First report for Platanthera brevicalcarata (Orchidaceae) in Korea

Platanthera brevicalcarata Hayata (Orchidaceae), previously known to be distributed from Taiwan to southern Japan, was found in the Jeju-do, Korea. This species is distinguished from the related taxon Platanthera chlorantha by its small plant size, white sepal with distinct one green vein, and short-cylindrical spur. The local name Young-ju-je-bi-ran is originated from the local name of Mt. Halla. The morphological characters and illustration of the species are provided with line drawing and photograph from the natural habitat.

Phomopsis seed decay (PSD) is a major fungal disease found in most soybean production areas of the world. PSD significantly reduces soybean yield and seed quailty, particularly when soybean plants are exposed to high humidity and high temperature during maturation. To certificate genetic segregation ratio for PSD, PI 417479 line(resistant) and Hwaeomputkong(susceptible) were crossed and examined their inheritance mode. Experiment was conducted at glasshouse of Soonchunhyang Univ., in Korea. Different PSD seed infection levels were induced in soybean seeds by artificially infected plants exposing to above 95% relative humidity and 30˚C. A sample of seeds harvested from inoculated plants was analyzed to determine percent PSD incidence. The patterns of resistant response between Hwaeomputkong and PI 417479 were different in their seed and pod shape. The F₁plants of the resistance is governed by the two dominant genes. This results indicate that a resistance to PSD is controlled by complementary genes.
지방자치단체의 지역기록화 전략 수립을 위한 수정델파이 연구

This study aimed to propose alternative approaches that should be prioritized in formulating a locality documentation strategy for local governments. To achieve this, we conducted an analysis of cases from five local governments, including Cheongju City, and extracted universal factors based on them. We then conducted a Modified Delphi survey. As a result, we identified the importance of 35 items across five areas, including the purpose and system of regional record localization, tasks, promoting factors, and the effects of regional record localization. Additionally, we clarified the responsibility of local governments for regional record localization and presented their roles and implementation strategies for fulfilling this responsibility. This study is significant in that it attempted an integrated and multilayered exploration based on the experiences and knowledge of record experts, considering the absence of standardized manuals for regional record localization. It is expected that this study can provide reference points for formulating regional record localization strategies for each local government in the future.
열대 약용식물 Clausena excavata의 RNA-seq 및 SSR 분석

Background : Several members of the genus Clausena have a great potential as a candidate for the identification of new drug lead molecules, but lack of their genomic information can be a hindrance for the verification of the genetic background for future use. To broaden and delve into the genomic features of this genus, Clausena excavata, an important medicinal plant in many Asian countries, was used for RNA-seq analysis. Methods and Results : A ten ㎍ of the total RNA was used for mRNA isolation using oligo-dT beads and random sheared mRNAs were used to prepare a cDNA library for a illumina hiseq 2500 analysis. In total, 17,580,456 trimmed clear reads from the illumina hiseq 2500 were used for de novo assembly using three assemblers, CLC genomics workbench, velvet-oases, and Trinity. A total of 16,638 non-redundant unigenes with an average length of 755 bp were generated by the assembly. The functional categorization of the identified unigenes by a gene ontology (GO) term resulted in 2,305 genes in the cellular component, 5,577 in the biological processes, and 8,056 in the molecular functions, respectively. The top sub-category in biological processes was the metabolic process with 4,374 genes. Among annotated genes, 3,006 were mapped to 123 metabolic pathways by KEGG metabolic pathway analysis tool. The search for simple sequence repeats (SSRs) resulted in 845 SSRs from 749 SSR-containing unigenes and the most abundant SSR motifs was AAG/CTT with 179 occurrences. Twelve SSR markers were tested for cross transferability among five Clausena species; eight of them exhibited polymorphism. Conclusion : In the present study, genomic resources of the genus Clausena were enriched through RNA-seq and SSR markers, which will serve as valuable resources for genomic/genetic studies of the genus Clausena and close relatives.
