A Prediction Model Based on the Risk Factors Associated with Pathological Upgrading in Patients with Early-Stage Gastric Neoplasms Diagnosed by Endoscopic Forceps Biopsy

        Zhao Yu Han,Zheng Yu,Sha Jie,Hua Hong Jin,Li Ke Dong,Lu Yu,Dang Yi Ni,Zhang Guo Xin 거트앤리버 소화기연관학회협의회 2023 Gut and Liver Vol.17 No.1

Background/Aims: The discrepancies between the diagnosis of preoperative endoscopic forceps biopsy (EFB) and endoscopic submucosal dissection (ESD) in patients with early gastric neoplasm (EGN) exist objectively. Among them, pathological upgrading directly influences the accuracy and appropriateness of clinical decisions. The aims of this study were to investigate the risk factors for the discrepancies, with a particular focus on pathological upgrading and to establish a prediction model for estimating the risk of pathological upgrading after EFB. Methods: We retrospectively collected the records of 978 patients who underwent ESD from December 1, 2017 to July 31, 2021 and who had a final histopathology determination of EGN. A nomogram to predict the risk of pathological upgrading was constructed after analyzing subgroup differences among the 901 lesions enrolled. Results: The ratio of pathological upgrading was 510 of 953 (53.5%). Clinical, laboratorial and endoscopic characteristics were analyzed using univariable and binary multivariable logistic regression analyses. A nomogram was constructed by including age, history of chronic atrophic gastritis, symptoms of digestive system, blood high density lipoprotein concentration, macroscopic type, pathological diagnosis of EFB, uneven surface, remarkable redness, and lesion size. The C-statistics were 0.804 (95% confidence interval, 0.774 to 0.834) and 0.748 (95% confidence interval, 0.664 to 0.832) in the training and validation set, respectively. We also built an online webserver based on the proposed nomogram for convenient clinical use. Conclusions: The clinical value of identifying the preoperative diagnosis of EGN lesions is limited when using EFB separately. We have developed a nomogram that can predict the probability of pathological upgrading with good calibration and discrimination value.

Transcriptional activation of insulin-like growth factor binding protein 6 by 17β-estradiol in SaOS-2 cells

        Yu-yan Zhao,Lei Guo,Xiao-juan Zhao,Hong Liu,Tian Lei,Dong-jie Ma,Xiao-yu Gao 생화학분자생물학회 2009 Experimental and molecular medicine Vol.41 No.7

Osteoblasts can synthesize the insulin-like growth factors (IGFs) and the IGF-binding proteins (IGFBPs), which may either enhance or attenuate IGF-stimulated bone cell proliferation. Since estrogen induced osteoblastic differentiation and proliferation through an estrogen- responsive gene in target cells, we investigated the effects of estrogen on IGFBP-6 expression in the human osteoblastic-like cell line SaOS-2. Expressions of IGFBP-6 protein and mRNA increased 2.8 and 2-fold, respectively, in the presence of 17-β-estradiol (E2) (0.01 to 1 μM) and estrogen receptor (ER) in SaOS-2 cells. On the other hand, E2 induced a 2-fold increase in SaOS-2 cell proliferation. To identify genomic sequences associated with estrogen responsiveness, the 5'-promoter region (-44 to +118) of the IGFBP-6 gene was cloned into a chloramphenicol acetyltransferase (CAT) reporter vector. E2 induced a 3-fold increase in CAT activity in SaOS-2 cells transiently transfected with this construct. Identification of the estrogen- responsive element (ERE) [5'-CCTTCA CCTG-3'] (-9 to +1) in this IGFBP-6 gene promoter region was confirmed using electromobility shift assays and deletion analysis. This functional ERE was important for E2-induced trans-activation of the IGFBP-6 gene. These results demonstrate that E2 exhibits a positive effect on IGFBP-6 gene transcription through estrogen- liganded ER binding to the functional ERE in the IGFBP-6 gene promoter in SaOS-2 cells. Intravenous Tenecteplase for Acute Ischemic Stroke Within 4.5–24 Hours of Onset (ROSE-TNK): A Phase 2, Randomized, Multicenter Study

