        In this paper, a reference model following control system using a fuzzy logic controller (FLC) is proposed. A reference model whose response does not overshoot and has a fast rise time is designed. A FLC is designed to follow as close as possible the response of the reference model. The proposed design method has the robustness and the optimal tracking property under modeling error, disturbance and parameter perturbations. The proposed designing method is compared using a PD controller throughout the experiment. The effectiveness of the proposed designing method is verified through computer simulation and experiments for the second-order and the first-order reference model.

        Chrysanthemum coronarium L. has long been used the appetite and constipation in oriental countries. We was investigated the effect on changes in serum glucose and lipid compositions of each fraction from Chrysanthemum coronarium by examing the induction of hyperglycemia by streptozotocin(STZ). Chrysanthemum coronarium butanol fraction treatment in STZ diabetic rats inhibited the rise of serum glucose concentration. Total serum total lipids, triglyceride and phospholipid levels in STZ-induced hyperglycemic rats were sihnificantly higher than those in ormal group treatment Chrysanthemum coronarium butanol fraction. In the STZ-induced diabetic group. the total serum cholosterol, VLDL-and LDL-cholosterol level, and the atherosclerotic index were higher, while the HDL-cholesterol level was lower when compared to the normal group. However, these changes were prevented by Chrysanthemum coronarium butanol fraction treatment. Treatment of Chrysanthemum coronarium butanol fraction significantly increased the activities of serum lipase compared to the STZ-treated group. But lactate dehydrogenase(LDH) was not significantly different in any of the groups. These results suggest that Chrysanthemum coronarium butanol fraction treatment suppresses STZ induced changes in serum and hepatic lipid composition by preventing serum glucose regulation.

        필라테스 운동이 여대생의 신체구성과 하지근력 및 척추 형태에 미치는 영향

        This study is to provide the preliminary data verifying Pilates effect analyzing the effect of the warm-up, exercise and warm-down were conducted for 50 minutes a day, three time a week and for ten week. Pilates exercises for female university students on their body compositions, changes in the spine and muscular strength. The study is to involve female university students, majoring in general department (non-Physical Education) who take liberal arts classes in K and N university, and it was distinguished between 9(PG: age(20.6±0.9), height (160.9±7.1), weight(53.9±4.5)) female university students who will participate in Pilates Exercise Program and 9(CG: age(22.2±1.4), height(159.9±5.4), weight(52.3±4.7)) female university students who won"t. First, There were insignificant differences in body composition factor. Second, There were significant differences in Peak torque from isokinetic muscular function factor, Total work and Average power. Third, There were insignificant differences in 3-dimension spine body type factor. Through the ten-week Pilates exercises for three times a week, there weren"t significant differences in the body composition and positive effect of isokinetic muscular function factor, and the 3-dimension spine body type was shown.

        Pseudomonas sp. JH007에 의한 DL-2-Chloropropionic Acid로부터 D-Lactic Acid의 생산

        DL-2-chloropropionic acid로부터 D-lactic acid를 생산하기 위하여, 토양으로부터 DL-2-chloropropionic acid를 유일한 탄소원 및 에너지원으로 이용할 수 있는 균주 80여종을 분리하였으며, 분리균주들로부터 DL-2-chloropropionic acid로부터 D-lactic acid 생산성이 우수하며, L-lactic acid를 생산성이 우수하며, L-lactic acid를 생산하지 않은 균주 JH-007을 선별하여 Pseudomonas sp.로 동정하였다. DL-2-chloropropionic acid로부터 D-lactic acid를 생산하기 위한 최적 조건을 조사하기 위하여 3 g/l의 DL-2-chloropropionic acid가 포함된 LB 배지에서 배양하여 수확한 후 수확된 균체를 효소원으로 사용하였다. D-lactic acid 생산시 최적 반응액 조건은 125 mM sodium carbonate buffer에서 휴지균체 10 g/l와 3 g/l의 DL-2-chloropropionic acid를 사용할 때였으며, 최적 반응 pH는 10.0, 최적반응 온도는 30℃이었다. 최적조건하에서 반응액에 1 g/l의 DL-2-chloropropionic acid를 간헐적으로 첨가하여 5시간 반응시켰을 때 5.72 g/l의 D-lactic acid가 생산되었으며, 전환율은 98.4%였으며, 광학순도는 99.8%이었다. For the production of D-lactic acid from DL-2-chloropropionic acid, about 80 strains of bacteria capable of assimilating DL-2-chloropropionic acid as a sole carbon and energy source were isolated from the soil. JH-007 strain that showed the higest productivity of D-lactic acid and didn't produce L-lactic acid from DL-2-chloropropionic acid was selected from them and identified as Pseudomonas sp. The optimal conditions for the production of D-lactic acid from DL-2-chloropropionic acid were examined. The resting cells of JH-007 cultured in LB medium containing 3 g/l of DL-2-chloropropionic acid were used as an enzyme source. The reaction mixtures for the maximal production of D-lactic acid were consist of 10 g/l of resting cells and 3 g/l of DL-2-chloropropionic acid in 125 mM sodium carbonate buffer. The optimal pH for the reaction was 10.0 and the optimal temperature was 30℃. When 1 g/l of DL-2-chloropropionic acid was added intermittently to the reaction mixture under the above condition, 5.72 g/l of D-lactic acid was produced after incubation of 5 hrs. This amount of D-lactic acid corresponded to a 98.4% yields and the optical purity was 99.8%.

