        Mapping of QTLs controlling content of fatty acid composition in rapeseed (Brassica napus)

        The improvement of fatty acid composition is one of the major goals of breeding in rapeseed (Brassica napus). The aim of this study was to provide more information on the genetic determination of fatty acid composition by investigating quantitative trait loci (QTLs). The study was based on two-year of field trials (in 2006 and 2007) with a population of recombinant inbred lines (RILs), which originated from a cross between GH06 and P174. The level of erucic acid (C22:1) was significantly negatively correlated with those of palmitic acid (C16:0), oleic acid (C18:1), linoleic acid (C18:2), linolenic acid (C18:3), and eicosenoic acid (C20:1) in both years. A total of 40 QTLs for six fatty acids were detected and most of them were clustered on linkage groups N8, N9, and N13. These results suggested strongly that there were significant correlations between the levels of fatty acid components and would be useful for the future improvement of breeding programs focused on fatty acids in rapeseed.

        Experimental and Numerical Investigations on the Spherical Steel Bearing Capacity of New Anti-Separation Design

        To solve the issues of slippage and dislodging caused by insufficient grout compactness in railway bridge spherical steel bearings, a novel kind of anti-separation spherical steel bearing is presented. Firstly, a comparative test model was meticulously designed and constructed for the bearing-stone assembly, encompassing both the traditional spherical steel bearing and the innovative anti-separation spherical steel bearing. Next, mechanical performance testing and finite element simulation analysis were completed. Finally, an empirical equation for the vertical load-strain fitting of the bearing was established. The results demonstrate that, under the maximum vertical static load, the convex spherical crown anti-separation design of the new anti-dislodging bearing is not sensitive to the bearing performance of the bearing-bedding stone combination. All indexes also meet the specifications. Under the action of cyclic load, the error of each parameter between finite element analysis and test results is less than 7%, which shows that the new anti- separation spherical bearing has higher bearing capacity and better fatigue resistance; the fitted value of the established bearing vertical load-strain calculation formula is in agreement with the test value by more than 0.95, which can predict whether the new anti-separation bearing fails.

        Genetic variation and phylogenetic relationships of the ectomycorrhizal Floccularia luteovirens on the Qinghai-Tibet Plateau

        Floccularia luteovirens, as an ectomycorrhizal fungus, is widely distributed in the Qinghai-Tibet Plateau. As an edible fungus, it is famous for its unique flavor. Former studies mainly focus on the chemical composition and genetic structure of this species. However, the phylogenetic relationship between genotypes remains unknown. In this study, the genetic variation and phylogenetic relationship between the genotypes of F. luteovirens in Qinghai-Tibet Plateau was estimated through the analysis on two protein-coding genes (rpb1 and ef-1α) from 398 individuals collected from 24 wild populations. The sample covered the entire range of this species during all the growth seasons from 2011 to 2015. 13 genotypes were detected and moderate genetic diversity was revealed. Based on the results of network analysis, the maximum likelihood (ML), maximum parsimony (MP), and Bayesian inference (BI) analyses, the genotypes H-1, H-4, H-6, H-8, H-10, and H-11 were grouped into one clade. Additionally, a relatively higher genotype diversity (average h value is 0.722) and unique genotypes in the northeast edge of Qinghai- Tibet plateau have been found, combined with the results of mismatch analysis and neutrality tests indicated that Southeast Qinghai-Tibet plateau was a refuge for F. luteovirens during the historical geological or climatic events (uplifting of the Qinghai-Tibet Plateau or Last Glacial Maximum). Furthermore, the present distribution of the species on the Qinghai-Tibet plateau has resulted from the recent population expansion. Our findings provide a foundation for the future study of the evolutionary history and the speciation of this species.

