Germ-line mutations at DNA repair loci confer susceptibility to colon cancer in hereditary non-polypopsis colorectal cancer. Somatic loss of DNA mismatch repair gene has been reported in a large variety of other tumor types. Replication errors(RERs) judged by microsatellite instability(MSI) and its associated mutations have been recognized as an important mechanism in various tumor types. To investigate associations between MSI and oral squamous cell carcinoma, the frequency of MSI using 12 microsatellite markers were analyzed for the series of oral tumors. Of 17 tumors, 8 cases(47%) did not show instability at any of the 12 loci; 5(29%) showed instability at 2~3 loci; and 4(24%) showed instability above 4 loci. The 4 cases showing widespread MSI did not differ from those without evidence of instability in terms of age at diagnosis, degree of differentiation, metastasis to lymph node, tumor location or the presence of mutations in the p53 tumor suppressor gene. DCC and D17S 796 were the most frequently detected in MSI analysis. There were no correlation between smoking and MSI frequency, instead, smoking was suggested to increase the mutation rate of p53 and development of oral carcinomas.

In order to breed the Korean native dog, sapsaree, which exist as a single population in KyungPook province, we are going to use DNA fingerprinting as a tool for studying the pedigree. In this experiment, 4 kinds of DNA probes, Exon 8, Intron B, M13, and pV47-2 were analysed by means of conventional Southern hybridization with Sapsaree genomic DNAs digested with restriction enzymes for their usefulness as probe. On the basis of their autoradiographic band patterns and resolution, pV47-2 was determined to be the best probe for studing the pedigree of sapsaree.

The Sapsaree is a nearly extincted Korean native dog whose historic habitat has included much of the Southeastern Korea. The only existing population of Sapsaree consists of 50 dogs living in the small area of Kyung-San Goon in Kyung-Pook Province. Field observations of the population indicated the presence of two distinct morphological phenotypes that are divided into Blue- and Yellow-Sapsaree. A RFLP (Restriction Fragment Length Polymorphism) analysis using pV47-2 probe and Sapsaree DNA indicated the existance of two distint genetic stocks concordant with the morphological phenotypes.

The objective of this study was to characterise the pattern of p53 mutations in bladder turmor. In this study, 25 bladder transitional cell carcinomas were analyed by immunohistochemistry (IHC) for p53 nuclear overexpression, and the results were compared with those of polymerase chain reaction-single strand conformational polymorphism (PCR-SSCP) analysis in exon 5-8 of the p53 gene and DNA sequencing analysis. 15 out of 20 cases (75%) showed p53 nuclear immunoreactivities on IHC. On PCR-SSCP analysis, 10 out of 25 cases(40%) had abnormal shifts on mobility. 62% of the mutations were in exon 8. Direct DNA sequencing analysis were performed in these 10 cases to confirm the presence of mutated p53 genes and to determine the type of mutations. Sixteen point mutations were detected in 10 cases. Tow specimens had double mutations and another two had triple mutations. G:C→A:T transitions were the most frequent patterns (62.5%). One mutation was a premature stop codon and two were silent mutations. There out of 10 had a point mutation at codon 285 (GAG/Glu→AAG/Lys) and two had at codon 280(GAG/Glu→AAG/Lys). One of 16 mutations was transition at hot spot codon 273 with CpG site. These results suggest that altered expressions and point mutations of p53 occured in all grade of bladder cancer, but are more associated with hight grade bladder tumors. To elucidate the carcinogenesis of bladder cancer, further studies should be carried out.

내시경 생검으로 얻은 위선암조직에서의 Microsatellite 불안정성 분석

Background/Aims : Microsatellite instability (MSI) is a genetic alteration observed frequently in gastric carcinoma (GC). To assess the role of microsatellite instability (MSI) in gastric cancer (GC), we investigated the incidence of MSI, especially widespread MSI (MSI-H). Methods : Endoscopic biopsy specimens from 59 gastric adenocarcinomas were analyzed for MSI at 13 loci. Paraffin-embedded GC samples from the same 59 GC were analyzed for p53 protein overexpression using immunohistochemical staining. Results: MSI was detected in 33/59 (56%). MSI-H in which over than 40% of investigated loci had MSI, was observed in 7 cases (12%) and low MSI (MSI-L), in which less than 40% of the loci had MSI, was observed in 26 cases (44%). All MSI-H found in 7 cases (12%) were located in distal stomach. There was no difference in the incidences of MSI and MSI-H according to the histology of Lauren's classification, TNM stage, presence of metastasis to lymph node and family history of GC. Overexpression of p53 protein was detected in 43/59 (73%) GC, but MSI-H was not related to the p53 overexpression. MSI in interferon-αR was observed more frequently in GC with family history than GC without family history (55% vs 3%)(p=0.0002). Conclusions : MSI and p53 mutation are involved frequently in oncogenesis of GC. MSI-H is frequently observed in distal GC, but is not related with tumor progression and family history of GC.

DNA지문법은 범죄감식, 친자감별, 가계분석, twin zygosity의 결정, 집단 유전학, 행동 생태학 등의 분야(jeffreys et al., 1985:Gill et al., 1985, 1987 : Lander, 1989:Neufeld and Colman, 1990)에서 널리 활용될 뿐만 아니라 멸종위기에 처한 동식물의 육종과 보존을 위해서도 없어서는 안되는 중요한 연구방법이 되었다.(Georges et al., 1988:Burke et al., 1989). 최근 미국에서는 멸종위기에 처한 퓨마와 매의 보존을 위해 이들 동물들에 대한 RFLP연구를 행했는데 현존집단내 혈통관계에 대한 중요한 정보를 얻을 수 있었다
