      • The synergistic effects of combined NaOCl, gamma irradiation and vitamin B<sub>1</sub> on populations of Aeromonas hydrophila in squid

        The present study investigated the synergistic disinfection effects of combined NaOCl/gamma irradiation and NaOCl/vitamin B<SUB>1</SUB> treatment against Aeromonas hydrophila ATCC 7966 in tryptic soy broth (TSB) and squid. The synergistic effects were not dependent on the concentration of NaOCl or the dose of gamma irradiation, but were dependent on the vitamin B<SUB>1</SUB> concentration. Synergistic values for NaOCl/gamma irradiation treatment in TSB and squid were -0.37 to 2.09 log<SUB>10</SUB> CFU/mL and 0.00-2.92 log<SUB>10</SUB> CFU/g, respectively. The largest synergistic values, 2.09 log<SUB>10</SUB> CFU/mL in TSB and 2.92 log<SUB>10</SUB> CFU/g in squid, were as a result of treatment with a combination of 40 ppm NaOCl and 1.0 kGy gamma irradiation and 80 ppm NaOCl and 2.0 kGy gamma irradiation, respectively. Synergistic values for NaOCl/vitamin B<SUB>1</SUB> treatment in TSB and squid were 0.59-2.98 log<SUB>10</SUB> CFU/mL and 0.06-1.35 log<SUB>10</SUB> CFU/g, respectively. The largest synergistic values, 2.98 log<SUB>10</SUB> CFU/mL in TSB and 1.35 log<SUB>10</SUB> CFU/g in squid, were as a result of treatment with a combination of 300 ppm NaOCl and 1000 ppm vitamin B<SUB>1</SUB> and 1000 ppm NaOCl and 1000 ppm vitamin B<SUB>1</SUB>, respectively. The results in the broth study suggest that the combination of 40 ppm NaOCl and 1.0 kGy gamma irradiation and 300 ppm NaOCl and 1000 ppm vitamin B<SUB>1</SUB> could be regarded as a potential optimum hurdle approach for application to real target foods and(or) food surfaces for the control of A. hydrophila. Moreover, the results in the food study indicate that the combination treatment of 80 ppm NaOCl and 2.0 kGy gamma irradiation and 100 ppm NaOCl and 1000 ppm vitamin B<SUB>1</SUB> could possibly be used in seafood production, processing, and distribution processes to enhance seafood safety.

        <P>Lentinula edodes, the popular shiitake mushroom, is one of the most important cultivated edible mushrooms. It is used as a food and for medicinal purposes. Here, we present the 46.1 Mb draft genome of L. edodes, comprising 13,028 predicted gene models. The genome assembly consists of 31 scaffolds. Gene annotation provides key information about various signaling pathways and secondary metabolites. This genomic information should help establish the molecular genetic markers for MAS/MAB and increase our understanding of the genome structure and function. (C) 2016 Elsevier B.V.         The giant magnetoimpedance (GMI) of Fe_(73.5-x)Mn_xSi_(13.5)B_9Nb₃Cu₁ (x= 1, 3, 5) alloys prepared by a rapid quenching technique has been measured to investigate the influence of the structural changes in the crystallization process as well as the changes of the soft magnetic properties such as permeability, coercivity, magnetic anisotropy, etc. after thermal treatment. Magnetoimpedance ratio (MIR) and incremental permeability ratio (PR) measurements were investigated in the frequency range of 1-10 ㎒ and at a fixed current of 10 ㎃. Ultrasoft magnetic behavior has been observed in the samples annealed at 560℃. The PR curves become narrower and sharper after heat treatment due to the decrease of anisotropy. The MIR and PR coincided with the softness of the magnetic properties of the thermally treated samples.