        인공 치아우식 발생 모델에서 디지털 방사선 공제술을 이용한 인접면 치아우식증의 진단

        Purpose : The purpose of the experiment was to evaluating the diagnostic ability of dental caries detection using digital subtraction in the artificial caries activity model. Materials and Methods : Digital radiographies of five teeth with 8 proximal surfaces were obtained by CCD sensor (Kodak RVG 6100 using a size #2). The digital radiographic images and subtraction images from artificial proximal caries were examined and interpreted. In this study, we proposed novel caries detection method which could diagnose the dental proximal caries from single digital radiographic image. Results : In artificial caries activity model, the range of lesional depth was 572-1,374 μm and the range of lesional area was 36.95-138.52mm². The lesional depth and the area were significantly increased with demineralization time (p<0.001). Furthermore, the proximal caries detection using digital subtraction radiography showed high detection rate compared to the proximal caries examination using simple digital radiograph. Conclusion : The results demonstrated that the digital subtraction radiography from single radiographic image of artificial caries was highly efficient in the detection of dental caries compared to the data from simple digital radiograph.

      • NFAT1 and JunB Cooperatively Regulate <i>IL-31</i> Gene Expression in CD4<sup>+</sup> T Cells in Health and Disease

        <P>IL-31 is a key mediator of itching in atopic dermatitis (AD) and is preferentially produced by activated CD4<SUP>+</SUP> T cells and Th2 cells. Although pathophysiological functions of IL-31 have been suggested in diverse immune disorders, the molecular events underlying <I>IL-31</I> gene regulation are still unclear. In this study we identified the transcription start site and functional promoter involved in <I>IL-31</I> gene regulation in mouse CD4<SUP>+</SUP> T cells. TCR stimulation–dependent IL-31 expression was found to be closely linked with in vivo binding of NFAT1 and JunB to the <I>IL-31</I> promoter. Although NFAT1 alone enhanced <I>IL-31</I> promoter activity, it was further enhanced in the presence of JunB. Conversely, knockdown of either NFAT1 or JunB resulted in reduced <I>IL-31</I> expression. NFAT1-deficient CD4<SUP>+</SUP> T cells showed a significant defect in <I>IL-31</I> expression compared with wild-type CD4<SUP>+</SUP> T cells. In agreement with these findings, mice subjected to atopic conditions showed much higher levels of IL-31, which were closely correlated with a significant increase in the number of infiltrated NFAT1<SUP>+</SUP>CD4<SUP>+</SUP> T cells into the AD ears. Amelioration of AD progression by cyclosporin A treatment was well correlated with downregulation of IL-31 expressions in CD4<SUP>+</SUP> T cells and total ear residual cells. In summary, our results suggest a functional cooperation between NFAT1 and JunB in mediating <I>IL-31</I> gene expression in CD4<SUP>+</SUP> T cells and indicate that interference with this interaction or their activity has the potential of reducing IL-31–mediated AD symptoms.</P>

