        The role of protein arginine-methyltransferase 1 in gliomagenesis

        Protein arginine methyltransferase 1 (PRMT1), a type-I arginine methyltransferase, has been implicated in diverse cellular events. We have focused on the role of PRMT1 in gliomagenesis. In this study, we showed that PRMT1 expression was up-regulated in glioma tissues and cell lines compared with normal brain tissues. The knock-down of PRMT1 resulted in an arrest in the G1-S phase of the cell cycle, proliferation inhibition and apoptosis induction in four glioma cell lines (T98G, U87MG, U251, and A172). Moreover, an in vivo study confirmed that the tumor growth in nude mouse xenografts was significantly decreased in the RNAi-PRMT1 group. Additionally, we found that the level of the asymmetric dimethylated modification of H4R3, a substrate of PRMT1, was higher in glioma cells than in normal brain tissues and decreased after PRMT1 knock-down. Our data suggest a potential role for PRMT1 as a novel biomarker of and therapeutic target in gliomas. [BMB Reports 2012; 45(8): 470-475]

        Propagation characteristics of longitudinal wave, shear wave and bending wave in porous circular nanoplates

        On the basis of nonlocal strain gradient theory, considering the material properties of porous FGM changing with thickness and the influence of moment of inertia, the wave equation of FG nano circular plate is derived by using the first-order shear deformation plate theory, by introducing dimensionless parameters, we transform the equations into dimensionless wave equations, and the dispersion relations of bending wave, shear wave and longitudinal wave are obtained by Laplace and Hankel integral transformation method. The influence of nonlocal parameter, porosity volume fraction, strain gradient parameters and power law index on the propagation characteristics of bending wave, shear wave and longitudinal wave in FG nano circular plate.

        New phenylpropanoids from Bulbophyllum retusiusculum

        Two new phenylpropanoids, retusiusines A (1) and B (2), and a pair of new phenylpropyl enantiomers, (±)-retusiusine C (3a and 3b), together with eight known compounds, dihydroconiferyl dihydro-p-coumarate (4), methyl 3-(4-hydroxyphenyl) propionate (5), 3-(4-hydroxyphenyl)-propanoic acid (6), dihydroferulic acid (7), methyl 3-(4-methoxyphenyl) propionate (8), 3-(3,4-dimethoxyphenyl)-2-propenal (9), trans-p-coumaric acid (10) and dihydroconiferyl alcohol (11), were isolated from the tubers of Bulbophyllum retusiusculum. The absolute configurations of the new compounds were determined by calculating their electronic circular dichroism (ECD), spectra and specific optical rotations and comparing the calculated values with the experimental data. Compound 2 exhibited potent antifungal activity against Candida albicans (16 μg/mL). Compound 3 showed moderate antibacterial activity against Bacillus subtilis (64 μg/mL).

        Wavelet Multiresolution Complex Network for Analyzing Multivariate Nonlinear Time Series

        <P>Characterizing complicated behavior from time series constitutes a fundamental problem of continuing interest and it has attracted a great deal of attention from a wide variety of fields on account of its significant importance. We in this paper propose a novel wavelet multiresolution complex network (WMCN) for analyzing multivariate nonlinear time series. In particular, we first employ wavelet multiresolution decomposition to obtain the wavelet coefficients series at different resolutions for each time series. We then infer the complex network by regarding each time series as a node and determining the connections in terms of the distance among the feature vectors extracted from wavelet coefficients series. We apply our method to analyze the multivariate nonlinear time series from our oil-water two-phase flow experiment. We construct various wavelet multiresolution complex networks and use the weighted average clustering coefficient and the weighted average shortest path length to characterize the nonlinear dynamical behavior underlying the derived networks. In addition, we calculate the permutation entropy to support the findings from our network analysis. Our results suggest that our method allows characterizing the nonlinear flow behavior underlying the transitions of oil-water flows.</P>

        Structure-based design, structure–activity relationship analysis, and antitumor activity of diaryl ether derivatives

        To identify novel therapeutic agents to treatcancer, we synthesized a series of diaryl ether derivatives. Structure–activity relationship studies revealed that thepresence of a chlorine or hydroxyl at the para-position onthe phenyl ring (5h or 5k) significantly enhanced antitumoractivity. Compound 5h had stronger growth inhibitory activityin HepG2, A549, and HT-29 cells than compound 5k,with IC50 values of 2.57, 5.48, and 30.04 lM, respectively. Compound 5h also inhibited the growth of other cells lines,including Hep3B, PLC/PRF5, SMMC-7721, HeLa, andA375, with IC50 values of 2.76, 4.26, 29.66, 18.86, and10.21 lM, respectively. The antitumor activity of compound5h was confirmed by a colony forming assay. Further,our results indicated that the antitumor activity ofcompound 5h may be mediated by enhancing expression ofp21 and cl-caspase3, and leading to apoptosis of cancercells.

      • Meta-analysis of Gene Expression Data Identifies Causal Genes for Prostate Cancer

        Prostate cancer is a leading cause of death in male populations across the globe. With the advent of gene expression arrays, many microarray studies have been conducted in prostate cancer, but the results have varied across different studies. To better understand the genetic and biologic mechanisms of prostate cancer, we conducted a meta-analysis of two studies on prostate cancer. Eight key genes were identified to be differentially expressed with progression. After gene co-expression analysis based on data from the GEO database, we obtained a co-expressed gene list which included 725 genes. Gene Ontology analysis revealed that these genes are involved in actin filament-based processes, locomotion and cell morphogenesis. Further analysis of the gene list should provide important clues for developing new prognostic markers and therapeutic targets.

