        돼지 H-FABP 유전자의 다형성 및 경제 형질과의 연관성 구명

        The purpose of this study was to detect association between genetic variation and economic trait in the porcine heart type fatty acid-binding protein gene as a candidate gene for the traits related with growth and meat quality in pigs. The H-FABP is a 15-kDa protein expressed in several tissues with high demand for fat metabolism such as cardiac and skeletal muscle and lactating mammary gland. H-FABP is small intracellular protein involved in fatty acid transport from the plasma membrane to the site of β-oxidation and/or triacylglycerol or phospholipid synthesis. In this study, H-FABP PCR-RFLP was performed in F_(2) population composed of 214 individuals form an intercross between Korean Native Boars and Landrace sows. PCR products form tow primer sets within H-FABP gene were amplified in 850bp and 700bp. Digestion of PCR products with the restriction digestion enzymes HaeⅢ and Hinf Ⅰ, revealed fragment length polymorphisms(RFL. Ps). The genotype frequencies from H-FABP/HaeⅢ was .29 for genotype DD, .53 for genotype Dd, and .15 for genotype dd, respectively. The genotype frequencies of HH, Hh, and hh from H-FABP(hinf Ⅰ was .38, .41, and .20, respectively, in the population.Relationships between their genotypes and economic traits were estimated. In H-FABP/HaeⅢ locus, there were specific genotypes(Dd and dd) associated with economic traits such as body weight. In H-FABP/Hinf Ⅰ Iocus, Genotypes of HH and Hh associated with growth traits such as body weights at 5, 12, and 30 week of age (p<.05 or p<.001) and back fat thickness, body fat including abdominal and trimmed fat (p<.001) and intramuscular fat(p<.05). The 'H'allele was positivecly associated with gaining of body weight and fatness deposition. In conclusion, a significant association of the H-FABP gene from its genetic variation was found on body weight, intramuscular fat and backfat thickness.

      • Expression and N-glycan analysis of human 90K glycoprotein in Drosophila S2 cells

        Human 90K (h90K; Mac-2-binding protein) glycoprotein is a potential pharmaceutical due to its inhibitory activity against cancer metastasis and expansion. Here, h90K glycoprotein was produced in insect Drosophila S2 cell system, and its N-glycan pattern was analyzed. A plasmid encoding h90K gene, fused with a hexahistidine tag under the control of Drosophila metallotionein promoter, was stably transfected into S2 cells. After copper sulfate induction, transfected S2 cells secreted recombinant h90K at a good expression level of 28mg/L in a 150-mL spinner flask culture. The purified recombinant h90K showed an apparent molecular weight of ~78kDa which was much smaller than that (~97kDa) of the natural h90K. Because de-N-glycosylated h90K appeared at ~60kDa protein band, it was suggested that the recombinant h90K from S2 cells has small N-glycans with about half the molecular weight (~18kDa) of N-glycans of the natural h90K. Through detail analyses using high-performance liquid chromatography and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry, the S2-derived recombinant h90K was confirmed that it has simple paucimannosidic structures containing two or three mannose residues with core fucose as the major (~79%) N-glycans.

      • 레이저 쇼크 피닝에 의한 H13/Cu 경사기능재료의 잔류응력 평가

        경사기능재료(Functionally Graded Materials)는 소재의 물성이 점진적으로 변화하는 것이 특징으로, 각 소재의 장점을 극대화 하는 것이 가능하여 많은 공학 분야에 적용될 수 있다. 본 연구에서는 DMT (Laser-aided Direct Metal Tooling) 공정을 통해 열전도성 사출 금형의 형상적응형 냉각회로 등에 적용 가능한 H13/Cu FGM 시편을 제작하였다. 레이저 기반 금속 프린팅으로 소재를 적층할 경우, 발생되는 열에 의해 제작된 시편의 균열 등을 유발하는 인장 잔류응력의 형성이 필연적이다. 따라서 본 연구에서는 적층 시편의 표면에 형성되는 인장잔류응력을 해소할 수 있는 방안으로 LSP (Laser Shock Peening) 공정 연구를 수행하였다. H13, Cu 소재의 무게 분율 20 wt%에 의해 6 종의 H13/Cu FGM 시편을 먼저 적층 제작하고, LSP 공정 전후의 시편에서 XRD 회절 패턴 분석(2θ 각도 변화 분석)을 통해 잔류응력을 측정하였다. 실험결과 LSP 공정 후 소재의 표면에 생성되는 압축잔류응력은 소재의 미세조직을 변형시키며, DMT 공정으로 생성된 인장잔류응력을 효과적으로 상쇄 시키는 것으로 판단된다.

      • 돼지 적출 신동맥에 있어서 Histamine receptor 의 특성

        To elucidate the physiological characteristics of histamine receptor of renal artery in pig. The effect of histamine and the effect of H₁-, A₂-receptor antagonist, Ha-agonist on the contractile response of histamine to isolated regal artery were investigated. The results were summarized as follows: 1. histamine caused the contraction of the pig isolated renal artery and the contractile responses increased between the concentration of histamine 10^(-6) and 10^(-3) M with a dose-dependent manner. 2. The contractile response induced by histamine was completely inhibited by the post-treatment with H₁-receptor antagonist, pyrilamine. 3. The contractile response induced by histamine was increased by the pre-treatment with H₂-receptor antagonist, cimetidine, and was powerfully inhibited by the pre-treatment with H₂-receptor agonist, impromidine. Our results were consistent with the hypothesis that histamine H₁-receptor induced contractile response was presented in the pig renal artery. However, indications for the additional presence of H₂-receptor induced relaxation responses were weak.

