      • Glutamate 억제제가 Pilocarpine에 의한 경련 및 기억장애에 미치는 영향

        목적 : Pilocarpine에 의한 경련 및 기억력 저하에 glutamate 수용체가 관련이 있는지 검토하였다. 대상 및 방법 : 생쥐에 pilocarpine을 투여하여 야기되는 경련 및 기억력저하가 glutamate 수용체 길항 약물인 MK-801 및 NBQX를 전처치하였을 때의 반응을 Morris Water Maze의 방법으로 관찰하였다. 결과 : MK-801과 NBQX를 전처치하고 pilocarpine을 투여하였는 바 pilocarpine에 의한 경련유발은 억제되지 않았으나, pilocarpine에 의한 reference memory와 working memory의 저하는 MK-801이나 NBQX를 처치로서 억제되었다. 결론 : Pilocarpine에 의한 경련작용에 비하여 기억력저허가 glutamte 수용체와 관련이 있는 것으로 보인다. The role of glutamate receptors in the genesis of pilocarpine-induced seizure and seizure-related memory deficit was studied using MK-801 and NBQX. After pretreatment with MK-801 or NBQX, pilocarpine was administered intraperitoneally. Both drugs had no effect on the behavioral seizure activity and onset of seizure activity. Memory was tested with reference and working memory version of Morris water maze at 20th day after pilocarpine treatment. Reference and working memory deficit was produced by pilocarpine. These deficit were prevented by MK-801 or NBQX, which were given in once a day for 4 day at 7th to 10th days after pilocarpine treatment. These results show that although MK-801 and NBQX do not have anticonvulsant property they can prevent pilocarpine-induced memory deficit.

        EFFECTS OF LIDOCAINE ON SOMATOSENSORY EVOKED POTENTIALS IN RAT VIBRISSA/BARREL CORTEX

        본 실험은 삼차신경 자극으로 발생되는 체성 감각 유발 전위에 대한 국소마취제의 효과를 관찰하였다. 나트륨 통로차단을 통하여 약리작용을 나타내는 것으로 알려져 있는 리도카인를 뇌 피질에 국소 투여한 후 삼차신경의 체성 감각유발 전위의 강도및 지연시간을 측정하였다. 케타민으로 마취된 흰쥐의 대측성 구레나룻 자극후 뇌의 체성 감각영역으로부터 기록되는 유발전위를 분석한 결과, 리도카인을 뇌 피질에 국소 투여시 유발전위의 강도 및 지연시간의 감소가 나타났으며, 필드 전위의 형태는 이상성(양극성 및 음극성) 혹은 삼상성(양극성, 음극성 및 양극성)의 파형으로 나타났다. 필드 전위의 발생 부위는 뇌 피질의 중대뇌동맥의 상행지 상방영역이었다. 본 실험에서 나타난 초기 전위변동은 피질판 상층에 존재하는 신경세포의 탈분극 과정에 의하여 생성되고 후기 전위 변동은 피질판 상층에 존재하는 신경세포의 탈분극 과정에 의하여 생성되고 후기의 전위 변동은 동일 영역의 하층 신경세포에서 과분극 혹은 재분극이 발생한 결과라고 유추된다. 따라서 삼차신경계의 체성 감각 영역에서는 피질 상층및 하층의 과립성 피라미드 세포의 순차적인 활성화에 의하여 기본적인 신경 회로망이 형성되어 있으며 생리적 자극으로 유발되는 필드 전위는 이러한 신경망를 통하여 발생될 것으로 사료된다.

        Perforated reinforced concrete beams for the passage of service ducts and piping can be minimized the story height and saved considerable materials and construction costs. With the lass section in beam, the reduction of reinforced concrete beam strengh and rigidity must be considered. Until now, shear test effects on reinforced concrete beams with web opening are more common than flexural test effects. Therefore, this laboratory programme was carried out on the perforated reinforced concrete beam subjected to pure bending under central loading. Tests were conducted on 9 beams to investigate deformation, cracking and strengh capacity of the perforated reinforced concrete beam and test resets were tabulated and analyzed comparing with those of solid beam.

