마우스 섬유아세포(3T3 fibroblast cells)에서 Insulin-like Growth Factor-I(IGF-I) 및 IGF Binding Protein-3 (IGFBP-3)이 세포증식에 미치는 영향

연구배경: 세포성장은 세포증식과 세포죽음의 균형에 의해 이루어 진다. 세포성장에 관여하는 여러 growth factor증 IGF-I은 IGF-IR와 결합하여 세포증식을 유발하는 mitogen으로 알려져 있다. 또한 IGF-I에 결합하는 IGFBPs중에 IGFBP-3는 혈액내에 가장 많은 carrier protein으로, IGF-I과 결합하여 IGF-I의 세포증식 효과를 증가 혹은 억제시킨다. 방 법: 3T3 fibroblast 세포를 이용하여 IGF-I과 IGF-IR transcripts를 northern blot으로 확인하고, IGF-I에 의한 mitogenic effect를 MTT assay 및 $^3H$-thymidine incorporation test로 관찰하고, IGF-I의 receptor인 IGF-IR의 활성화를 보기 위해 intracellular $\beta$-subunit의 tyrosine kinase domain의 phosphorylation을 western blot으로 관찰하였다. 또한 IGFBP-3가 3T3 세포에서 mitogenic effect에 미치는 영향을 보기 위해 anti-IGFBP-3와 ${\alpha}IR_3$을 단독 및 병용투여하여 관찰하였다. 결 과: 3T3 세포는 IGF-I와 IGF-IR의 mRNA expression을 보였으며, IGF-I을 투여시 IGF-IR의 intracelluar cytoplasmic protein인 $\beta$-subunit의 tyrosine kinase domain을 phosphorylation시켜 활성화시키며, 5%, 1% serum-containing media에서 세포증식을 보였으나, serum-free media에서는 세포증식을 보이지 않았다. 또한 anti-IGFBP-3 투여와 ${\alpha}IR_3$과 anti-IGFBP-3를 병용투여시는 세포종식이 각각 2배이상 증가하였으나, ${\alpha}IR_3$을 4시간 전처치후 ${\alpha}IR_3$과 anti-IGFBP-3를 병용투여시는 anti-IGFBP-3에 의한 세포종식이 보이지 않은 것으로 보아 IGF -I/IGFBP-3 결합에서 분리되는 free IGF-I어l 의해 세포증식이 유도된 것으로 생각된다. 결 론: IGF-I은 IGF-IR를 phosphorylation시켜 mitogenic effect를 보이며, IGFBP-3는 IGF-I의 mitogenic effect를 억제하는 것으로 생각된다. Background: Cell growth is a balance between cell proliferation and cell death. Insulin-like growth factor-I(IGF-I), which binds IGF-I receptor(IGF-IR), mediates cellular proliferation as a potent mitogen. IGF binding protein-3(IGFBP-3) as a circulating major IGFBP can inhibit or enhance the effects of IGF-I on cellular growth by binding IGFs. Methods: We investigated the expressions of mRNA of IGF-I and IGF-IR by northern blot and phosphorylation of IGF-IR with the treatment of IGF-I by western blot in 3T3 fibroblast cells. The cellular proliferations of 3T3 cells with the treatments of IGF-I were evaluated using $^3H$-thymidine incorporation and MTT assay. Also to observe the effect of IGFBP-3 on cellular proliferation, 3T3 cells were treated with anti-IGFBP-3 and ${\alpha}IR_3$(monoclonal antibody to IGF-IR) alone or in combination. Results: Our results demonstrated that 3T3 cells showed mRNA expressions of IGF-I and IGF-IR and the IGF-I increased phosphorylation of IGF-IR. The treatments of 3T3 cells with IGF-I increased cellular proliferation in 5 % and 1 % seruma-containing media, not in serum-free media. The addition of anti-IGFBP-3 to neutralize IGFBP-3 showed 2-fold increase of cellular proliferation, and also co-incubation of anti-IGFBP-3 and ${\alpha}IR_3$ together showed similar increase of cellular proliferation in 3T3 cells. Interestingly, when the cells were pretreated with ${\alpha}IR_3$ for 4 hr, prior to the simultaneous addition of ${\alpha}IR_3$ and anti-IGFBP-3, anti-IGFBP-3-mediated cellular proliferation was decreased to control level. All of these results suggest that free IGF-I released from IGF-I/IGFBP-3 complex would be involved in the cellular proliferation. Conclusion: IGF-I is a mitogen through the activation of IGF-IR in 3T3 cells, and IGFBP-3 could be a potent inhibitor for IGF-I action by binding IGF-I.

Profile Index에 따른 Panelipse Ⅱ 방사선 상층의 변화

This research was made to investigate the change of the image layer and of the vertical and horizontal magnification on the Panelipse radiographic image by the control of profile index. Using the Panelipse, a series of 60 exposures were taken with the steel balls placed in the holes of the plastic model plate, and then evaluated by 4 observers. Two points were assigned for a reading of sharp, one for middling sharp, and zero for unsharp. Each ball image then could be given a total reader score of 0 to 8. The author analyzed the image layer as defined by a sharpness score of 6 or more. The results obtained were as follows: As the profile index was increased, the shape of the image layer was not changed, and the width of the image layer was increased, and the position of the layer shifted away from the rotation center. As the profile index was increased, the ranges of vertical and horizontal magnification was increased, especially the ranges of horizontal magnification was greater than that of vertical magnification. 저자는 Panelipse Ⅱ의 상층 및 상의 확대변화를 관찰하기 위하여 모형판에 금속구를 위치시켜 3가지 Profile Index에서 각기 20장씩 방사선사진을 촬영하여 관찰한 결과를 다음과 같이 요약할 수 있었다. Profile Index의 증가에 따른 Panelipse Ⅱ 방사선상의 상층에 있어 모양의 변화는 없었고, 폭은 더 넓어졌으며, 회전축중심에서 외측으로 상층은 이동하였다. 방사선상에서 수평 수직확대는 공히 Profile Index가 증가함에 따라 그 범위가 커졌으며 이는 수평확대에 있어 수직확대보다 현저하였다.

