Tanshinone II-A Inhibits Angiogenesis through Down Regulation of COX-2 in Human Colorectal Cancer

Angiogenesis plays a significant role in colorectal cancer (CRC) and cyclooxygenase-2 (COX-2) appears to be involved with multiple aspects of CRC angiogenesis. Our aim was to investigate the inhibitory effects of Tan II-A (Tanshinone II-A, Tan II-A) on tumor growth in mice, as well as alteration of expression of COX-2 and VEGF in CRC. We established the mice xenograft model of C26 CRC cell line, and injected 0.5, 1, 2mg/kg of Tan II-A and 1mg/kg of 5-FU in respectively in vivo. Then, we assayed tumor weight and volume, and evaluated microvascular density and expression of VEGF. COX-2 promoter and COX-2 plasmids were transfected into HCT-116 cells, followed by detection of COX-2 promoter activity by chemiluminescence, and detection of COX-2 mRNA expression by fluorescence quantitative PCR. Taken together, the results showed Tan II-A could inhibit tumor growth and suppress the VEGF level in vivo. HCT-116 cell experiments showed marked inhibitory effects of Tan II-A on COX-2 and VEGF in a dose-dependent manner. The results indicate that Tan II-A can effectively inhibit tumor growth and angiogenesis of human colorectal cancer via inhibiting the expression level of COX-2 and VEGF.
Effect of height-to-width ratio on composite wall under compression

Double skin composite walls are increasingly popular and have been applied to many safety-related facilities. They come from the concept of composite slabs. Conventional connectors such as shear studs and binding bars were used in previous studies to act as the internal mechanical connectors to lock the external steel faceplates to the concrete core. However, the restraint effects of these connectors were sometimes not strong enough. In this research, a recently proposed unique type of steel truss was employed along the wall height to enhance the composite action between the two materials. Concrete-filled tube columns were used as the boundary elements. Due to the existence of boundary columns, the restraints of steel faceplates to the concrete differ significantly for the walls with different widths. Therefore, there is a need to explore the effect of height-to-width ratio on the structural behavior of the wall. In the test program, three specimens were designed with the height of 3000 mm, the thickness of 150 mm, and different widths, to simulate the real walls in practice. Axial compression was applied by two actuators on the tested walls. The axial behavior of the walls was evaluated based on the analysis of test results. The influences of height-to-width ratio on structural performance were evaluated. Finally, discussion was made on code-based design.

Double skin composite walls offer structural and economic merits over conventional reinforced concrete counterparts in terms of higher capacity, greater stiffness, and better ductility. This paper investigated the axial behavior of double skin composite walls with steel truss connectors. Full-scaled tests were conducted on three specimens with different height-to-thickness ratios. Test results were evaluated in terms of failure mode, load-axial displacement response, buckling loading, axial stiffness, ductility, strength index, load-lateral deflection, and strain distribution. The test data were compared with AISC 360 and Eurocode 4 and it was found that both codes provided conservative predictions on the safe side.

Purpose: Urapidil is putatively effective for patients with hypertension and acute heart failure, although randomized controlled trials thereon are lacking. We investigated the efficacy and safety of intravenous urapidil relative to that of nitroglycerin in older patients with hypertension and heart failure in a randomized controlled trial. Materials and Methods: Patients (>60 y) with hypertension and heart failure were randomly assigned to receive intravenous urapidil (n=89) or nitroglycerin (n=91) for 7 days. Hemodynamic parameters, cardiac function, and safety outcomes were compared. Results: Patients in the urapidil group had significantly lower mean systolic blood pressure (110.1±6.5 mm Hg) than those given nitroglycerin (126.4±8.1 mm Hg, p=0.022), without changes in heart rate. Urapidil was associated with improved cardiac function as reflected by lower N terminal-pro B type natriuretic peptide after 7 days (3311.4±546.1 ng/mL vs. 4879.1±325.7 ng/mL, p=0.027) and improved left ventricular ejection fraction (62.2±3.4% vs. 51.0±2.4%, p=0.032). Patients given urapidil had fewer associatedadverse events, specifically headache (p=0.025) and tachycardia (p=0.004). The one-month rehospitalization and all-cause mortality rates were similar. Conclusion: Intravenous administration of urapidil, compared with nitroglycerin, was associated with better control of blood pressure and preserved cardiac function, as well as fewer adverse events, for elderly patients with hypertension and acute heart failure.

The red stigmas of saffron are one of the mostexpensive spices in the world and serve as a traditionalChinese medicine. More saffron has been cultivated inChina, and different drying technologies have been studied. However, a comprehensive and comparative analysis ofdifferent drying approaches has not been well studied. Inthis study, we compared electric oven and vacuum ovendrying approaches on saffron. We found saffron was driedquicker under high vacuum drying mode with high temperatureand the quicker drying rate provided, the moreopen microstructural interstices on the saffron surface. Both methods were best fit to Midilli and Kucuk model. Besides, the coloring, aroma and bitterness strength afterdrying showed the similar results. In sum, our data suggestedthe optimal drying temperature was 100 C for20 min for two evaluated methods, however consideringthe machine cost, the electric oven drying would be the firstchoice.

BACKGROUND: Human umbilical cord mesenchymal stem cells (hUCMSCs) have emerged as promising therapy for immune and inflammatory diseases. However, how to maintain the activity and unique properties during cold storage and transportation is one of the key factors affecting the therapeutic efficiency of hUCMSCs. Schisandrin B (SchB) has many functions in cell protection as a natural medicine. In this study, we investigated the protective effects of SchB on the hypothermic preservation of hUCMSCs. METHODS: hUCMSCs were isolated from Wharton’s jelly. Subsequently, hUCMSCs were exposed to cold storage (4 C) and 24-h re-warming. After that, cells viability, surface markers, immunomodulatory effects, reactive oxygen species (ROS), mitochondrial integrity, apoptosis-related and antioxidant proteins expression level were evaluated. RESULTS: SchB significantly alleviated the cells injury and maintained unique properties such as differentiation potential, level of surface markers and immunomodulatory effects of hUCMSCs. The protective effects of SchB on hUCMSCs after hypothermic storage seemed associated with its inhibition of apoptosis and the anti-oxidative stress effect mediated by nuclear factor erythroid 2–related factor 2 signaling. CONCLUSION: These results demonstrate SchB could be used as an agent for hypothermic preservation of hUCMSCs.
