        새로운 아드리아마이신유도체 DA-125 의 조혈기독성과 함암효과에 미치는 G-CSF 의 영향

        The present study was designed to evaluate the effects of a recombinant human granulocytecolony stimulating factor (G-CSF) on leukopenia and tumor growth in mice treated with DA-125, an adriamycin (ADM) derivative. In normal mice, single intravenous injection of DA-125 produced transient leukopenia accompanied with weight loss and splenic atrophy in a dose-related manner. However, subcutaneous administration of G-CSF (5 ㎍/head) for 5 consecutive days after DA-125 resulted in a significantly elevated nadir of leukocyte counts and facilitation of recovery from the leukopenia. To investigate the effect of G-CSF on antitumor effects of DA-125, ADM (12 mg/kg) or DA-125 (40 mg/kg) was administered to Colon-26 murine adenocarcinoma-bearing Balb/c mice with G-CSF. Regardless of treatment with G-CSF, DA125 and ADM markedly retarded the growth of implanted tumor, though they failed to increase mean survival time of tumor-bearing mice. These results suggest that G-CSF is able to not only ameliorate, but reconstitute DA-125-induced myelosuppression without affecting its antitumor potential.

        기후변화와 우리 삶의 존립방식

        Implications of climate change is of a permanent change of relationships between Human beings and Nature. We are responsible for regulating of impacts by human beings on nature as it is mankind itself who has been changing the grand cycle of nature. First of all, it is necessary for the causes of global warming to be eliminated and the controls preventing its resulting consequences to be well-equiped. A myriad of measures is of feasibility and needs less costs than expected, rather having positive effects without adverse influences on climate change. Among these can be included such as the removal of subsidies inviting the consumptions of fossil fuels, the protection of forests and the enlargement of means of public transportation etc. But efforts by individual State or within separate regions may have the distinctive limitations which have being derived from the fact that there are the considerable disparity of the historical responsibilities as well as a sharp difference in each State's capacity to cope with. Therefore, the adjustments and agreements of international community might be mandatory. Irrespective of the climate change negotiations, things, inter alia, are urgent that the reversion of one's values and determined practices. Especially, it is required to depart away from the alledged dogma of ??the unlimited growth is possible??

        에탄올 - 유발 위점막손상에 대한 애엽추출물 (DA-9601) 의 방어효과 및 기전에 관한 연구

        Protective effect of DA-9601, an extract of Artemisia Herb, against ethanol-induced gastric mucosal injury was evaluated in rats. In the prophylactic study, DA-9601 exhibited total protection (99.4%) against absolute ethanol-induced gastropathy. And the protective effect of DA-9601 lasted up to 2 hours, which was longer than those of other contemporary mucoprotectants. In the treatment study, DA-9601 significantly facilitated the healing of 70% ethanol-induced mucosal damage, which was superior to cetraxate, a commonly used anti-ulcer drug. The mechanisms of mucoprotection of DA-9601 were also assessed. DA9601 increased the release of prostaglandin E₂ from murine neutrophils in a dose-dependent manner in vitro. The cytoprotective effect of DA-9601 against ethanol-induced mucosal damage was significantly diminished by the concommitant injection of N_w-nitro-L-arginine methyl ester (L-NAME, 5 mg/kg, i.v.), a non-specific nitric oxide (NO) synthase inhibitor, while it was not affected by preinjection of indomethacin (5 mg/kg, s.c.), a prostaglandins-depletor. And it was found that DA-9601 significantly enhanced adaptive cytoprotective action of 10% ethanol against absolute ethanol (56.9±6.5 vs 23.0±3.3 mm², p<0.05, mean±SEM), though its exact underlying mechanism remains to be clarified. The present findings demonstrate that DA-9601 exerts gastroprotective actions for the stomach against ethanol through several different underlying mechanisms, in which prostanglandins and NO are involved. In conclusion, the results obtained suggest that DA-9601 can be useful both in prevention and treatment of ethanol-induced gastric damage.

