        Lead acetate and Bojungiki-tang(BIT) extract, a herbal restorative were treated p.o. 40mg/kg and 500mg/kg respectively and concurrently. and examined efects of metabolic enzymes of rats. The results obtained in this study are as follows: 1. The value of protein concentration in lead-poisoned group showed significant increse while the group of concurrent administration with BIT whowed slight decrease. 2. The value of aniline hydroxylase activities in lead-poisoned group showed 32.37% of increse while the group of concurrent administration with BIT showed 3.91% at 4th week. 3. The value of TBA-value in lead-poisoned group showed 66.7% of incresed while the group of concurrent adiministration with BIT whowed 7.72% at 4th week. 4. The value of glucose-60phosphate activities in lead-poisoned group showed a little decrease. These results on the whole indicated that concurrent administration with BIT showed significant recovery from the toxicity in lead-treated group.

      • 玄胡索중 3級鹽基成分의 分離와 定量에 관한 硏究

        A new application of specrophotometry for the isolation and determination of tertiary base in Corydalis Tuber was investigated. In order to establish the method for isolation and determination of tertiary base in the Corydalis Tuber. Tertiary base was isolated from Corydalis Tuber by organic solvent. When tertiary base was added to a solution containing cobaltous ion and excess thiocyan ion, yellowish-green, 1,2-dichloroethane-solube(base)_2Co(SCN)_4 precipitate. Supportive evidence for identification could be obtained by complexation with tetrathiocyanatocobaltate(Ⅱ) ion. The ultraviolet-visible spectrum of complex vompound in 1,2-dichloroethane exhibits maxima at 282nm and 624nm. The tertiary base was isolated from compex compound and identified as glaucine by UV-Visible spectrum and TLC. A tertiary base in 1,2-dichloroethane scanned from 200nm to 400nm qualitatively exhibits the same absorbance characteristics at identical wavelength as does isolated and concomitantly measured solution of a complex compound. The tertiary base components present in comples compound have been resolved on an absorbance solvent. And it is studied that the differential determination of tertiary base in Corydalis Tuber by spectrophometry. The tertiary base is determined by absorbance of colored products that is produced to reaction of tetrathiocyanatocobaltate(Ⅱ) ion under determination of tertiary base was possible to the range of glaucine 0.100mg/ml-1.000mg/ml in 1,2-dichloroethane. (maximum wavelength; 624nm) The calibration curve showed that the coefficient of variation was 0.9999% for glaucine. Also reproducibility tests showed that the coefficient of variation was 0.9999%. Arepid, specific and precise method using UV-Visible spectrophotometry and TLC has been developed for the isolation and determination of tertiary base in the Corydalis Tuber. The method was applicable to the isolation and determination of tertiary base in Corydalis Tuber or it's extract.

      • Berberine 含有生藥의 β-cyclodextrin 포접화합물의 生體利用率에 관한 硏究

        To increase the bioavailability of berberine inclusion complex were prepared and studied in this experiment. Inclusion complexes of berberine(BER) and methanol extract of Coptis Rhizomia(MEX) with B-cyclodextrin were prepared. The solubilities of BER and MEX were 0.14% and 1.79%, while those of BECD and MECD were 0.36 and 3.22% respectively. The lipid-water partition coefficients of Ber-B-CD indlusion complex(BECD) and MEX-B-CD inclusion complex(MECD) were more increased than BER and MEX respectively. The absorption rates of BECD and MECD in stomach, duodenum, and jejunum of rats were compared with those of BER and MEX, which were more increased than BER and MEX respectively. The times required for the maximum serum concentration fo berberine from BER, MEX, BECD and MECD in mice were 30 minutes after oral administration. The maximum serum concentrations of berberine from BECD and MECD were higher than those from BER and MEX respectively. BECD was shown more increased antibacterial activity to all the gram positive bacteria, gram begative bacteria and mold than BER,MECD was shown more increased antibacterial activity to only S. epidermidis, K. pneumoniae, E. coli than MEX. The dissolutions of BECD and MECD were higher than BER and MEX respectiely. Therefore, it suggest that preparing of inclusion complex brings to prevention of coprecititated formation and improvement fo bioavailability of berberine.

      • 정향 수침액에 의한 아나필락시의 억제 효과

        The current study evaluates the capacity of Caryophylli Flos water extract(CFWE) to regulate immediate allergic reaction by control of histamine release. CFWE was found to exhibit a ingibitory activity on the compound 48/80-induced anaphylaxis. CFWE inhibited the serum histamine release induced in anaphylaxis by compound 48/80. CFWE was also showed a dose-depent activity on the conpound 40/80-induced histamine release from the highly purified population of Alcian Blue-positive peritoneal mast cells. This result suggest that CFWE may be useful for the prevention and treatment of allergy related disease.

      • 형광검출 역상 액체크로마토그래피에 의한 Histamine의 정량

        Histamine을 정확하고 신속하게 정량하기 위해 9.Fiuorenylmethyl chloroformate를 형광유도체화제로 하여 역상 HPLC법으로 정량하였다. 히스타민을 형광유도체화할 때 반응액의 pH, 반응시간, 형광유도체화제의 농도 등 최적 반응 조건을 검토하였다. 이 방법으로 히스타민을 분석한 결과 0.1~0.5ug/ml의 농도범위에서 상관계수가 0.922인 양호한 직선성을 나타내었으며 검출한계는 0.01??/ml였다. A rapid and simple method for the determination of histamine by reverse-phase high performance liquid chromatography with fluorescence detection was established. 9-Fluorenylmethyl chloroformate(FMOC) was used as fluorescent derivative reagent. The optimum conditions for the derivatiation such as pH, reaction time and he concentration of FMOC were investigated. Linearity of calibration curve was obtained between 0.1ug/ml/and 0.5ug/ml(r=0.922) and the limit of detection was 0.01ug/ml.

