Ginsenoside Rg1 activates ligand-independent estrogenic effects via rapid estrogen receptor signaling pathway

        Gao, Quan-Gui,Zhou, Li-Ping,Lee, Vien Hoi-Yi,Chan, Hoi-Yi,Man, Cornelia Wing-Yin,Wong, Man-Sau The Korean Society of Ginseng 2019 Journal of Ginseng Research Vol.43 No.4

Background: Ginsenoside Rg1 was shown to exert ligand-independent activation of estrogen receptor (ER) via mitogen-activated protein kinase-mediated pathway. Our study aimed to delineate the mechanisms by which Rg1 activates the rapid ER signaling pathways. Methods: ER-positive human breast cancer MCF-7 cells and ER-negative human embryonic kidney HEK293 cells were treated with Rg1 ($10^{-12}M$, $10^{-8}M$), $17{\beta}$-estradiol ($10^{-8}M$), or vehicle. Immunoprecipitation was conducted to investigate the interactions between signaling protein and ER in MCF-7 cells. To determine the roles of these signaling proteins in the actions of Rg1, small interfering RNA or their inhibitors were applied. Results: Rg1 rapidly induced $ER{\alpha}$ translocation to plasma membrane via caveolin-1 and the formation of signaling complex involving linker protein (Shc), insulin-like growth factor-I receptor, modulator of nongenomic activity of ER (MNAR), $ER{\alpha}$, and cellular nonreceptor tyrosine kinase (c-Src) in MCF-7 cells. The induction of extracellular signal-regulated protein kinase and mitogen-activated protein kinase kinase (MEK) phosphorylation in MCF-7 cells by Rg1 was suppressed by cotreatment with small interfering RNA against these signaling proteins. The stimulatory effects of Rg1 on MEK phosphorylation in these cells were suppressed by both PP2 (Src kinase inhibitor) and AG1478 [epidermal growth factor receptor (EGFR) inhibitor]. In addition, Rg1-induced estrogenic activities, EGFR and MEK phosphorylation in MCF-7 cells were abolished by cotreatment with G15 (G protein-coupled estrogen receptor-1 antagonist). The increase in intracellular cyclic AMP accumulation, but not Ca mobilization, in MCF-7 cells by Rg1 could be abolished by G15. Conclusion: Ginsenoside Rg1 exerted estrogenic actions by rapidly inducing the formation of ER containing signalosome in MCF-7 cells. Additionally, Rg1 could activate EGFR and c-Src ER-independently and exert estrogenic effects via rapid activation of membrane-associated ER and G protein-coupled estrogen receptor.

      • <i>Orobanche cernua</i> Loefling Attenuates Ultraviolet B-mediated Photoaging in Human Dermal Fibroblasts

        Gao, Wei,Wang, Yu-shuai,Qu, Zheng-yi,Hwang, Eunson,Ngo, Hien T. T.,Wang, Ying-ping,Bae, Jahyun,Yi, Tae-hoo Wiley (Blackwell Publishing) 2018 Photochemistry and photobiology Vol.94 No.4

<P>UV radiation is the primary cause of skin photoaging, which results in an increase in matrix metalloproteinases and degradation of collagen. Developing new natural antioxidant as photoprotective agents has become a popular area of research. Orobanche cernua Loefling is a parasitic plant that is rich in phenylethanoid glycosides (PhGs). This study investigated the photoprotective effects of the ethanolic extract of Orobanche cernua Loefling (OC) and its principal component acteoside on UVB-induced photoaging as well as their underlying molecular mechanisms in normal human dermal fibroblasts (NHDFs). Biological testing demonstrated that OC and acteoside possessed significant photoprotective activities, reducing MMP and IL-6 levels while improving type-I procollagen synthesis in UVB-irradiated NHDFs. Further study showed that the protective mechanisms were the improvement of transcription factor Nrf2-mediated antioxidant defensive system, suppression of MAPK/AP-1 and activation of the TGF-/Smad pathway. Together, our results suggested that OC might be a promising antiphotoaging agent against UV radiation-induced skin damage.</P>

Magnus Effect over Aerodynamic Components of Spinning Missile

Yi Gao,Juanmian Lei,Jintao Yin 한국항공우주학회 2022 International Journal of Aeronautical and Space Sc Vol.23 No.3

