The root of Zingiber officinale R. was extracted with 80% aqueous MeOH, and the concentrated extract was fractionated with EtOAc, n-BuOH and water phases. The EtOAc fraction exhibited most potent antioxidant activity among them, and two active compounds were isolated by activity-guided separation using repetitive silica gel column chromatography. On the bases of spectral data and the chemical characteristics, the structure of the compounds were determined as 6-shogaol and 12-shogaol, respectively. The EC_(50) values of them were evaluated as 26.4 μM and 22.5 μM, respectively, where these values were higher than the commercial antioxidants, such as, BHA, BHT and α-tocopherol.
Insecticidal Alkaloids from the Seeds of Macleaya cordata on Cotton Aphid (Aphis gossypii)

Alcohol extracts from the seeds of Macleaya cordata have shown a significant insecticidal effect against the growth of the cotton aphid, Aphis gossypii Glover. In this study, we aimed to isolate the principal component of M. cordata to manifest its insecticidal effect on A. gossypii. The dried seeds of M. cordata were extracted with 80% aqueous methanol (MeOH), and the concentrated extracts were partitioned successively with ethyl acetate (EtOAc), n-butyl alcohol (n-BuOH), and $H_2O$. During the search for an insecticidal compound against the aphids, activity-guided fractionation led to the isolation of two alkaloids from an n-BuOH fraction through repeated silica gel column chromatographic separations. Based on NMR, MS, and IR spectroscopic data, the chemical structures of the compounds were determined to be 8-hydroxydihydrochelerythrine (1) and 8-methoxydihydrosanguinarine (2) This is the first study in which 8-methoxydihydrosanguinarine was isolated from M. cordata. 8-Hydroxydihydrochelerythrine (1) and 8-methoxydihydrosanguinarine (2) decreased the survival of the cotton aphid by $76.1{\pm}7.9$ and $73.6{\pm}14.6%$ at 100 ppm, respectively.
Real-Time Lane Detection Based on Deep Learning

As the research and development of autonomous vehicles has become more active, lane detection technologies for providing road information have become key elements. There are limits to detecting lanes in dynamic driving environments in conventional machine vision research, as the approaches are generally dependent on expert scenarios and fi ne-tuned heuristics. Deep learning has shown good performance in classifying target information with this distribution of nonlinear data; thus, many studies have actively applied deep learning to lane detection. However, most of these studies have focused on improving the accuracy, rather than on the operating speed. For the work reported herein, a benchmarking deep-learning framework for lane detection was applied with lightened feature extraction modules and decoder modules. These were used to compare performances and to present an indicator for selecting a model for optimizing real-time performance and accuracy. The VGG-16, MobileNet, and Shuffl eNet networks were used for the encoder module, whereas frontend dilation and UNet were used for the decoder module. The limitations of the benchmarking framework were analyzed, and perspective loss concepts were applied to the processing of the network using front-view images to ensure improvements in the accuracy and operating speed. All of the candidate networks obtained objective performance indicators based on a large-scale benchmark dataset (TuSimple) and network training with a dataset collected and verifi ed via performance on public roads in Singapore

<P>Mice lacking the IL-1R-associated kinase 4 (IRAK4) are completely resistant to LPS-induced endotoxic disorder or the TLR9 agonist CpG DNA plus D-galactosamine-induced acute liver injury (ALI), whereas wild-type strains succumb. However, translational drugs against sepsis or ALI remain elusive. Lonicerae flos extract is undergoing the clinical trial phase I in LPS-injected healthy human volunteers for sepsis treatment. In the current study, chlorogenic acid (CGA), a major anti-inflammatory constituent of lonicerae flos extract, rescued endotoxic mortality of LPS-intoxicated C57BL/6 mice, as well as ameliorated ALI of LPS/D-galactosamine-challenged C57BL/6 mice. As a mechanism, CGA inhibited various TLR agonist-, IL-1 alpha-, or high-mobility group box-1-stimulated autophosphorylation (activation) of IRAK4 in peritoneal macrophages from C57BL/6 or C3H/HeJ mice via directly affecting the kinase activity of IRAK4, a proximal signal transducer in the MyD88-mediated innate immunity that enhances transcriptional activity of NF-kappa B or AP-1. CGA consequently attenuated protein or mRNA levels of NF-kappa B/AP-1 target genes encoding TNF-alpha, IL-1 alpha, IL-6, and high-mobility group box-1 in vivo under endotoxemia or ALI. Finally, this study suggests IRAK4 as a molecular target of CGA in the treatment of innate immunity-related shock and organ dysfunction following insult of various TLR pathogens from bacteria and viruses.</P>

Banha (Pinellia ternata) is a herbal medicine which is composed of alkaloid substances, homegentisic acid, 3,4-dihydroxybenzaldehyde and 3,4-diglycosilic benzaldehyde and is mainly prescribed in cough with dyspnea, gastric retention, distention and pain of the thorax and abdomen, lymph node tuberculosis, carbuncle, and furuncle. The use of raw Banha has been prohibited in pregnant woman because of it's strong toxicity. In this study, to investigate whether Banha affects the antioxidant system of male reproductive organs, we analyzed the mRNA expression patterns of glutathione peroxidase (GPx) family (type 1-4) in the reproductive organs of rats exposed to Banha. Banha extracts (0.05, 0.1, 0.2, 0.5, 1, and 2 g/㎏ body weight) were treated to 6 week-old rats for 4 weeks per oral. Total RNAs were extracted from testis, epididymis, prostate, and seminal vesicle and then RT-PCR was perfomed with the specific primers of each GPx enzyme. In epididymis and prostate, GPx1 mRNA were decreased by Banha treatment except for 2 g/㎏ treatment in prostate. However, GPx1 mRNA in seminal vesicle was increased by Banha treatment. GPx2 mRNA was increased in testis and seminal vesicle. GPx4 (PHGPx) mRNA was decreased in seminal vesicles by 0.1 and 1 g/㎏ treatment of Banha. These findings indicate that Banha may affect the antioxidant system of male reproductive organs, although further study is necessary.

Juvenile rheumatoid arthritis (JRA), an autoimmune disease, was characterized by chronic synovitis and associated with various extra-articular manifestations. Abnormal hematologic findings have been reported in all form of JRA, especially anemia due to chronic disease or iron deficiency. Dysplastic changes were rarely noted in the peripheral blood and bone marrow. We experienced a 15-year-old female patient with pauciarticular JRA who have pancytopenia in peripheral blood and a number of dysplastic changes in bone marrow, and present the case here with brief review of literatures.(Korean J Med 59:587-590, 2000)
