Molecular analysis of TMC1 gene in the Korean patients with nonsyndromic hearing loss

Hereditary nonsyndromic hearing loss (NSHL) is a highly heterogeneous disorder in humans. Mutations of the transmembrane channel‐like (TMC1) gene have been identified as the genetic cause for both autosomal recessive (DFNB7/11)and autosomal dominant (DFNA36) nonsyndromic hearing loss. To evaluate the spectrum and frequency of mutation(s)caused by TMC1 gene in the Korean population, we have performed sequencing analysis of the PCR products amplified from genomic DNA of each proband in 193 unrelated families showing 30 autosomal dominant and 163 autosomal recessive inheritance patterns. As a result, we identified eight different novel sequence variations for the first time in this study,respectively. However, none of these showed co‐segregation of phenotype in the families. Therefore, our study suggests that the TMC1 gene is not the cause of nonsyndromic hearing loss in the Korean population.
Analysis of eco-corridor usage status of mammals in Odaesan National Park

We investigated species, ecological pathway type, and mammal size using two eco-tunnels from trail-camera data. The analyses of the camera data showed that 12 mammalian species from 8 families of 5 orders used Jin-gogae eco-corridor (bridge type) and 14 mammalian species from 9 family 5 orders were found at Odaesan eco-corridor (tunnel type). At both eco-corridors, water deer used eco-corridor most frequently, and then secondly racoon dog did. The analyses by mammal size (small, medium, and large) showed that large-sized mammals were found in high ratio at the bridge type of the eco-corridors. In case of medium-sized mammals, utilization ratio of the tunnel-typed corridor was 42%, whereas that of the bridge-typed corridor was 24%, showing 1.8 times higher usage.
Analysis of monitoring Road-kill in Odaesan National Park

We surveyed mammalian roadkill at the National Route 6 (Jin-gogae to Songcheon, 18㎞) and a local road 446 (Odaesan National Park office to Sangwon-temple, 12㎞) in Odaesan National Park. Data of the surveys were analyzed by year, month, and species. Roadkills of 226 individuals were found at the survey areas. Of them, the road-killed 131 mammal individuals, 44 bird individuals, 40 reptile individual and 11 amphibian individuals were found during the survey period. High ratio of the road-kills was observed in mammals than the other animals. Among mammalian species, high ratio of the road-kills was observed in squirrels. There were road-kills of three endangered species, containing otters, yellow-throated marten and leopard cats. According to the analyses of annual road-kills, roadkills of 104 cases were occurred and highly frequent in 2014, following by roadkills of 81 cases in 2015, and then those of 41 cases in 2016. Most ases of the roadkills occurred in summer (June to September). The results will contribute to establishing management for reduction of roadkill in Odaesan National Park.
쇼펜하우어 의지론의 도덕교육적 함의 : 불교 수행론과의 관련에서

‘칸트의 물자체는 바로 의지’라고 요약되는 쇼펜하우어의 철학은 칸트 철학의 기반 위에서 시작되는 것이지만, 쇼펜하우어는 자신의 ‘의지론’을 통하여 ‘물자체’와 ‘선의지’를 중심으로 하는 칸트 철학의 입장을 수정한다. 칸트의 인식론과 윤리학은 인식과 도덕적 실천의 ‘논리적 조건’을 밝히는 과정을 보여준다는 점에서 메타피직스의 전형에 해당한다. 그러나 인식론과 윤리학이 그것을 읽는 사람의 도덕적 성장을 꾀할 수 있으려면 그것들은 인식과 도덕에 관한 ‘형식적 설명’ 이상으로 양자의 관련과 이상적 상태에 도달하기 위한 노력을 구체적으로 다룰 수 있어야 한다. 쇼펜하우어의 의지론은 칸트가 말한 물자체가 우리 자신의 존재와 삶에 어떻게 관련되는가를 보여준다. 의지는 ‘의지 그 자체’(Wille)와 ‘삶에의 의지’(Wille zum Leben) 사이의 구분을 기반으로 하는 쇼펜하우어의 의지론은 불교 수행론의 진여-육바라밀의 관계에 비추어 볼 때 칸트의 메타피직스를 극복하려는 ‘메타프락시스’로서의 의미를 더욱 뚜렷하게 드러내며, 그의 염세주의 또한 의지론의 논리적 확장이요, 교육받은 인간이 나타낼 수 있는 삶의 자세로 해석될 수 있다. Schopenhauer’s main philosophical theory can be summed up in a single statement ‘Kant’s thing-in-itself is Will’. By this statement he launches his own philosophy in Kantian base, but he modifies Kant’s main theories of epistemology and ethics elaborated in CPR and CPrR. In his view, Kant’s epistemology and ethics only deals with the logical presuppositions of knowing and moral practice, and if they are to be able to bring moral growth for a person who reads it, they should deal with specifically the efforts to reach the ideal state not just the ‘formal explanations’. Schopenhauer’s Will is a concept that shows how Kant’s ‘thing-in-itself’ related to our own existence and life. By this concept of Will, knowledge and morality can be related with each other, while these two have nothing to do with each other in Kant’s philosophy. Schopenhauer s theory of Will can reveal its meaning as ‘metapraxis’ more clearly given the relation between Jin-yeo and Yookbaramil in the theory of Buddhist practice.
알코올의 섭취가 흰쥐의 혈액과 간의 지질대사에 미치는 영향

