      • Oil Contents and Fatty Acid Composition of Korean Perilla (Perilla ocimoides L.) Collections

        This study was carried out to obtain informations on the variations of oil content and fatty acid composition among 90 Korean perilla collections. Average oil content of 90 perilla collections was 44.2% with a range from 29.7% to 61.9%. Perilla collections with late-maturing, super-large seed and gray seed coat showed higher oil content than other types in general. Average saturated fatty acid content in perilla oil was 9.0% with a range from 8.2% to 10.7%, while average unsaturated fatty acid content varied from 89.3% to 91.8% with a mean of 91.0%. Contents of saturated and unsaturated fatty acids were not related to maturity. There were no differences in the contents of saturated and unsaturated fatty acids according to maturity. Linoleic acid and linolenic acid contents were the highest in the super large-sized group(15.5%) and in the large-sized seed group(61.4%), respectively, while contents of fatty acids among the perilla collections were variable with different seed coat colors. Most of the traits studied were not significantly correlated with oil content, but linoleic acid($\omega$ 수식 이미지-6) content was negatively correlated (r=-0.217*) with linolenic acid(ω-3) content.

      • 김치에서 분리한 Lactobacillus sp. JC-7과 Lactobacillus acidophilus 88간의 Electrofusion 최적조건 설정

        김치 발효 숙성기간을 연장하고 신선도를 오랫동안 유지 할 수 있는 김치발효 starter의 개발을 목적으로 최적숙성기에 있는 김치로부터 bacteriocin생성능이 없는 유산균을 분리하여 동정하였으며, bacteriocin생성능이 우수한 L.acidophilus 88을 융합시키기 위해 electrofusion에 대한 최적 조건을 검토하였다. 분리균주는 Lactobacillus속으로 동정되어 Lactobacillus sp. JC-7이라 명명하였다. Electrofusion에 의해 생성된 융합주를 식별하기 위해 streptomycin(2.5㎎/ml)에 내성을 나타내는 Lactobacillus sp. JC-7변이주와 kanamycin(600㎍/ml)에 내성을 나타내는 L.acidophilus 88변이주를 분리하였다. Electrofusion을 100V/㎝, 120msec(72ohms,1670 capacitance) 에서 수행했을 때 융합효율이 가장 양호하였으며 전기장의 세기와 시간이 중가할수록 융합 효율이 현저히 감소하였다. 2가 양이온은 농도가 중가할수록 대체적으로 융합효율을 감소시키는 경향을 나타내었으나 1mM MGCl_2에서는 대조군에 비해 융합효율이 약간 증가하였다. PEG매개에 의한 융합법의 융합효율을 비교한 결과, 융합효율은 chemical fusion<electrofusion<electrofusion+20% PEG순을 나타내었다. Evaluation of Optimum Conditions for the Electrofusion between Lactobacillus sp. JC-7 Isolated from Kimchi and Lactobacillus acidophilus 88. Young-Bae Jo, Hyun-Jung Choi, Hyung-Suk Baik and Hong-Ki Jun*. Department of Microbiology, Pusan National University, Pusan 609-735,Korea-A lactic acid bacterium was isolated from kimchi. The isolated strain was identified as the genus Lactobacillus thrugh its morphological characteristics and named as Lactobacillus sp. JC-7. The optimum conditions for the electrofusion between streptomycin(2.5㎎/ml)resistant mutant of Lactobacillus acidophilus 88 and kanamycin(600㎍/ml) resistant mutant Lactobacillus sp. JC-7 were evaluated. The highest number of fusants were obtained at a capacitance value of 120msec(1670㎌), a field strength of 100V/㎝,and a pulse controller setting of 72Ω. The potimum pH of elecroporation buffer was 7.5 and the concentration of divalent cation was 1mM MG^2+. Electrofusants were efficiently obtained by addition 20% polyethylene glycol to electroporation buffer. The yield of fusion was better than that of using polyethylene glycol mediated chemical induction.