        Wang Lu,Dai Ying-Jie,Cui Yu,Zhang Hong,Jiang Chang-Hao,Duan Ying-Jie,Zhao Yong,Feng Ye-Fang,Geng Shi-Mei,Zhang Zai-Hui,Lu Jiang,Zhang Ping,Zhao Li-Wei,Zhao Hang,Ma Yu-Tong,Song Cheng-Guang,Zhang Yi,Ch 대한뇌졸중학회 2023 Journal of stroke Vol.25 No.3

Background and Purpose Intravenous tenecteplase (TNK) efficacy has not been well demonstrated in acute ischemic stroke (AIS) beyond 4.5 hours after onset. This study aimed to determine the effect of intravenous TNK for AIS within 4.5 to 24 hours of onset. Methods In this pilot trial, eligible AIS patients with diffusion-weighted imaging (DWI)-fluid attenuated inversion recovery (FLAIR) mismatch were randomly allocated to intravenous TNK (0.25 mg/kg) or standard care within 4.5–24 hours of onset. The primary endpoint was excellent functional outcome at 90 days (modified Rankin Scale [mRS] score of 0–1). The primary safety endpoint was symptomatic intracranial hemorrhage (sICH). Results Of the randomly assigned 80 patients, the primary endpoint occurred in 52.5% (21/40) of TNK group and 50.0% (20/40) of control group, with no significant difference (unadjusted odds ratio, 1.11; 95% confidence interval 0.46–2.66; P=0.82). More early neurological improvement occurred in TNK group than in control group (11 vs. 3, P=0.03), but no significant differences were found in other secondary endpoints, such as mRS 0–2 at 90 days, shift analysis of mRS at 90 days, and change in National Institutes of Health Stroke Scale score at 24 hours and 7 days. There were no cases of sICH in this trial; however, asymptomatic intracranial hemorrhage occurred in 3 of the 40 patients (7.5%) in the TNK group. Conclusion This phase 2, randomized, multicenter study suggests that intravenous TNK within 4.5–24 hours of onset may be safe and feasible in AIS patients with a DWI-FLAIR mismatch.

      • Acupuncture reduces nicotine-induced norepinephrine release in the hypothalamus via the solitary NMDA receptor/NOS pathway

        Liu, Hong Feng,Zhao, ZhengLin,Zhang, Jie,Wu, Yi Yan,Jiao, Yu,Wu, Tong,Kim, Sang Chan,Lee, Bong Hyo,Fan, Yu,Lee, Chul Won,Kim, Young Woo,Yang, Chae Ha,Zhu, Xiao Dong,Zhao, Rong Jie Elsevier 2019 Neuroscience Letters Vol.705 No.-

Abstract

Noradrenergic projections from the nucleus tractus solitarius (NTS) to the hypothalamic paraventricular nucleus (PVN) are involved in nicotine (Nic) dependence. Nic induces hypothalamic norepinephrine (NE) release through N-methyl-D-aspartate receptors (NMDARs) and nitric oxide in the NTS. However, acupuncture attenuates Nic withdrawal-induced anxiety. Therefore, this study investigated the effects of acupuncture on Nic-induced hypothalamic NE release. Rats received an intravenous infusion of Nic (90 μg/kg, over 60 s) and extracellular NE levels in the PVN were determined by in vivo microdialysis. Immediately after Nic administration, the rats were bilaterally treated with acupuncture at acupoint HT7 (Shen-Men) or PC6 (Nei-Guan), or a non-acupoint (tail) for 60 s. Acupuncture at HT7, but not at PC6 or the tail, significantly reduced Nic-induced NE release. However, this was abolished by a post-acupuncture infusion of either NMDA or sodium nitroprusside into the NTS. Additionally, acupuncture at HT7, but not the control points, prevented Nic-induced plasma corticosterone secretion and inhibited Nic-induced increases in the phosphorylation of neuronal nitric oxide synthase (nNOS) and endothelial NOS in the NTS. These findings suggest that acupuncture at HT7 reduces Nic-induced NE release in the PVN via inhibition of the solitary NMDAR/NOS pathway.