To reveal the flow mechanism of the Magnus effect of the spinning missile, the flow field under supersonic conditions was numerically simulated based on the unsteady Reynolds-time-averaged Navier–Stokes equation, and the implicit dual-time stepping method and sliding mesh method were used. Validation was performed to ensure the reliability of the numerical methods. The results of the numerical simulation and the wind-tunnel experimental data coincided quite well. The aerodynamic characteristics of various configurations while spinning were calculated. The effect of aerodynamic components and aerodynamic interference between components on the Magnus effect were analyzed. The results indicate that for the projectile body, the interference of the canard enlarges the time-averaged side force; when the slenderness ratio becomes large, from the view of the base, the left leeward separation vortex is close to the surface, resulting in a low-pressure region, and the direction of the side force and yawing moment are changed. The fin installation angle can weaken the body Magnus effect to some extent. For the projectile fin, the influence of leeward separation vortices on the fin depends on their relative positions. The fin installation angle can weaken and even cause the reversal of the side force direction.

      • Systematic Investigation of the Wavelength-Dependent Upconversion Enhancement Induced by Single Plasmonic Nanoparticles

        Yi, Gao,Moon, Byeong-Seok,Wen, Xu,Kim, Young-Jin,Kim, Dong-Hwan American Chemical Society 2018 The Journal of Physical Chemistry Part C Vol.122 No.24

<P>A convenient method was developed to systematically study the wavelength-dependent upconversion enhancement induced by single plasmonic nanoparticles to overcome unavoidable variations of nanoparticle geometry in practical synthesis techniques. Gold nanospheres and gold nanotriangles coupled to an upconversion nanoparticle monolayer were selected to compare emission and excitation resonance couplings, respectively. The emission intensity of a upconversion nanoparticle monolayer coupled with gold nanospheres (i.e., emission coupling) was found to be slightly lower than that of a reference upconversion nanoparticle monolayer, independent of pumping power. In turn, the emission intensity of a upconversion nanoparticle monolayer coupled with a gold nanotriangle (i.e., excitation coupling) showed an enhancement factor of the emission intensity of about 3.26 at low pumping power, which decreased to 0.48 at high pumping power because of a local heating effect. Our method could provide a convenient strategy for massive data collection of coupled upconversion nanoparticles and plasmonic nanoparticles in a single-particle level and a guideline for systematic investigations involving plasmonic nanoparticles.</P> [FIG OMISSION]</BR>

Performance assessment for liner shipping industry

        Zi-Yi Gao,Okan Duru,Emrah Bulut,Shigeru Yoshida 인하대학교 정석물류통상연구원 2013 JOURNAL OF INTERNATIONAL LOGISTICS AND TRADE Vol.11 No.2

This paper investigates the performance assessment of liner shipping industry and presents a multi-dimensional evaluation framework to ensure both financial and non-financial monitoring. The traditional performance assessment approach is based on the financial indicators such as ratio analysis, but it is limited to the fiscal perspective. The meaning of performance has dramatically changed and non-financial (and intangible) assets increased their importance in recent years. Under these circumstances, the multi-attribute performance assessment methods play a critical role to combine many aspects of the business. Balanced Scorecard (BSC) is originally developed for the multi-attribute performance assessment and its philosophy on business process evaluation pioneered the importance of key performance indicators and the quality management issues including the internal customer. Service quality and the business performance assessment are some of the hot issues in the liner shipping industry and the long term competitiveness is a critical concern in the recent liner shipping business. The BSC method is utilized to ensure a cumulative analysis of the short/long and tangible/intangible indicators of performance and computes the weight of each criterion by using Fuzzy-AHP method in the liner shipping industry.

      • Investigation of a Novel IGCT Module for DC Circuit Breaker

        Qiang Yi,Yifei Wu,Yi Wu,Fei Yang,Zhihui Zhang,Chong Gao 전력전자학회 2019 ICPE(ISPE)논문집 Vol.2019 No.5

Power semiconductors for DC circuit breaker (DCCB) have attracted great attentions due to their fast and arc-less operation under full-range current-breaking. Since the robustness of Integrated Gate-Commutated Thyristor (IGCT) is the highest among all full-control power semiconductors, it becomes the perfect power semiconductor candidate for DC circuit breaker. In this paper, the principle and operation of hybrid circuit breaker are introduced. Technical analysis of full-control power semiconductor is conducted and IGCT is the best choice for HCB. Then, A novel power semiconductor circuit based on two IGCTs, diode-bridge, snubber-circuit and MOV is proposed and designed both topologically and structurally. The selection of full-control power semiconductor is discussed and fast recover diode must be used to ensure reliability. Furthermore, the internal current distribution is analyzed. Finally, a test of a single-stage IGCT module prototype demonstrates successful current-breaking at over 10kA and below 4kV, which is promising in the area of fault protection in future MVDC distribution system.