        해마 theta리듬과 pyramidal neuron의 세포내 특성과의 상관관계

        Eledctrophysiological phenomena of pyramidal cells in the CAI area of the dorsal hippocampus were recorded from and filled with neurobiotin in anesthetized rats. The electropharmacological properties of membrane as well as the cellular-synaptic generation of rhythmic slow activity (theta)were examined. The intracellular response characteristics of these pyramidal cells were distinctly different from responses of inteneurons. Pyramidal cells had a high resting membrane potential, a low input resistance, and a large amplitude action potential. A afterhyperpolarization was followed a single action potential. Most of pyramidal cells did not display a spontaneous firing. Pyramidal cells displayed weak inward rectification and anodal break excitation. The slope of the frequency-current relation was 53.4 Hz/nA for the first interspike interval and 15.9 Hz/nA for the last intervals, suggesting the presence of spike frequency adaptation. Neurobiotin-filled neurons showed pyramidal morphology. Cells were generally bipolar dendritc processes ramifying in stratum lacunosum-moleculare, radiatum, and oriens. Commissural stimulation discharged pyramidal cells, followed by excitatory and inhibitory postsynaptic potentials (EPSPs and IPSPs). The frequency of theat-related membrane potential oscillation was voltage-independent in pyramidal neurons. At strong depolarization levels (less than 30 mV) pyramidal cells emitted sodium spike oscillation, phase-locked to theta. The observations provide direct evidence that theta-related rhythmic hyperpolarization of principal cells is brought by the rhythmically discharging interneurons. Furthermore, the findings in which interneurons were also paced by rhythmic inhibitory postsynaptic potentials during theta suggest that they were periodically hyperpolarized by their GABAergic septal afferents.

        여수시 관광개발 활성화 중요도 분석

        Taking the Yeosu Expo 2012 as the opportunity, there had been the inflow of the population into the Yeosu City and a landmark of the city had been formed. And, by leading to the visits by the tourists from Korea and overseas, the city has been solving the limitations of the region and the urban tasks called the invigoration of the finance of the local government and of the economy of the regional residents. Yeosu had been able to pull up the social infrastructure and the social network that had fallen behind due to the holding of the marine expo within a short time period. And, through this, the Yeosu City had prepared a stepping stone for making the leap by one phase. As a result of an AHP analysis, firstly, regarding both the level of the general importance and the level of the general compensation importance, the item of the marine, waterside tourist destination appeared as the absolute priority. Therefore, it is judged that tourist destinations that have been proceeded with at the present and the tourist destinations that are scheduled to be proceeded with, including the Ungcheon Marina Port, the formation of a seashore yachting course, the building of a marine education institute for the youths, the expansion of the Dolsan Marine Ecology Center, the formation of a tourism belt linked with Dolsan, the formation of the Namdo Food Street, etc., must be formed early, and the efforts must be made for a plan for managing these. It has been understood that, as seen in the results of this research, regarding the tourism development of the Yeosu City, centered on the Yeosu City, the maintenance of the linkages and the relationships with Dolsan, Goheung, and the Suncheon City, which are the surrounding regions, is an important variable with regard to achieving the growth. Also, the attention is paid to achieving the invigoration of the regional society and the making of the individual s Yeosu City into an exclusive brand through the formation of a tourism belt in addition to the Namdo foods.

