        Hydrogen sulfide, a gaseous signaling molecule, elongates primary cilia on kidney tubular epithelial cells by activating extracellular signal-regulated kinase

        Primary cilia on kidney tubular cells play crucial roles in maintaining structure and physiological function. Emerging evidence indicates that the absence of primary cilia, and their length, are associated with kidney diseases. The length of primary cilia in kidney tubular epithelial cells depends, at least in part, on oxidative stress and extracellular signal-regulated kinase 1/2 (ERK) activation. Hydrogen sulfide (H2S) is involved in antioxidant systems and the ERK signaling pathway. Therefore, in this study, we investigated the role of H2S in primary cilia elongation and the downstream pathway. In cultured Madin-Darby Canine Kidney cells, the length of primary cilia gradually increased up to 4 days after the cells were grown to confluent monolayers. In addition, the expression of H2S-producing enzyme increased concomitantly with primary cilia length. Treatment with NaHS, an exogenous H2S donor, accelerated the elongation of primary cilia whereas DL-propargylglycine (a cystathionine γ-lyase inhibitor) and hydroxylamine (a cystathionine-β-synthase inhibitor) delayed their elongation. NaHS treatment increased ERK activation and Sec10 and Arl13b protein expression, both of which are involved in cilia formation and elongation. Treatment with U0126, an ERK inhibitor, delayed elongation of primary cilia and blocked the effect of NaHS-mediated primary cilia elongation and Sec10 and Arl13b upregulation. Finally, we also found that H2S accelerated primary cilia elongation after ischemic kidney injury. These results indicate that H2S lengthens primary cilia through ERK activation and a consequent increase in Sec10 and Arl13b expression, suggesting that H2S and its downstream targets could be novel molecular targets for regulating primary cilia

        몇가지 사료 첨가제가 계란의 특정성분에 미치는 영향

        The experiment was conducted to determine the effects of various commercial feed additives on specific components in shell egg and to develop a new type of brand egg. Forty-four week old ISA Brown layers were randomly assigned to 8 treatments. Hens of each treatment were 300 and treatment was replicated 3 times. Experimental period was 10 weeks. The 8 treatments were as follows: astarich^ⓡ 2%(B), astarich^ⓡ 5%(C), chitin+chitosan 2%(D), omega-3 powder 2%(E), pyrogreen 1%(F), greenpia 0.2%(G), hydrogenated soy oil 3%(H) and commercial layer feed(A). Eggs were collected at day 0, 7, 14, 40 and 70 for chemical analyses. Chemical analyses were done for vitamins A and E, cholesterol and fatty acid profile of egg yolks. During the experimental period, the contents of vitamin A increased overall. Astarich^ⓡ 5%(C) group showed the highest amounts of vitamin A (11.14±3.93 ㎍/g yolk) whereas commercial layer feed goup(A) showed the lowest amounts of vitamin A(8.65±1.97 ㎍/g yolk). Vitamin E contents were significantly different among treatments at day 7, 40, and 70 (p$lt;0.05). Mean value of vitamin E (99.66±8.30 ㎍/g yolk) was the highest in omega-3(E) group and the lowest in pyrogreen (F) group (53.49±11.36 ㎍/g yolk). Yolk cholesterol contents tended to increase, although the fluctuation existed somewhat depending on the day of measurement. The highest value of yolk cholesterol(11.71±0.50㎎/g yolk) was observed in astarich^ⓡ 2%(B) group and the Lowest value(9.45±2.32㎎/g yolk) in pyrogreen (F) group. The mean compositions of yolk fatty acids during the experimental period were 37.3∼39.6% for saturated fatty acids(SFA), 39.3∼41.3% for monounsaturated fatty acids(MUFA), 16.2∼18.9% for omega-6 fatty acids and 2.20∼5.36% for omega-3 fatty acids. In general, the concentrations of SFA and omega-6 fatty acids decreased, while those of MUFA tended to increase. On the other hand, eicosapentaenoic acid(C_(20:5) ω6, EPA) and docosapentaenoic acid(C_(22:5) ω3, DPA) were detected only in astarich^ⓡ 5%(B) and omega-3(E) groups where the docosahexaenoic acid(C_(22:6) ω3, DHA) concentration was over twice higher than those of other group. The study proved both astarich^ⓡ and omega-3 powder were suitable to produce brand eggs specially high in certain components.