        An Adaptation Method in Noise Mismatch Conditions for DNN-based Speech Enhancement

        The deep learning based speech enhancement has shown considerable success. However, it still suffers performance degradation under mismatch conditions. In this paper, an adaptation method is proposed to improve the performance under noise mismatch conditions. Firstly, we advise a noise aware training by supplying identity vectors (i-vectors) as parallel input features to adapt deep neural network (DNN) acoustic models with the target noise. Secondly, given a small amount of adaptation data, the noise-dependent DNN is obtained by using L2 regularization from a noise-independent DNN, and forcing the estimated masks to be close to the unadapted condition. Finally, experiments were carried out on different noise and SNR conditions, and the proposed method has achieved significantly 0.1%-9.6% benefits of STOI, and provided consistent improvement in PESQ and segSNR against the baseline systems.

        Inhibitory effects of piceatannol on human cytomegalovirus (hCMV) in vitro

        Human cytomegalovirus (hCMV) is a ubiquitous herpesvirus, which results in the establishment of a latent infection that persists throughout the life of the host and can be reactivated when the immunity is low. Currently, there is no vaccine for hCMV infection, and the licensed antiviral drugs mainly target the viral enzymes and have obvious adverse reactions. Thus, it is important to search for compounds with antihCMV properties. The present study aimed to investigate the suppressive effects of piceatannol on hCMV Towne strain infection and the putative underlying mechanisms using human diploid fibroblast WI-38 cells. Piceatannol supplementation prevented the lytic changes induced by hCMV infection in WI-38 cells. Furthermore, piceatannol suppressed the expression of hCMV immediate-early (IE) and early (E) proteins as well as the replication of hCMV DNA in a dose-dependent manner. Moreover, hCMV-induced cellular senescence was suppressed by piceatannol, as shown by a decline in the senescence-associated β-galactosidase (SA-β-Gal) activity and decreased production of intracellular reactive oxygen species (ROS). p16INK4a, a major senescence-associated molecule, was dramatically elevated by current hCMV infection that was attenuated by pre-incubation with piceatannol in a dose-dependent manner. These results demonstrated that piceatannol suppressed the hCMV infection via inhibition of the activation of p16INK4a and cellular senescence induced by hCMV. Together, these findings indicate piceatannol as a novel and potent anti-hCMV agent with the potential to be developed as an effective treatment for chronic hCMV infection.

        Isolation and Characterization of Parthenogenetic Embryonic Stem (pES) Cells Containing Genetic Background of the Kunming Mouse Strain

        Parthenogenetic embryonic stem (pES) cells could provide a valuable model for research into genomic imprinting and X-linked diseases. In this study, pES cell lines were established from oocytes of hybrid offspring of Kunming and 129/Sv mice, and pluripotency of pES cells was evaluated. The pES cells maintained in the undifferentiated state for more than 50 passages had normal karyotypes with XX sex chromosomes and exhibited high activities of alkaline phosphatase (AKP) and telomerase. Meanwhile, these cells expressed ES cell molecular markers SSEA-1, Oct-4, Nanog, and GDF3 but not SSEA-3 detected by immunohistochemistry and RT-PCR. The pES cells could be differentiated into various types of cells from three germ layers in vitro by analysis of embryoid bodies (EBs) with immunohistochemistry and RT-PCR, and in vivo by observation of pES cell-derived teratoma sections. Therefore, the established pES cell lines contained all features of mouse ES cells. This work provides a new strategy for isolating pES cells from Kunming mice, and the pES cell lines could be applied as the cell model in research into genomic imprinting and epigenetic regulation of Kunming mice.