      • 사람 위선암에서 cathepsin L의 발현증가

        cathepsin L은 lysosomal cysteine 단백분해효소로서 기저막(basement membrane)과 세포외기질(extracellular matrix)을 파괴하여 암세포의 침윤과 전이에 중요한 역할을 하는 물질로 알려져 있다. 이러한 cathepsin L에 대한 mRNA 발현도를 5개의 사람 위선암(gastric adenocarcinoma) 세포주와 5명의 위선암 환자에서 채취한 조직에서 방사능으로 표지된 cathepsin L특이 cDNA를 사용한 Northern blot법으로 측정하였다. 위암의 전이병소에서 수립한 세포주인 SNU-5, SNU-16, MKN-45와 Kato Ⅲ에서는 cathepsin L mRNA가 발현되었으나 원발병소에서 수립한 AGS 세포주에서는 mRNA의 발현이 관찰되지 않았다. 5명의 위암 환자에서는 원발병소, 전이가 확인된 임파절 및 암 근처 정상 위점막에서 각각 조직을 채취하여 cathepsin L mRNA의 발현을 측정하였다. 원발병소와 전이병소에서는 모두 cathepsin L mRNA가 발현되었으나 정상 위점막조직에서는 전예에서 mRNA 발현이 관찰되지 않았다. 한편 mRNA의 발현도는 1예에서는 전이병소가 원발병소에 비해 높았으나, 2예에서는 전이병소에서 발현도가 낮았으며, 나머지 2예에서는 원발병소와 전이병소 사이에 차이가 없어, 원발병소와 전이병소 사이에 mRNA의 발현도의 일관성 있는 경향은 관찰되지 않았다. 이상의 결과는 cathepsin L은 위암의 발생과 진행에 있어 암세포의 침윤과 전이를 촉진하는 것 이외에 또 다른 역할을 할 가능성을 시사하고 있다고 사료된다. Cathepsin L, a lysosomal cysteine protease, is known to play an important role in cancer invasion and metastasis by degrading the components of basement membrane and extracellular matrix. The mRNA expression of cathepsin L was determined by Northern blot analysis using a radiolabeled cDNA specific for cathepsin L in five human gastric adenocarcinoma cell lines and five surgical specimens of primary gastric adenocarcinomas, their metastatic lymph nodes and matched adjacent normal mucosae. The mRNA of cathepsin L was expressed in all of the four cell lines established from the metastatic sites, SNU-5, SNU-16, MKN-45 and Kato Ⅲ, while not detected in one cell line established from the primary site, AGS. The mRNA was expressed in all of the five primary and five metastatic cancer specimens tested, while it was not detected in all matched normal mucosae. The intensities of the mRNA expressions, however, did not show the consistent pattern between primary sites and metastatic lymph nodes. These results suggest that cathepsin L may have the other function in addition to facilitation of the invasion and metastasis during the development and progression of stomach cancer.

      • 동아의 체중조절 효과

        본 연구에서 선발된 17종의 천연물을 Sprague- Dawley rat에 급여한 결과, 체중 증가, 에너지 소비, fat pads 중량, 간의 triglyceride 함량, atherogenic index 그리고 혈중 글루코오스의 함량 등이 감소하는 경향을 나타내었다. 특히 동아를 구강으로 섭취시킨 결과, 체중 증가, 에너지 소비, fat pads 중량, 그리고 혈중 글로코오스의 함량등이 감소하였으나, 간의 triglyceride 함량은 오디와 송엽의 열수 추출물보다 적게 감소하였다. 동아, 오디, 그리고 송엽을 혼합하여 사용한 결과, 각각의 원료를 사용할 경우보다 비만이 개선되는 것으로 나타났다. This study attempted to evaluate the effectiveness of function of Wax Gourd for obesity and the related disease of adult people can be developed by the combination of the 17 food materials screened in this study. The experiments using Sprague-Dawley male rats show that seventeen kinds of food material inhibited the increase of body weight gain, energy intake, fat pads weight, liver triglycerides content, atherogenic index, and blood glucose concentration induced by cafeteria diet. Oral administration of wax gourd decreased most body weight gain, energy intake, fat pads weight, and blood glucose concentration induced by cafeteria diet. Oral administration of wax gourd decreased most body weight gain, energy intake, fat pads weight, and blood glucose concentration. Liver triglycerides content was the lowest in the group administered mulberry stem extract and the group administered pine leaves showed the lowest atherogenic index. The combined administration of wax gourd, mulberry stem extract and pine leaves improved more the obesity and related symptoms than the individual administrations.

