        The Education of Multimedia in the kindergarten become more and more important to young children these days. However most educators didn't integrate multimedia activities to their other parts of the classroom. The purpose of this study is to recognize the importance of multimedia education for young children and promote teacher's positive role. Accordingly, this study examined computer activity, software, use of internet and the factors the influence teacher to integrate computer technology into the teaching and learning process. The results of this study are as follows, 1) Multimedia should be understood by the concept of multimedia computer and interacting environment. 2) The important thing in computer activity is not controversy about computer effects but how we connect computer activity into children's life well. 3) Developmentally appropriate software is essential part of successful computer activities. 4) Developmentally appropriate internet program may serve as an interesting, creative platform in the acquisition of general skills. 5) External factors and intern릴 factors should be considered together for early multimedia education and integrate multimedia activity to children's environment.

        유산균들의 콜레스테를 저하성, 내산성, 내담즙성, 항생제 내성 비교

        기능성 요구르트의 제조에 사용되는 유산균은 인체에 유익한 생리활성과 우수한 생존능력을 보유하고 있는 것이 바람직하다. 본 연구에서는 다양한 유산균주의 산과 담즙 및 항생제에 대한 내성을 조사하였으며 in vitro에서 콜레스테롤 저하능을 조사하였다. In vitro에서 콜레스테롤 저하능이 우수한 13균주를 선발하여 이들의 동결건조 분말을 고 콜레스테롤 식이의 실험쥐에 투여하였다. 그 결과 8균주는 비 투여구보다 유의적으로 18.3~27.3%의 콜레스테롤 저하능을 나타냈다(P≤0.05). 이들 중에서 Bifidobacterium infantis AM-220, Lactobacillus AM-245, Streptococcus MA-1의 3균주를 이용하여 요구르트를 제조한 후 동결건조 분말을 이용하여 급여실험을 수행한 결과, 총 콜레스테롤과 LDL 함량이 유의적으로 감소하는 것으로 나타났다. For a probiotic yoghurt it is desirable to utilize lactic acid bacteria with a high survival rate and beneficial function to human beings. We have examined a variety of lactic acid bacteria to assess the acid and bile tolerance and antibiotic resistance. In addition, an in vitro culture experiment was performed to evaluate their ability to reduce cholesterol levels in the growth medium. Thirteen strains were selected from in vitro cholesterol assays and fed to Sprague-Dawley rats with a high-cholesterol diet. Among the 13 strains tested, 8 strains were shown to reduce serum cholesterol levels significantly after 24 days of administration in vivo. Rats were fed lyophilized yoghurt powder fermented with a combination of 3 selected strains: Bifidobacterium infantis AM-220, Lactobacillus AM-245, and Streptococcus MA-1. The levels of total cholesterol and low density lipoprotein were significantly lower (p≤0.05) in rats fed the yoghurt powder compared with control group. These studies suggest that yoghurt fermented with appropriately selected lactic acid bacteria may have a anticholesterolemic effect.

        H9c2 심근 세포주에서 외인성 nitric oxide가 허혈에 의한 세포 독성에 미치는 영향

        Background: Nitric oxide(NO) is known to have protective effects on an ischemic heart and to exert triggering effects on ischemic preconditioning. However, the effects of NO during the ischemic period have not been investigated. To investigate the role of exogenous nitric oxide in a model of ischemic heart cell death, we studied the effects of ischemic preconditioning and ischemia in a normal and an ischemic buffer. Methods: Rat cardiac myoblast cells(H9c2) were cultured in a normal and an ischemic buffered medium. For the ischemic culture of heart cells, the cells were cultured in a dessicator with GasPak for 5 hrs. In ischemic preconditioning, the cells were pretreated with ischemic buffer for 5 min and then perfused with normal medium for 30 min. For the measurement of the cytotoxicity, a MTT(3-4-Sdimethylthiazol-2-yl-2,5-diphenyl tetrazolium bromide) assay was performed. A DAPI(4',6-diamidino-2-phenylindole dihydrochloride) staining procedure and a flow cytometry analysis were performed to confirm apoptotic cell death by ischemia. Results: Cell viability, as determined by using a MTT assay, showed that the preconditioned group treated with NO showed more cell death than with the not-preconditioned groups in both normal and ischemic buffers. But, In normal medium and not-preconditioned groups, NO showed protective effect according to the concentrations(100,1000μM) . No treatment with NO produced the different results. In normal medium, the protective effect of ischemic preconditioning was demonstrated, but no protective effect of ischemic preconditioning could be seen in the case of the ischemic buffer. The DAPI staining and flow cytometry analysis of heart cells showed characteristic apoptotic features. Conclusion: NO added in the ischemic phase had deterious effects on heart cells. Ischemic preconditioning was more harmful than ischemia alone. The toxicity of the cells was characteristic apoptosis.

