Objectives: To assess knowledge of HPV and attitudes towards HPV vaccination among the general female population, government officials, and healthcare providers in China to assist the development of an effective national HPV vaccination program. Methods: A cross-sectional epidemiologic survey was conducted across 21 urban and rural sites in China using a short questionnaire. 763 government officials, 760 healthcare providers, and 11,681 women aged 15-59 years were included in the final analysis. Data were analyzed using standard descriptive statistics and logistic regression. Results: Knowledge of HPV among the general female population was low; only 24% had heard of HPV. Less than 20% of healthcare providers recognized sexually na$\ddot{i}$ve women as the most appropriate population for HPV vaccination. There was high acceptance of the HPV vaccine for all categories of respondents. Only 6% of women were willing to pay more than US $300 for the vaccine. Conclusions: Aggressive education is necessary to increase knowledge of HPV and its vaccine. Further proof of vaccine safety and efficacy and government subsidies combined with increased awareness could facilitate development and implementation of HPV vaccination in China.

Background: Evidence supporting an association between the 8q24 rs4242382-A polymorphism and prostate cancer (PCa) risk has been reported in North American and Europe populations, though data from Asian populations remain limited. We therefore investigated this association by clinical detection in China, and meta-analysis in Asian, Caucasian and African-American populations. Materials and Methods: Blood samples and clinical information were collected from ethnically Chinese men from Northern China with histologically-confirmed PCa (n=335) and from age-matched normal controls (n=347). The 8q24 (rs4242382) gene polymorphism was genotyped by polymerase chain reaction-high-resolution melting analysis. We initially analyzed the associations between the risk allele and PCa and clinical covariates. A meta-analysis was then performed using genotyping data from a total of 1,793 PCa cases and 1,864 controls from our study and previously published studies in American and European populations, to determine the association between PCa and risk genotype. Results: The incidence of the risk allele was higher in PCa cases than controls (0.222 vs 0.140, $P=7.3{\times}10^{-5}$), suggesting that the 8q24 rs4242382-A polymorphism was associated with PCa risk in Chinese men. The genotypes in subjects were in accordance with a dominant genetic model (ORadj=2.03, 95%CI: 1.42-2.91, $Padj=1.1{\times}10^{-4}$). Presence of the risk allele rs4242382-A at 8q24 was also associated with clinical covariates including age at diagnosis ${\geq}65$ years, prostate specific antigen >10 ng/ml, Gleason score <8, tumor stage and aggressive PCa, compared with the non-risk genotype ($P=4.6{\times}10^{-5}-3.0{\times}10^{-2}$). Meta-analysis confirmed the association between 8q24 rs4242382-A polymorphism and PCa risk (OR=1.62, 95%CI: 1.39-1.88, $P=1.0{\times}10^{-5}$) across Asian, Caucasian and African American populations. Conclusions: The replicated data suggest that the 8q24 rs4242382-A variation might be associated with increased PCa susceptibility in Asian, Caucasian and African American populations. These results imply that this polymorphism may be a useful risk biomarker for PCa in multi-ethnic populations.
Susceptibility Loci Associations with Prostate Cancer Risk in Northern Chinese Men

Background: KLK3 gene products, like human prostate-specific antigen (PSA), are important biomarkers in the clinical diagnosis of prostate cancer (PCa). G protein-coupled receptor RFX6, C2orf43 and FOXP4 signaling plays important roles in the development of PCa. However, associations of these genes with PCa in northern Chinese men remain to be detailed. This study aimed to investigate their impact on occurrence and level of malignancy. Methods: All subjects were from Beijing and Tianjin, including 266 cases with prostate cancer and 288 normal individuals as controls. We evaluated associations between clinical covariates (age at diagnosis, prostate specific antigen, Gleason score, tumor stage and aggressive) and 6 candidate PCa risk loci, genotyped by PCR- high resolution melting curve and sequencing methods. Results: Case-control analysis of allelic frequency of PCa associated with PCa showed that one of the 6 candidate risk loci, rs339331 in the RFX6 gene, was associated with reduced risk of prostate cancer (odds ratio (OR) = 0.73, 95% confidence interval (CI) =0.57-0.94, P = 0.013) in northern Chinese men. In addition, subjects with CX (CC+TC) genotypes had a decreased risk for prostrate cancer compared to those carrying the TT homozygote (OR =0.64, 95% CI = 0.45- 0.90, P = 0.008). The TT genotype of 13q22 (rs9600079, T) was associated with tumor stage (P=0.044, OR=2.34, 95% CI=0.94-5.87). Other SNPs were not significantly associated with clinical covariates in prostate cancer (P > 0.05). Conclusions. rs339331 in the RFX6 gene may be associated with prostate cancer as a susceptibility locus in northern Chinese men.