        본 논문은 주식투자를 문제영역으로 하는 전문가 시스템 설계와 구현 연구에 관한 것이다. 주식투자와 관련된 기존 프로그램들의 대부분이 데이타베이스, 통계분석, 시장모델 등에 관한 것이었으나 본 논문에서 구현하는 주식투자 전문가 시스템인 ASIE(Artificial Stock Investment Expert) 시스템은 주식투자 분야의 투자 전문지식을 조직화해서 주식투자 전문가 시스템을 구축하고, 이를 사용자들에게 제공하여 효율적이고 합리적인 투자를 할 수 있도록 도와주는 것을 목표로 한다. ASIE 시스템은 현재의 상황에 대한 사용자의 의견을 입력으로 받아 지식베이스에 들어 있는, 투자에 경험이 많은 전문인의 경험적 지식으로 추론을 하여 여러가지 투자 조건에 알맞는 적절한 투자 종목을 선정해 준다. Design and implementation of an expert system for stock investment is described. Existing programs related to stock investments are mostly about database management, statistical analysis systems or mathematical model of stock markets, instead the ASIE(Artificial Stock Investment Expert)system described is an expert system, which systematizes human expert knowledges for stock investment and provides inferred decision alternatives to users for better investment decisions. The ASIE system incorporates the user's opinions about current situations and performs inferences using the formalized knowledges in knowledge-base together with other data in the database and finally composes the preferred portfolio suitable for the various investment conditions and user's personal preferences.

        산란계의 연령과 면역주기에 따른 난황 중의 Streptococcus mutans 특이 항체 함량

        Streptococcus mutans-specific IgY content change, laying rate, egg weight and weight change were measured for 17-week and 30-week old hens. Vaccinations with Streptococcus mutans were made two times(eight week interval), three times(four week interval) and five times(two week interval), respectively. It was observed that the laying rate of vaccinated hens was likely lower than that of non-vaccinated group. No effect on body weight by vaccination was found out. Egg weight did not show a certain tendency by vaccination. Anti-S. mutans IgY started to be detected two weeks after the lst vaccination for 30-week old hens. It was not detected for non-vaccinated group. The antibody activity was consistently detected after 8 weeks from the last vaccination. The measurement of total IgY and S. mutans-specific IgY in the egges from vaccinated hens revealed that IgY tended to increase with the number of vaccination. S. mutans-specific IgY content of five-time vaccinated l7-week hens was much higher than that of 30-week old hens. To obtain steady amount of specific IgY, multiple vaccination with two week interval was recommended.

        Production of Hydrogen-Rich Syngas from Dimethyl Ether by Using a Microwave Steam Plasma

        Dimethyl ether (DME) is disintegrated by a microwave steam-plasma raising the gas temperature in a reaction chamber of 100 liters in a swirl-type gasifier. With additional heating of DME by partial oxidation, the gasifier temperature reaches 1250 °C. The initial breakdown species of DME molecules are hydrogen, carbon monoxide and methane, which eventually disintegrate to CO and H2 by steam modification at temperatures higher than 1200 °C. The relative concentrations of hydrogen, carbon monoxide and carbon dioxide produced solely from DME modification by steam were 67.5 percent, 30.3 percent and 2.2 percent, respectively, achieving a conversion rate of 88 percent at a temperature of 1058 °C. The total energy efficiency of the present experiment was 81 percent.