인천 숭의 평화 시장 재생을 위한 숭의 창작 공간 설계 제안

숭의 평화 시장은 1971년 문을 연 남구의 대표적인 재래시장으로 이용되었으나 대형 마트의 입점과 시설이 낙후되고 상인들의 고령화 등의 이유로 점차 활기를 잃기 시작했다. 본 논문은 건축적, 사회적 이유로 점차 재래시장의 모습이 사라지고 있는 숭의 평화 시장의 재생방안을 제안한다. 이전에 창작 공간으로 재기를 꿈꿨으나 실패한 모습을 보고 건축적 문제가 크다고 분석한 후 창작 공간에 맞는 설계를 계획하는 것을 목적으로 하며 그 결과로 숭의 평화 시장뿐만 아니라 주변 지역의 활성화가 될 수 있도록 기대한다. Sungi-Conventional Market was used as a representative conventional market in Nam-gu, Incheon. Gradually, It sarted to lose vigor neighbor of hypermarketw and facilities are outdated and traders of aging. This study is suggest a way of playing the market that disappearing figure of traditional market. Previous dreamed of a resurgence in the creative space. After a review at the sight failed architectural problem analysis aims to design a plan for the creative space. As a result, we expect to be the activation of the market as well as the surrounding area sungi-conventional market.

본 연구는 응급의료서비스품질의 발견과 환자가치, 환자만족, 재이용의도에 영향을 미치는 인과적 관계를 파악하는데 있다. 이를 토대로 차후에 병원의 응급의료진과 행정관계자에게 응급의료서비스의 중요성에 대한 인식을 제고시키고, 응급의료서비스품질의 향상 및 발전방안을 제공하기 위한 의미있는 자료를 제공하고자 한다. 본 연구의 이론적인 시사점에 대한 논의는 다음과 같다. 첫째, 본 연구의 실증적 분석결과, 종합병원의 응급의료서비스품질은 응급의료프로세스, 의료진 태도, 사후의료프로세스, 의료시설 및 환경, 병원이미지, 의료진 역량의 여섯 개 차원으로 구성됨을 확인할 수 있었다. 총 효과 측면에서 이들 요인들이 환자만족과 재이용의도에 미치는 영향을 살펴본 결과, 6개 차원 중 병원이미지, 의료시설 및 환경, 의료진 태도가 환자만족과 재이용의도에 유의한 영향을 미치고 있는 것으로 나타났다. 본 연구는 몇 가지 한계점을 가지며 향후 추가적인 연구를 필요로 한다. This research was designed to investigate causal relationship among emergency medical service, patient value, patient satisfaction, reuse intention. On the basis of research result, we intended to offer significant information that can improve and develop emergency medical service quality. Theoretical implications of this research are as follows. First, as a result of empirical analysis, we found that emergency medical service qualities of general hospital could be six components such as emergency medical process, attitude of medical staff, follow-up medical process, medical facilities and environment, image of hospital, and ability of medical staff. In the view point of total effects, analysis result shows that image of hospital, medical facilities, and attitude of medical staff are positively related to patient satisfaction and reuse intention. Result of research has some limitation to be solved through additional study in future.

We have fabricated the PDMS/glass micro reactor for the biochemical reaction. To evaluate the proposed micro reactor, we performed the Restriction Enzyme(R.E) reaction and the Polymerase Chain Reaction(PCR). The R.E reaction and PCR using the microbiochip were successfully done with less time and sample compared to the conventional way.

N/A The positional effect on ear oximetry was studied in 39 patients with unilateral pleural effusion, 1) The mean values of oximetric SaO2, in the supine, sitting, normal side dependent, and effusion side dependent positions were 94.7±2.4, 96.7±2.4, 94.8±2.7, and 93.2±2.6%, respectively. Oximetric SaO2, values were highest in the sitting position and lowest in the effusion side dependent position. 2) The mean values of oximetric SaO2 in the sitting and normal side dependent positions were significantly higher than those in the effusion side dependent position in small and total pleural effusion (p<0,05). 3) Larger positional differences of oximetric SaO2 were found in patients with a small pleural effusion, 4) The mean values of oximetric SaO2, after aspiration were not significantly higher than those before aspiration (p>0.05). We suggest that the patients with a small pleural effusion has a higher SaO2, in the sitting and normal side dependent positions than that in the effusion side dependent position.

It"s been over 60 years since the Korean war. As time goes on, North and South are changing. The man who experienced Korean war got old or passed away. This generation who have never experienced the war and heard about before are living in this land. They don"t feel the need to unify north and south and they are living under the belief which the peaceful life last forever. However, is it alright to live like this? In this project, Emotions which experienced Korean war and lived hard through the period of recovery give a shape and make a space. Seeing these spaces meditate the past and give a time which understand rightly the present to visitor. Futhermore, The present we face. have an optional alternative plan about future and the space warn the future which not far out of the past.