Highlights

Acupuncture inhibited nicotine-induced hypothalamic norepinephrine release.

Acupuncture diminished nicotine-induced plasma corticosterone secretion.

Acupuncture reversed nicotine-induced phosphorylation of solitary nNOS and eNOS.

NMDA abolished acupunctural effects on nicotine-induced norepinephrine release.

NO abolished acupunctural effects on nicotine-induced norepinephrine release.

Whole Cell-mediated Biocatalytic Synthesis of Helicid Cinnamylate and Its Biological Evaluation as a Novel Tyrosinase Inhibitor

        Rong-ling Yang,Xi Chen,Yu-ye Song,Qian-lin Zhu,Muhammad Bilal,Yu Wang,Zheng Tong,Ting-ting Wu,Zhao-Yu Wang,Hong-zhen Luo,Xiang-jie Zhao,Ting-ting He 한국생물공학회 2022 Biotechnology and Bioprocess Engineering Vol.27 No.3

Tyrosinase inhibitors are clinically effective for treating some dermatological disorders related to melanin hyperpigmentation. Accordingly, the discovery and development of tyrosinase inhibitors have great value in the pharmaceutical and cosmetic industry. Here, a novel tyrosinase inhibitor, 6′-O-cinnamoyl-helicid (helicid cinnamylate) was successfully synthesized by a simple and effective biocatalytic approach with Aspergillus oryzae cells. Investigation of the effects of several key variables on helicid cinnamylate synthesis found that the reaction conversion, reaction rate and regioselectivity reached 99%, 9.40 mM/h and > 99%, respectively, at the optimal conditions with anhydrous acetone as the solvent, whole-cell concentration of 40 mg/mL, and the molar ratio of vinyl cinnamate to helicid of 10 at 45°C. The whole-cells retained 68.87% of its initial activity after reusing for seven batches, indicating a potent application potential in non-aqueous biocatalytic systems. It was worth noting that helicid cinnamylate demonstrated a more potent tyrosinase inhibitory activity with an IC50 value of 3.55 mM than helicid (IC50 = 4.48 mM) and arbutin (IC50 = 5.48 mM), which suggest that helicid cinnamylate could be developed as a more potential tyrosinase inhibitor. In conclusion, this study provides a novel whole-cell catalytic approach for the synthesis of helicid cinnamylate and insight into its application as a tyrosinase inhibitor.

      • Expression of Connexin 43 and E-cadherin Protein and mRNA in Non-small Cell Lung Cancers in Chinese Patients

        Zhao, Jun-Qiang,Sun, Fang-Jie,Liu, Shan-Shan,Yang, Jun,Wu, Yu-Quan,Li, Gui-Shan,Chen, Qing-Yong,Wang, Jia-Xiang Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.2

Aim: Connexin 43 (Cx43) and E-cadherin are important biomarkers related with cancer. Their expression at protein and mRNA levels was here investigated in 50 primary lung carcinoma tissues and 20 samples of adjacent normal tissue of Chinese patients with non-small cell lung cancer (NSCLC). Methods: Protein and mRNA expression were evaluated by ABC immunohistochemistry and RT-PCR. Results: (1) The positive expression rates of Cx43 and E-cadherin protein were higher in the adjacent normal tissues than those in the primary lung carcinoma tissues; (2) the positive expression rates of Cx43 and E-cadherin protein decreased with NSCLC progression; (3) the expression of E-cadherin protein was not related with the pathological type of NSCLC; and (4) the relative quantity of the Cx43 or E-cadherin mRNA expression was correlated with the the histological type, clinical stage, cancer cell differentiation and the lymph node metastasis. Conclusion: The data suggested that the Cx43 and E-cadherin are reduced with NSCLC progression, and might be important biomarkers for judging the metastasis and prognosis.

