        Discrimination of neutrons and gamma-rays in plastic scintillator based on spiking cortical model

        In this study, a spiking cortical model (SCM) based n-g discrimination method is proposed. The SCMbased algorithm is compared with three other methods, namely: (i) the pulse-coupled neural network (PCNN), (ii) the charge comparison, and (iii) the zero-crossing. The objective evaluation criteria used for the comparison are the FoM-value and the time consumption of discrimination. Experimental results demonstrated that our proposed method outperforms the other methods significantly with the highest FoM-value. Specifically, the proposed method exhibits a 34.81% improvement compared with the PCNN, a 50.29% improvement compared with the charge comparison, and a 110.02% improvement compared with the zero-crossing. Additionally, the proposed method features the second-fastest discrimination time, where it is 75.67% faster than the PCNN, 70.65% faster than the charge comparison and 38.4% slower than the zero-crossing. Our study also discusses the role and change pattern of each parameter of the SCM to guide the selection process. It concludes that the SCM's outstanding ability to recognize the dynamic information in the pulse signal, improved accuracy when compared to the PCNN, and better computational complexity enables the SCM to exhibit excellent n-g discrimination performance while consuming less time.

      • The High Expressed Serum Soluble Neural Cell Adhesion Molecule, a High Risk Factor Indicating Hepatic Encephalopathy in Hepatocelular Carcinoma Patients

        Objective: To investigate whether the expression of serum soluble neural cell adhesion molecule (sNCAM) is associated with hepatic encephalopathy (HE) in hepatocelular carcinoma (HCC) patients. Materials and Methods: The Oncomine Cancer Microarray database was used to determine the clinical relevance of NCAM expression in different kinds of human cancers. Sera from 75 HCC cases enrolled in this study were assessed for expression of sNCAM by enzyme linked immunosorbent assay (ELISA). Results: Dependent on the Oncomine Cancer Microarray database analysis, NCAM was down regulated in 10 different kinds of cancer, like bladder cancer, brain and central nervous system cancer, while up-regulated in lung cancer, uterine corpus leiomyoma and sarcoma, compared to normal groups. Puzzlingly, NCAM expression demonstrated no significant difference between normal and HCC groups. However, we found by quantitative ELISA that the level of sNCAM in sera from HCC patients with HE ($347.4{\pm}151.9ng/ml$) was significantly more up-regulated than that in HCC patients without HE ($260.3{\pm}104.2ng/ml$), the p-value being 0.008. sNCAM may be an important risk factor of HE in HCC patients, the correlation coefficients was 0.278 (P<0.05) on rank correlation analysis. Conclusions: This study highlights that up-regulated level of serum sNCAM is associated with HE in HCC patients and suggests that the high expression can be used as an indicator.

        A Rapidly New-typed Detection of Norovirus Based on F0F1-ATPase Molecular Motor Biosensor

        In order to adapt port rapid detection of food borne norovirus, presently we developed a new typed detection method based on F0F1-ATPase molecular motor biosensor. A specific probe was encompassed the conservative region of norovirus and F0F1-ATPase within chromatophore was constructed as a molecular motor biosensor through the “ε-subunit antibody-streptomycinbiotin- probe” system. Norovirus was captured based on probe-RNA specific binding. Our results demonstrated that the Limit of Quantification (LOQ) is 0.005 ng/mL for NV RNA and also demonstrated that this method possesses specificity and none cross-reaction for food borne virus. What’s more, the experiment used this method could be accomplished in 1 h. We detected 10 samples by using this method and the results were consistent with RT-PCR results. Overall, based on F0F1-ATPase molecular motors biosensor system we firstly established a new typed detection method for norovirus detection and demonstrated that this method is sensitive and specific and can be used in the rapid detection for food borne virus.

        Transient virus-induced gene silencing of MaBAM9b efficiently suppressed starch degradation during postharvest banana fruit ripening

        The genetic basis of metabolic pathways that operate during fruit ripening needs to be understood before the nutritional value of the banana can be improved. The banana is a typical starch conversion fruit, and β-amylase is a key enzyme that may play an important role in starch degradation during the ripening process. Musa acuminata β-amylase 9b (MaBAM9b) is closely related to starch degradation. However, its exact function in starch degradation has not been demonstrated in banana. Stable genetic transformation to identify gene function is a time- and energy-consuming process. Thus, an efficient and rapid method is needed for functional identification. Virus-induced gene silencing (VIGS) is a reverse-genetics method based on RNA-mediated antiviral plant defense that has been used to rapidly identify gene functions in plants. The aim of this study was to optimize a transient VIGS system and functionally elucidate MaBAM9b in postharvest banana fruit. Using 2- to 4-mm-thick fruit slices, vacuum infiltration of suspensions of Agrobacterium strains carrying TRV1 and TRV2-MaBAM9b, 0.5% iodine-potassium-iodide (I2-KI) staining for 150 s, and 1:3 TRV1:TRV2-MaBAM9b cultivation at 30 mmHg for 30 s achieved an optical density (OD) of 0.8 at 600 nm; after being incubated on Murashige and Skoog (MS) media for 5 days (d), starch degradation was efficiently suppressed during postharvest banana fruit ripening, as determined by I2-KI staining, total starch content, β-amylase activity, soluble sugar content, and endogenous MaBAM9b expression. The system described here is particularly useful for studying genes and networks involved in starch conversion in fruits, which alone would not produce a visual phenotype. This system will provide a platform for functional genomics and fruit quality improvement in banana.