Purpose We sought to investigate the effectiveness and safety of dabrafenib in children with BRAFV600E-mutated Langerhans cell histiocytosis (LCH). Materials and Methods A retrospective analysis was performed on 20 children with BRAFV600E-mutated LCH who were treated with dabrafenib. Results The median age at which the patients started taking dabrafenib was 2.3 years old (range, 0.6 to 6.5 years). The ratio of boys to girls was 2.3:1. The median follow-up time was 30.8 months (range, 18.9 to 43.6 months). There were 14 patients (70%) in the risk organ (RO)+ group and six patients (30%) in the RO– group. All patients were initially treated with traditional chemotherapy and then shifted to targeted therapy due to poor control of LCH or intolerance to chemotherapy. The overall objective response rate and the overall disease control rate were 65% and 75%, respectively. During treatment, circulating levels of cell-free BRAFV600E (cfBRAFV600E) became negative in 60% of the patients within a median period of 3.0 months (range, 1.0 to 9.0 months). Grade 2 or 3 adverse effects occurred in five patients. Conclusion Some children with BRAFV600E-mutated LCH may benefit from monotherapy with dabrafenib, especially high-risk patients with concomitant hemophagocytic lymphohistiocytosis and intolerance to chemotherapy. The safety of dabrafenib is notable. A prospective study with a larger sample size is required to determine the optimal dosage and treatment duration.
Antennal UDP-glycosyltransferase genes in the coffee white stemborer, Xylotrechus quadripes

The antenna of Xylotrechus quadripes is the principle olfactory organ that is subjected to a large number of endogenous and exogenous compounds. The gene families associated with the detoxification of these compounds are essential for the adaptive evolution of insect defensive strategies. However, knowledge on uridine diphosphate (UDP)-glycosyltransferases (UGTs) of X. quadripes is unavailable. Here, we characterized 30 UGT genes identified from an antennal transcriptome of X. quadripes. Among them, 16 UGT genes encoding 508–527 amino acids shared the full-length sequences and signal peptides in N-terminus. Multiple sequence alignment revealed that X. quadripes UGTs had a variable N-terminus and a conserved C-terminus. Phylogenetic analysis showed that X. quadripes UGTs were classified into ten sub-families with the largest UGT one of UGT352 (nine genes) and a strict single copy of UGT50 within coleopteran species. Gene structural analysis indicated that coleopteran UGT50s underwent intron gains or losses. Expression profile revealed that all studied X. quadripes UGTs were transcribed in the antennae of both sexes, some of which exhibited sex-biased expression including UGT2, UGT6, UGT20 and UGT27 in females as well as UGT3, UGT11 and UGT12 in males. In addition, most of UGTs were widely expressed in other tissues, indicating their functional diversities in this beetle. Together, these findings provide valuable information for further functional studies of UGTs in X. quadripes, especially their roles in olfaction.
Mapping of QTLs controlling content of fatty acid composition in rapeseed (Brassica napus)

The improvement of fatty acid composition is one of the major goals of breeding in rapeseed (Brassica napus). The aim of this study was to provide more information on the genetic determination of fatty acid composition by investigating quantitative trait loci (QTLs). The study was based on two-year of field trials (in 2006 and 2007) with a population of recombinant inbred lines (RILs), which originated from a cross between GH06 and P174. The level of erucic acid (C22:1) was significantly negatively correlated with those of palmitic acid (C16:0), oleic acid (C18:1), linoleic acid (C18:2), linolenic acid (C18:3), and eicosenoic acid (C20:1) in both years. A total of 40 QTLs for six fatty acids were detected and most of them were clustered on linkage groups N8, N9, and N13. These results suggested strongly that there were significant correlations between the levels of fatty acid components and would be useful for the future improvement of breeding programs focused on fatty acids in rapeseed.

The multi functionalization of isotactic polypropylenes (iPP) with glycidyl methacrylate (GMA) was carried out by in situ chlorinating graft copolymerization (ISCGC), in which chlorine (Cl2) is used as a radical initiator as well as a radical scavenger. The molecular weight measurement indicates that functionalized iPP with controlled degradation could be obtained because the unstable radicals that induce iPP degradation could be terminated quickly by chlorine. Owing to the unique allyl-containing structure of GMA, multi functionalized iPP could be synthesized under the effect of chlorine, which contained both epoxy groups and C=C double bonds as well as trace amount of chlorine atoms in the functionalized polymer structure. The structure of both the functionalization polymer and homopolymer formed in this system were analyzed by FTIR and ^1H NMR. The mechanism of grafting reaction, iPP chain degradation, and GMA homopolymer formation scheme in ISCGC were proposed. The thermal properties of the functionalized polymer are also discussed.
