Streptomces sp. MET 0515의 균체외 Alginate lyase의 정제 및 특성

알긴산분해효소(EC 4. 2. 2. 3)를 균체외로 생산하는 새로운 방선균을 전라남도 완도의 연안토양으로부터 분리하고 이 균주를 Streptomyces sp. MET 0515라 명명하였다. 이 균주가 생산하는 균체외 알긴산분해효소를 아세톤 침전, 음이온 교환 크로마토그래피(Q-Sepharose and DEAE-Sepharose), 젤 크로마토그래피(Sephacryl S-200 HR gel filtration chromatography)를 이용하여 정제하였다. 이 효소의 최적 활성 온도과 pH는 각각 $70^{\circ}C$와 pH 7.5이고, 온도 안정성은 $0-50^{\circ}C$, pH 안정성은 pH 6.0-9.0였다. 이 효소는 $Mn^{2+}$ 이온 첨가시에는 활성이 증가하였으며 1mM $Fe^{3+}$, 1mM EDTA, 1mM $Zn^{2+}$ 이온 첨가시에는 활성이 억제되었다. 또한 이 효소는 poly-alpha 1,4-L-guluronate와 poly-beta 1,4-D-mannuronate두 종류의 기질에 대하여 활성을 나타냈다. 따라서 본 효소는 다른 미생물 기원의 효소와 비교해 볼 때 Streptomyces sp.가 생산하는 첫번째 효소로 인정되었다. We isolated a new extracellular alginate lyase-producing microorganism, which displayed alginate-depolymerizing activity in plate assays, from coastal soils in Wando, Jeollanam-do, Korea. This alginate-depolymerizing bacterium belonged to the genus Streptomyces and it was named Streptomyces sp. MET 0515. An extracellular alginate lyase(ALY1) secreted by Streptomyces sp. MET 0515, was purified to homogeneity by a combination of acetone precipitation, anion-exchange chromatography (Q-Sepharose and DEAE-Sepharose) and Sephacryl S-200 HR gel filtration chromatography. Its molecular mass was 26 kDa as determined by SDS-PACE analysis. The enzyme had an optimal temperature of $70^{\circ}C$ for its activity, and was most active at pH 7.5. The thermal and pH stability were $0-50^{\circ}C$, and pH 6.0-9.0, respectively. The enzyme activity was stimulated by 1mM $Mn^{2+}$, and inhibited by 1mM $Fe^{3+}$, 1mM EDTA and 1mM $Zn^{2+}$. Preliminary analysis of substrate specificity showed that this alginate lyase had activity on both poly-alpha 1,4-L-guluronate and poly-beta 1,4-D-mannuronate in the alginate molecule.
기능성 소화불량증 환자의 피로도와 기혈수 변증의 상관성 분석

Objectives: The aim of this study was to investigate the correlation between fatigue degree and Comprehensive diagnosis of Qui Xui Shui in patients with functional dyspepsia. There again, we investigated an association between scores obtained from Comprehensive diagnosis of Qui Xui Shui and state of skin sympathetic tone or cardiovascular reactivity. Methods: Subjects were 56 patients with Functional dyspepsia and 8 healthy People. Fatigue degree was assessed by questionnaires consisting of subjective complaints of fatigue. Comprehensive diagnoais of Qui Xue Shui was investigated by questionnaire and physical examination, total score and composition score were calculated from the symptom score. Skin sympathetic tone was measured by Ryodoraku Score(RS) and Cardiovascular Reactivity(CV Reactivity) was checked by Pulse diagnostic apparatus. All patients were divided into two groups by RS $40{\mu}A$(below and above) or by CV Reactivity(decreased and increased or not decreased). Results: There was highly significant correlation between Fatigue Degree and total score and each composition score of Comprehensive diagnosis of Qui Xui Shui. And total score of patients was significantly higher than that of health controls : in the patients, females than males. Significance related with the level of RS $40{\mu}Aor$ the state of CV Reactivity were not remarkable in both patients and health control group. Conclusions: Fatigue degree seems to have a special feature reflecting the state of Comprehensive Diagnosis of Qui Xui Shui in patients with functional dyspepsia.
과채류껍질 추출물의 학습능력수행 및 기억력증진에 미치는 효과