      • 세계산 Rumiceae Dum.족 (마디풀과)의 진화계통학적 연구

        세계산 Rumiceas Dum.족을 구성하는 4속(Rumex속의 4절, Rheum속의 3절, Oxyria속, Emex속) 분류군간의 진화적 유연관계에 대한 가설을 제공하고, 형질진화의 경향을 밝히기 위한 분자분식(cladistic analysis)을 수행하였다. 최소절약의 원리를 이용하여 24개의 두단계 혹은 다단계 형질을 분석하였다. 분석 결과 계통수의 길이는 36, 일관성지수는 0.714, 보존지수는 0.843인 4개의 등가질약 계동수가 산출되었다. Rumex속의 4개 절과 Emex속은 하나의 Clade를 구성하며, 이는 여러 개의 공유파생형질(익상수과, 오용 존서, 만곡의 배, 등)에 의해 지지된다. 결과적으로 Emex속은 Rumex속의 하나의 절(section)로 처리함이 타당하다. 또한 Oxyria속은 3개의 독립파생형질(화사수:2개, 렌즈형의 수과, 4개의 화피핀)에 의해 다른 속과 잘 구별되었다. Cladistic analysis of four genera (4 sections of the genus Rumex, 3 sections of the genus Rheum, Oxyria and Emex) of the tribe Rumiceae Dum. was undertaken to provide hypotheses about evolutionary relationships and to elucidate patterns of character evolution. A set of 24 binary and multistate morphological characters were analyzed using maximum parsimony algorithms. With the given data set. four equally parsimonious trees 36 tree length with a consistency index 0.714 and a retention index of 0.843 were produced. Four sections of Rumex and Emex came together as one clade and was strongly supported by several synapomorphies (e.g.. winged achene. trichome present. curved embryo. etc.). Thus: it is considered that Emex is better treated as one of section in Rumes. Oxyria is also well distinguished by its automorphic characters (by having 2 styles. lenticular achene. 4 perianth segment number).

        pH순환 모델에서 과포화 용액의 초기 우식 법랑질에 대한 재광화 효과

        Dental caries is the most common oral disease. There are many factors contributing to its development. but complete understanding and prevention are not fully know. However, it is possible to remineralize the early enamel carious lesion by fluoride containing remineralization solution. Recently the pH-cycling model has been used to examine the effect of fluoride solution on remineralization of artificial caries in vitro as it can closely simulate the conditions encountered in vivo within a carefully controlled enviroment.

      • 에폭시 수지를 혼합한 아스팔트 방식재의 특성

        Epoxy resins were mixed with asphalt solution in the range of 0 - 60phr for the total components. The blending properties, curing properties, and surface properties for mixtures were investigated experimentally. Compatability of epoxy resins with asphalt were relatively good over mixture temperature 50℃. Interpenetrated polymer networks(IPNs) were partially formed between epoxy resins and asphalt. Increase of hardness was depended on the crosslinking density of epoxy resins and oxidation of asphalt. Defects of coated surface including crater, pinhole, and crack could be improved by adding epoxy resins 20 phr.

      • 벽시계틀 소재로 사용되는 목재와 플라스틱의 접착력 향상에 관한 연구

        Wood and plastic were used as elemental materials of the frame of clock. In this research, it was studied to improve adhesive strength between this materials by new adhesion process. Vinyl or acryl emulsion(H-403, H-422, Y-840, Y-850) was used as the adhesive. The adhesive properties was tested experimantally on various partial process including sanding, cleaning of sanded surface and pressing of bonded areas. The results obtained in this experiments were follows : 1) Y-850 represented most excellent adhesive properties. 2) The maximum of adhesive strength could be obtained by sanding with sand paper #600, pressing of 0.3∼0.5kg/㎠, and cleaning with acetone. In that case, increase of 20% in adhesive strength was showed than obtained by traditional process.