      • Topical tacrolimus for the treatment of atopic dermatitis with truncal lesion

        Background: Topical tacrolimus is an effective anti-inflammatory therapy for acute and chronic state of atopic dermatitis (AD) in both adults and children. Topical tacrolimus has particular use at sensitive areas like face, anogenitals, and skin folds, where have greater adverse risk of topical corticosteroids. However, many AD patients experience aggravated symptoms in trunk as well as flexural areas. Objective: The aim of this study was to investigate efficacy and safety of topical tacrolimus for AD patients with truncal lesion. Methods: An open-label, single arm, multicenter (20 centers), observational study was conducted. AD patients (aged ≥ 2 years) with truncal lesion were applied topical tacrolimus ointment twice daily during 4 weeks. The primary efficacy end point was the change from baseline in local Eczema Area and Severity Index (EASI) of trunk at day 28. The secondary end points were the change from baseline in patient global assessment (PGA) score and itch VAS (visual analogue scale) of trunk at day 28. In addition, variations of local EASI, PGA, and itch VAS of trunk during study period were compared with those of whole body. All adverse events were recorded for investigation period. Results: Total 291 patients were recruited in this study, and 176 patients (99 males and 77 females, mean age: 18.3 years, range 2~59) completed the full 4-week treatment course. By the end of treatment, median local EASI of trunk (0.93±0.74) was significantly decreased than those at the baseline (1.74±0.8) ( p < 0.001). PGA score of trunk (1.71±1.15) and itch VAS of trunk (2.61±2.19) at day 28 were also profoundly decreased compared with baseline (2.96±1.07 and 5.15±2.47, respectively). The percent improvement from baseline in PGA and itch VAS of trunk was significantly greater compared with those of whole body (p=0.048 and p=0.013, respectively). There was no serious adverse events except local reactions including burning sensation (n=17), itching sensation (n=8), and erythema (n=1). Conclusion: Topical tacrolimus is an effective and safe therapy for truncal lesion of AD patients.

The secondary end points were changes in the patient global assessment (PGA) score and itch visual analogue scale (VAS) score of the trunk between baseline and day 28. Results: Two hundred and ninety- one patients were recruited, and 176 patients completed the full 4-week treatment course. By the end of the treatment, the mean local EASI of the trunk (2.2±4.71) was significantly decreased from that at baseline (4.71±4.03, p<0.001). PGA (1.71±1.15) and itch VAS score of the trunk (2.61±2.19) on day 28 were also profoundly decreased compared with the baseline (2.96±1.07 and 5.15±2.47, respectively). No serious adverse events were observed during the study period. Conclusion: Topical tacrolimus is an effective and safe therapy for truncal lesions in AD patients.       • Establishment of Baculovirus Infected Insect Cell Line Expressing Porcine Salivary Lipocalin(SAL1) Protein

        Salivary lipocalin (SAL1) is a member of the lipocalin protein family that has a property to associate with many lipophilic molecules. The importance of SAL1 during pregnancy in pigs has been suggested by our previous study which has shown that SAL1 is expressed in the uterine endometrium in a cell type- and implantation stage-specific manner and secreted into the uterine lumen. However, function of SAL1 in the uterus during pregnancy in pigs is not known. To understand SAL1 function in the uterus during pregnancy, we generated recombinant porcine SAL1 protein in an insect cell line. Porcine SAL1 cDNA was cloned into a baculovirus expression vector using RT-PCR and total RNA from uterine endometrium on day 12 of pregnancy, and the expression vector was used to generate recombinant Bacmid containing the SAL1 gene. The recombinant Bacmid was then transfected Sf9 cell to produce recombinant baculovirus. By infecting Sf9 cell with recombinant baculovirus, we established a SAL1-expressing insect cell expression system. Immunoblot analysis confirmed SAL1 expression in the infected cells. Recombinant SAL1 produced by the Sf9 cell line will be useful for understanding physiological function of SAL1 during pregnancy in pigs.

      • Platelet-activating factor-induced NF-κB activation enhances VEGF expression through a decrease in p53 activity

        <P><B>Abstract</B></P><P>We investigated the role of p53 in nuclear factor (NF)-κB dependent, platelet-activating factor (PAF)-induced vascular endothelial growth factor (VEGF) expression. Transfected NF-κB subunits in ECV304 cells increased the tumor necrosis factor-α promoter activity, which was completely inhibited by p53. Transfected p53 increased p53RE promoter activity, which was completely inhibited by NF-κB subunits, indicating that cross-regulation occurs between NF-κB and p53. PAF-induced increase in VEGF expression was correlated with decreased p53 activity. These data suggest that NF-κB-dependency of the PAF-induced increase in VEGF expression is due to decreased p53 activity, which is reciprocally regulated by increased NF-κB activity.</P>