본 연구는 광범위한 식품 용도에서 독성을 일으키지 않고 오랫동안 사용되어 온 과일껍질은 질기고 거친 식감과 떫은 맛이 많은 관능성의 문제로 껍질의 섭취를 꺼려하여 폐기물로서 버려지고 있지만, 과일껍질은 무기질, 비타민, 폴리페놀과 같은 유용한 생리활성 성분이 다량 함유되어 있다. 따라서 우리나라 국민들이 가장 일반적으로 소비하는 과일류 중 과일껍질 유래의 과피 부분을 쉽게 분리할 수 있는 사과 또는 배의 과채류 껍질추출물을 활용하여 최근 문제시 되고 있는 알츠하이머성 치매를 예방 및 치료 할 수 있는 학습능력 수행 또는 기억력 증진에 미치는 효과에 대해 평가하였다. 그 결과, 사과껍질과 배껍질의 혼합 추출물은 사과껍질과 배껍질의 추출물 각각에 비하여 아세틸콜린에스테라제의 억제활성이나 스코폴라민 유도 동물 모델의 수미로 실험에서 학습능력 수행방 및 기억력 증진효과가 더욱 현저하였다. 이는 추출물에 함유된 성분들 간의 상호작용에 의한 상승효과에 기인한 것으로 해석할 수 있었다. 이를 통하여 볼 때 과채류 껍질 추출물은 아세틸콜린에스테라제 억제, 단기기억, 장기기억 활성 촉진으로 콜린성 신경계를 자극하여 기억 및 학습증진에 매우 효과적으로 작용하는 항치매 물질로 사료된다. In the present study, we assessed the effecs of fruit and vegetable bark resources rich in beneficial substances on the learning ability and memory improvement. It functions to inhibit the activity of acetylcholinesterase in brain tissue and improve cognitive functions in a simulation model of cognitive impairment induced by scopolamine. Hence, it can promote memory and learning ability. The formulation of the fruit and vegetable bark uses the extract of fruit and vegetable peels, which has long been used without causing toxicity in a wide range of food applications. Therefore, it can be used safely without a risk of side effects, even in the case of a long-term administration for the preventive purpose. Furthermore, the present study is a very beneficial study in the environment-friendly aspect in association with recycling of resources as it is based on the novel efficacies of fruit and vegetable barks which have been conventionally disposed as a refuse of fruits due to their poor sensory qualities despite the content of beneficial substances.
기능성 소화불량증 환자의 피로도에 대한 체표 교감신경 활성 및 심혈관 반응의 연계성 (양도락과 맥진검사의 진단적 가치)

Background & Object: The aim of this study was to investigate the association of degree of fatigue and gastric motility, measured by EGG, with skin sympathetic tone or cardiovascular reactivity in patients with functional dyspepsia. Methods: Subjects were 56 patients with Functional dyspepsia and eight healthy people. Degree of fatigue was assessed by questionnaires consisting of subjective complaints of fatigue. Skin sympathetic tone was measured by Ryodoraku Score and Cardiovascular Reactivity was checked by Pulse diagnostic apparatus. Gastric motility was estimated by EGG. First, all patients were divided into two groups by Ryodoraku Score $40{\mu}A$(below and above). Second, they were subdivided into two groups by Cardiovascular Reactivity(decreased and increased or not decreased). Estimates were made on the extent differences of degree of fatigue or state of gastric motility in each group. Results: 1. Fatigue scores was significantly higher in females and in the Ryodoraku-Score-below-$40{\mu}A$ group. It was higher in the decreased cardiovascular reactivity group than the increased group, but to no significanct extent. Also, gastric motility was better in the Ryodoraku-Score-above-$40{\mu}A$, group than in the below group. Conclusions: These results suggest that degree of fatigue and gastric motility are associated with skin sympathetic tone, but not associated with cardiovascular reactivity, and that $40{\mu}A$ is a useful cutoff point in Ryodoraku Score for assessing degree of fatigue in functional dyspepsia patients.
