        Schisandrin B Improves the Hypothermic Preservation of Celsior Solution in Human Umbilical Cord Mesenchymal Stem Cells

        BACKGROUND: Human umbilical cord mesenchymal stem cells (hUCMSCs) have emerged as promising therapy for immune and inflammatory diseases. However, how to maintain the activity and unique properties during cold storage and transportation is one of the key factors affecting the therapeutic efficiency of hUCMSCs. Schisandrin B (SchB) has many functions in cell protection as a natural medicine. In this study, we investigated the protective effects of SchB on the hypothermic preservation of hUCMSCs. METHODS: hUCMSCs were isolated from Wharton’s jelly. Subsequently, hUCMSCs were exposed to cold storage (4 C) and 24-h re-warming. After that, cells viability, surface markers, immunomodulatory effects, reactive oxygen species (ROS), mitochondrial integrity, apoptosis-related and antioxidant proteins expression level were evaluated. RESULTS: SchB significantly alleviated the cells injury and maintained unique properties such as differentiation potential, level of surface markers and immunomodulatory effects of hUCMSCs. The protective effects of SchB on hUCMSCs after hypothermic storage seemed associated with its inhibition of apoptosis and the anti-oxidative stress effect mediated by nuclear factor erythroid 2–related factor 2 signaling. CONCLUSION: These results demonstrate SchB could be used as an agent for hypothermic preservation of hUCMSCs.

        The evaluation of online course of Traditional Chinese Medicine for Medical Bachelor, Bachelor of Surgery international students during the COVID-19 epidemic period

        Background: During the COVID-19 epidemic period, Traditional Chinese Medicine (TCM) course for international students of Medical Bachelor, Bachelor of Surgery (MBBS) program in Zhejiang University has shifted from traditional classroom to online environment. This study aimed to investigate MBBS international students’ perception on online TCM course, and to assess the online learning efficacy. Methods: A total of 84 MBBS international students attending course of “Basic Traditional Chinese Medicine” during 2020 academic years at Zhejiang University were enrolled in this study. A quantitative questionnaire was respectively completed before and after the TCM course using a pretest–post-test design. By means of two online learning platforms, Learning in ZJU and DingTalk, TCM course was broadcast in both live and archived format to students. Results: A total of 48 participants completed both baseline and follow-up questionnaires. The majority of participants preferred face-to-face classroom learning (26, 54.17% of total) when compared with online learning. Students felt that the course had brought in much benefits (mean 3.88, SD 0.87), and they were satisfied with the course content (mean 3.83, SD 0.95). Students’ TCM related knowledge and their behaviors of discussion and consulting were significantly improved by online TCM course (all P < 0.001). Students’ awareness of the necessity of TCM education and their feeling of difficulty in learning TCM were significantly strengthened (P = 0.042, 0.025, respectively). Conclusion: Online learning is a good alternative for TCM course of MBBS international students when classroom learning is suspended, whereas it cannot replace the need for onsite and face-to-face learning.

      • Involvement of GRP78 in the Resistance of Ovarian Carcinoma Cells to Paclitaxel

        Background: Glucose regulated protein 78 (GRP78) is a type of molecular chaperone. It is a possible candidate protein that contributes to development of drug resistance. We first examined the involvement of GRP78 in chemotherapy-resistance in human ovarian cancer cell. Materials and Methods: The expression of GRP78 mRNA and protein were examined by RT-PCR and western blotting, respectively, in human ovarian cancer cells line (HO-8910). Sensitivity of HO-8910 to paclitaxel was determined with methyl thiazolyl tetrazolium (MTT). Suppression of GRP78 expression was performed using specific small-interfering RNA (siRNA) in HO-8910 cells, and cell apoptosis was assessed by flow cytometry. Statistical analysis was performed using the SPSS 15.0 statistical package. Results: HO-8910 cells, with high basal levels of GRP78, exhibited low sensitivity to paclitaxel. The mRNA and protein levels of GRP78 were dramatically decreased at 24h, 48h and 72h after transfection and the sensitivity to paclitaxel was increased when the GRP78 gene was disturbed by specific siRNA transfection. Conclusions: The results suggested that high GRP78 expression might be one of the molecular mechanisms causing resistance to paclitaxel, and therefore siRNA of GRP78 may be useful in tumor-specific gene therapy for ovarian cancer.