      • Arachidonic Acid 12-lipoxygenase (ALOX12) Promotes Pulmonary Fibrosis

        Background 12(S)-hydroxyeicosatetraenoic acid (12(S)-HETE) is product of arachidonic acid metabolism through the enzyme of arachidonic acid 12-lipoxygenase (ALOX12) in human platelets or leukocytes. ALOX12 plays an important role in inflammation and oxidation and is involved in development of various human diseases. However, the role of 12(S)-HETE and ALOX12 in the pathogenesis of idiopathic pulmonary fibrosis (IPF) remains unexplored. We investigated the role of ALOX12 in IPF. Methods The level of 12-HETE was measured in human plasma (healthy control = 40, IPF = 76) via liquid chromatography-mass spectrometry (LC-MS/MS). The function of 12-HETE and ALOX12 were evaluated in TGF-β1-stimulated human lung fibroblast (MRC-5 cell, primary lung fibroblast) using ALOX12 inhibitor or silencing RNA (siRNA). The anti-fibrotic effects of ALOX12 inhibitor were evaluated in bleomycin-induced mice model of pulmonary fibrosis. Results The level of 12-HETE was significantly increased in plasma of IPF patients compared with controls. 12-HETE increased protein expression levels of collagen I and fibronectin in human lung fibroblast. ALOX12 levels were increased in lung tissues of patients with IPF and human lung fibroblasts stimulated by TGF-β1 compared with those of controls. ALOX12 inhibitor and ALOX12 specific siRNA downregulated protein expression levels of collagen I and fibronectin increased by TGF-β1 in human lung fibroblasts. In addition, ALOX12 inhibitor decreased TGF- β1 induced phosphorylation of Smad2/3 in MRC-5 cell. In bleomycin treated mice, ALOX12 inhibitor decreased levels of hydroxyproline in the lung compared of control mice. Conclusions Our findings suggest that inhibition of ALOX12 may have anti-fibrotic effects on pulmonary fibrosis, suggesting that ALOX12 is implicated as a potential therapeutic target in IPF.

        Right hepatic vein stenosis at anastomosis in patients after living donor liver transplantation: optimal Doppler US venous pulsatility index and CT criteria--receiver operating characteristic analysis.

        <P>PURPOSE: To establish optimal Doppler ultrasonographic (US) venous pulsatility index and computed tomographic (CT) criteria for right hepatic vein (RHV) stenosis after living donor liver transplantation (LDLT) and to compare accuracies of these methods by using receiver operating characteristic (ROC) analysis. MATERIALS AND METHODS: This retrospective study was approved by an institutional review board; informed consent was waived. Eighty patients (48 men, 32 women; mean age, 51.5 years +/- 9.2 [standard deviation]) underwent Doppler US and CT within 8 days of hepatic venography following right lobe LDLT between October 2006 and September 2008. At venography, RHVs were classified into a stenosis or nonstenosis group. At Doppler US, venous pulsatility index was defined as the difference between maximum and minimum frequency shifts divided by maximum frequency shift. At CT, diameters of anastomosis and RHV were measured; percentage of stenosis was calculated. Mean Doppler US and CT parameters in the two groups were compared; ROC analysis was performed. RESULTS: There were 30 stenotic and 50 nonstenotic RHVs. Mean venous pulsatility index and mean anastomosis diameter were significantly lower and mean percentage of stenosis was significantly higher in the stenosis than the nonstenosis group (P < .001 each). Optimal cutoffs for venous pulsatility index, anastomosis diameter, and percentage of stenosis were 0.16, 3.7 mm, and 47%, respectively. Sensitivity and specificity were 86.7% and 68.0% for venous pulsatility index, 96.7% and 88.0% for anastomosis diameter, and 96.7% and 86.0% for percentage of stenosis, respectively. At ROC analysis, anastomosis diameter (P = .002) and percentage of stenosis (P = .003) were significantly more accurate than venous pulsatility index. CONCLUSION: CT is more accurate than Doppler US for RHV stenosis after LDLT, with venous pulsatility index as the sole sonographic criterion. Patients suspected of having RHV stenosis at Doppler US may benefit from CT to reduce unnecessary venography.</P>