        야생참깨에서 agrobacterium rhizogenes 유도 모상근의 배양조건

        In order to investigate optimized culture conditions of hairy roots of Sesamum schinzianum (wild sesame) induced by infection of Agrobacterium rhizogenes, ATCC 15834 and MAFF03-01724 were cultured in various media, sucrose concentrations and pH levels. As the results, best growth condition of hairy roots induced by Agrobacterium rhizogenes strains was obtained in 1/2 MS medium free of hormone, 3% sucrose and pH 8.0. Hairy roots induced by Agrobacterium rhizogenes MAFF03-01724 showed better growth of extensive lateral root branches than that of ATCC 15834. In opine analysis, hairy roots induced by Agrobacterium rhizogenes ATCC 15834 was detected as agropine type while mikimopine in that of MAFF03-01724.

        지베렐린 처리가 땅콩 鍾間 交雜 親和性에 미치는 影響

        Interspecific crosses between three peanut cultivars(Arachis hypogaea) having different plant type, and four wild species, Arachis monticola, Arachis correntina, Arachis applisippla, Arachis rigonii were made by gibberellin(GA₃) treatment and the GA₃ effect on the interspecific crossability and growth of pollen tube in pistil was investigated. Pollen germination on the stigma and penetration of the pollen tubes into the style were increased by GA₃ treatment on the flower base. The GA₃ treatment increased pegging and pod formation up to 47% and 1.6% higher than those of control, respectively. Interspecific-crossability was varied with the combination of cultivar and wild species. Combination of Shipung-type variety Daekwangtangkong×wild species and Spanish-type variety Jinpungtangkong×wild species showed about 2% higher crossability than those of Virginia type variety Namdaetangkong×wild species. Arachis monticola among wild species showed the highest crossability with peanut cultivars, while Arachis correntina did the lowest. This result suggested that interspecific crossbility become higher in the cross between autoplyploids rather than those between allopolyploids.

        참깨의 비닐피복 기계화 일관작업 체계 연구

        This study was carried out to develop and investigate integrated mechanization system on polyethylene(P.E.) film mulching culture in sesame. Field trials were carried out to develop optimum mechanics for each step of manual operation in sesame culture and demonstrated those developed mechanics of sesame dibbling vinyl mulcher, sesame binder, sesame thresher and sesame grader at the farmer, s field of Hwasong (Kyunggi-do) in 1994 and 1995. Newly developed sesame dibbling vinyl mulcher brought saving manpower 280 hrs /ha for sowing and P.E. film mulching with it's 98% of labour saving for the harvesting operation of sesame. Sesame Binder showed 93% labour saving for cutting and binding with only 22 hrs /ha compared to 330 hrs /ha that of conventional. Sesame thresher was appeared to have 85% effects of labour saving for threshing with completely dry sesame bundles at one time by 23 hrs /ha compared to 151 hrs /ha that of conventional. Sesame grader was appeared to have 72% effects for sesame grain grading with 12 hrs /ha compared to that of conventional 54 hrs /ha. Grain yield of integrated mechanization culture system showed 6% higher than that of coventional. Integrated mechanization culture system (sesame dibbling vinyl mulcher + sesame binder + sesame thresher + sasame grader) showed 62% of labour saving effects through whole steps of sesame culture by 472 hrs /ha compared to that of conventional 1, 230 hrs /ha.

        Ac/Ds 삽입 변이체를 이용한 벼 유전자 기능 연구

        Rice is the staple food of more than 50% of the worlds population. Cultivated rice has the AA genome (diploid, 2n=24) and small genome size of only 430 megabase (haploid genome). As the sequencing of rice genome was completed by the International Rice Genome Sequencing Project (IRGSP), many researchers in the world have been working to explore the gene function on rice genome. Insertional mutagenesis has been a powerful strategy for assessing gene function. In maize, well characterized transposable elements have traditionally been used to clone genes for which only phenotypic information is available. In rice endogenous mobile elements such as MITE and Tos (Hirochika. 1997) have been used to generate gene-tagged populations. To date T-DNA and maize transposable element systems has been utilized as main insertional mutagens in rice. A main drawback of a T-DNA scheme is that Agrobacteria-mediated transformation in rice requires extensive facilities, time, and labor. In contrast, the Ac/Ds system offers the advantage of generating new mutants by secondary transposition from a single tagged gene. Revertants can be utilized to correlate phenotype with genotype. To enhance the efficiency of gene detection, advanced gene-tagging systems (i.e. activation, gene or enhancer trap) have been employed for functional genomic studies in rice. Internationally, there have been many projects to develop large scales of insertionally mutagenized populations and databases of insertion sites has been established. Ultimate goals of these projects are to supply genetic materials and informations essential for functional analysis of rice genes and for breeding using agronomically important genes. In this report, we summarize the current status of Ac/Ds-mediated gene tagging systems that has been launched by collaborative works from 2001 in Korea.