      • ANXA2 Regulates the Behavior of SGC-7901 Cells

        ANXA2, a member of the annexin family, is overexpressed and plays important roles in tumor development. However, the significance of ANXA2 expression in gastric carcinoma has not been clarified.To elucidate its roles in growth of gastric cancer, ANXA2 expression in SGC-7901 cells was inhibited with a designated siRNA, then cell proliferation, cell cycling, apoptosis and motility were determined by MTT assay, flow cytometry, Hoechst 33342 staining and wound healing assay, respectively. To further assess the behavior of ANXA2 deleted SGC-7901 cells, changes of microstructures were observed under fluorescence microscopy, laser scanning confocal microscopy and electron microscopy. We found that inhibition of ANXA2 expression caused cell proliferation to decrease significantly with G1 arrest, motility to be reduced with changes in pseudopodia/filopodia structure and F-actin and ${\beta}$-tubulin expression, and apoptosis to be enhanced albeit without significance. At the same time, ANXA2 deletion resulted in fewer pseudopodia/filopodia, non-stained areas were increased, contact inhibition among cells reappeared, and expression of F-actin and ${\beta}$-tubulin was decreased, with induction of polymerized disassembled forms. Taken together, these data suggest that ANXA2 overexpression is important to maintain the malignancy of cancer cells, and this member of the annexin family has potential to be considered as a target for the gene therapy of gastric carcinoma.

        DFT and Time-dependant DFT Investigation of eLectronic Structure, Phosphorescence and Electroluminescence Properties of Iridium (III) Quinoxaline Complexes

        Quantum-chemistry study was explored to investigate the electronic structures, absorption and phosphorescence mechanism, as well as electroluminescence (EL) properties of three red-emitting Ir(III) complexes, $(fpmqx)_2Ir$(L) {fpmqx=2-(4-fluorophenyl)-3-methyl-quinoxaline; L=triazolylpyridine (trz) (1); L=picolinate (pic) (2) and L=acetylacetonate (acac) (3)}. The calculated results show that the HOMO distribution for 1 is mainly localized on trz moiety due to its stronger ${\pi}$-electron acceptor ability, and HOMO for 2 and 3 is the combination of Ir d- and phenyl ring ${\pi}$-orbital. The higher phosphorescence yields and differences among 1-3 are investigated in this paper. In addition, the reasons of higher EL efficiency of 2 than 1 and 3 have been rationalized. 3개의 적색 발광 Ir(III) 착물들인 $(fpmqx)_2Ir$(L) {fpmqx=2-(4-fluorophenyl)-3-methyl-quinoxaline; L=triazolylpyridine (trz) (1); L=picolinate (pic) (2) and L=acetylacetonate (acac) (3)}의 전자 구조, 흡수 및 인광 메커니즘, 전기 발광(EL) 특성을 양자화학적으로 연구하였다. 계산 결과에 따르면, 1의 HOMO는 강한 ${\eth}$-전자 받개 능력을 갖는 trz 부분에 편재되어 있으며, 2와 3의 HOMO는 Ir d-오비탈 과 페닐 고리 ${\pi}$-오비탈의 결합이라는 것을 나타내었다. 이 논문에서는 1-3사이의 인광 수득률과 차이에 대하여 연구하였으며, 1과 3보다 2의 EL 효율이 더 큰 이유를 합리적으로 설명하였다.

        Anatomical Study of the Accessory Tendon of the Extensor Hallucis Longus Muscle and Its Clinical Application

        Background: The accessory tendon of the extensor hallucis longus (ATEHL) muscle is a common abnormal structure, and its clinical significance remains debatable. In this study, we provide the incidence of the ATEHL and characterize its morphological types in Asian cadavers and investigate its clinical applications. Methods: The tendons from 50 adult cadaveric feet, fixed in 10% formalin, were analyzed. We measured the length and width of both the ATEHL and the extensor hallucis brevis (EHB). Results: All dissected specimens had an ATEHL. The first metatarsophalangeal joint was surrounded by an accessory tendon that inserted onto the joint capsule and the dorsal base of the proximal phalanx. We classified the ATEHL into 3 types based on their directions. Differences in ATEHL type based on sex were not statistically significant. Conclusions: We found an ATEHL in all cadaveric specimens in this study. We surmise that the ATEHL acts as an antagonist with the EHB when the toe is extending, which might help prevent the occurrence of hallux valgus deformity.
