</SUB> (α) and NO (β), and the time-average fraction of NO included in the NOx feed stream (ξ). The results demonstrated that the amounts of NO<SUB>2</SUB> produced over PBO catalysts by the combichem method under transient conditions correlate well with those from a conventional packed-bed reactor under steady-state conditions. Among the PBO formulations examined, La<SUB>0.5</SUB>Ag<SUB>0.5</SUB>MnO<SUB>3</SUB> has been identified as the best chemical formulation for oxidation of NO to NO<SUB>2</SUB> by the present combichem method and also confirmed by the conventional packed-bed reactor tests.         네트워크를 이용한 온실 감시 시스템의 개발

        this study was carried out to design, construct, and test a greenhouse monitoring system for the environment and status of control devices in a greenhouse from a remote site using internet. The measuring items selected out of many environmental factors were temperature, humidity, solar radiation, CO_2, SO_x, NO_x concentration, EC, pH of nutrient solution, the state of control devices, and the image of greenhouse. The network system consists of the three kinds of monitors named the Group Monitor and the Server Monitor and the Server Monitor. The results of the study are summarized as follows. 1.The measuring module named the House Monitor, which is used to watch the state of the control device and the environment of the greenhouse, was developed to a embedded monitoring module using one chip microprocessor. 2.For all measuring items, the House Monitor showed a satisfactory accuracy within the range of ±0.3%FS. The House Monitors were connected to the Group Monitor by communication method of RS-485 type and could operate under power and communication fault condition within 10 hours. The Group Monitor was developed to receive and display measurement data received from the House and to control the greenhouse environmental devices. 3.The image of the plants inside greenhouse were captured by PC camera and sent to the Group Monitor. the greenhouse manager was able to monitor the growth state of plants inside greenhouse without visiting individual greenhouse. 4.Remote monitoring the greenhouse environment and status of control devices was implemented in a client/server environment. The client monitor of the greenhouse manager at a remote site or other greenhouse manager was able to monitor the greenhouse environment and the state of control devices from the Server Monitor using internet.

        Objectives: The purpose of the study was to identify the relationships between cybersex addition, loneliness and relationship satisfaction among Korean college students. Methods: 293 students were conveniently selected. The instruments used in this study were cyber-sexual addiction index, loneliness scale and relationship assesment scale. ANOVA, t-test and Pearson correlation coefficient with SPSS 22.0 Version were used to analyze the data. Results: 8.9% of the participants had moderate or severe addiction to cybersex. The level of cybersex addiction is significantly influenced by gender, period of relationship, time taking to sexual relations, contact number of cyber pornography and contact time of cyber pornography. Significant association was observed between cybersex addition, loneliness and relationship satisfaction. Conclusions: The factors mentioned in this study as well as loneliness and relationship satisfaction were important factors. Therefore, future studies on this topics need to develope interventions to reflect cultural and physical character of college students.

      • 유방암 환자의 말초혈액에서 역전사효소연쇄중합반응을 이용한 Human Mammaglobin 측정의 임상적 유용성

        Background: The mammaglobin gene encodes a novel protein that is secreted from the mammalian epithelium of normal breast tissue as well as malignant breast cancer tissues. In order to ascertain the prognostic value of mammaglobin gene in breast cancer patients, we measured the expression of human mammaglobin (hMAM) by RT-PCR method in various stages of breast cancer patients. Methods: Peripheral blood samples from forty healthy volunteers and 114 breast cancer patients were obtained. Peripheral blood stem cells (PBSC) collected for the purpose of autologous stem cell transplantation in five patients with metastatic breast cancer and ten patient with high risk for relapse and no evidence of disease were used for hMAM assay. Results: All samples from peripheral blood of forty healthy individuals (twenty males and twenty females) were negative for hMAM, whereas 43 of 114 samples (38%) from breast cancer patients were positive for hMAM mRNA. All the normal breast tissues were positive for hMAM mRNA. hMAM mRNA expression was detected in 11 of 42 (26%) in breast cancer patients who underwent for curative resection and had no evidence of disease, in 8 of 25 (34%) with chemo-sensitive relapsed disease, and in 16 of 32 (53%) with chemo-refractory progressive disease. Eight (53%) samples from peripheral blood of 15 breast cancer patients with metastatic disease at diagnosis were positive for hMAM. Three (20%) samples from peripheral blood stem cells of 15 breast cancer patients for high dose chemotherapy were positive for hMAM. Conclusion : In contrast to healthy volunteers, hMAM transcripts were detected in the peripheral blood of breast cancer patients. The frequency of hMAM expression in peripheral blood was correlated with the clinical stages of disease, but, was not significant. The contamination of hMAM expressing cells in the stem cell pool warrants additional effective purging method before the transplantation. The clinical relevance of hMAM RT-PCR-based tumor cell detection in the peripheral blood of breast cancer patients should be further evaluated in prospective studies.