      • Organic-inorganic hybrid photocatalyst for carbon dioxide reduction

        <P>Efficient hybrid photocatalysts for carbon dioxide reduction were developed from dye-sensitized TiO2 nanoparticles and their catalytic performance was optimized by ternary organic/inorganic components. Thus, the hybrid system consists of (E)-2-cyano-3-(5'(5 ''-(p-(diphenylamino)phenyl)thiophen-2 ''-yl)thiophen-2'-yl)-acrylic acid as a sensitizer and fac-[Re(4,4'-bis(diethoxyphosphorylmethyl)-2,2'-bipyridine)(CO)(3)Cl] as a reduction catalyst (ReP), both of which have been fixed onto TiO2 semiconductors (s-TiO2, h-TiO2, d-TiO2). Mott-Schottky analysis on flat-band potential (E-fb) of TiO2 mesoporous films has verified that Efb can be finely modulated by volume variation of water (0 to 20 vol%). The increase of added water resulted in substantial positive shifts of Efb from -1.93 V at 0 vol% H2O, to -1.74 V (3 vol% H2O), to -1.56 V (10 vol% H2O), and to -1.47 V (20 vol% H2O). As a result, with addition of 3-10 vol% water in the photocatalytic reaction, conversion efficiency of CO2 to CO increased significantly reaching a TON value of similar to 350 for 30 h. Catalytic activity enhancement is mainly attributed to (1) the optimum alignment of Efb by 3-10 vol% water with respect to the E-ox* of the dye and E-red of ReP for smooth electron transfer from photo-excited dye to ReP via the TiO2 semiconductor and (2) the water-induced acceleration of chemical processes on the fixed ReP. In addition, the energy level was further tuned by variation of the dye and ReP amounts. We also found that the intrinsic properties of TiO2 sources (morphology, size, agglomeration) exert a great influence on the overall photocatalytic activity of this hybrid system. Implications of the present observations and reaction mechanisms are discussed in detail.</P>

      • 대학생 커플 간의 성 의사소통 양상에 따른 성 태도, 성적 자율성 및 성 만족도의 상관관계에 관한 연구

        Purpose: The purpose of this study was to investigate sexual communication among college student couples and to analyze the relationship among sexual communication, sexual attitude, sexual autonomy and sexual satisfaction. Methods: Using a descriptive correlation study design, 281 college student couples were recruited through convenience random sampling from September 18th to October 3rd, 2014. Data were analyzed using average, standard deviation, percentage, t-test, ANOVA, Scheffe test, Duncan’s test and Pearson’s correlation using the SPSS 21.0 program. Results: In both genders, it is identified that high scores for sexual communication is related to sexual openness (male: R= .347, p<0.01 / female: R=.337, p<0.01) higher sexual satisfaction (male: R= .349, p<0.01 / female: R=.336, p<0.01) and sexual autonomy (male: R= .336, p<0.01 / female: R=.364, p<0.01). The same outcome is found when each male and female are grouped as a couple. Among sub-categories of sexual attitude, sexual pleasure was related to sexual communication the most in male (R=.337, p<0.01), female groups (R=.317, p<0.01) and couples (R=.402, p<0.01). Among sub-categories of sexual satisfaction, psychological factor was related to sexual communication the most in the male group (R=.342, p<0.01) and interactive factor was related to sexual communication the most in the female group (R=.317, p<0.01) and couples (R=.402, p<0.01). Conclusion: The significance of this research is that the relations of sexual communication with the other three dependent variables among each gender and couples are shown. In addition, this study suggests the importance of sexual communication between couples which will lead to healthier and better quality of sexual relationships. However, with the lack of sufficient advanced research, there are limitations of analyzing each variables divided by demographic characteristics.

        돼지 염색체 6번의 연관지도 및 양적형질 유전자좌위 탐색

        The objective of this study was to identify the quantitative traits loci(QTL) for economically important traits such as growth, carcass and meat quality on pig chromosome 6. A three generation resource population was constructed from cross between Korean native boars and Landrace sows. A total of 240 F_(2) animals were produced using intercross between 10 boars and 31 sows of F_(1) animals. Phenotypic data including body weight at 3 weeks, backfat thickness, muscle ?, shear force and crude protein level were collected from F_(2) animals. Animals including grandparents(F_(0)), parents(F_(1)) and offspring(F_(2) were genotyped for 29 microsatellite markers and PCR-RFLP marker on chromosome 6. The linkage analysis was performed using CRI-MAP software version 2.4(Green et al., 1990) with FIXED option to obtain the map distances. The total length of SSC6 linkage map estimated in this study was 169.3cM. The average distance between adjacent markers was 6.05cM. For mapping of QTL, we used F_(2) QTL. Analysis Servelt of QTL express, a web-based QTL mapping tool(http://qtl.cap.ed.ac.uk). Five QTLs were detected at 5% chromosome-wide level for body weight of 3 weeks of age, shear force meat ? at 24 hours after slaughtering, backfat thickness and crude protein level on SSC6.

      • 교차 배열된 지르코늄관 간격 측정을 위한 전압 평면에서의 신호해석

        The eddy current(EC) signals were evaluated for gap measurement of cross aligned zirconium tubes for preventing the contact which might be the safety concern. Liquid lnjection Nozzle(LIN) tube and Calandria tube(CT) were cross-aligned horizontally and LIN could be detected by inserting the EC probe into pressure tube (PT). Signal analysis on the voltage plane was applied to describe the position and shape of possible defects using several frequencies. Test variables are lift-off, PT thickness variation, and gap variation between PT and CT All the EC measurements in the laboratory were done in variance with the gap spacings and showed the possibility to evaluate the gap between CT. and LIN tube by analyzing the signal on voltage plane. Electromagnetic technique could be a potential tool for inspecting the inter-tube gap in CANDU reactor.
