        우리나라 일부 석면사업장의 석면폐 유병률

        Asbestos industry has been in operation over 60 years in Korea. However, the prevalence of asbestosis has not been yet reported. With rapid turn-over of workers, previous cross-sectional studies of current workers on the job could not find cases with exposures long enough for the development of asbestosis. This study was done to evaluate asbestosis prevalence of those worksites with operation history of more than 20 years. In total, 139 workers from 5 worksites were examined. Asbestos industries covered in the study include 2 asbestos textile, 1 brake lining, and 2 ship repairing worksites. Chest x-ray was taken from all workers and read by two experts familiar with pneumoconiosis classification according to 1980 ILO guidelines. Those with findings compatible with asbestosis were further ckecked with high resolution computerized tomography (HRCT). Pulmonary function tests were done according to ATS guidelines, and occupational and previous medical history was taken through a standardized interview. Air-borne asbestos was measured according to NIOSH method 7400. The air-borne asbestos concentrations ranged from 0.2-1.3 f/cc for asbestos textile, from 0.7-1.0 f/cc for brake lining, and from 6.3-7.8 f/cc during asbestos removal at ship repairing worksite. Of the 139 workers 25 had abnormal chest radiographic findings, and 10 of them had findings compatible with pneumoconiosis. When work history and current asbestos measurements were accounted, 9 workers who had more than 10 years of asbestos exposure history showed chest radiographic findings of pneumoconiosis with Finally, 4 workers showed finding of pulmonary fibrosis and/or pleural thickening at HRCT, and 2 of them had restrictive lung function changes. The study results showed that, among 139 subjects, there were 4(3%) definite asbestosis cases confirmed with HRCT. The prevalence of probable asbestosis was 7% for 10-14 years of exposure, 13% for 15-19 years of exposure, and 23% for 20 or more years of exposure. The prevalence of compensable asbestosis with abnormal lung function was 4-6% for those with 15 or more years of exposure.

        Adapative control theory or parameter tuning of PID controller is mainly limited in linear systems. But in this paper, using a nonlinear mapping capability of NNS, we derive a realtime tuning method of PID controller based on a Back-Propagation method of single GMDP NN. A variety of neural models, especially higher-order networks, are known to computationlly powerful for complex application. While they have advantages over traditional multilayered perceptrons, the uniformity in their network structures, learning algorithms and loop time creates practical problems. Thus this paper evaluates the capability between MLP and GMDP for performing velocity control. Backpropagation is applied to the GMDP as learning rule.

      • Digital Microfluidic Approach for Efficient Electroporation with High Productivity: Transgene Expression of Microalgae without Cell Wall Removal

        <P>A unique digital microfluidic electroporation (EP) system successfully demonstrates higher transgene expression than that of conventional techniques, in addition to reliable productivity and feasible integrated processes. By systematic investigations into the effects of the droplet EP conditions for a wild-type microalgae, 1 order of magnitude higher transgene expression is accomplished without cell wall removal over the conventional bulk EP system. In addition, the newly proposed droplet EP method by a droplet contact charging phenomena shows a great potential for the integration of EP processes and on-chip cell culture providing easy controllability of each process.       • A rare case of abnormal vaginal bleeding caused by vulva venous hemangioma: a case report

        Haemangioma is a benign formation originating from vascular tissue which usually occurs in childhood. A hemangioma can occur anywhere on the body but, vulva venous hemangiomas are extremely rare in nulliparous women. A 25-year-old woman with abnormal vaginal bleeding with genital ulcerative lesion for six months presented to the hospital. Vaginal examination revealed a 1.5X1 cm papillary mass with ulcerated surface on the vaginal fourchette. Speculum examination revealed a cervix with an apparently normal epithelium and external os with no pathological discharge. The mass was visually suspected to be a precancerous lesion or malignancy. The mass was successfully removed by complete excision and biopsy showed ulcerative lesion with chronic active inflammation and granulation tissue and compatible with squamous papilloma. One month later, the same lesion with bleeding was growing on vaginal fourchette and, excision was performed. The pathologic finding was vulva hemangioma. The review of primary vulva lesion was diagnosed with hemangioma and, the second lesion was recurred hemangioma. Vulva hemangiomas with ulcerative lesion are very rare and only small number of cases are reported in literature. It is necessary to carry out differential diagnosis of premalignant or malignant lesions.