      • HYDRA 및 PLANARIA의 X-RAY 傷害에 미치는 數種物質 處理後의 影響

        1. Hydra에 X­ray 4,500 r을 照射한 後 NaCN, SC(NH_2)_2, streptomycin 및 MgCl_2 溶液에 繼續的으로 處理하여 出芽 및 再生에 미치는 影響을 調査하여 다음과 같은 結果를 얻었다。 (a) Hydra bulgaris는 4,500 r으로 出芽 및 再生이 甚히 抑制되었다。 (b) MgCl_2 및 streptomycin은 非照射區 Hydra의 出芽 및 再生을 促進시켰다。 (c) MgCl_2 및 streptomycin은 照射區 Hydra의 出芽 및 再生로 促進시켜 X­ray傷害에 對한 保護作用을 나타내었다。 2. 4,500 r의 X­ray에 照射된 Planaria를 MgSO_4, NaNO_2, SC(NH_2)_2 및 Penicilline의 4種 溶液에 處理하여 兩眼再生 所要 時間을 測定 結果는 아래와 같다。 (a) 4,500 r의 X­ray는 兩眼在生에 時間的인 抑制作用을 갖는다. (b) MgSO_4, NaNo_2, SC(NH_2)_2의 3種 物質은 照射區 兩眼再生에 미친 X­ray의 傷害作用을 保護하고, 反面 非照射區 兩眼再生에는 何等 影響이 없다。 (c) Penicilline은 X­ray 傷害에 對한 保護效果가 全혀 없고, 反面 非照射區 兩眼在生을 顯著히 促進시킨다。 1) Hydras were treated in sodium cyanide, thiourea, streptomycin and magnecium chloride each in standard solution after exposure to 4,500 r X-rays. The rates of budding and regeneration in each solutions were determined. It was found that: a) Forty-five hundred r inhibited the budding and regeneration of Hydra bulgaris significantly. b) Streptomycin and magnesium chloride stimulated budding and regeneration of non-irradiated Hydras. c) Magnesium chloride and streptomycin significantly stimulated budding and regeneration of irradiated Hydras, and showed the protective effects against X-rays injury. 2) Planarias were placed continuously in the solutions of magnesium sulfate, sodium nitrite, thiourea and penicilline each in standard solution after exposure to 4,500 r X-rays. The rates of regeneration of Planarias in each solutions were determined. The results obtained were as follows. a) 4,500 r of X-rays inhibited the regeneration of Planaria significantly. b) Magnesium sulfate, sodium nitrite and thiorea modified the inhibitory effect of the radiation and were not effective at all on the non-irradiated planarian regeneration. c) Penicilline did not modify the inhibitory effect the radiation, but stimulated the regeneration of non-irradiated Planaria.

        Alkylphloroglucinol derivatives and triterpenoids with soluble epoxide hydrolase inhibitory activity from Callistemon citrinus

        <P>Phytochemical analysis of the leaves and stems of Callistemon citrinus (Curtis) Skeels led to the isolation of two new alkylphloroglucinols, gallomyrtucommulone E and F (1 and 2), along with four other known alkylphloroglucinol derivatives, gallomyrtucommulone A (3), endoperoxide G3 (4), myrtucommulone B (5), callistenone B (6) and five known triterpenoids, induding betulinic acid (7), 3 beta-acetylmorolic acid (8), 3 beta-hydroxy-urs-11-en-13(28)olide (9), diospyrolide (10) and ursolic acid (11). The structures of the natural compounds were determined from the spectroscopic evidences including 1D-/2D-NMR and HR-MS spectrometry. All the isolated compounds were assessed for the effects on the sEH inhibitory activity. The acylphloroglucinols myrtucommulone B (5)/callistenone B (6) (in mixture), and two triterpenoids, ursolic acid (11) and 3 beta-hydroxy-urs-11-en-13(28)-olide (9) displayed strong inhibition of sEH activity, with IC50 values of 0.7, 112 and 24.8 mu M, respectively.         주성분 분석을 이용한 거세한우의 체형분류에 관한 연구

