한우 유전자칩(Hanwoo 50K chip)을 이용한 전라북도 한우의 연관불평형 분석

This study was conducted with the Hanwoo SNP chip (50k_v1) analysis to investigate genotypes of SNPs of Hanwoo (Korean native cattle) in Jeollabuk-do and extent of linkage disequilibrium (LD) among SNP markers. Genomic DNA for genotyping was extracted from the hair root samples of Hanwoo cows (n=1,523). A total of 52,195 SNPs were obtained and 11,056 unnecessary SNPs were removed through the QC process, and a total of 41,139 SNPs were used for LD analysis. The total length of the SNP markers used in the analysis was 2,500 Mb, the mean of minor allele frequency was 0.27 and the average interval distances of adjacent SNP markers by each chromosome was 0.0463 to 0.0725. The results of this study which confirmed by using Hanwoo in Jeollabuk-do, showed a pattern similar to previously reported studies for Hanwoo. The r 2 value was 0.234 for the SNP distance between 0 and 50 Kb and was 0.068 for distance between 150 and 200 Kb. Therefore this study could be utilized as baseline data using genetic improvement for Hanwoo industry.

Chikso(brindle cow) is a subspecies of Hanwoo, and is one of the Korea’s native breeds. It has the characteristics of being strong and strong against disease. The black stripes around its body resemble that of a tiger. In this study, 9,663 Chikso in South Korea were classified into 4 groups based on their birth year. The polymorphism analysis of Chikso whole groups revealed the average number of alleles in each marker (number of alleles), the expected heterozygosity (HEX), the observed heterozygosity (HOB), the polymorphism information content (PIC), and the FIS mean as 16.2, 0.769, 0.759, 0.737 and 0.0061, respectively. For group D, with the birth year 2018-2020, the HEX and HOB were 0.767 and 0.760 respectively and the PIC was 0.734. The average observed heterozygosity (HOB) for each generation of the Chikso group was confirmed to be 0.753, and it was confirmed that it gradually increased from group A (0.749) to group C (0.751), and increased sharply in group D (0.760). The average of the PIC was found to be 0.732, and the most recently born group, group D (2018-2020), had the highest value of 0.734. It was confirmed that the FIS value also gradually decreased with the recent generation. The result of this study are expected to be used for enhancing the efficiency of Hanwoo Industry.

Electricity generation is the leading cause of global warming all over the world. Most of our electricity comes from coal, nuclear, and other non-renewable power plants. Producing energy from these resources takes a severe toll on our environment which is like polluting our air, land and water. Moreover, Korea does not have the resources much. so we are facing with needing to cut down consumption of Electricity. Therefore, The objective of this study is to find out how we can reduce the cost of energy. And this study is going to proceed on comparing real measurement with simulation results using Energy Plus program. we are currently expecting that if we used more efficient glass window or package air conditioner, we could get a lot of energy savings than before.

To develop new cephalosporin antibiotics with improved antibacterial activities, a series of 7beta-[2-(2-aminothiazol-4-yl)-(Z)-2-(1-carboxy-1-methylethoxyimino)acetamido]-3-[5-(heterocycle)thiomethylpyrrolidin-3-ylthio]methyl-3-cephem-4-carboxylic acid (14-18) having aminothiazol carboxymethylethoxyimino group on the C-7 position and (heterocycle) thiomethyl pyrrolidinthiomethyl group on the C-3 position of the cephem ring were synthesized. These compounds were tested for antimicrobial activity in vitro against Gram(+) and Gram(-) bacteria. Compounds 15 and 16 showed remarkable antibacterial activity against Salmonella typhimurium TV119 and Alcalienes faecalis KCTC1004, but most of compounds showed lower activity than cefotaxime.

The issue of carbon neutrality has become more serious around the world. In particular, the livestock industry has been an industry with very high carbon emissions. The government provided support and subsidies to livestock farms and businesses to help them reduce carbon emissions. This assistance aimed to encourage the adoption of environmentally friendly practices and technologies, ultimately promoting carbon-efficient production. The livestock industry has also been researching technology development to reduce carbon emissions through improved productivity. The purpose of this study was to more accurately identify the carbon emissions status of the Hanwoo industry. We analyzed the carbon footprint status and trends of the Hanwoo by region. For this purpose, traceability information and slaughter information of Hanwoo slaughtered from 2015 to 2022 were collected and analyzed. We determined the impact of average carcass weight and slaughter age trends on carbon emissions. Through ANOVA analysis, it was confirmed that there was a significant change in the carbon footprint of each region (p<0.01).
The characteristics of bovine satellite cells with highly scored genomic estimated breeding value

Background: The grading of Hanwoo (Korean native cattle) is based on four economic traits, and efforts have been continuously made to improve the genetic traits associated with these traits. There is a technology to predict the expected grade based on the 4 economic genetic SNP characteristics of Korean cattle calves using single nucleotide polymorphism (SNP) technology. Selection of highly proliferative, self-renewing, and differentiating satellite cells from cattle is a key technology in the cultured meat industry. Methods: We selected the Hanwoo with high and low-scored of genomic estimated breeding value (GEBV) by using the Hanwoo 50K SNP bead chip. We then isolated the bovine satellite cells from the chuck mass. We then conducted comparative analyses of cell proliferation, immunocytochemistry, qRT-PCR at short- and long-term culture. We also analyzed the differentiation capability at short term culture. Results: Our result showed that the proliferation was significantly high at High scored GEBV (Hs-GEBV) compared to Low scored GEBV (Ls-GEBV) at short- and long-term culture. The expression levels of Pax3 were significantly higher in Hs-GEBV bovine satellite cells at long-term culture. However, there were no significant differences in the expression levels of Pax7 between Hs- and Ls-GEBV bovine satellite cells at short- and long- term culture. The expression levels of MyoG and MyHC were significantly high at Ls-GEBV bovine satellite cells. Conclusions: Our results indicated that selection of bovine satellite cells by Hanwoo 50K SNP bead chip could be effective selection methods for massive producing of satellite cells.
한우 산업 축군의 친자검정 유전자형 데이터를 이용한 모니터링 분석

In this study, the genetic diversity of the commercial Hanwoo population was monitored through the analysis of genotype information of a total of 33,300 heads. Set1 is a group that has the dam's genotype. Set2 is a group without dam’s genotype. As a result, it was confirmed that the genetic diversity of the analysis group was well maintained. The locus with the highest diversity identified is TGLA53. On the other hand, the locus with the lowest diversity was identified for TGLA126. Of the mismatched individuals with the dam’s genotype, 58% identified only one KPN. Monitoring using genetic information is necessary for the development of the Hanwoo industry and the enhancement of the efficiency of improvement projects.

The paternity test technique using microsatellite markers can accurately determine whether the pedigree father is true or false. However, through this study, it was confirmed that pedigree errors may occur because paternity tests using microsatellite marker genotypes estimate multiple fathers rather than finding one father. For this reason, it was found that in more than 76% of cases it was impossible to correct pedigree errors. However, if the snp chip is used, it is possible to accurately estimate one real father and correct the pedigree error. This study is thought to be used as a very effective means for improving Korean cattle(Hanwoo) because it enables lineage management and genome-based genetic ability evaluation.

It has found that the using library base on criteria of a foreign country resulted in problem of reliability. In order to obtain reliable result, it was necessary to develop the simulation input library based on domestic condition. So, this study was conducted for making the library in accordance with domestic condition when doing simulation modeling about the exist and new buildings. The range of library were consist of material, facility systems, schedules and weather data. And Energy Plus program was used when making library. We hope that the developed library will contribute to reliability when simulating exist and new buildings.