        Simultaneous Stability and Path Following Control for 4WIS4WID Autonomous Vehicles Based on Computationally Efficient Offset Free MPC

        The steady-state error problem of autonomous vehicle MPC-based motion control has not been effectively solved for a long time. This problem is more serious for lateral and longitudinal coupling control problems of vehicles with over-actuated configurations. Based on our newly designed general offset-free MPC (OF-MPC) solver and the TMeasy tire model, a steady-state error free control strategy for simultaneous stability and path following control of four-wheel steering and four-wheel drive vehicles is proposed. OF-MPC uses the disturbances term to describe the model mismatch and external disturbances, then uses the Kalman filter to observe the disturbances, and finally considers the disturbances in the optimization stage to realize the control without steady-state error. Realtime simulation results show that OF-MPC can solve model mismatch and external disturbances problems, and the steady-state error free control is realized. The simulation results of the double lane change maneuver show that the OF-MPC dynamic control performance is also better than the traditional MPC (TRA-MPC), which is more obvious when the vehicle is at the stability boundary and under various constant or time-varying disturbances. Regardless of the dimensions and complex constraints of this problem, real-time performance is still guaranteed, thanks to the proposed OF-MPC. When the horizon length is 100, the average time consumption is only about 15 milliseconds.

        Clinical and Pathologic Features of Multifocal and Multicentric Breast Cancer in Chinese Women: A Retrospective Cohort Study

        Purpose: This study aims to analyze the clinical-pathological characteristics of multifocal and multicentric breast cancer (MMBC) in Chinese women. Methods: Sixty-seven cases with MMBC were randomly collected and reviewed at seven hospitals in representative districts of China during 1999 to 2008. Results: The incidence of MMBC in breast cancer in China was 1.75%. Compared to those with unifocal breast cancer, women with MMBC were more likely to have larger tumor size, lymph node metastasis (59.70% vs. 45.62%) and stage III to IV (46.26% vs. 21.10%). The peak age at onset of MMBC was 40 to 49 years old and has been gradually increasing during 1999 to 2008. Most of the MMBC women were treated with surgery and adjuvant therapy. Conclusion: In China, the incidence of MMBC in breast cancer is significantly lower than that in Western countries. Compared to unifocal breast cancer, MMBC is biologically more aggressive. Most MMBC women underwent mastectomy, instead of breast conservation surgery.

        High yield engineered nanovesicles from ADSC with enriched miR-21-5p promote angiogenesis in adipose tissue regeneration

        Background: Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) have been found to have a great potential for soft tissue repair due to various biological functions, including pro-angiogenesis and low immunogenicity. However, the low yield and heterogeneity of MSC-EVs limited their clinical transformation. This study was designed to develop a novel adipose-derived stem cell engineered nanovesicles (ADSC-NVs) with high production and explore its pro-angiogenetic effect and application in adipose tissue regeneration. Methods: Adipose-derived stem cell-derived extracellular vesicles (ADSC-EVs) were isolated from an EVs-free culture medium for human ADSCs (hADSCs). ADSC-NVs were prepared by sequentially extruding ADSCs followed by iodixanol density gradient ultracentrifugation and were compared with ADSC-EVs in morphology, size distribution, protein contents and yield. The pro-angiogenetic effect of ADSC-NVs in different doses (0, 5, 20 and 80 μg/mL) in vitro was determined using transwell assay, tube formation assay, western blot and qRT-PCR. In vivo, BALB/c nude mice were administered injection of a mixture of fat granules and different dose of ADSC-NVs and grafts were harvested at 12 weeks post-transplantation for further analysis. By analyzing the weight and volume of grafts and histological evaluation, we investigated the effect of ADSC-NVs in vessel formation and adipose tissue regeneration. Results: Our results showed yield of purified ADSC-NVs was approximately 20 times more than that of ADSC-EVs secreted by the same number of ADSCs. In vitro, both ADSC-NVs and ADSC-EVs exhibited a dose-dependent proangiogenetic effect, despite their distinct miRNA profiles. These effects of ADSC-NVs may be mediated by enriched miR-21-5p via PTEN inhibition and PI3K/p-Akt signaling activation. Furthermore, after a mixed injection of ADSC-NVs, vessel formation and adipose regeneration were observed in vivo in fat implants. Conclusions: Our study developed a potent alternative of ADSC-EVs. ADSC-NVs have a high pro-angiogenesis potential and can be used as cell-free therapeutic biomaterials in soft tissue regeneration.

        Dielectric and Conductivity Properties of Poly(L-lactide) and Poly(L-lactide)/Ionic Liquid Blends

        Dielectric relaxation spectroscopy (DRS) was employed to study the dielectric and conductivity propertiesof poly(L-lactide) and poly(L-lactide)/ionic liquid (IL) blends. The experimental dielectric data were analyzedwithin the formalism of complex permittivity. The results were discussed in terms of AC conductivity, αN relaxationcorresponding to the longest normal mode motion, α relaxation originating from the segmental mode motion, andDC conductivity. The results revealed that the motion of the polymer chains governs the charge carrier transport. The temperature dependence of the relaxation times follows the Vogel-Tammann-Fulcher (VTF) equation. Theincorporation of IL into the matrix accelerates the segmental (α) and normal mode (αN) motions and the conductivity,and increases the charge carrier movement of IL following the longest αN motion of chains, which led to an increasein the relaxation strength of the αN relaxation.