        1997년부터 2001년까지 (사)한국종축개량협회의 전국한우능력평가대회에 출품된 642두의 거세한우의 도축 전까지 4회에 걸쳐 주요 10개 부위의 체형 측정치를 조사하여 주성분 분석을 이용하여 체형을 분류하였으며, 각 체형 측정치의 주성분 지수와 도체 형질과의 상관관계를 분석하였으며, 그 결과를 요약하면 다음과 같다. 1. 1차, 2차, 3차 및 4차 체형 측정치의 principal component(이하 P.C) 1의 분산은 각각 7.610, 8.297, 7.269 및 5.736로서 전체분산 중 76.0%, 83.0%, 72.7% 및 57.4%를 나타낼 수 있었고, P.C 1, P.C 2, P.C 3의 합은 전체분산 중 각각 86.69%, 90.49%, 84.62% 및 77.26%를 나타났다. 2. 각 차수별 체형 측정치의 P.C 1의 모든 계수가 양(+)이어서 일반적으로 좋은 체구의 크기를 나타내었으며, 체구의 크기에 가장 크게 영향을 미치는 형질은 흉심(0.328∼0.339)과 요각폭(0.325∼0.341)이였다. 또한 P.C 2, P.C 3 및 P.C 4는 각 차수별로 다양하게 나타났다. 3. 1차, 2차, 3차 및 4차 체형 측정치의 주성분 지수들과 도체형질간의 상관계수 추정에서 대부분의 분산을 차지하는 P.C 1은 도체중(0.539∼0.755), 일당증체량(0.256∼0.564), 등지방두께(0.227∼0.280) 및 배최장근단면적(0.187∼0.344)간에 정(+)의 상관을 보였으며, 육량지수(-0.246∼-0.110)와는 부(-)의 상관을 보였고, 근내지방도(0.066∼0.099)와는 대부분 유의적인 상관을 보이지 않거나 낮은 상관을 보였고, P.C 2, P.C 3 및 P.C 4는 주성분 지수들의 값에 따라 도체형질과 다양한 상관관계를 보였다. 단일 체형형질과 도체형질간의 상관 그리고 P.C 1의 주성분 지수와 도체형질과의 상관간에는 큰 차이가 없었다. Data were consisted of the ten body measurements (withers height, rump height, body length, chest depth, chest width, rump width, rump length, thurls width, hipbone width and chest girth) of 642 steers (Korean cattle), which was entered in the National Beef Quality Contest hosted by the Korea Animal Improvement Association from 1997 to 2001. A principal components analysis was used to classify the body types of the steers, and estimate the correlations between carcass traits and principal components for the body measurements of the first, second, third and fourth period, respectively. The first principal component of body measurements at the first, second, third and fourth period accounted for 76.0%, 83.0%, 72.7% and 57.4% of the total variance, respectively. The sum of first, second and third principal component at each period accounted for 86.69%, 90.49%, 84.62% and 77.26% of the total variance, respectively. At each period, all the first principal component of the body measurements were positive and it generally showed large framed body shape. The size of body was influenced mostly by chest depth(0.328∼0.339) and rump length(0.325∼0.341). The second, third and fourth principal component at the each period were various. There were positive correlations between principal components index of each period and carcass traits such as carcass weight(0.539∼0.755), average daily gain(0.256∼0.564), backfat thickness(0.227∼0.280), and eye muscle area(0.187∼0.344). The correlation with yield grade index(-0.246∼-0.110), however, was negative. The correlation with marbling score(0.066∼0.099) was low or statistically insignificant. According to principal component indexes of the second, third, and fourth components, the correlations with the carcass traits were various. There were no large differences between the correlations of the single body measurement trait with the carcass traits and the correlations of the first principal component indexes with the carcass traits.

        The effects of serum chemical values on the indidence of retained fetal membrane (R. F. M) was evaluated in dairy cows raised in Kyoung gi-do from May through September 1989. The results were as follows ; Data for serum chemical showed that 1. The values of total protein, glucose, cholesterol, GOT, GPT in the blood serum were significantly higher in the R. F. M. group.( ?? ) 2. The value of calcium in the blood serum was significantly lower in the R. F. M. group. ( ?? ) 3. The values of phosphorus, magnesium, Ca/P ratio in the blood serum were not any significantly differences.

        조제시 정제분할에 의한 중량편차

        Although original tablets are almost fit in weight variation test, in many cases, tablets are practically splitted for dose control in dispensing. These splitted tablets are considered secondary cause of weight variation. So, in this study tablets were classified into three groups by the average weight, first group was below 0.12g(A), second was more than 0.12g below 0.3g(B) and third was more than 0.3g(C). Each group consists of four kinds of tablet and weight variation after tablet splitting was calculated. As a result, average violation rate of weight variation test was 60% for A group, 42.5% for B group, 55% for C group. And the relation of hardness of tablet was the higher rate of weight variation. In conclusion, split of tablet for dose control in dispending was absolutely a cause of dose mistake. Especially drugs having side effect or toxic effect should be carefully when pharmacists split of tablet for dose control in dispending was absolutely a cause of dose mistake.         Poly-sgRNA/siRNA ribonucleoprotein nanoparticles for targeted gene disruption

        <P>Clustered regularly interspaced short palindromic repeats (CRISPR)-associated protein-9 nuclease (Cas9) can be used for the specific disruption of a target gene to permanently suppress the expression of the protein encoded by the target gene. Efficient delivery of the system to an intracellular target site should be achieved to utilize the tremendous potential of the genome-editing tool in biomedical applications such as the knock-out of disease-related genes and the correction of defect genes. Here, we devise polymeric CRISPR/Cas9 system based on poly-ribonucleoprotein (RNP) nanoparticles consisting of polymeric sgRNA, siRNA, and Cas9 endonuclease in order to improve the delivery efficiency. When delivered by cationic lipids, the RNP nanoparticles built with chimeric poly-sgRNA/siRNA sequences generate multiple sgRNA-Cas9 RNP complexes upon the Dicer-mediated digestion of the siRNA parts, leading to more efficient disruption of the target gene in cells and animal models, compared with the monomeric sgRNA-Cas9 RNP complex.
