The purpose of this research was to investigate the mechanism of inhibitory action of Glycyrrhizin(GZ) on the proliferation of mouse leukemia cell-line, L1210 cells. The cytotoxic activity was tested using a colorimetric tertrazolium assay(MTT assay), the apoptosis was tested using flow cytometry. Nitric oxide(NO) production form mouse peritoneal macrophage was tested using a Griess method and the phagocytosis of human polymorphonuclear cells was tested using a lucigenin chemiluminescence. GZ ingibited the proliferation of L1210, BALB/c 3T3 cells and mouse thymocytes at 50 ug/ml/ GZ did not affect nitric oxide production from mouse peritoneal macrophages in vitro, but ingibited nitric oxide production from lipopolysaccharide and y-interferon treated macrophages. Macrophages of GZ-administered mice accelerated NO production. The proliferation of L1210 cells apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. The apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. GZ increased the phagocytosis of human polymorphonuclear cells. These results suggest that GZ inhibit the proliferation of L1210 cells not only as a direct cytotoxic agent o tumor cells, but also by the enhancement of NO production and phagocytic activity.

L-Theanine, found in green tea leaves has been shown to positively affect immunity and relaxation in humans. There have been many attempts to produce L-theanine through enzymatic synthesis to overcome the limitations of traditional methods. Among the many genes coding for enzymes in the L-theanine biosynthesis, glutamylmethylamide synthetase (GMAS) exhibits the greatest possibility of producing large amounts of production. Thus, GMAS from Methylovorus mays No. 9 was overexpressed in several strains including vectors with different copy numbers. BW25113(DE3) cells containing the pET24ma::gmas was selected for strains. The optimal temperature, pH, and metal ion concentration were 50℃, 7, and 5 mM MnCl<sub>2</sub>, respectively. Additionally, ATP was found to be an important factor for producing high concentration of L-theanine so several strains were tested during the reaction for ATP regeneration. Baker’s yeast was found to decrease the demand for ATP most effectively. Addition of potassium phosphate source was demonstrated by producing 4-fold higher L-theanine. To enhance the conversion yield, GMAS was additionally overexpressed in the system. A maximum of 198 mM L-theanine was produced with 16.5 mmol/l/h productivity. The whole-cell reaction involving GMAS has greatest potential for scale-up production of L-theanine.

The purpose of this research was to investigate effects of Sa-Kunja-Tang(SKT) on immune cells of L1210 cell-transplanted mice. The apoptosis and T lymphocytes subopoulation were tested using a flow cytometry, and the proliferation was tested using a MTT assay. Nitric oxide production from mouse peritoneal macrophage was tested using a Griess reagents, and the phagocytic activity of mouse peritoneal macrophage was tested using a lucigenin chemiluminescence. SKT suppressed apoptosis of T-lymphocytes induced by L1210 transplantation. SKT decreased nitric oxide production from mice peritoneal macrophages increased by L1210 transplantation, and the phagocytic activity decreased by L1210 transplantation. These results suggest that SKT suppresses T lymphocyte apoptosis and macrophage activity in L1210 transplanted mice.

Background: Diagnosis and management of neuropathic cancer pain (NCP) is diffi cult and complicated. This study investigated current practice in the diagnosis and management of NCP and its impact on quality of life (QoL). Methods: Patients with cancer pain with visual analogue scales (VAS) =1 from 28 hospitals in Korea were enrolled in this cross-sectional observational study. If the patient obtained a score =4 from neuropathic pain diagnostic questionnaire (DN4), he/ she was diagnosed with NCP. The clinical characteristics, pain severity measured by Brief Pain Inventory-Short Form (BPI-SF), and the QoL measured by EuroQoL EQ-5D Health Questionnaire were evaluated. Results: Among 1,505 enrolled patients, 885 patients (58.8%) had moderate to severe pain (VAS=4). Neuropathic cancer pain (when DN4 =4) was identified in 556 patients (36.9%). Among those NCP patients, 254 patients (45.7%) were treated with NCP-targeted therapies (administering opioid or non-opioids analgesics with adjuvant analgesics). Among entire patients, opioids were used in 1,005 patients (66.8%) and 205 patients (13.6%) were not prescribed any analgesic. Anti-depressants and anti- convulsants were prescribed for 91 (6.0%) and 349 (23.2%) patients respectively. Patients with NCP had more severe pain and their pain interference level with daily living was higher than those without NCP. The QoL measured by EQ-5D index score was signifi cantly worse in patients with NCP than those without NCP (Table 1). Conclusions: This large-scaled survey shows that NCP has deteriorative effect on pain and QoL in patients having cancer pain. Current status of management for cancer pain is not satisfactory including NCP in Korea. *This study was partially presented at the MASCC/ISOO International symposium on supportive care in cancer, 26 - 28 June 2014, Miami, USA.
PEX5 regulates autophagy via the mTORC1-TFEB axis during starvation