To investigate the effects of ethanol on the lipid metabolism in blood and liver, twenty one male Sprague Dawley rats had orally taken the 25% alcohol and 50% alcohol by 5g per Kg of weight during 3 weeks after the adjusting period. For the energy balance, 85% sucrose and 45% sucrose was added in control group and 25% alcohol group each. The weight was reduced by alcohol treatment according to the levels, otherwise the weight of liver and heart was little increased in alcohol groups rather than in control without significance. Moreover, RBC, WBC, hemoglobin and hematocrit were increased by alcohol treatment according to the levels without significance. Otherwise, adjusted bilirubin levels by 100g of weight was significantly increased by alcohol treatment dose dependently. Alcohol induced RBC degradation and increased circulated bilirubin because of liver damage. For blood lipid profiles, blood cholesterol was increased in 50% of alcohol group compared to that of 25% and control groups, otherwise, blood triglycerides (TG) was increased by 30% in 25% alcohol groups rather than in control but not significantly different. The hepatic lipid composition, cholesterol levels in alcohol treatment groups was lower than those in control. The hepatic TG was not influenced by alcohol treatment. The two reasons of decreasing hepatic lipids were suggested by 85% sucrose of control for energy balance more synthesized cholesterol and TG than those in 25 or 50% alcohol groups, and by cholesterol and TG could not be synthesized because of liver damage from alcohol.
Bupivacaine과 ropivacaine이 Xenopus oocyte에 발현된 HERG 전류에 미치는 영향

Bupivacaine is an amide-type local anesthetic widely used for regional anesthesia. Ropivacaine is developed as a less cardiotoxic alternatives to bupivacaine. In the present study, we have analyzed the effects of bupivacaine and ropivacaine on HERG currents expressed in Xenopus oocytes. Bupivacaine and ropivacaine(3∼1,000μM) blocked HERG currents in a concentration dependent manner. EC_(50) was 26.1±3.1μM(n_(R) 0.65±0.04) and 43.5±7.9μM(n_(H) 0.99±0.13) in bupivacaine and ropivacaine, respectively. Bupivacaine and ropivacaine did not affect the activation and deactivation kinetics of HERG channels. However, the drugs decreased the slope conductance measured from fully activated current-voltage relationship curves. These results suggest that bupivacaine and ropivacaine have a similarinhibitory effect on HERG channels, which could be a possible cellular mechanism of LQT or ventricular arrythmia by the drugs.
Flavobacterium meningosepticum이 생산하는 Carotenoids의 동정 및 생리활성

Some carotenoids isolated from F. meningosepticum showed 3 different bands on thin layer chromatogram, one of which was the dominant. The dominant band had 455 nm of λmax, showed negative result on epoxide test, 20 nm of basochromic shift on ketone test, and absorption band characteristic of hydroxyl group on IR spectrum, which means that the main component is hydroxyechinenone. Also, the carotenoids had ROS scavenging activity and antioxidant activity. The carotenoids showed no cytotoxic activity. So it was found out that this carotenoids could be used in industry.