        A high performance liquid chromatographic method developed for the determination of polyamines(putrescine, spermidine and spermine) in the serum of normal human and cancer patients. The benzoyi chloride derivatives of polyamines are separated on a uBondapak C_18 reverse-phase column with methanol/water(50v/50v) as the mobile phase. The results show that the mean level of polyamines in concer patients serum is much higher than that in normal human serum.

      • 사군자탕이 항암제를 투여한 마우스의 면역세포에 미치는 영향

        The purpose of this research was to investigate effects of Sa-Kunja-Tang(SKT) on immune cells of antitumor drugs administered mice. The apoptosis and T lymphocytes subpopulation were tested using a flow cytometry, and the proliferation was tested using a MTT assay. The administration of etoposide. vincristine or doxorubicin increased the apoptosis of T-lymphocytes, but the action of doxorubicin was decreased by the administration of SKT. The administration of etoposide or vincristine decreased helper T and cytotoxic T cells population of T lymphocytes, but the action of vincristine was recovered by the administration of SCT. The administration of etoposide, vincristine or doxorubicin decreased the proliferation of T-lymphocytes, but the action of doxorubicin was increased by the administration of SKT. These results suggest that SKT has a regulative function of T-lymphocytes in anti-tumor drugs administered mice.

      • 黃蓮解毒湯의 製劑化 및 品質管理에 관한 硏究

        A traditional prescription, Hwangryean-haedog-tang(HRHT) was composed of Coptidis Rhizoma, Phellodendri Cortex, Scutellariae Radix, Gardeniae Fructus(1.5:1.5:3.2) and it has been used in the treatment of hypertension. insomnia, mental disorder, medicinal poisoning, crapulence. The precipitation was formed during the preparation of decoction of HRHT. This precipitatiion was very slightly soluble in water. Because this precipitated reaction decrease the effect of a gerbal medicine, the content of berberine and baicalin was determined in the precipitated product and supernatant. Berberine which active ingredient of Coptidis Rhizoma, Phellodendri Cortex was identified and determined by HPLC using u-Bondapak C_18 column with acetonitrile:pH 5.2 phosphate buffer(60:40v/v). Baicalin which active ingredient of Scutellariae Radix was identified and determined by HPLC using u-Bondapak C_18 column with 0.5% phosphoric acid:acetonitrile(73:27v/v). The content of erberine in the coprecipitated product and supernatant were 11.6% and 1.5% and the content of baicalin were 20.7% and 2.4% respectively. To ingibit the precipitated reaction of HRHT. Coptidis Rhizoma and Phellodendri Cortex were mixed and extracted whth hot water, it were made into gastric-coated granule, Scutellariae Radix and Gardeniae Fructus mixed and extracted, it were made into enteric0coated granule.

      • Effect of Biphenyl Dimethyl Dicarboxylate on the Immunosuppression of Ketokonazole(Ⅰ)

        The present study was undertaken to investigate the effect of biphenyl dimethyl dicarboxylate (PMC) on the immunosuppression of ketoconazole in ICR mice. PMC at dose of 6 mg/kg was administered orally to the mice daily for 14 consecutive days. Ketoconazole was suspended in RPMI 1640 medium and also orally administered at 160 mg/kg/day 2 hrs after the administration of PMC. Mice were immunized and challenged with sheep red blood cells (SRBC). The results of the present study are summarized as follows: 1. A gain of body weight and the relative weights of spleen and liver were significantly increased by the combination of PMC and ketoconazole, as compared with those in mice treated with ketoconazole alone. 2. Splenic plaque forming cells (PFC), hemagglutination (HA) titers and delayed-type hypersensitivity (DTH) reaction to SRBC were greatly enhanced by the combination of PMC and ketoconazole compared with treatment of ketoconazole alone. These values were similar to those of control mice.

      • L1210 세포증식에 대한 Glycyrrhizin의 억제작용 기전

        The purpose of this research was to investigate the mechanism of inhibitory action of Glycyrrhizin(GZ) on the proliferation of mouse leukemia cell-line, L1210 cells. The cytotoxic activity was tested using a colorimetric tertrazolium assay(MTT assay), the apoptosis was tested using flow cytometry. Nitric oxide(NO) production form mouse peritoneal macrophage was tested using a Griess method and the phagocytosis of human polymorphonuclear cells was tested using a lucigenin chemiluminescence. GZ ingibited the proliferation of L1210, BALB/c 3T3 cells and mouse thymocytes at 50 ug/ml/ GZ did not affect nitric oxide production from mouse peritoneal macrophages in vitro, but ingibited nitric oxide production from lipopolysaccharide and y-interferon treated macrophages. Macrophages of GZ-administered mice accelerated NO production. The proliferation of L1210 cells apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. The apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. GZ increased the phagocytosis of human polymorphonuclear cells. These results suggest that GZ inhibit the proliferation of L1210 cells not only as a direct cytotoxic agent o tumor cells, but also by the enhancement of NO production and phagocytic activity.