        DNA분석기법을 이용한 한국재래산양육의 판별

        재래산양의 유전자원 보존과 유전적 개량을 위하여 재래산양과 수입산양의 genomic DNA의 유전적 다형을 분석하기 위하여 수행되었으며 재래산양과 수입산양의 유전적 판별 분석은 RAPD기법을 이용하였으며 공시품종은 재래산양 30두, 재래산양 교잡종 10두, 수입산양 10두를 이용하였다 재래 산양육과 수입 산양육의 판별을 위한 시료는 재래 산양육 10두와 수입 산양육 10두를 이용하였다. 이들로부터 얻어진 결과를 요약하면 다음과 같다. 1. 재래산양, 수입산양 및 재래산양 교잡종으로부터 추출된 genomic DNA는 전기영동에 의해 약 23kb 크기의 DNA을 얻을 수 있었으며 UV spectrophotometer를 이용하여 흡광도 A 260과 A280의 비율로 측정한 결과, 그 비율이 175~210의 범위로 순도는 비교적 양호한 결과를 얻었다. 2. 순수 재래산양의 유전자원의 보존을 위한 재래산양의 유전자 감식여부를 탐색하기 위하여 약 110 여종의 random primer를 이용한 RAPD 기법에 의하여 재래산양, 수입산양 및 교잡종의 다형성을 분석한 결과 random primer OPO-19(5'-CAA ACG TCG G-3')를 이용하였을 때 재래산양에서만 396bp에서 band 가 나타났으며 수입산양과 교잡종에서는 vand가 나타났으며 수입산양과 교잡종에서는 band가 나타나지 않았다. 3. 또한, 재래 산양육과 수입 산양육의 유전적인 차이를 구명하기 위한 RAPD 기법에 의한 genomic DNA의 다형성 분석에 있어서도 random primer OPO-19(5'-CAA ACG TCG G-3')를 사용하였을 때 396bp에서 재래 산양육에서는 band가 나타났지만, 수입 산양육에서는 band가 나타나지 않았다. This study was carried out to analyze the genetic polymorphisms of genomic DNA of blood and meat for conservation of the genetic resources and genetic improvement of Korean Native goat. The genetic identification between Korean Native goat and imported goat was examined using RAPD(random amplified polymorphisms DNAs) analysis with 30 Korean Native goat, 10 hybrid, 10 imported goat. 10 Korean native goat meat and 10 imported goat meat. The results obtained from this study were summarized as follows. 1. Genomic DNA from Korean native goat, hybrid and imported goat could be obtained above about 23kb size using 0.5% agarose gel electrophoresis and the ratio of optical density at 260nm to that at 280nm was between 1.7 and 2.0 using UV spectrophtometer instrument. 2. In the results of the gene identification between Korean Native goat and hybrid, and imported goat using RAPD methods with random primer of 110 kinds, only Korean native goat showed a specific band at about 369bp using a random primer OPO-19 (5’-CAA ACG TCG G-3’), but imported goat and hybrid not showed. 3 Also, in the results of the gene identification between Korean Native goat meat and imported goat meat using RAPD methods with random primer, Korean native goat only showed a specific band at about 369bp using a random primer No 19(5’-CAA ACG TCG G-3’). but imported goat not showed.

        스트렙토마이신 정맥주사를 이용한 폐결핵의 치료의 대안적 접근방법

        Streptomycin is currently utilized to treat advanced tuberculosis in Korea. However, the usefulness of intravascular streptomycin is not fully understood. In this study, streptomycin was administered intravenously in 30 patients. Intravenous administration of streptomycin may be a useful route of administration without undesirable toxicities.

      • 초전도 한류기를 적용한 계통의 3상 단락사고에 대한 EMTDC 시뮬레이션

        We have performed an EMTDC simulation for the current limiting effects of a superconducting fault current limiter (SFCL). A three-phase fault in the 154 kV transmission line between the gaepo and Sungnam substations produced a fault current of up to 60kA. The SFCL with 100 Ω impedance after quench limited the current to 17kA within a half cycle. This limited current is well below the upper limit of a circuit breaker, suggesting the impedance of the SFCL in the transmission line is sufficient.

        This paper achieves a research about remote control of robot based on voice recognition. Through real-time remote control and wireless network capabilities of an unmanned remote-control experiments and Home Security / exercise with an unmanned robot, remote control and voice recognition and voice transmission are possible to transmit on a PC using a microphone to control a robot to pinpoint of the source. Speech recognition can be controlled robot by using a remote control. In this research, speech recognition speed and direction of self-driving robot were controlled by a wireless remote control in order to verify the performance of mobile robot controller.

      • Reversible and Irreversible Responses of Defect-Engineered Graphene-Based Electrolyte-Gated pH Sensors

        <P>We have studied the role of defects in electrolyte-gated graphene mesh (GM) field-effect transistors (FETs) by introducing engineered edge defects in graphene (Gr) channels. Compared with Gr-FETs, GM-FETs were characterized as having large increments of Dirac point shift (similar to 30-100 mV/pH) that even sometimes exceeded the Nernst limit (59 mV/pH) by means of electrostatic gating of H+ ions. This feature was attributed to the defect-mediated chemisorptions of H+ ions to the graphene edge, as supported by Raman measurements and observed cycling characteristics of the GM FETs. Although the H+ ion binding to the defects increased the device response to pH change, this binding was found to be irreversible. However, the irreversible component showed relatively fast decay, almost disappearing after 5 cycles of exposure to solutions of decreasing pH value from 8.25 to 6.55. Similar behavior could be found in the Gr-FET, but the irreversible component of the response was much smaller. Finally, after complete passivation of the defects, both Gr-FETs and GM-FETs exhibited only reversible response to pH change, with similar magnitude in the range of 68 mV/pH.</P>