<P> Defects in the PEX5 gene impair the import of peroxisomal matrix proteins, leading to nonfunctional peroxisomes and other associated pathological defects such as Zellweger syndrome. Although PEX5 regulates autophagy process in a stress condition, the mechanisms controlling autophagy by PEX5 under nutrient deprivation are largely unknown. Herein, we show a novel function of PEX5 in the regulation of autophagy via Transcription Factor EB (TFEB). Under serum-starved conditions, when PEX5 is depleted, the mammalian target of rapamycin (mTORC1) inhibitor TSC2 is downregulated, which results in increased phosphorylation of the mTORC1 substrates, including 70S6K, S6K, and 4EBP-1. mTORC1 activation further suppresses the nuclear localization of TFEB, as indicated by decreased mRNA levels of TFEB, LIPA, and LAMP1. Interestingly, peroxisomal mRNA and protein levels are also reduced by TFEB inactivation, indicating that TFEB might control peroxisome biogenesis at a transcriptional level. Conversely, pharmacological inhibition of mTOR resulting from PEX5 depletion during nutrient starvation activates TFEB by promoting nuclear localization of the protein. In addition, mTORC1 inhibition recovers the damaged-peroxisome biogenesis. These data suggest that PEX5 may be a critical regulator of lysosomal gene expression and autophagy through the mTOR-TFEBautophagy axis under nutrient deprivation. </P>

Background: Pioglitazone is known to improve insulin sensitivity, glycemic control in patients with T2DM. In spite of these advantages of pioglitazone, there are also concerns about the risk of adverse events. In practice, the usual initial and maintaining dosages of pioglitazone are 15mg although the maximal recommended dosage is 30mg in Korea. we planned to analyze the ef. cacy and safety of pioglitazone when dose was up-titrated to 30mg in subjects with type 2 diabetes mellitus with inadequate glycemic control on 15mg of pioglitazone retrospectively. Methods: This is a single-arm, retrospective, observational study. Subjects (45 diabetes patients) must had been on a stable dose of 15 mg of pioglitazone for at least 3 months before dose titration to be eligible to enter the study. The combined treatments with stable doses of metformin or/and sulfonylurea will be allowed. Results: HbA1c were decreased in 3 months and 6 months after 10mg pioglitazone treatment (HbA1c, base line :8.4 ± 0.84 vs 3 months : 8.1 ± 0.8 vs 6 months : 7.8 ± 0.8, P < 0.01). Body Weight increased in 3 months and 6 months after 10mg pioglitazone treatment. ( n = 44, Body weight, base line : 71.3± 7.9 vs 3 months : 70.8 ± 7.7 vs 6 months : 71.3 ± 8.2, P < 0.01). Conclusions: Korean diabetes patients with 10mg pioglitazone use showed favorable metabolic effect for glycemic control, but dose up titration of pioglitazone increased the disadvantage of increasing the weight.

Hand-foot-and-mouth disease, a highly contagious viral infection, occurs more common in children than in adults. However, there was a recent outbreak of Coxsackievirus A6-induced infection with an atypical presentation among the adult population. Stevens–Johnson syndrome is a severe mucocutaneous disease characterized by extensive necrosis and detachment of the epidermis, and this condition is commonly caused by medications. Herein, we describe a 30-year-old male patient taking allopurinol for the management of gout. The patient presented with numerous erythematous papules, vesicles, and patches with mucosal eruptions on the whole body, oral mucositis, and fever, and he was finally diagnosed with hand-foot-and-mouth disease.

A series of four sodium α-sulfo fatty vinyl ester oligomers including sodium α-sulfo lauric acid vinyl ester oligomer, sodium α-sulfo myristic acid vinyl ester oligomer, sodium α-sulfo palmitic acid vinyl ester oligomer and sodium α-sulfo stearic acid vinyl ester oligomer were examined for surface activities such as surface tension, foaming power, foam stability, emulsifying power, dispersion effect, solubilization of orange OT. The critical micelle concentration(CMC) was also evaluated. Consequently, these sodium α-sulfo fatty acid vinyl ester oligomers were shown to have a good cohesive power and dispersion effect.