      • Promotion of osteoblastic differentiation and osteogenic transcription factor expression on a microgroove titanium surface with immobilized fibronectin or bone sialoprotein II

        <P>We demonstrate that a composite surface of microgroove titanium (Ti) with immobilized fibronectin (FN) or bone sialoprotein II (BSP2) promotes osteoblastic differentiation and osteogenic transcription factor expression in human bone marrow-derived mesenchymal stem cells (MSCs). Comparisons made between smooth microgrooves, microgrooves with silanization and microgrooves with matrix protein (FN or BSP2)-immobilization Ti surfaces revealed a significant promotion of in vitro osteogenic activity and osteoblastic differentiation at various timelines of culture. An even more significant increase was verified on microgrooves with a matrix protein-immobilization Ti surface in 28 d time-dependent gene expression of the main osteogenic transcription factors, such as ARF4, FRA1, RUNX2, and OSX. As a result, a synergestic effect regarding the promotion of osteogenic transcription factor expression and osteoblastic differentiation in the matrix protein-microgroove Ti composite surface was confirmed. From a multiple regression analysis using various timelines of osteogenic culture as independent variables, day 13 was verified as the most prominent influential timeline for the promotion of osteoblastic differentiation induced by the matrix protein-microgroove Ti composite surface. The FN-or BSP2-microgroove Ti composite surface resulting from silanization can strongly induce the promotion of osteoblastic differentiation in human MSCs. The proposed surface is expected to be useful in the development of a variety of osteogenic biomaterial surfaces.</P>

      • Digital Electrophoresis of Charged Droplets

        <P>A digital microfluidic system based on a direct electric charging and subsequent electrophoretic manipulation of droplets is made by simple fabrication at low cost. Digitally controlled two-dimensional droplet motions are realized by digital polarity control of an array of electrodes. By independent control of droplets and colorimetric detection, the coalescence and mixing of droplets is analyzed quantitatively. The gelation of sodium alginate and the crystallization of calcium carbonate by multiple droplet translations and coalescence and the actuation of glassy carbon beads are demonstrated to show the versatile manipulation capability of the proposed technology.       • Transfection of Jurkat T cells by droplet electroporation

        <P><B>Abstract</B></P> <P>A droplet electroporation system has been successfully demonstrated for transfection of Jurkat T cell with much higher transfection efficiency (66%) than that of conventional system (11.4%) with the advantages of superior cell viability, comparable throughput, and user-friendly interface. Because the productivity of the proposed system is much greater than previous microfluidic systems, flow cytometry is used for the analysis of transfection efficiency. The high transfection efficiency and cell viability of the droplet electroporation system is mainly attributed to small size which requires lower voltage and provides concentrated environment. The successful transfection of Jurkat cell using the droplet electroporation system broadens the applicability of the proposed technology to medical field. The implication of the present work and the future development direction for a fully automated cell engineering platform are discussed.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Our previously developed electroporation system has been applied to transfection of Jurkat T cell. </LI> <LI> Effects of voltage and time on the transfection were investigated. </LI> <LI> Higher transfection efficiency than that of conventional system was accomplished. </LI> </UL> </P>

        디지털 미세유체를 이용한 미세녹조류 형질전환에서의 세포벽의 영향 분석

        디지털 미세유체 전기천공 시스템을 활용하여 미세녹조류에 대한 형질전환 실험을 통해 기존 상용화 장치 대비 높은 유전자 전달 효율과 세포 생존도를 확인하였다. 전기천공의 주요 파라미터인 인가전압 및 인가시간 변화를 주며 세포벽이 존재하는 세포종과 세포벽이 없는 세포종에 대한 비교 실험을 수행하였다. 이를 통해 식물 세포의 전기천공에서 세포벽의 역할은 단순히 유전체의 전달을 방해하는 부정적 요소로만 작용하지 않는 다는 사실을 확인하였다. 병렬화 및 온 칩 세포 배양 등을 통해 제안된 디지털 전기천공 기술이 향후 새로운 청정 형질전환 방법으로써의 가능성도 확인하였다. Digital microfluidic electroporation system was used for the transformation of microalgae and we have obtained higher transformation efficiency and viability than that of conventional method. Key parameters of electroporation such as pulse voltage, number, and duration time were systematically investigated for two different microalgal strains with and without cell wall. We have found that cell wall does not always have negative effects on the gene transformation of microalgae. Parallel processing of proposed digital microfluidic electroporation was demonstrated together with on chip culture of microalgae.
