연구목적 : 세포의 면역 기능 조절에 관여하는 CTLA-4의 유전자 다형성을 분석하여 주요 우울장애와의 면역 유전학적 관련성을 알아보고자 하였다. 방 법 : DSM-Ⅳ진단 기준에 따라 주요 우울장애로 진단된 환자 77명을 선정하였으며 가톨릭대학교 의과대학 조혈모세포은행에서 보유하고 있는 149명의 정상 한국인 자료를 대조군으로 사용하였다. 전혈에서 DNA를 추출하였으며 중합효소반응으로 CTLA-4유전자 exon 1 부위를 증폭한 후 SSCP방법으로 유전?? 형별 분석하였다. 결 과 : 대조군과 주요 우울장애 환자군 사이의 CTLA-4 유전자형과 대립유전자의 발현 빈도를 비교시 유의한 차이가 없었다. 결 론 : 본 연구 결과 exon 1 CTLA-4 유전자형 및 대립유전자의 발현빈도는 주요 우울장애 환자군과 정상 대조군 사이에 차이가 없어, exon 1 영역은 후보 유전자로서의 가능성이 없었다. 향후 연구에서는 다양한 변인들에 따라 대상군을 임상적으로 동질성을 지닌 집단으로 세분하여 조사대상 유전자와의 관련성에 대한 연구가 이루어져야 하겠다. Objectives : This study was carried out to explore the relationship between major depressive disorder and CTLA-4 which is related to the immunologic function such as T cell regulation. Methods : Among the korean patients diagnosed as major depressive disorder according to DSM-Ⅳ, 77 patients without neurological illness, hormonal disorder, or comorbid mental illness were selected. The stored data of 149 normal Koreans from the Catholic Hemopoietic Stem Cell Bank of Korea, were used as a normal control group. The data of Korean control group were compared with those of the studies of different ethnics. DNA was extracted from whole blood and the exon I region of CTLA-4 gene was amplified by polymerase chain reaction. Gene typing was performed by using SSCP and then, the results were assessed. Results : There were no significant differences in genotype frquencies of CTLA-4*G/G, CTLA-4*G/A, and CTLA-4*A/A between the patients with major depressive disorder and the control group in Korean population(48.1% vs 46.3%, 41.6% vs 39.6%, 10.3% vs 14.1%, respectively).There were no significant differences in allelic frequencies of CTLA-4*G and CTLA-4*A between the patients with major depressive disorder and the control group in Korean population(68.8% vs 66.1%, 31.2% vs 33.9%, respectively). Conclusion : Considering negative result for the association of the exon I polymorphism of CTLA-4 gene with major depressive disorder in this study, the exon I polymorphism does not appear to be possible candidate gene for major depressive disorder. Moreover, further systematic researches including diverse clinical variables would required.
A Study on the Mixture Formation Process of Evaporating Diesel Spray by Offset Incidence Laser Beam

This paper analyzes heterogeneous distribution of branch-like structure at the downstream region of the spray. The liquid and vapor phase of the spray are obtained using a 35mm still camera and CCD camera in order to investigate spray structure of evaporating diesel spray. There have been many studies conducted on diesel spray structure but have yet only focused on the analyses of 2-D structure. There are a few information which is concerned with 3-D structure analysis of evaporating spray. The heterogeneous distribution of droplets in inner spray affects the mixture formation of diesel spray and the combustion characteristics of the diesel engines. In this study, the laser beam of 2-D plane was used in order to investigate 3-D structure of evaporating spray. The incident laser beam was offset on the central axis of the spray. From the analysis of images taken by offset laser beam, we will examine the formation mechanism of heterogeneous distribution of the diesel spray by vortex flow at the downstream of the spray. The images of liquid and vapor phase of free spray are simultaneously taken through an exciplex fluorescence method. Through this, the branch-like structure consisting of heterogeneous distribution of the droplets forms high concentrated vapor phase at the periphery of droplets and at the spray tip.

The porcine reproductive and respiratory syndrome virus (PRRSV) has six structural proteins which encoded by ORFs 2 to 7 are designated as GP2, 3, 4, 5, M and N, repectively. In this study, we determined the expression of each protein using novel transfer vector, pBmKSK4 which has the polyhedrin promoter of BmNPV and 6xHis tag. The recombinant transfer vector was co-transfected into Bm5 cells along with bBpGOZA DNA. Recombinant virus was purified by plaque assay and amplified in Bm5 cells. Expression of each protein was identified by SDS-PAGE and Western blot analysis using anti-6xHis monoclonal antibody. The expression levels of the structural proteins in Bm5 cells were stronger than the expression system using pBacPAK9 transfer vector in Sf21 cells. As expected, GP5 was expressed at low levels from its structural properties and its toxicity for cells. In addition, each recombinant protein was purified using Ni-NTA spin columns. The ability to produce each protein in the baculovirus system indicates that these could be major candidates for the development of a vaccine against PRRSV.

We investigated the effects of fermentation filtrates from Rubus coreanus on the function of the male reproductive system. We performed an ex vivo study to determine if the candidate compounds relax isolated New Zealand white rabbit corpus cavernosum, which were precontracted by phenylephrine (5 × 105 M). The results reveal that the filtrates of the reddish-purple (FRRC) and green (FGRC) R. coreanus exerted concentration-dependent relaxing effects, leading to median effective concentrations of 4.53 mg/mL and >10 mg/mL, respectively. For the in vivo study, male ICR mice were orally administered FRRC or FGRC (100 or 500 mg/kg) for 28 days, and the reproductive organ weights, serum testosterone level, cauda epididymal sperm counts, and motility were analyzed. Both the FRRC and FGRC had no significant effect on the reproductive organ weights; however, FRRC (100 or 500 mg/kg) enhanced testosterone levels and especially sperm counts at the higher dose (500 mg/kg). In comparison, FGRC increased hormone levels and sperm counts at a relatively low dose (100 mg/kg). In summary, it is proposed that the crude fermentation filtrates of ripe R. coreanus have positive effects on the function of the male reproductive system by triggering a penile erection, enhancing serum testosterone levels, and increasing epididymal sperm counts.

