간세포암종에서 TPEF의 Methylation에 의한 불활성화

Background : Abnormalities of genomic methylation patterns have been shown to play a role in the development of carcinoma, and the silencing of tumor suppressor genes is related to local de novo methylation. Methods : Using methylation specific arbitrarily primed-Polymerase Chain Reaction (Ms AP-PCR), we identified a 322 bp sequence that contained a 5’ un-translated and exon1 regions of the TPEF gene. To evaluate the inactivation of the TPEF gene through hypermethylation in hepatocellular carcinoma (HCC), we investigated the correlation between methylation patterns and TPEF expression in tumor tissues of human HCC and cell lines via a Combined Bisulfite Restriction Assay (CoBRA) and RT-PCR. Results : A dense methylation pattern of the TPEF was detected in most cell lines, as well as in 10 of the 14 (71.4%) HCC tissues. In addition, loss of heterozygosity (LOH) from the TPEF gene was observed in 5 of the 14 (36%) HCC tissues. Furthermore, RT-PCR analysis revealed TPEF expression in 5 of 8 (62.5%) cell lines. Finally, treatment with a demethylating agent, 5-Aza- 2’-deoxycitidine (5-AzaC), increased the expression of TPEF mRNA. Conclusion : These results indicate that inactivation of the TPEF gene through hypermethylation may be a mechanism by which tumorigenesis occurs in HCC.
유방 침윤성 관암종의 CD44 발현도와 예후 인자들과의 관계

CD44 is a family of transmembrane glycoproteins involved in cell-cell and cell-matrix interactions. Expression of CD44 isoforms (splice variants) has been shown to be associated with poor prognosis in several human cancers. We evaluated the expression patterns of the CD44 isoform (CD 44 splice variant v6) in infiltrating ductal carcinoma of the breast by immunohistochemical and RT-PCR method. Paraffin embedded blocks from seventy-five cases of mastectomized samples were analyzed immunohistochemically using monoclonoal antibody against CD44v6. CD44v6 was detected in fifty-seven cases(76%) of the tumor samples. Adjacent normal myoepithelial cells and ductal epithelial cells revealed focal positive reaction to CD44v6. Thirty-two cases (80.0%) withe lymph nodal metastasis revealed overexpression of CD44v6 monoclonal antibody, but twenty-five cases (71.4%) without nodal metastasis also showed positive reaction to CD44v6 monclonal antibody, and there is no statisticallly significant value. Other prognostic factors of infiltrating ductal carcinoma, such as tumor size, histologic grade and hormonal receptors did not show any significant correlation with CD44v6 expression. The RT-PCR studies for 9 cases of infiltrating ductal carcinoma showed the same band patterns both in the normal and tumor tissues. From the above results, it is concluded that the expression of CD44v6 is not a valuable prognostic marker of infiltrating ductal carcinoma of breast.(Korean J Pathol 1997;31: 635~643)

The Sapsaree is a nearly extincted Korean native dog whose historic habitat has included much of the Southeastern Korea. The only existing population of Sapsaree consists of 50 dogs living in the small area of Kyung-San Goon in Kyung-Pook Province. Field observations of the population indicated the presence of two distinct morphological phenotypes that are divided into Blue- and Yellow-Sapsaree. A RFLP (Restriction Fragment Length Polymorphism) analysis using pV47-2 probe and Sapsaree DNA indicated the existance of two distinct genetic stocks concordant with the morphological phenotypes.

In order to breed the Korean native dog, sapsaree, which exist as a single population in KyungPook province, we are going to use DNAfingerprinting as a tool for studying the pedigree. In this experiment, 4 kinds of DNA probes, Exon 8, Intron B, M13, and pV47-2 were analysed by means of conventional Southern hybridization with Sapsaree genomic DNAs digested with restriction enzymes for their usefulness as probe. On the basis of their autoradiographic band patterns and resolution, pV47-2 was determined to be the best probe for studing the pedigree of sapsaree.

DNA지문법은 범죄감식, 친자감별, 가계분석, twin zygosity의 결정, 집단 유전학, 행동 생태학 등의 분야(jeffreys et al., 1985:Gill et al., 1985, 1987 : Lander, 1989:Neufeld and Colman, 1990)에서 널리 활용될 뿐만 아니라 멸종위기에 처한 동식물의 육종과 보존을 위해서도 없어서는 안되는 중요한 연구방법이 되었다.(Georges et al., 1988:Burke et al., 1989). 최근 미국에서는 멸종위기에 처한 퓨마와 매의 보존을 위해 이들 동물들에 대한 RFLP연구를 행했는데 현존집단내 혈통관계에 대한 중요한 정보를 얻을 수 있었다