        Effect of ZnO on the microstructures and piezoelectric properties of K0.5Na0.5NbO3 ceramics

        Lead-free ZnO added K0.5Na0.5NbO3 piezoelectric ceramics have been prepared by a conventional solid state process. The XRD results show that all the K0.5Na0.5NbO3 ceramics with various ZnO contents are single phase with an orthorhombic perovskite structure. The bulk density obtained by the Archimedes method indicates that the addition of ZnO improves the density of K0.5Na0.5NbO3 ceramics under the same sintering conditions. Especially, at a level of 1 mol% ZnO, the sample sintered at 1100 oC for 1h exhibits the highest bulk density (ρ = 4.28 g/㎤) and shows optimal piezoelectric properties, d33 = 135 pC/N and kp = 0.40.         MAPK4 silencing in gastric cancer drives liver metastasis by positive feedback between cancer cells and macrophages

        Liver metastasis is a major cause of death in gastric cancer patients, but the underlying mechanisms are poorly understood. Through a combination of in vivo screening and transcriptome profiling followed by quantitative RT-PCR and tissue array analyses, we found that mitogen-activated protein kinase 4 (MAPK4) downregulation in gastric cancer tissues from patients is significantly associated with liver metastasis and poor prognosis. The knockdown of MAPK4 in gastric cancer cells promotes liver metastasis in orthotopic mouse models. MAPK4 depletion in gastric cancer cells induces the secretion of macrophage migration inhibitory factor (MIF) to polarize tumor-associated macrophages (TAMs) in orthotopic xenograft tumors. Moreover, TAMs activate epithelial–mesenchymal transition of gastric cancer cells to suppress MAPK4 expression, which further increases MIF secretion to polarize TAMs. Taken together, our results suggest a previously undescribed positive feedback loop between cancer cells and macrophages mediated by MAPK4 silencing that facilitates gastric cancer liver metastasis.

        Hydrogen sulfide inhibits the growth of Escherichia coli through oxidative damage

        Many studies have shown that hydrogen sulfide (H2S) is both detrimental and beneficial to animals and plants, whereas its effect on bacteria is not fully understood. Here, we report that H2S, released by sodium hydrosulfide (NaHS), significantly inhibits the growth of Escherichia coli in a dose-dependent manner. Further studies have shown that H2S treatment stimulates the production of reactive oxygen species (ROS) and decreases glutathione (GSH) levels in E. coli, resulting in lipid peroxidation and DNA damage. H2S also inhibits the antioxidative enzyme activities of superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) and induces the response of the SoxRS and OxyR regulons in E. coli. Moreover, pretreatment with the antioxidant ascorbic acid (AsA) could effectively prevent H2S-induced toxicity in E. coli. Taken together, our results indicate that H2S exhibits an antibacterial effect on E. coli through oxidative damage and suggest a possible application for H2S in water and food processing.

        <i>Chd7</i> Is Critical for Early T-Cell Development and Thymus Organogenesis in Zebrafish

        <P>Coloboma, heart defect, atresia choanae, retarded growth and development, genital hypoplasia, ear anomalies/deafness (CHARGE) syndrome is a congenital disorder affecting multiple organs and mainly caused by mutations in CHD7, a gene encoding a chromatin-remodeling protein. Immunodeficiency and reduced T cells have been noted in CHARGE syndrome. However, the mechanisms underlying T lymphopenia are largely unexplored. Herein, we observed dramatic decrease of T cells in both chd7knockdown and knockout zebrafish embryos. Unexpectedly, hematopoietic stem and progenitor cells and, particularly, lymphoid progenitor cells were increased peripherally in nonthymic areas in chd7-deficient embryos, unlikely to contribute to the T-cell decrease. Further analysis demonstrated that both the organogenesis and homing function of the thymus were seriously impaired. Chd7 might regulate thymus organogenesis through modulating the development of both neural crest cell-derived mesenchyme and pharyngeal endoderm-derived thymic epithelial cells. The expression of faxn1, a central regulator of thymic epithelium, was remarkably down-regulated in the pharyngeal region in chd7-deficient embryos. Moreover, the T-cell reduction in chd7-deficient embryos was partially rescued by overexpressingfoxnl, suggesting that restoring thymic epithelium may be a potential therapeutic strategy for treating immunodeficiency in CHARGE syndrome. Collectively, the results indicated that chd7 was critical for thymic development and T-lymphopenia in CHARGE syndrome may be mainly attributed to the defects of thymic organogenesis. The current finding may benefit the diagnosis and therapy of T lymphopenia and immunodeficiency in CHARGE syndrome.</P>
