Cholesterol is an essential component for maintaining health; however, excessive consumption can lead to diseases. Thus, continuous monitoring of cholesterol content is important in food research. The cholesterol analysis method used in Korea follows the Korean Food Standards Codex. As this method uses gas chromatography, derivatization of the sample is required, and analysis time is more than 30 min. Kim developed a new method using liquid chromatography; however saponification by the non-heat saponification method is insufficient. To address these limitations, a new cholesterol analysis method was developed and verified. The correlation coefficients for the cholesterol standard (STD) were maintained above 0.99. The limit of detection and limit of quantitation of cholesterol STD were 2.41 μg/mL and 7.31 μg/mL, respectively. The accuracies for cholesterol were 92.21-99.02%. The developed analytical method was also verified using three standard reference materials, and their accuracies were 93.71-97.09%. In addition, the cholesterol content of fishes was analyzed, and the values were compared with those recorded by the United States Department of Agriculture. Our results suggest that this method could be used as a new analytical method for cholesterol in seafood.

This study was performed to provide c1arity on the environmental distribution of Vibrio vulnificus oysters, biological characteristics, distribution of biotypes and serotypes, the detection of the cytolysin gene by PCR, repetitive sequence-based PCR (rep-PCR) analysis and antimicrobial susceptibility of the isolates from oysters and patients in Korea. ν vulnificus was isolated from 67 (38.5%) of the 175 samples of oysters collected at a shoreline in the Kunsan city area in July and August, 2002. Oyster and patient isolates had similar biochemical characteristics, i.e., all were positive on an ONPG test, but not for sucrose. When comparing the identification of V. vulnificus obtained with the API 20E assay and serotyping, 87 API 20E profiles with both a separation test on the colony of TCBS agar and an ONPG test were obtained with 84 profiles (96.6%), but three isolates were not identifiable with the API 20E system. API 20E code numbers showed 13 different biotypes in 87 isolates. The lrighest rate of 48 (55.2%) isolates of API 20E code numbers was obtained with code No. 5146105 (biotype N). Oyster is이ates belonged to tlrirteen serotypes and patient strains belonged to 9 serotypes. Prevalent serotypes were 012, and 04A, 16 (18.6%) and 13 (15.1 %) from oysters, and 52 (67.5%) and 7 (9.1%) of 04A and 04AB from patients, respectively. 163 isolates of V. vulnificus cytolysin gene from oysters and patients were positive by PCR. 30 V. vulnificus isolates were divided into 5 genotypes using rep-PCR analysis. Only three bands have all of the isolates from the oysters by rep-PCR. on an antimicrobial susceptibility test they were susceptible to tetracyc1ine, chloramphenicol and ciprofloxacin, but none were susceptible to colistin.

<P>In this study, we investigated the effects of the potential probiotic Bacillus subtilis CSY191 on the fatty acid profiles of Cheonggukjang, a fermented soybean paste, prepared using new Korean brown soybean cultivars, protein-rich cultivar (Saedanbaek), and oil-rich cultivar (Neulchan). Twelve fatty acids were identified in the sample set-myristic, palmitic, palmitoleic, stearic, oleic, vaccenic, linoleic, alpha-linolenic, arachidic, gondoic, behenic, and lignoceric acids-yet, no specific changes driven by fermentation were noted in the fatty acid profiles. To further explore the effects of fermentation of B. subtilis CSY191, complete profiles of volatiles were monitored. In total, 121, 136, and 127 volatile compounds were detected in the Saedanbaek, Daewon (control cultivar), and Neulchan samples, respectively. Interestingly, the content of pyrazines-compounds responsible for pungent and unpleasant Cheonggukjang flavors-was significantly higher in Neulchan compared to that in Saedanbaek. Although the fermentation period was not a strong factor affecting the observed changes in fatty acid profiles, we noted that profiles of volatiles in Cheonggukjang changed significantly over time, and different cultivars represented specific volatile profiles. Thus, further sensory evaluation might be needed to determine if such differences influence consumers' preferences. Furthermore, additional studies to elucidate the associations between B. subtilis CSY191 fermentation and other nutritional components (e.g., amino acids) and their health-promoting potential are warranted. Background: The usefulness of additional examination of Primovist-enhanced magnetic resonance images (MRI) during initial staging workup in hepatocellular carcinoma (HCC) patients who are candidates for curative treatment with no suspicious intrahepatic HCC lesion by liver dynamic computed tomography (CT) remains to be elucidated. Methods: A consecutive 104 patients who were initially diagnosed as having definite HCC without any suspicious HCC lesions by liver dynamic CT at our institution between December 2009 and May 2012 were enrolled. All of them showed Barcelona Clinic Liver Cancer (BCLC) stages 0 or A. We evaluated whether an additional examination of Primovist- enhanced MRI could affect on BCLC stage, directly comparing to liver dynamic CT. Results: Newly detected intrahepatic abnormal findings on Primovist-enhanced MRI were observed in 41 (39.4%) of the 104 patients. Eighteen (17.3%) patients had truly new HCCs, but 6 patients of initial BCLC stage A remained at this stage despite an increase in tumor number. Of the 104 patients, 31 (29.8%) and 73 (70.2%) had BCLC stages 0 and A HCC before Primovist-enhanced MRI, respectively, and 26 (25.0%), 71 (68.3%), and 7 (6.7%) patients had BCLC stages 0, A, and B HCC after Primovist-enhanced MRI, respectively. In 12 (11.5%) of the 104 patients, BCLC stage changed from 0 to A (5/31, 16.1%) or from A to B (7/73, 9.6%). Conclusions: Primovist-enhanced MRI can provide additional information regarding the detection of new intrahepatic HCC lesions during initial staging workup in patients with BCLC 0 or A stage, but with no suspicious HCC lesion by liver dynamic CT.
비교 유전자 보합법을 이용한 한국인 갑상선 유두상 암종에서의 유전자 변화에 관한 연구

Transformation and progression of cancers are thought to be caused by an accumulation of complex genetic alterations, but little is known about specific genetic changes. In this study, the authors have undertaken a genome-wide screening to detect changes in 12 cases of papillary thyroid carcinoma which is the most common subtype of thyroid cancer among Koreans. Comparative genomic hybridization (CGH) was used to screen for DNA sequence gains and losses across all human chromosomes. Biotin-labeled tumor DNA and digoxigenin-labeled normal DNA were hybridized to normal metaphase cells. The fluorescence signals were captured by fluorescence microscope after detection by avidin FITC and antidigoxigenin rhodamine. Then, the ratio of fluorescence was calculated by an image analyzer. The CGH results showed gains on chromosomes 1q(25%), 13q(25%), 17q(25%), 7p(17%), 18q(17%), 17p(8%) and 7q(8%). The Deletions were on chromosomes 22q(25%), 19p(8%), 19q(8%), 20p(8%), and 20q(8%). Usual amplification sites of genome were 1q, 13q, and 17q. The most common deletion site was 22q. The results indicate that the amplification on chromosome 1q, 13q, and 17q and the deletion on chromosomes 22q are the most frequent genetic alterations in papillary thyroid carcinoma among Koreans. The CGH was thought to be very useful in the screening of genetic alterations of solid tumors.