<P><B>Background</B></P><P>Improvements in mobile telecommunication technologies have enabled clinicians to collect patient-reported outcome (PRO) data more frequently, but there is as yet limited evidence regarding the frequency with which PRO data can be collected via smartphone applications (apps) in breast cancer patients receiving chemotherapy.</P><P><B>Objective</B></P><P>The primary objective of this study was to determine the feasibility of an app for sleep disturbance-related data collection from breast cancer patients receiving chemotherapy. A secondary objective was to identify the variables associated with better compliance in order to identify the optimal subgroups to include in future studies of smartphone-based interventions.</P><P><B>Methods</B></P><P>Between March 2013 and July 2013, patients who planned to receive neoadjuvant chemotherapy for breast cancer at Asan Medical Center who had access to a smartphone app were enrolled just before the start of their chemotherapy and asked to self-report their sleep patterns, anxiety severity, and mood status via a smartphone app on a daily basis during the 90-day study period. Push notifications were sent to participants daily at 9 am and 7 pm. Data regarding the patients’ demographics, interval from enrollment to first self-report, baseline Beck’s Depression Inventory (BDI) score, and health-related quality of life score (as assessed using the EuroQol Five Dimensional [EQ5D-3L] questionnaire) were collected to ascertain the factors associated with compliance with the self-reporting process.</P><P><B>Results</B></P><P>A total of 30 participants (mean age 45 years, SD 6; range 35-65 years) were analyzed in this study. In total, 2700 daily push notifications were sent to these 30 participants over the 90-day study period via their smartphones, resulting in the collection of 1215 self-reporting sleep-disturbance data items (overall compliance rate=45.0%, 1215/2700). The median value of individual patient-level reporting rates was 41.1% (range 6.7-95.6%). The longitudinal day-level compliance curve fell to 50.0% at day 34 and reached a nadir of 13.3% at day 90. The cumulative longitudinal compliance curve exhibited a steady decrease by about 50% at day 70 and continued to fall to 45% on day 90. Women without any form of employment exhibited the higher compliance rate. There was no association between any of the other patient characteristics (ie, demographics, and BDI and EQ5D-3L scores) and compliance. The mean individual patient-level reporting rate was higher for the subgroup with a 1-day lag time, defined as starting to self-report on the day immediately after enrollment, than for those with a lag of 2 or more days (51.6%, SD 24.0 and 29.6%, SD 25.3, respectively; <I>P</I>=.03).</P><P><B>Conclusions</B></P><P>The 90-day longitudinal collection of daily self-reporting sleep-disturbance data via a smartphone app was found to be feasible. Further research should focus on how to sustain compliance with this self-reporting for a longer time and select subpopulations with higher rates of compliance for mobile health care.</P>
Neuraminidase Inhibitors from the Fruiting Body of Phellinus igniarius

During our ongoing investigation of neuraminidase inhibitors from medicinal fungi, we found that the fruiting bodies of Phellinus igniarius exhibited significant inhibitory activity against neuraminidase from recombinant H3N2 influenza viruses. Two active compounds were isolated from the methanolic extract of P. igniarius through solvent partitioning and Sephadex LH-20 column chromatography. The active compounds were identified as phelligridins E and G on proton nuclear magnetic resonance (1H NMR) and electrospray ionization mass measurements. These compounds inhibited neuraminidases from recombinant rvH1N1, H3N2, and H5N1 influenza viruses, with IC50 values in the range of 0.7~8.1 μM.

<P>The single-bath electrochemical deposition of CuInSe2 often leads to short-circuit behavior of the resulting solar cells due to,lhe high shunt conductance. In this, study, in amattemp't to resolve this problem, the influence of the Se precursor concentration (C-se) on electrodqpOsited CuInSe2 films and solar cell devices is examined in the C-Se range of 4.8 to 12:0 mM in selenite-based aqueous solutions ':containing Cu and In chlorides along with sulfarnic acid (H3NSO3) and potassium hydrogen phthalate (C8H5KO4) additives. As C-se increases,, the CuInSe2 layers become porous; and the grain, growth of the CuInSe2 phase is restricted,-while the parasitic shunting problem wag-markedly alleviated, as unambiguously demonstrated by measurements of the local current distribution. Due to these ambivalent influences, an optimal value of cse that achieves the best quality of the films for high-efficiency solar cells is identified. Thus, the.device prepared with 5.2 mM C-Se exhibits a power-conversion efficiency exceeding 10% with greatly improved device parameters, such as the shunt conductance and the reverse saturation current. The rationale of the present approach along-with the physicochemical,origin of its conspicuous impact on the resulting devices is discusSed in-conjunction with the electro-crystallization mechanism-of the CuInSe2 compound.</P>

Infitration of the kidneys in acute 1ymphocytic leukemia is thought to cause rarely functional imparment or lead to renal failure. When renal failure occurs in acute lymphocytic leukemia(ALL), it is secondary to complications such as tumor lysis syndrome, uric acid nephropathy, obstruction, haemolysis and glomerulone-phritis. We report a 56 year-old female patient with ALL who presented with renal failure and was found to have extensive infitration of the kidney with leukemic infiltrates. The absence of other causes of renal failure and the presence of large kidneys with extensive infiltration of the interstitum with tumor cells makes it highly likely that renal failure was caused by leukemic infiltration.