        벼 種間交雜 O. sativa L.×O. grandiglumis Prod.에서 戾交雜과 胚珠培養의 效率性

        For the successful production of F₁ plants and backcrossed progenies, various factors concerning seed-setting and plant recovery from ovules were evaluated in the interspecific cross between Hwaseongbyeo (AA) and O. grandiglumis (CCDD). Ovule culture time was quite critical for plant regeneration from cultured ovules with its highest rate at 15 days after pollination. Plants were not recovered from ovules less than 7 days or more than 21 days after pollination. Hormones applied to plants during booting stage significantly affected seed-setting and plant regeneration. For the seed-setting, GA₃ was most effective with single or combined treatments. 2, 4-D proved quite effective for plant regeneration, showing 80% regeneration rate in 2, 4-D 110ppm treatment and 85% in combined treatment, 2, 4-D 110ppm+NAA 50ppm+Kinetin 75ppm. When F₁ was used as female parent for backcross, seed-setting was quite low due to the severe shattering and abnormal embryo development. Non-clipping and successive pollination method was proved quite effective in preventing of shattering, resulting in elevated seed-setting and plant regeneration compared with the conventional clipping method. Embryo development was increased by successive backcross, reaching the highest rate of 6.3% at BC₃F₁ generation. The same trend was observed for plant regeneration rate, 2, 4% at BC₂F₁ and 100% at BC₃F₁ generation.

      • Network Analysis of Acupuncture Points Used in the Treatment of Low Back Pain

        <P><I>Background</I>. The appropriate selection of acupoints is fundamental to obtain a therapeutic effect from clinical acupuncture. <I>Objective</I>. Using a network analysis method, we investigated the acupoints that are combined to treat low back pain (LBP). <I>Methods</I>. To analyze the patterns of the combinations of acupoints, we used acupoint information from clinical trials to calculate the modified mutual information (MI) value, integrated these data, and visualized the network. <I>Results</I>. Based on the highest MI values, we found two different types of acupoint pairs used in the treatment of LBP: pairs of distant acupoints and pairs of local acupoints. Using modular analysis, we found that three acupoint modules were applied in the treatment of LBP: local acupoints, distant acupoints along the meridian, and distant acupoints based on the symptom differentiations. <I>Conclusion</I>. Using the modified MI technique, we provide a systematic framework for the acupoint combination network, and reveal how the technique of acupoint combination is used in the treatment of LBP. Application of this knowledge in acupuncture research may help clarify the mechanisms underlying acupuncture treatment at the systems level, bridging the gap between traditional medicine and modern science.</P>

      • RESEARCH PAPER : Docetaxel-Loaded Thermosensitive and Bioadhesive Nanomicelles as a Rectal Drug Delivery System for Enhanced Chemotherapeutic Effect

        PURPOSE: To investigate the potential of thermosensitive and biadhesive nanomicelles in improving the bioavailability of docetaxel (DCT) and its chemotherapeutic effect. METHOD: DCT-loaded nanomicelles were prepared by emulsufication and characterized in terms of physico-chemical and visco-elastic parameters. The optimzed formulation was evaluated for in vivo localization, pharmacokinetic and anti-tumor efficacy. RESULTS: The hydrodynamic size of DCT-loaded nanomicelles was approximately 13 nm and thenanomicelles exhibited a sufficient gelation strength (9250 mPa·s) and bioadhesive force (2100 dyn/cm²) to be retained in the upper part of rectum. We observed a high rectal bioavailability of 29% DCT compared to that following oral administration in rats, as it successfully evaded the multidrug efflux transporters and hepatic first-pass metabolism. Plasma concentration around ∼50 ng/mL was maintained throughout the study period (12 h) while Taxotere® attained subtherapeutic range within 4 h of drug administration. Results also revealed that the rectally administered DCT-loaded nanomicellesexhibited a significant anti-tumor effect (200 mm³) with a reduced toxicity profile when compared to orally administered DCT (950 mm³). Furthermore, histological study showed that the rectal mucosa was completely intact with no signs of irritation upon treatment with DCT-loaded nanomicelles. CONCLUSIONS: Taken together, our novel thermosensitive and biadhesive nanomicellesdemonstrated the ability to improve the bioavailability and chemotherapeutic potential of DCT in vivo. To the best of our knowledge, this is the first report describing the rectal delivery of DCT-loaded nanomicelles.
