A total of 499 stomachs consisted of 393 slaughter and 106 breeding pigs from industrial farm was examined for ulcerative lesions grossly and microscopically. The relationship between the ulcerative lesions and stomach contents was also examined. 1. The majority of stomachs has one or more lesions of ulcerative process, including parakeratosis(64.1%), erosions(10.2%), acute ulcers(2.8%), chronic ulcers(1.4%), and scars(0.8%). 2. The incidence and the degree of the esophagogastric lesions in the breeding pigs were similar to those of the slaughter pigs. But ulcerative lesions of the fundic lesions in the breeding pigs were found to be more remarkable than those of the slaughter pigs. 3. The stomach contents of the pigs having gastric ulcers were mostly fluid in nature, which were dry in the normal stomach. 4. Histologically, muscular hypertrophy, connective tissue hyperplasia and vascularity were shown in the ulcerative lesions of the esophagogastric portion. Thrombosis, necrosis and granulation tissue formation were found frequently.
Antisense DNA를 사용한 Cdk2조절에 의한 암세포의 성장 억제

암 세포의 두드러진 특징은 세포 증식 조절능력이 없다는 것이다. 세포 증식 주기의 조절에서 G1기와 S기에서 중요한 역할을 담당하는 Cdk2의 발현 억제를 통해서 세포 주기 진행의 차단과 그에 의한 세포 성장억제를 관찰하고자 하였다. HeLa 세포로부터 세포 조절 인자 Cdk2 유전자의 발현을 RT-PCR을 통해 관찰하였고, HeLa 세포 mRNA로부터 Cdk2 번역 개시부분의 Cdk2 cDNA을 합성하여 뉴클레아제에 안정하고 고 효율성의 폐쇄형 단일가닥 환형 antisense DNA (Cdk2 antisense, Cdk2 sense)를 제작하였으며, HeLa 세포에 농도 별로 실험군으로 Cdk2 antisense, 대조군으로 Cdk2 sense, control 단일가닥 DNA을 LipofectAMINE PLUS^TM와 복합체를 형성하여 transfection하였을 때 실험군에서 Cdk2 mRNA가 특이적으로 감소하는 것을 확인 할 수 있었다. 또한 Cdk2 mRNA 감소에 의한 세포주기의 진행차단은 세포의 성장을 억제하였다. 따라서 암 치료제의 개발을 위해 antisense을 이용한 유전자 치료가 수행될 수 있음을 나타낸다. Cancer is characterized by deregulated cell cycle. Recent studies on cell cycle have demonstrated that some proteins, which regulate cell proliferation, may also be involved in oncogenesis. Critical transitions in a cell cycle are regulated by sequential activation of a series of cyclins and cyclin-dependent kinases (Cdks). Cdks are catalytic subunits of a family of serine/threonine protein kinase complexes, and are involved in the progression of cell cycle at various stages through G1, S, G2 and M phases. Cdk2 is essential for transition from G1 to S phase of mammalian cell cycle and overexpressed in cancer cells. In this study, we examined the effects of antisense to Cdk2 with a novel structure. Circular antisense, targeting a large area containing the translation start site of the Cdk2 cDNA, was constructed. When HeLa cells were treated with Cdk2 antisense, dose-dependent decrease of Cdk2 mRNA was observed. Cancer cell growth was found to be inhibited by more than 50% determined by cell counting assays. However, cell growth was not significantly inhibited, when treated with sense and control single strand DNA. These results suggest that Cdk2 plays an important role in the cancer cell proliferation and circular antisense DNA can be employed in the development of molecular antisense drugs for cancer.
Potential Role of Immunodiagnosis for Pulmonary Tuberculosis Using Induced Sputum Cells

Purpose: To evaluate the diagnostic utility and predictors for determinate results of an enzyme-linked immunospot assay using induced sputum cells (IS ELISPOT) for a rapid diagnosis of pulmonary tuberculosis (TB). Materials and Methods: Subjects suspected of pulmonary TB who had either sputum acid fast bacilli smear-negative or not producing sputum spontaneously were prospectively enrolled. ELISPOT assay was performed using cells from induced sputum. Results: A total of 43 subjects, including 25 with TB (TB group) and 18 with non-TB disease(non-TB group) were enrolled. Results of IS ELISPOT were determinate in only 17/43 (39%) subjects, but all of determinate results were consistent with the final diagnosis. Of the 43 sputum samples, 11 (26%) were inadequate to perform IS ELISPOT. Of 32 adequate sputum samples, the proportion of determinate resultswas significantly higher in the TB group (75%, 15/20) than in the non-TB group (17%, 2/12) (p=0.002). The status of active TB was a unique predictor but smear positivity was not a significant predictor for determinate results. In addition, sensitivity of IS ELISPOT (75%, 9/12) in smear negative TB was higher than that of TB-polymerase chain reaction (25%, 3/12). Conclusion: IS ELISPOT showed relatively high diagnostic value and accuracy in the TB group, independent of smear positivity. IS ELISPOT may provide additional diagnostic yield for microbiologicaltools in the rapid diagnosis of smear-negative TB.

Purpose: Guinea pig is one of the most suitable animal models for Mycobacterium tuberculosis (M. tb) infection since it shows similarities to pulmonary infection in humans. Although guinea pig shows hematogenous spread of M. tb infection into the whole body, immunological studies have mainly focused on granulomatous tissues in lungs and spleens. In order to investigate the time-course of disease pathogenesis and immunological profiles in each infected organ, we performed the following approaches with guinea pigs experimentally infected with M. tb over a 22-week post-infection period. Materials and Methods: We examined body weight changes, M. tb growth curve, cytokine gene expression (IFN-γ and TNF-α), and histopathology in liver, spleen, lungs and lymph nodes of infected guinea pigs. Results:The body weights of infected guinea pigs did not increase as much as uninfected ones and the number of M. tb bacilli in their organs increased except bronchotracheal lymph node during the experimental period. The gene expression of IFN-γ and TNF-α was induced between 3 and 6 weeks of infection; however, kinetic profiles of cytokine gene expression showed heterogeneity among organs over the study period. Histophathologically granulomatous lesions were developed in all four organs of infected guinea pigs. Conclusion: Although IFN-γ and TNF-α gene expression profiles showed heterogeneity, the granuloma formation was clearly observed in every organ regardless of whether the number of bacilli increased or decreased. However, this protective immunity was accompanied with severe tissue damage in all four organs, which may lead to the death of guinea pigs.