        Bt 벼의 토양미생물상 영향 비교평가

        경제적 및 농업적 장점은 유전자 변형 작물 재배면적의 증가를 가져왔다. 그러나 유전자 변형 작물의 상업적 재배전에 유전자 변형 작물의 인간건강 및 환경에 미칠 잠재적 위해성에 대한 면밀한 검토가 필수적이다. 본 연구에서는 Bt벼의 토양미생물 군집에 미치는 영향을 조사하였다. 토양화학성분을 분석한 결과, Bt벼와 낙동벼 근권토양 간 화학 성분의 유의성 있는 차이는 없는 것으로 조사되었다. 재배전, 재배초기, 최고 분얼기의 토양미생물 군집밀도를 조사했을 때 Bt벼 근권토양의 세균, 방선균, 진균 군집밀도는 낙동 벼와 유사한 수준으로 나타났다. 시기별 DGGE 분석결과 Bt 벼 근권토양 전체미생물상은 낙동벼와 차이가 없는 것으로 조사되었다. Pyrosequencing을 통한 Bt벼와 낙동벼의 미생 물 군집조성을 조사한 결과 주요 미생물상 분포에 있어서도 매우 유사한 양상을 나타내었다. 위의 결과들을 종합해볼 때 Bt 재배에 따른 토양미생물상에 미치는 영향은 미미한 것으로 사료된다. 수확 후 벼 잔존물이 토양환경에 미치는 영향에 대해서는 좀 더 연구가 진행되어야 할 것이다. The cultivation of genetically modified (GM) crops has increased due to their economic and agronomic advantages. Before commercialization of GM crops, however, we must assess the potential risks of GM crops on human health and environment. The aim of this study was to investigate the possible impact of Bt rice on the soil microbial community. Microbial communities were isolated from the rhizosphere soil cultivated with Bt rice and Nakdong, parental cultivar and were subjected to be analyzed using both culture-dependent and molecular methods. The total counts of bacteria, fungi, and actinomycetes in the rhizosphere of transgenic and conventional rice were not significantly different. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that the bacterial community structures during cultural periods were very similar each other. Analysis of dominant isolates in the rhizosphere cultivated with Bt and Nakdong rice showed that the dominant isolates from the soil of Bt rice and Nakdong belonged to the Proteobacteria, Cloroflexi, Actinobacteria, Firmicutes, and Acidobacteria. These results indicate that the Bt rice has no significant impact on the soil microbial communities during cultivation period. Further study remains to be investigated whether the residue of Bt rice effect on the soil environment.

        새로운 괴립구 콜로니 자극인자 ( rhG-CSF ) DA-3030 의 국소자극성에 관한 연구

        As a series of safety studies of DA-3030, a new rhG-CSF, its local irritancy was examined in the rabbits after the following treatment; application into the conjunctival sac of the eye(singie), subcutaneous injection(single), intramuscular injection(single), and intravenous injection(8-day repeated). In addition, paravenous irritation of DA-3030 was investigated in mice. The results obtained were as follows. 1. In the result of ocular irritation test, 0.03% solution of DA-3030 could be considered as a non-irritating material. 2. The local irritation of DA-3030 by an injection of 0.5 ml of its solution subcutaneously or intramuscularly was negligible and not so much different from that of saline. 3. In the vascular irritancy test, macro- and microscopic observations revealed that the irritating activity of DA-3030 in blood vessels was not different from that of saline when they were injected once a day into vein retroauricularis of rabbits for 8 days. 4. The paravenous administration of DA-3030 did not induce any abnormal changes at injection sites except mild swelling in 1 mouse at 3 hours after injection which was thought to be due to slow absorption. The above-mentioned results suggest that DA-3030 has no irritating activity when injected through intravenous or subcutaneous route for clinical practice as 0.03% solution.