      • Research on PN Code Acquisition Strategy

        PN code acquisition is the key to synchronization in direct-sequence spread-spectrum(DSSS) systems. Aiming at the problems in the process of PN code acquisition, the block diagram of acquisition strategy is illustrated firstly. Then, with the choice of different search mode, determination mode and verification mode in the block diagram, the detection probabilities and false alarm probabilities are presented separately, and the false alarm probability of threshold detection method, the detection probability and false alarm probability of maximum-selection determination method are deduced, and all kinds of acquisition strategies are given. Finally, Simulation results shows that the detection probabilities and false alarm probabilities in theory and experiment is consistent with different acquisition strategy, and so does the validation of detection probabilities by maximum-selection method in multiple dwell times.

        Deficiency of Follistatin-Like Protein 1 Accelerates the Growth of Breast Cancer Cells at Lung Metastatic Sites

        Purpose: Follistatin-like protein 1 (FSTL1) is a secreted glycoprotein that has been shown to play a role in various types of cancer. However, the clinical significance and function of FSTL1 in breast cancer have not been reported. We investigated the role of FSTL1 in breast cancer in this study. Methods: Enzyme-linked immunosorbent assays, western blot analysis, and reverse transcription polymerase chain reaction were used to monitor the expression of FSTL1 in breast cancer tissue and in serum samples from breast cancer patients. We employed a 4T1 breast cancer model and Fstl1+/− mice for in vivo studies. Hematoxylin and eosin staining, western blot analysis, and RNA sequencing were used to analyze the effect of FSTL1 on primary tumor growth and lung metastasis. Results: We demonstrated that the expression of FSTL1 is reduced in both the breast cancer tissue and the serum of breast cancer patients. We showed that reduced levels of FSTL1 in serum correlate with elevated expression of Ki-67 and epidermal growth factor receptor (EGFR) in cancer tissues. Moreover, lowered expression of FSTL1 was associated with decreased survival in breast cancer patients. Experiments on the Fstl1+/− mouse model established that FSTL1 deficiency had no effect on primary tumor growth, but increased the lung metastases of breast cancer cells, resulting in reduced survival of tumor-bearing mice. RNA sequencing found significantly reduced expression of Egln3 and increased expression of EGFR in Fstl1+/− mice. Thus, our results suggest that FSTL1 may affect the expression of EGFR through Egln3, inhibiting the proliferation of breast cancer cells at lung metastatic sites. Conclusion: In conclusion, we suggest a suppressor role of FSTL1 in breast cancer lung metastasis. Furthermore, FSTL1 may represent a potential prognostic biomarker and a candidate therapeutic target in breast cancer patients.

      • A Segmentation Model Combining Local and Global Energy Based on Local Entropy

        Image segmentation based partial differential equation is very popular in image analysis and computer vision. This article presents a geometric active contour model based on local entropy for segmentation. In the proposed method, energy functional item of this model contains local energy and global energy items, and uses local entropy of each point on the contour line to determine the proportion of each kind of energy. The combined method allows the contour for the rapid evolution and convergence to the final edge after iterations. Experiment results on both synthetic and real images demonstrate the efficiency and accuracy of this model.

        Generation of Quantum Phase Gates via an Unconventional Geometric Phase Shift in an Ion-Trap System

        We propose a scheme to generate a quantum phase gate via an unconventional geometric phase shift in an ion-trap system. Since the gate time is only related to the frequency of the vibrational mode, the present scheme, based on the resonant interaction of the trapped ions with a standing-wave laser beam, requires a much shorter gate time than the scheme based on the resonant interaction of the atoms and the cavity. Also the heating effect of the vibrational mode is negligible under certain conditions.         In the network environment, supply chain management has greatly reduced the product development cycle, reduce the inventory. With the continuous development of information technology, e-commerce logistics platform has become the main factor affecting the development of logistics industry. In this paper, the authors research on the E-commerce platform performance and green supply chain based on data mining and SVM. The green supply chain considers the environmental problems in every link of the supply chain, and promotes the coordinated development of economy and environment. The result shows that the most critical factor that affects the satisfaction of consumer to B2C e-commerce platform is the accurate, complete and reliable logistics service.

      • Personnel Localization Algorithm of Prison Supervision System Based on RFID

        The particularity of the prison decides the importance of its security system, and how to improve the accuracy of personnel positioning for prison monitoring system is the problem to be solved to realize intelligent prison monitoring management. RFID positioning technology with non line-of-sight transmission and large transmission range is widely applied in the personnel positioning system. RFID technology is used to design the monitoring and management system of prison in this paper. The Trilateral Ranging Method was adopted to realize personnel positioning. RSSI positioning algorithm its technology is relatively simple was adopted when calculating the trilateral distance and correcting the errors in order to improve the positioning precision. Simulation test results show that the positioning precision for the distance between a single reader and the tag is increased by 2.17% on average, up to 4.54% after the error correction. The personnel location coordinates, X direction average error is 2.96%, average error is 3.12%, Y direction, and meeting the requirements of the system design.