      • Therapeutic targeting of subdural medulloblastomas using human neural stem cells expressing carboxylesterase

        The prognosis of medulloblastoma has improved significantly because of advances in multi-modal treatments; however, metastasis remains one of the prognostic factors for a poor outcome and is usually associated with tumor recurrence. We evaluated the migratory potential and therapeutic efficacy of genetically engineered human neural stem cells (NSCs) that encode a prodrug enzyme in the subdural medulloblastoma model. We genetically modified HB1.F3 (F3) immortalized human NSCs to express rabbit carboxylesterase (rCE) enzyme, which efficiently converts the prodrug CPT-11 (Irinotecan) into an active anti-cancer agent (SN-38). To simulate clinical metastatic medulloblastomas, we implanted human medulloblastoma cells into the subdural spaces of nude mice. rCE expressing NSCs (F3.rCE) were labeled with fluorescence magnetic nanoparticle for in vivo imaging. The therapeutic potential of F3.rCE was confirmed using a mouse subdural medulloblastoma model. The majority of intravenously (i.v.) injected, F3.rCE cells migrated to the subdural medulloblastoma site and a small number of F3.rCE cells were found in the lungs, pancreas, kidney and liver. Animals that received F3.rCE cells in combination with prodrug CPT-11 survived significantly longer (median survival: 142 days) than control mice that received F3.rCE cells only (median survival: 80 days, P<0.001) or CPT-11 only (median survival: 118 days, P<0.001). In conclusion, i.v. injected F3.rCE NSCs were able to target subdural medulloblastomas and demonstrate therapeutic efficacy. Our study provides data that supports further investigation of stem-cell-based gene therapy against metastatic medulloblastomas.

        Epidemiological Investigation of Deaths Among Radiation Workers in Nuclear Power Plants of Korea

        Many epidemiological studies showed increases of cancer risk in humans following radiation exposure. But, these results mainly came from high dose and high rate radiation exposure and the risk related low dose exposure still remains in question. To overcome the problem about statistical power due to limitation of study population, IARC is processing international collaborative study for pooling data analysis. In Korea, reconstructed cohort of NPP workers based on dosimetry records has followed since 1994. Based on this cohort, we introduce epidemiological study about NPP workers in korea and investigate the exposure level and death cases in NPP workers exposed to protracted low-dose radiation.

        In this paper, we demonstrate that it is possible to develop an optimum ESD protection device by comparing the relative ESD robustness of each device, based on the monitoring of the peak temperature correlated with the peak power density. The methodology, supported by measurement data, could give an extra degree of freedom to the IC designer confronted with the troublesome ESD phenomenon.

        In the present study, the problem of an infinite anisotropic plate with an inclined crack subjected to a biaxial load is analyzed. It is assumed that the material is ideal homogeneous anisotropic body. By considering the effect of the load applied parallel to a horizontal plane of the plate, the distribution of stresses at the crack tip is reanalyzed. In addition the problem of predicting crack propagation in anisotropic solids which is a subject of considerable practical importance is examined. The effect of load biaxiality on the direction of initial crack extension is made explicitly.