Effects of Three Types of Resistant Starch on Intestine and Their Gastric Ulcer Preventive Activities in vivo

        Yu Qian,Xin Zhao,Gui-Jie Li,Kai Zhu,Huayi Suo,Peng Sun 한국응용생명화학회 2013 Applied Biological Chemistry (Appl Biol Chem) Vol.56 No.6

To examine the effects of three types of resistant starch (RS) on intestinal metabolites, structure and their gastric injury preventive activities, Sprague-Dawley mice were fed diet containing 15% RS for 4 weeks. Response relationships among three types of RS (RS2, RS3, and RS4) food intake, body weight gain, food efficiency, wet weight of cecum (with or not with contents), pH, ammonia production, and short-chain fatty acids (SCFAs) concentration of cecal contents as well as intestinal structure were investigated. All three types of RS had effects on food intake, weight control, lowering of pH, ammonia production in cecal contents, increasing the wet weight of cecum (with or without contents), changes in SCFAs concentration of cecal contents, and physiological structure of small intestine and cecum compared to the control group (common starch-fed rats). RS3group showed minimum weight gain and food efficiency, compared to both the control group and the other two types of RS diet. RS3 group showed more significant lowering of pH,ammonia production, and SCFAs of cecal contents. In addition, all three types of RS shortened villous height, and muscle and mucosal thickening in small intestine, resulting in significantly decreased villous height and mucosal thickness, whereas significantly increased muscle thickness in the cecum. However, they did not cause detectable pathological changes in the small intestine and cecum. Animal model was also used to check its gastric ulcer preventive effects. The gastric ulcer level was significantly down-regulated by RS3, demonstrating its antiinflammatory properties. RS3 reduced the levels of serum proinflammatory cytokines of IL-6, IL-12, TNF-α, and IFN-γcompared to RS2 and RS4. RS3 increased the somatostatin (SS)and vasoactive intestinal peptide (VIP), decreased the motillin (MOT) and substance P (SP) serum levels. These results suggest that RS3 shows the best gastric ulcer preventive effect.

      • Streptomyces xinghaiensis sp. nov., isolated from marine sediment.

        Zhao, Xin-Qing,Li, Wen-Jun,Jiao, Wen-Ce,Li, Yan,Yuan, Wen-Jie,Zhang, Yu-Qin,Klenk, Hans-Peter,Suh, Joo-Won,Bai, Feng-Wu Society for General Microbiology 2009 International journal of systematic and evolutiona Vol.59 No.11

A novel actinomycete, strain S187(T), was isolated from a marine sediment sample collected from Xinghai Bay, Dalian, China. Growth occurred on ISP medium 2 containing 0-9 % NaCl and at pH 6.0-9.0 and 10-45 degrees C. The cell wall of strain S187(T) contained the isomer ll-diaminopimelic acid as the diagnostic diamino acid. The predominant menaquinones were MK-9(H(6)) (40.8 %), MK-9(H(8)) (38.2 %) and MK-9(H(2)) (8.8 %). The major fatty acids were iso-C(16 : 0) (29.6 %), anteiso-C(15 : 0) (14.0 %) and anteiso-C(17 : 0) (11.6 %). Cells contained phosphatidylethanolamine, phosphatidylinositol, phosphatidylglycerol, phosphatidylinositol mannosides and one unknown phospholipid. The G+C content of the genomic DNA was 72.01 mol%. The 16S rRNA gene sequence of the isolate had similarities of 98.1 and 97.5 % with those of Streptomyces flavofuscus NRRL B-8036(T) (=DSM 41426(T)) and Streptomyces albiaxialis DSM 41799(T), respectively, showing that the novel strain should be assigned to the genus Streptomyces. DNA-DNA hybridizations with the two above-mentioned Streptomyces species showed 31.4 and 46.9 % relatedness, respectively. Moreover, the three strains differed in some physiological and biochemical properties. Thus, on the basis of phenotypic and genotypic analyses, it is proposed that strain S187(T) represents a novel species of the genus Streptomyces, for which the name Streptomyces xinghaiensis sp. nov. is proposed; the type strain is S187(T) (=NRRL B-24674(T)=CCTCC AA 208049(T)=KCTC 19546(T)).
